Effects of the Constituents of Paeonia lactiflora Root on Arachidonate and NO Metabolism

  • In order to establish the anti-inflammatory cellular mechanism of the paeony root(Paeonia lactiflora, Pall, Paeoniaceae), the constituents including paeoniflorin, albiflorin, (+)-catechin, paeonol, benzoic acid and methyl gallate were evaluated for their effects on arachidonate and NO metabolism. Among the compounds tested, only paeonol weakly inhibited cyclooxygenase-2-mediated $PGE_2$ production from LPS-treated RAW 264.7 cells. (+)-Catechin and methyl gallate weakly inhibited inducible nitric oxide synthase-mediated NO production from the same cell line. In particular, methyl gallate significantly inhibited 5-lipoxygenase from RBL-l cells with an $IC_{50}$ of 8.4 ${\mu}M$. These results suggest that the inhibition of these components on arachidonate and NO metabolism may contribute at least in part to anti-inflammatory mechanism of the paeony root.

Comparison of Hydroxyl Radical, Peroxyl Radical, and Peroxynitrite Scavenging Capacity of Extracts and Active Components from Selected Medicinal Plants

  • The ability of 80% ethanol extracts from five medicinal plants, Aralia continentalis, Paeonia suffruticosa, Magnolia denudata, Anemarrhena asphodeloides, and Schizonepeta tenuifolia, to neutralize hydroxyl radical, peroxyl radical and peroxynitrite was examined using the total oxyradical scavenging capacity (TOSC) assay. Peroxyl radical was generated from thermal homolysis of 2,2'-azobis(2-methylpropionamidine) dihydrochloride (ABAP); hydroxyl radical by an iron-ascorbate Fenton reaction; peroxynitrite by spontaneous decomposition of 3-morpholinosydnonimine N-ethylcarbamide (SIN-1). The oxidants generated react with $\alpha$-keto-$\gamma$-methiolbutyric acid (KMBA) to yield ethylene, and the TOSC of the substances tested is quantified from their ability to inhibit ethylene formation. Extracts from P. suffruticosa, M. denudata, and S. tenuifolia were determined to be potent peroxyl radical scavenging agents with a specific TOSC (sTOSC) being at least six-fold greater than that of glutathione (GSH). These three plants also showed sTOSCs toward peroxynitrite markedly greater than sTOSC of GSH, however, only P. suffruticosa revealed a significant hydroxyl radical scavenging capacity. Seven major active constituents isolated from P. suffruticosa, quercetin, (+)-catechin, methyl gallate, gallic acid, benzoic acid, benzoyl paeoniflorin and paeoniflorin, were determined for their antioxidant potential toward peroxynitrite, peroxyl and hydroxyl radicals. Quercetin, (+)-catechin, methyl gallate, and gallic acid exhibited sTOSCs 40~85 times greater than sTOSC of GSH. These four components also showed a peroxynitrite scavenging capacity higher than at least 10-fold of GSH. For antioxidant activity against hydroxyl radical, methyl gallate was greatest followed by gallic acid and quercetin. Further studies need to be conducted to substantiate the significance of scavenging a specific oxidant in the prevention of cellular injury and disease states caused by the reactive free radical species.

Simultaneous detection of five biocides in household products by gas chromatography-mass spectrometry

  • A gas chromatography-mass spectrometric method was developed for determining 5-chloro-2-methyl-4-isothiazolin-3-one (CMIT), 2-methyl-4-isothiazolin-3-one (MIT), 1,2-benzisothiazolin-3-one (BIT), 3-iodo-2-propynyl butyl carbamic acid (IPBC) and benzoic acid (BA) in household products. A 0.5 g sample was placed in a test tube and dissolved with 5 mL water, 5 mL methylene chloride and 1.0 mL methanol. The solution was extracted by ultra-sonication followed by mechanical shaking using the salting out effect. Under the established condition, the lowest quantification limits of all analytes were in the range of 0.04-10 mg/kg and their relative standard deviations were less than 8.0 %. The method was used to analyze 10 household products. As a result of analyzing 10 household products, MIT was detected in the range of 1.2-3.5 mg/kg in 3 of 10 samples, CMIT was detected in the range of 2.6-8.2 mg/kg in 3 of 10 samples, and BA was detected in the range of 5.0-15 mg/kg in 4 of 10 samples. Meanwhile, BIT and IPBC were not detected in any of the products. It has been shown that this method can be used for the simultaneous determination of biocides with various physical and chemical properties in household products.

Screening of the Antioxidant Defense Systems from Parthenocisuss tricuspidata PLANCH (담쟁이덩굴의 항산화 방어계의 탐색)

  • The non-enzymatic antioxidants and antioxidant enzyme from the extracts of Parthenocissus tricuspidata PLANCH. were examined in order to utilize natural product for cerchemopreventive agents. The antioxidant potential and enzyme activities on plant positions in the extracts of Parthenocisuss tricuspidata PLANCH. showed considerable differences. The antioxidant activity of the leaf extracts by Ethyl acetate fractions of Parthenocisuss tricuspidata PLANCH. was the highest among three positions ($7.57\mu\textrm{g}/m\ell$). The highest activities showed in S-5 (in leaf), S-4 (in stem) and S-3 (in root) fraction by Silicagel column chromatography and the antioxidant activity showed, in purified extract of each positions, $7.06\mu\textrm{g}/m\ell$ (in leaf), $6.99\mu\textrm{g}/m\ell$ (in stem) and $12.39\mu\textrm{g}/m\ell$ (in root) respectively. The activities of DPPH by LH-20 column chromatography revealed much higher than those by silica-gel column chromatography. These were identified as the phenolic compounds known as antioxidant compounds such as Benzoic acid(Gallic acid), 1-methyl-3-(2-phenylethen) benzene, phloroglucinol and 1,2-dihydroxy-4-(1-propyl)benzene by GC/MS. POD activities in the stem and root were higher than in the leaf. SOD activity was highest in the leaf, stem and root activity was comparatively low. Especially, SOD activity in leaf was over 2 times higher than root.

Synthetic Conditions of an Aspartame Precursorby Immobilized Thermolysin (고정화 Thermolysin을 사용한 아스파탐 전구체의 최적 합성조건 선정)

  • N-Benzoyl-L-aspartyl-L-phenylalanine methyl ester(BzAPM), a novel aspartame precursor, was investigated for its enzymatic synthesis by immobilized thermolysin using a water-miscible organic solvent system. The substrate used were N-benzoyl-L-aspartic acid(BzAsp) and L-phenylalanine methyl ester (PheOMe). Synthetic conditions such as substrates concentration, temperature, pH, and some metallic ions were varied to study their effects on BzAPM synthesis. The synthetic reaction rate increased linearly as the PheOMe concentration increased at a constant concentration of BzAsp(100 mM), and the maximum reaction rate was obtained at BzAsp concentration of 200 mM when 300 mM PheOMe was used. The optimum pH and temperature were found to be 6.1 and $40^{\circ}C$, respectively. The metallic ions such as $Zn^{2+},\;Mg^{2+},\;Mn^{2+},\;Fe^{2+},\;Pb^{2+}\;and\;Cu^{2+}$ at 5 mM level showed inhibitory effect on BzAPM synthesis, while $Co^{2+}$ and $Ca^{2+}$ ion increased synthesis. $Co^{2+}$ ion was also found to have synergistic effect with $Ca^{2+}$ ion. Benzoic acid, L-phenylalanin and NaCl showed inhibitory effect.

Two Phenolic Compounds Isolated from Umbilicaria esculenta as Phospholipase $A_2$ Inhibitors (석이로부터 분리한 페놀성 화합물의 phospholipase $A_2$ 저해활성)

  • Two depsides, medicinal herb products isolated from the methanol extract of Umbilicaria esculenta, inhibited human synovial fluid Phospholipase $A_2\;(PLA_2)$ ($IC_{50}$ of 0.22 and 0.26 mM, respectively). In the course of screening for antiinflammatory compounds from natural products, we successfully isolated two depsides $PLA_2$ inhibitory compounds, Orcinol and methyl orsellinic acid. The compounds were identified as orcinol and methyl orsellinic acid on the basis of various NMR studies including $^1H,\;^{13}C$ and DEPT experiments.

Studies on the Production of (10-Deacetyl) Baccatin III in Cell Cultures of Taxus baccata Pendula (주목세포 배양에 의한 (10-Deacetyl) Baccatin III 생산 연구)

  • Enhanced production of (10-deacetyl) baccatin III and related taxanes was observed in suspension cultures of Taxus baccata Pendula. six % of initial glucose and sucrose concentration increased 10-deacetyl baccatin III production 3.5 and 2.5 times, respectively. Methyl jasmonate, as an elicitor, increased taxane production. Time course changes of taxane production after methyl jasmonate addition showed that baccatin II and 10-deacetyl baccatin III were detected first and paclitaxel, 10-deacetyl taxol and cephalomanine were produced in sequence. Feeding experiments with $500{\mu}M$ of benzoic acid increased 10-deacetyl baccatin III production 10 times. Baccatin III production was also increased 8 times by feeding of $500{\mu}M$ of lysine as a precursor.

  • The interaction of tetraphenylphthalic anhydride with o-chlorotoluene under Friedel-Craft condition gives 2-(4-chloro-3-methyl)benzoyl-3,4,5,6-tetraphenyl benzoic acid(1), which on reaction with hydrazine derivatives gave phthalazinones (2a-d). The behaviour of (2a) towards carbon electrophiles and carbon nucleophiles has been investigated. The chlorophthalazinones (4a) also has been synthesized from the action of $PCl_5/POCl_3$ on (2a). The behaviour of (4a) towards nitrogen, and oxygen nucleophiles also have been described.

Studies on the Separation and Identification of Acids in Izmir Tobacco Leaves (Izmir 잎담배 중 Acids 성분의 분리 및 확인에 관한 연구)

  • This study was carried out to separate and identify the acidic compounds in tobacco leaves. Izmir tobacco leaves was extracted with isopropyl ether and the extract was concentrated. The concentrate was extracted with 6% NaHCO3 aqueous solution. The aqueous extract was acidified with sulfuric acid, and extracted with diethyl ether. The acidic material was fractionated on silicic acid column using a benzene-methanol mixture with a stepwise increasing methanol concentration. The resulting fractions were esterified with diazomethane, and then identified by GC, GC/MS using SPB -5 fused silica capillary column. Most of acidic compounds in Izmir tobacco leaves were elected from fraction B which was benzene-methanol(98 : 2) mixture on silicic acid column chromatography. The identified acidic compounds of Izmir tobacco leaves were 18 saturated acids, 8 unsaturated acids, 5 dicarboxylic acids, 13 aromatic acids and 7 terpenoid acids. The major acidic compounds of lzmir tobacco leaves were 2- methylbutanoic, 3-methyl butanoic, 3- methylpentanoic, hexanoic, nonanedioic, phenylacetic, benzoic, 4- methoxybenzoic, 3, 5- dimethoxybenzoic, methoxycinnamic and 3, 4- dimethoxycinnamic acid.   • The bioassay-guided fractionation of protective agents against sepsis-induced lethality from the root bark of Paeonia suffruticosa led to the isolation of ten known compounds: paeonol (1), 2,5-dihydroxy-4-methoxyacetophenone (2) methyl 3-hydroxy-4- methoxybenzoate (3), acetovanillone (4), benzoic acid (5), benzoylpaeoniflorin (6), paeonoside (7), paeoniflorin (8), oxypaeoniflorin (9) and apiopaeonoside (10). Among them, 4 exhibited the highest survival rate in a dose-dependent manner (100% with a dose of 30 mg/kg versus 16.7% for the control experiment) and showed reduction of plasma alanine aminotransferase (ALT) value on the in vivo assay model of sepsis induced by LPS/GalN.
