Characterization of Acid Phosphatase from Welsh Onion (파의 Acid Phosphatase의 특성)

  • Acid phosphatase (EC 3.1.3.2) from Welsh onion was partially purified by Sephacryl S-200 gel filtration and CM-Sepharose CL-6B ion exchange chromatography. The optimum pH and temperature of acid phosphatase from green onion were pH 5.5 and $60^{\circ}C$, respectively. The enzyme was the most stable at pH 6.0 and unstable above pH 9.0. The activation energy of the enzyme was determined to be 4.86kcal/mole. The enzyme utilized p-nitrophenyl phosphate most as a best substrate among tested possible substrates, while 5'-GMP and 5'-IMP were poor substrates for the enzyme. $K_{m.app.}$ of the enzyme with p-nitrophenyl phosphate as a substrate was identified as 0.87mM. Among metal ions and inhibitors tested, $Cr^{+++},\;Zn^{++},\;Cu^{++}$, molybdate and metavanadate ions inhibited the enzyme reaction drastically.

Early Changes after Death of Plaice, Paralichthys olivaceus Muscle -3. Effect of Killing Methods on Changes in Content of ATP and Its Related Compounds and Lactate- (넙치(Paralichthys olivaceus)육의 사후 조기 변화 -3. 치사 방법이 ATP 관련 물질과 유산 함량의 변화에 미치는 영향-)

  • To clarify the effects of killing methods on biochemical changes of plaice, Paralichthys olivaceus muscle at early period after death, cultured plaices were killed by the following four different methods; 1. spiking at the brain instantly. 2. letting them to die in the air. 3. dipping in sea water including anesthetic. 4. electrifying in sea water. Immediately after death, the changes in ATP and its related compounds, ATP breakdown, and IMP or lactate accumulation rates of muscle during storage at $5^{\circ}C$ were studied. ATP in samples killed by letting and electrifying were decomposed more rapidly than spiking and dipping samples. The rate constant of ATP breakdown were $0.429h^{-1}$ for samples killed by electrifying, $0.224h^{-1}$ for letting samples, $0.195h^{-1}$ for spiking samples, and $0.167h^{-1}$ for dipping samples. The maximum speed and content of IMP or lactate accumulation were showed in samples killed by electrifying among the all killing methods. The rate constant of lactate accumulation were $2.256h^{-1}$ for samples killed by electrifying, $1.123h^{-1}$ for letting samples, $0.534h^{-1}$ for spiking samples, and $0.526h^{-1}$ for dipping samples. From the results above, it was revealed that electrifying in sea water could accelerate ATP breakdown and accumulation of IMP or lactate among the all killing methods. The other hand, dipping in sea water including anesthetic delayed those changes.

Enzymatic Method for Measuring ATP Related Compounds in Fish Sauces (효소법에 의한 액젓중의 ATP 관련물질 측정)

  • HPLC method usually has been used for the determination of ATP and its related compounds in fish muscle and fish sauce. But, total amount of ATP related compounds in fish sauce is determined less than that of fish muscle. In order to establish the extract analysis method for ATP related compounds in fish sauce, a new enzymatic method was developed and compared with existing HPLC method. Fish sauce was extracted with chilled perchloric acid and neutralized to Ph 7.0 with potassium hydroxide solution, the extract was used as sample analyzed by HPLC as usual. On the other hand, for sample analyzed by enzymatic method, 1 ml extract solution was pipetted into test tube. To the tube, 0.5ml of mixed suspension adenosinedeaminase (4U), nucleosidephosphorylase (0.02U) and xanthineoxidase (0.03U) suspended in 2.0ml of 1/15 M sodium phosphate buffer solution pH 7.6 and 1.5ml deionized water wereadded for the decomposition of IMP, HxR and Hx to uric acid at $37^{\circ}C$ for 40 minutes. Total uric acid was determined by measuring optical density at 290nm. In HPLC method, salt decreased the total amount of ATP related compounds by $13.6\~16.2\%$ at $2.5\%$ concentration, but no effect in enzymatic method. IMP, HxR and Hx were detected at 254nm, while uric acid at only 290nm. The ratio of the total amount of ATP related compounds by HPLC method was about $45\%$ of that by enzymatic method in fish sauce. Form these results, enzymatic method is more accurate and simple than HPLC method for analysis of ATP related compounds in fish sauce.

  • This study was directed to define the taste compounds of crucian carp, Carassius caressius, free amino acids, nucleotides and their related compounds, organic bases, sugars, organic acids and minerals in the extracts of crucian carp were analyzed, and then followed by sensory evaluation of synthetic extracts prepared from 44 pure chemicals on the basis of the analytical data. Taste panel assessments of synthetic extracts prepared with each extractive component omitted were carried out by a triangle difference test, and changes in taste profile were assessed. In free amino acid composition, histidine was dominant occupying $46\%$ of the total free amino acids. The other abundant free amino acids were glycine, lysine, alanine and taurine. As for the nucleotides, IMP was dominant showing about $80\%$ of the total of nucleotides. The most abundant organic base was total creatinine. The content of betaine was poor and TMAO were trace in content. The main organic acids were succinic, propionic, butyric and valeric acid. Small amount of glucose, fructose and inositol were detected and ribose and arabinose were trace in content $K^+,\;Na^+,\;PO_4^{3-}\;and\;Cl^-$ were found to be the major ions and small amount of $Ca^{++}\;and\;Mg^{2+}$ were deleted. Judging from the results of omission test, the major components which contribute to produce the taste were serine, glutamic acid, lysine, arginine, tyrosine, phenylalanine, IMP, $Na^+,\;K^+\;and\;PO_4^{3-}$.

  • In order to elucidate the taste compounds of wild eel, Anguilla japonica, free amino acids, nucleotides and their related compounds, organic bases, sugars, organic acids and minerals were analyzed, and then followed by organoleptic test of synthetic extract prepared on the basis of the analytical data. Taste panel assessments of synthetic extracts prepared with each extractive component omitted were carried out by a triangle difference test, and changes in profile were assessed. In free amino acid composition, lysine was dominant occupying $25\%$ of total free amino acid. The other abundant amino acids were glycine, arginine, alanine and histidine. From the results of analysis of nucleotides, IMP was dominant showing about $70\%$ of total nucleotides while ATP, ADP, AMP, inosine and hypoxanthine were low in content. Among organic bases total creatinine was abundant. The amount of betaine was 24 mg/100 g. The main organic acid were butyric acid, valeric acid and succinic acid. As for the sugars, glucose and inositol were 1-2 mg/ 100g in content. $K^+,\;Na^+,\;PO_{4}^{3-}\;and\;Cl^-$ were found to be the major ions. From the results of omission test the major components which contribute to produce the taste were glycine, serine, glutamic acid, IMP, $Na^+,\;K^+,\;Cl^-,\;PO_{4}^{3-}$, lysine, alanine, isoleucine, aspartic acid and creatinine.

Among organic bases, total creatinine was abundant and its nitrogen content occupied more than $50\%$ of the total extractive nitrogen. The amount of betaine was 18mg/100g and trace amount of trimethylamine and trimethylamine oxide were detected. The main organic acids were succinic, butyric, propionic and valeric acid. On the other hand, oxalic, fumaric, maleic, tartaric and citric acids were analyzed in trace. As for the sugars, glucose was found to be the most abundant monosaccharide. Extremely small amounts of fructoae, inositol were also detected and ribose and arabinose were trace in content. $K^+,\;Na^+,\;PO_{4}^{3-}$ and $Cl^-$ were found to be the major ions and small amount of $Ca^{2+}$ were defected. The synthetic extract, prepared with about 40 pure chemicals based on the analytical data satisfactorily revealed the natural taste of the original extract except slight difference in meaty taste and mildness. From the results of omission test the major components which contribute to produce the taste were serine, IMP, succinic acid and $PO_{4}^{3-}$.

The analysis of useful components in Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium (팽이버섯 자실체, 팽이버섯 균사체 및 동충하초 균사체의 유용성분 분석)

  • Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium were analyzed for their proximate composition, protein-bound polysaccharide, nucleic acid and amino acids. The content of ash and crude fiber in F. velutipes fruit body were higher than F. mycelium and C. militaris mycelium. C. militaris mycelium showed the highest crude fat content while F. velutipes fruit body had lowest. Nitrogen free extract content of the samples varied from 56.8% in F. velutipes fruit body to 61.9% in F. velutipes mycelium. The compositions of total protein and total free sugars of protein-bound polysaccharide were found to be significant differences for all samples. Nucleic acid related compounds were identified the 5'-GMP, 5'-XMP, 5'-IMP in all samples. The content of total nucleic acids were high in the orders of F. velutipes myclial (286.71 mg%), F. velutipes fruit body(187.36 mg%) and C. militaris mycelial(76.85 mg%). The highest content of 5'-GMP was found in F. velutipes fruit body. The most nucleic acid of F. velutipes mycelial and C. militaris mycelial were the 5'-XMP. As for the analysis of total amino acids, seventeen amino acids were identified by HPLC and the major amino acid was glutamic acid in all samples. The content of total amino acids were high in the orders of F. velutipes fruit body(19,919 mg%), F. velutipes mycelium(19,018 mg%) abd C. militaris mycelium(18,965 mg%). We determined the developing new food product such as amino acid drink and amino acid containing food using extracts of Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium.

Nonlinear distortion compensation of LD and optical intensity modulator using predistorter (전치왜곡기를 이용한 LD 및 광변조기의 비선형 왜곡 보상)

  • Nonlinear distortion introduced by the optical transmitter using a direct modulation of laser diode(LD) and an indirect modulation of optical modulator can limit considerably the performance of analog optical link system such as subcarrier multiplexing system. In this paper, we have compensated the nonlinear distortion of LD and optical modulator using electrical predistorter. In the case of LD, two-tone experiments using 877.5 MHz and 882.5 MHz have shown reduction of 19 dB in third-order intermodulation product(IMP$_3$). And in the case of optical modulator, we have achieved nearly 21 dB reduction in IMP3 through two-tone experiments using 1.8975 GHz and 1.9025 GHz. The spurious free dynamic ranges of LD and optical modulator are also improved by 5 dB/Hz$^{2}$ 3/ and 7 dB/Hz$^{2}$ 3/, respectively.

Taste Compounds of Abalone, Haliotis diversicolor japonica (오분자기의 정미성분)

  • Abalone, Haliotis diversicolor japonica, is one of the most palatable sea foods in Jeju-Do, Korea : however relatively little is known about the characteristics of its taste. This study was undertaken to establish the basic data for evaluating the taste compounds in the extract of abalone including nucleotides and their related compounds, free amino acids, trimethylamine oxide, trimethylamine and total creatinine. The amount of AMP was $2.21{\mu}mole/g$ while IMP and inosine was trace in content. In the extracts, taurine, arginine and glycine were abundant holding $39.9\%,\;25.4\%,\;14.0\%$ of total free amino acids content respectively, but tyrosine and phenylalanine was trace in content. TMAO, TMA and total creatinine were poor in content marking $5.4mg\%,\;1.0mg\%\;and\;5.0mg\%$. It was presumed that the characteristic flavor of abalone was mainely attributed to free amino acids. Organic acids, nucleotides and their related compounds played auxiliary role in the characteristic flavor of abalone.

Basic Studies on Developing Equipment for Waterless Transportation of Live Fish (활어의 무수 수송 장치 개발을 위한 기초적 연구)

  • This study was undertaken to get basic data for the cold-waterless transportation of live fish. The optimal cold temperature of plaice, Paralichthy olivaceus was determined by checking the changes of dissolved oxygen and ammonia in the sea water and the survival times during storage at various temperatures. After determination of optimal temperature for transportation, the changes of serum components and muscle components of live plaice were also carried out during storage at cold($5^{\circ}C$)-waterless conditions and the recovery conditions($10\%$ density at $15^{\circ}C$). At higher storage temperature, decreases in dissolved oxygen and the increases in ammonia in seawater were observed. In addition, the survival time was short at low temperature($0^{\circ}C\;and\;3^{\circ}C$). Almost all of the serum components(hemoglobin, glucose, LDH, GOT and GPT) of live plaice gradually increased during storage in cold-waterless conditions, and then those values decreased to the initial levels after $3{\sim}10hrs$ storage in conditions of recovery. The concentration of ATP in the muscle steadily decreased during storage in cold-waterless conditions. The contents of ADP and IMP seemed to be directly related to the extent of ATP breakdown. ADP and IMP thus showed a gradual increase during storage. The level of lactate in the muscles gradually increased during these storage times, also. On the other hand, the levels of those components in the muscle entirely recovered to their original levels within $3{\sim}6hrs$ storage after they were returned to conditions of recovery. The ratio of ATP to the ATP and its related compounds${ATP/(ATP+ADP+AMP+IMP){\times}100}$ in the muscle showed $45\%$ after 18hrs storage in cold-waterless conditions. Otherwise, ratios returned to their original levels within $3{\sim}6hrs$ of storage in recovery conditions.
