Identification of Foreign Objects in Soybeans Using Near-infrared Spectroscopy (근적외선 분광법을 이용한 콩과 이물질의 판별)

  • The objective of this research was to classify intact soybeans and foreign objects using near-infrared (NIR) spectroscopy. Intact soybeans and foreign objects were scanned using a NIR spectrometer equipped with scanning monochromator. NIR spectra of intact soybeans and foreign objects in the wavelength range from 900 to 1800 nm were collected. The classification of intact soybeans and foreign objects were conducted by using partial least-square discriminant analysis (PLS-DA) and soft independent modelling of class analogy (SIMCA) multivariate methods. Various types of data pretreatments were tested to develop the classification models. Intact soybeans and foreign objects were successfully classified by the PLS-DA prediction model with mean normalization pretreatment. These results showed the potential of NIR spectroscopy combined with multivariate analysis as a method for classifying intact soybeans and foreign objects.

Construction of Logic Trees and Hazard Curves for Probabilistic Tsunami Hazard Analysis (확률론적 지진해일 재해도평가를 위한 로직트리 작성 및 재해곡선 산출 방법)

  • Due to the difficulties in forecasting the intensity and the source location of tsunami the countermeasures prepared based on the deterministic approach fail to work properly. Thus, there is an increasing demand of the tsunami hazard analyses that consider the uncertainties of tsunami behavior in probabilistic approach. In this paper a fundamental study is conducted to perform the probabilistic tsunami hazard analysis (PTHA) for the tsunamis that caused the disaster to the east coast of Korea. A logic tree approach is employed to consider the uncertainties of the initial free surface displacement and the tsunami height distribution along the coast. The branches of the logic tree are constructed by reflecting characteristics of tsunamis that have attacked the east coast of Korea. The computational time is nonlinearly increasing if the number of branches increases in the process of extracting the fractile curves. Thus, an improved method valid even for the case of a huge number of branches is proposed to save the computational time. The performance of the discrete weight distribution method proposed first in this study is compared with those of the conventional sorting method and the Monte Carlo method. The present method is comparable to the conventional methods in its accuracy, and is efficient in the sense of computational time when compared with the conventional sorting method. The Monte Carlo method, however, is more efficient than the other two methods if the number of branches and the number of fault segments increase significantly.

A Comparative Study on the Physical, Psychological, and Social Well-being of Elderly Women in Rural Areas Depending on their Participation in Swimming Activities (수영 활동 참여에 따른 농촌 지역 여성노인의 신체적·정신적·사회적 건강상태 비교)

  • The purpose of this study is to compare the physical, psychological, and social well-being of elderly women living in rural areas depending on their participation in swimming activities. Study sample consisted of 41 elderly women residing in the G province of C region who participated in swimming activities at least 3 times per week for one or more years, and 43 elderly women of the same area who did not participate in swimming activities. Data was collected over the course of 6 weeks. Analysis of the data showed that there are significant differences between participants and non-participants of swimming activities in their physical well-being(pain, flexibility), psychological well-being(perceived health status, depression), and their social well-being(strengthened interpersonal relations). In other words, participants of swimming activities, compared to non-participants, were in better physical, psychological, and social conditions. Thus, active encouragement and support must be in place for elderly women in rural areas to engage in swimming activities by utilizing local swimming facilities.

A High-Throughput Method Based on Microculture Technology for Screening of High-Yield Strains of Tylosin-Producing Streptomyces fradiae

  • Tylosin is a potent veterinary macrolide antibiotic produced by the fermentation of Streptomyces fradiae; however, it is necessary to modify S. fradiae strains to improve tylosin production. In this study, we established a high-throughput, 24-well plate screening method for identifying S. fradiae strains that produce increased yields of tylosin. Additionally, we constructed mutant libraries of S. fradiae via ultraviolet (UV) irradiation and/or sodium nitrite mutagenesis. A primary screening of the libraries in 24-well plates and UV spectrophotometry identified S. fradiae mutants producing increased yields of tylosin. Mutants with tylosin yield 10% higher than the wild-type strain were inoculated into shake flasks, and the tylosin concentrations produced were determined by high-performance liquid chromatography (HPLC). Joint (UV irradiation and sodium nitrite) mutagenesis resulted in higher yields of mutants with enhanced tylosin production. Finally, 10 mutants showing higher tylosin yield were re-screened in shake flasks. The yield of tylosin A by strains UN-C183 (6767.64 ± 82.43 ㎍/ml) and UN-C137 (6889.72 ± 70.25 ㎍/ml) was significantly higher than that of the wild-type strain (6617.99 ± 22.67 ㎍/ml). These mutant strains will form the basis for further strain breeding in tylosin production.

Patterns of categorical perception and response times in the matrix scope interpretation of embedded wh-phrases in Gyeongsang Korean (경상 방언 내포문 의문사의 작용역 범주 지각 양상과 반응 속도 연구)

  • This study investigated the response time and patterns of categorical perception of the wh-scope of an embedded clause with the non-bridge verb, "gung-geum hada 'wonder'," in the matrix verb phrase in Gyeongsang Korean. Using the same procedure as Yun (2022), 72 responses and response times for each stimulus were collected from 24 participants over the course of three trials. The stimuli were recorded readings of 40 speakers (20 male, 20 female). Context was provided to induce a matrix scope interpretation of the embedded wh-phrase in the target sentence. We sorted the 40 stimuli according to the number of matrix scope responses each received, and charted the response times for each stimulus. Although there was considerable overlap for the different types of wh-scope interpretations, there was a clear difference in categorical perception between the matrix and embedded scopes. The 24 participants also differed in their categorical perceptions. The results suggested that response time and wh-scope interpretation were not directly related and that two main weighted factors affected wh-scope interpretation: morpho-syntactic constraints and prosodic structural integrity. The weighting of each of these factors was inversely correlated and varied among subjects.

Hospital-based home care reinbursement and service use for the elderly (노인의 의료기관 가정간호 급여청구 및 서비스 이용 현황)

  • The purpose of this study was to address the current status of hospital-based home care(HBHC). We analyzed the data on HBHC from national electronic data information of Health Insurance Review Agency. Beside, we surveyed 75 hospital-based home care agency. In 2006, 20,343 elderly(64.0% from all HBHC user) used 333,889 visits(76.8%from all visits). Medical diagnosis was composed of circulatory disease including cerebrovascular diseases 41.3%, endocrine system disease including Diabetes mellitus 10.4%, neoplasm 9.7%. Some of subjects used HBHC in excess of maximum covered 8 visits a month by National Health Insurance, decubitus 7.0%, the cancer 5.4%, the diabetes 2.5%, the hypertension 1.1%, and the stroke 0.9%. This results will contribute to expand the coverage of hospital-based home care by National Health Insurance. There was distribution difference in medical diagnosis and nursing intervention between HBHC and Public health center-based home care(PBHC) subjects. Therefore, HBHC subjects had more severe medical diagnosis, and were intervened more injections, examinations, than PBHC subjects. These differences must be considered to set up functional role among the three types of home visit care.

Production of a New Biosurfactant by a New Yeast Species Isolated from Prunus mume Sieb. et Zucc.

  • Biosurfactants reduce surface and interfacial tension due to their amphiphilic properties and are an eco-friendly alternative for chemical surfactants. In this study, a new yeast strain JAF-11 that produces a biosurfactant was selected using drop collapse method, and the properties of the extracts were investigated. The nucleotide sequences of the strain were compared with closely related strains and identified based on the D1/D2 domain of the large subunit ribosomal DNA (LSU) and internal transcribed spacer (ITS) regions. Neodothiora populina CPC 39399T, the closest species with strain JAF-11, showed a sequence similarity of 97.75% for LSU and 94.27% for ITS, respectively. The result suggests that the strain JAF-11 represents a distinct species that cannot be assigned to any existing genus or species in the family Dothideaceae. Strain JAF-11 produced a biosurfactant reducing the surface tension of water from 72 mN/m to 34.5 mN/m on the sixth day of culture and the result of measuring the critical micelle concentration (CMC) by extracting the crude biosurfactant was found to be 24 mg/l. The molecular weight 502 of the purified biosurfactant was confirmed by measuring the fast atom bombardment mass spectrum. The chemical structure was analyzed by measuring 1H nuclear magnetic resonance (NMR), 13C NMR, and two-dimensional NMRs of the compound. The molecular formula was C26H46O9, and it was composed of one octanoyl group and two hexanoyl groups to myo-inositol moiety. The new biosurfactant is the first report of a compound produced by a new yeast strain, JAF-11.

Growth Characteristics according to GA3 Treatment of Angelica acutiloba (Siebold & Zucc.) Kitagawa (일당귀의 GA3 처리에 따른 생육특성)

  • 본 연구의 목적은 약용과 식용으로서의 활용도가 높은 일당귀[Angelica acutiloba (Siebold & Zucc.) Kitagawa]의 GA3농도에 따른 생육특성을 확인하고, 최적의 재배기술 개발 및 지속가능한 안정적인 작물생산에 대한 기초자료의 제공에 있다. 연구재료는 강원도 태백시에 위치하는 일당귀 재배농가에서 2021년 10월에 채종한 종자를 구입하였고, 영주시 풍기읍에 위치한 연구온실에서 연구를 수행하였다. 일당귀의 종자를 증류수에 48시간 침지 후 GA3농도 4조건(50 ppm, 100 ppm, 500 ppm, 1,000 ppm)에 24시간 처리 후 72구 포트에 파종 후 출아율과 생육특성을 조사하였다. 연구결과 일당귀 종자는 16일이 경과한 시점부터 출아를 시작하였고, 500 ppm처리구에서 73.3%로 가장 높은 출아율을 나타냈다. 생육특성조사는 출아 후 40일(1차)과 80일(2차) 경과된 시점에서 총 2회를 수행하였는데 1차 생육조사에서는 주근길이와 직경을 제외한 모든 생육특성에서 처리구별 생육의 차이가 확인되지 않았으나 2차 생육조사에서는 지상부와 지하부의 모든 생육특성에서 처리구별 생육의 차이가 확인되었다. 잎수(6.3개)와 주근길이(4.26 cm)는 500 ppm처리구에서 가장 높은 생육량을 나타냈고, 지상부 높이(21.81 cm), 잎길이(9.0 cm), 잎너비(10.77 cm), 주근직경(5.71 mm)은 1,000 ppm처리구에서 가장 높은 생육량을 나타냈다. 따라서 GA3 처리에 따른 일당귀의 생육특성은 생육초기보다 후기로 갈수록 발현량이 높게 나타나는 것으로 판단된다. 또한 저농도의 GA3처리보다 500-1,000 ppm 사이의 처리농도가 적합한 것으로 판단되며, 추후 일당귀의 유용성분 분석을 통한 이화학적 연구가 추가된다면 최적의 재배기술 확립을 통한 안정적인 작물 생산에 기여할 수 있을 것이라 사료된다.

Physio-chemical Characteristics of Starch Extracted from the Root of Pteridium spp. According to Cultivation Year (고사리 재배년차에 따른 뿌리 전분의 이화학적 특성)

  • 우리나라의 고사리 재배면적은 2018년 기준 5,406ha 수준을 보이고 있으며 전체 산채류 재배면적에서 가장 높은 비중을 차지하고 있다. 고사리(Ferns)는 고사리 속(Pteridium spp.)에 속하는 양치식물의 총칭으로서 우리나라에는 22과 70속 272종이 분포되어 있는 것으로 보고되었다. 고사리의 어린 순에는 가식부 100g 당 칼슘 15.0mg, 칼륨 185.0mg 등이 함유되어 있으며 골다공증, 심혈관 질환 등에 효과가 있고 식이섬유로 인해 변비 예방에도 유용한 것으로 알려져 있다. 고사리 뿌리에서 추출한 전분은 중국 및 일본 등지에서 면류나 제과용으로 이용되고 있어 고사리 재배의 부가가치를 향상시킬 수 있는 방안으로 기대되고 있다. 본 연구에서는 고사리의 재배년차간에 뿌리 전분의 생산성과 이화학적 특성을 조사하기 위해 2020년 3월 26일 각 재배년차별로 굴취한 뿌리를 세척 후 고무망치로 파쇄하고 수침 방법을 사용해 추출한 전분의 이화학적 특성을 조사하였다. 재배년차간 고사리 뿌리의 생산성은 5년생과 7년생에서 3,000 kg/10a 이상으로 유의하게 높은 수량을 보였다. 세척한 고사리 뿌리를 24시간 sodiun metasulfite 4% V/W 용액에 수침하여 전분을 추출한 후 24시간 간격으로 3회에 걸쳐 진탕, 침전과 세척의 과정을 거친 후 조사한 전분 추출 수율은 뿌리 생체중 대비 6.8~7.6% 수준이었으며 재배년차간 차이는 없었다. 추출한 전분의 색도분석에서 L 값은 재배년차가 높아질수록 낮아지는 경향을 보였으나 a 값은 3.7~4.8, b 값은 13.5~15.9로서 뚜렷한 차이를 보이지 않았다. 고사리 뿌리전분의 호화특성에서 최고점도는 3년생 고사리 뿌리에서 추출한 전분이 557.4 RVU로 높은 값을 보였으나 재배년차간 통계적 유의성을 인정할 수는 없었으며, 최저검도에서는 5년생에서 추출한 전분이 269.9RVUfh 낮은 경향을 보였다. 또한 최대 점도 시간은 5.0~5.2hr로 재배년차간 차이는 없었으며 호화 저지온도는 67.0~68.9℃ 범위로 유의한 차이를 보이지 않았다. 고사리 뿌리에서 채취한 전분의 무기물 함량을 조사한 결과 황의 함량은 1년생에서 0.081%로 유의하게 높았으며, 인산 함량은 3년생에서 72mg/kg으로 가장 높았고 1년생에서 31mg/kg으로 가장 낮은 값을 나타냈다. 칼륨의 함량은 7년생에서 추출한 전분에서 376.3mg/kg으로 높았으나 재배년차 간 뚜렷한 경향을 보이지 않았으며 칼슘, 마그네슙 및 철분 함량에서는 5년생 전분에서 유의하게 높은 경향을 보였다.

  • 껍질째 먹는 청포도 품종인 '샤인머스켓'은 망고포도라고 불릴 정도로 단맛이 강하고 풍부한 머스켓향을 가지는 것이 특징이다. 또한 포도알이 크고 무핵처리에 의해 씨가 제거되어 소비자 만족도가 높아 최근 각광받고 있는 포도 품종이다. 샤인머스켓에 대한 국내연구는 주로 재배조건에 따른 과실특성 위주로 보고되고 있어, 본 연구는 소비가 늘고 있는 샤인머스켓의 기능성에 대한 기초자료를 제시하고자 부위별 추출조건에 따른 총폴리페놀, 총플라보노이드 함량 및 항산화 활성을 조사하였다. 샤인머스켓은 2019년 11월에 경북 상주지역에서 생산된 것으로, 과피와 과육의 2가지로 나누어 동결건조하였으며, 추출용매(증류수, 에탄올, 메탄올)와 추출용매농도(30%, 50%, 70%, 100%) 조건에 따라 시료 무게대비 10배의 용매를 가한 후 초음파방법으로 2시간 추출하였으며, 이를 2회 반복하였다. 부위별 총폴리페놀과 총플라보노이드 함량은 과육보다 과피에서 유의적으로 높게 나타났으며, 항산화 활성 또한 과피에서 월등히 높은 값을 나타냈다. 추출조건에 따른 생리활성 물질의 함량과 항산화 활성은 과육과 과피에서 다르게 나타났는데, 과육의 경우 메탄올 추출 시 활성이 높게 나타난 반면, 과피는 에탄올 추출 조건에서 활성이 더 높게 나타났다. 과육의 경우, 70% 메탄올 추출 조건에서 가장 높은 활성을 보였으며, 총폴리페놀 0.99 mg CE/g dw, 총플라보노이드 0.31 mg CE/g dw, DPPH 라디칼 소거능 2.94 mM TE/g dw, ABTS 라디칼 소거능 6.82mM TE/g dw, ORAC 11.40 mM TE/g dw, FRAP 4.92 mM Fe(II)/g dw이었다. 이와 달리 과피는 30% 에탄올 추출 조건에서의 활성이 유의적으로 높게 나타났으며, 총폴리페놀 10.23 mg CE/g dw, 총 플라보노이드 5.22 mg CE/g dw, DPPH 라디칼 소거능 44.72 mM TE/g dw, ABTS 라디칼 소거능 123.25 mM TE/g dw, ORAC 208.38 mM TE/g dw, FRAP 96.24 mM Fe(II)/g dw이었다. 이상의 결과를 볼 때, 껍질째 샤인머스켓을 섭취하는 것은 기능성 측면에서 유익하며, 추후 부위를 나누지 않은 샤인머스켓을 대상으로 추출조건 설정 및 기능성 평가에 대한 연구를 진행할 예정이다.
