10-(치환)-레보푸록사신 유도체의 합성 및 항균 작용 (Synthesis and Antimicrobial Activity of 10-Substituted Levofloxacin Derivatives)

  • A series of (-)-9-fluoro-2,3-dihydro-3(S)-methyl-10-(4-substituted-1-piperazinyl)-7-oxo-7H-pyrido [1,2,3-de] [1,4]-benzoxazine-6-carboxylic acid (5-8) was synthesized and evaluated for antibacterial activity against gram(+) and gram(-) bacteria. These synthesized compounds showed a lower activity than levofloxacin.

  • Bioisostere approach has been shown to be useful to augment potency or to modify certain physiological properties of a lead compound. Based upon well documented bioisosterism, an isosteric replacement of benzene ring of 4-hydroxy-2-quinolone compound (L-695902) with a thiophene moiety was carried out to prepare the title compounds, 4-hydroxy-6-oxo-6,7-dihydro-thieno[2,3-b] pyrimidines 15. The resulting bioisosteric compounds 15 were evaluated for their antagonistic activity (birding assay) for NMDA receptor glycine site.

  • 담수답 토양중에서 배양된 Alachlor는 그 분해산물로 1-formyl-2, 3-dihydro-7-ethylindole, 2,6-diethylaniline, 2,6-diethylacetanilide, 2,6-diethyl-N-(methoxymethyl) acetanilide, 2-hydroxy-2', 6'-diethyl-N-(methoxymethyl) acetanilide, 그리고 삼종(三種)의 미확인 화합물을 생성하였다. 환(環)-$^{14}C$ 표식(標識) Alachlor의 사용으로 입증된 바와 같이 토양 현탁액중에서 Alachlor의 수용성 분해산물은 배양기간이 증가함에 따라 증가하였고 Rhizoctonia solani의 배양에서도 유사한 결과를 얻었다. Streptomyces lavendulae Ru3340-8은 주요분해산물로 2-hydroxy-2', 6'-diethyl-N-(methoxymethyl) acetanilide를 최고 25%까지 생성하는 반면 Bacillus brevis IFO 3331, Bacillus cruciviae, 그리고 Pseudomonas putida는 그것을 생성하지 않았다.

The Rhizomes of Acorus gramineus and the Constituents Inhibit Allergic Response In vitro and In vivo

  • The rhizomes of Acorus gramineus have frequently been used in traditional medicine mainly for sedation as well as enhancing brain function. In this study, the anti-allergic activity of A. gramineus was investigated. The 70% ethanol extract of the rhizomes of A. gramineus was found to inhibit the allergic response against 5-lipoxygenase (5-LOX)-catalyzed leukotriene (LT) production from rat basophilic leukemia (RBL)-1 cells and ${\beta}$-hexosaminidase release from RBL-2H3 cells with $IC_{50}$'s of 48.9 and > $200{\mu}g/ml$, respectively. Among the 9 major constituents isolated, ${\beta}$-asarone, (2R,3R,4S,5S)-2,4-dimethyl-1,3-bis (2',4',5'-trimethoxyphenyl)tetrahydrofuran (AF) and 2,3-dihydro-4,5,7-trimethoxy-1-ethyl-2-methyl-3-(2,4,5-trimethoxyphenyl)indene (AI) strongly inhibited 5-LOX-catalyzed LT production in A23187-treated RBL-1 cells, AI being the most potent ($IC_{50}=6.7{\mu}M$). Against ${\beta}$-hexosaminidase release by antigen-stimulated RBL-2H3 cells, only AI exhibited strong inhibition ($IC_{50}=7.3{\mu}M$) while ${\beta}$-asarone and AF showed 26.0% and 39.9% inhibition at $50{\mu}M$, respectively. In addition, the ethanol extract of A. gramineus showed significant inhibitory action against the hapten-induced delayed hypersensitivity reaction in mice by oral administration at 200 mg/kg. Therefore, it is suggested that A. gramineus possesses anti-allergic activity and the constituents including ${\beta}$-asarone and AI certainly contribute to the anti-allergic activity of the rhizomes of A. gramineus.

  • 뇨시료 중 amineptine의 (dihydro-10, 11-dibenzo[a, d] cycloheptenyl-5-amino-7-heptanoic acid) metabolites를 분석하기 위한 최적조건을 찾기 위하여 pH 변화에 따른 추출률과 세가지의 유도체화시약에 대한 반응성을 조사해본 결과 pH는 9.5, 유도체화시약은 carboxylic acid group에 MSTFA로 반응 시켰을 경우 좋은 결과를 나타내었다. GC/MS를 이용하여 amineptine을 복용한 사람의 뇨를 분석한 결과 amineptine과 그 대사물질인 dihydro-10, 11-dibenzo[a, d] cycloheptenyl-5-amino-5-pentanoic acid ($C_5$-metabolite)와 $C_5$-metabolite의 lactamized product인 ${\delta}$-lactam을 확인하였다. Amineptine과 그 metabolite들을 GC/MS-SIM mode로 분석하기 위한 monitoring ion들은 m/z 192를 공통 이온으로 선정하였으며, 각각의 분자이온을 선정하였다. Amineptine의 excretion study 결과, amineptine, ${\delta}$-lactam 및 $C_5$-metabolite는 4시간이내에 70-90%가 배설되었고 20시간 이내에 거의 배설이 완결되었다.

  • 새로운 항균제 및 항종양제를 합성하기 위하여 norfloxacin(8) 또는 ciprofloxacin(9)이 pteroic acid의 C-9위치에 치환되고 C-2 위치에 아미노기 대신 $CH_3$기가 치환된 새로운 pteroic acid 유도체 13a와 13b를 합성하였다. 이는 출발 물질인 norfloxacin과 ciprofloxacin의 piperazine N-4 위치에 2-amino-3-cyano-5-chloro-methylpyrazine(20)을 결합하여 1-alkyl(ethyl, cyclopropyl)-6-fluoro-1,4-dihydro-4-oxo-7-[[4-N- (2-amino-3-cyanopyrazin-5-yl)methyl]piperazin-1-yl]-3-quinolinecarboxytic acid(12a, 12b)를 합성하였다. 이를 각각 acetamidine. HCl과 고리화시켜 C-2 위치에 아미노기 대신 $CH_3$기가 치환된 새로운 pteroic acid 유도체 135와 13b를 각각 76.2%와 82.8%의 수율로 합성하였다. 그리고 이들 화합물에 대한 항균활성의 측정은 Pseudomonas aeruginosa ATCC9027을 포함하여 Gram-positive와 Gram-negative bacteria에 대하여 검토한 결과 합성한 화합물 13a와 13b는 일반적으로 norfloxacin보다 낮은 항균활성을 나타냈다.
