Methicillin, Cefamandole, Oxytetracycline에 의한 사람 호중구 Elastase의 변화 (Antibiotics; Methicillin, Cefamandole and Oxytetracycline, Can Modulate the Activity of Human Neutrophil Elastases)

  • 사람 혈액속의 elastase와 관련된 질병에 대한 연구는 다양한 저해제의 개발을 동반해 왔으며, 최근 항생제도 그 관심대상이 되고 있다. 두 단계의 액체 크로마토그래피를 거쳐 얻은 고순도의 elastase에 12종의 항생제를 처리하였다. 세포벽합성 저해제로 알려져 있는 penicillin계와 cephalosporin계 항생제를 각각 3종씩 처리한 결과, methicillin과 cefamandole은 10mM 농도에서 elastase 활성을 50% 이상 저해하였지만, 나머지는 거의 10% 미만이었다. 단백질합성 저해제 중 oxytetracycline의 elastase에 대한 저해효과는 10mM 농도에서 95% 이상으로 매우 탁월하였으며 $(IC_{50}=0.3mM)$, gentamicin도 50% 이상 저해하였으나, 다른 aminoglycoside나 chloramphenicol은 역시 10%미만이었다. 실험해 본 항생제 가운데, oxytetracycline, cefamandole, methicillin, gentamicin 등은 elastase에 대한 강력한 저해제였으며, 그 작용기전은 항생제의 알려진 약리학적 기전과는 다른 차원의 모델임이 분명하였다.

Anti-inflammation effect of Exercise and Korean red ginseng in aging model rats with diet-induced atherosclerosis

  • BACKGROUND/OBJECTIVES: The aim of this study was to investigate the effects of exercise (EX) and Korean red ginseng (KRG) on inflammation mechanism in aging model rats with diet-induced atherosclerosis. MATERIALS/METHODS: Forty-eight male Sprague-Dawley rats were divided into 6 groups: Young control (Y-C), Aging control (A-C), A-C with HFD (AHF), AHF with EX (AHF-EX), AHF-EX with KRG (AHF-EX+RG), and AHF with KRG (AHF-RG). Aging was induced by D-gal (100mg/kg) and atherosclerosis was induced by HFD (60% fat) for 9 weeks. The experimental rats were performed swimming (60 min/day, 5 days/week) and supplied KRG orally (dose of 200 mg/kg) for 8 weeks. All rat aorta samples were harvested for biochemical and immunohistochemical analyses. REULTS: The EX and KRG supplementation significantly inhibited body weight and levels of TC, TG, LDL-C, and enhance of HDL-C compared with untreated AHF groups. AHF-EX, AHF-EX+RG, and AHF-RG group showed a decreased plasma CRP and increase plasma NO activities compared to AHF group. In addition, these groups revealed reduced 4-HNE, NF-kB, TNF-, ${\alpha}$, IL-6, COX-2, ICAM-1, VCAM-1 and enhanced eNOS expression in the aorta. CONCLUSION: These results suggest that EX alone, KRG alone, and combined treatment of EX and KRG may be an effective anti-inflammatory therapeutic for the atherosclerosis, possibly acting via the decreased of CRP and pro-inflammation proteins and the increased NO and eNOS.

천연 소재 조성물의 3.3.3 System Diet Program이 각 단계별 신체조성, 혈청 지질 개선 효과 및 Leptin 수준에 미치는 영향에 관한 연구 (The Effects of Programmed Dietary System of a Mixture of Natural Supplements on the Composition, Serum Lipids and the Leptin Levels)

  • This study was to investigate the effects of a silk peptide, and prune and raspberry mixture on the body fat, serum lipid composition and fat distribution in Korean women during the time they received these supplements. During the 4 month period of this research (June to October, 2002), 75 women (average age 22.9 yr) were selected as subjects. Their nutrient intake was investigated by use of a questionnaire and the 24-hour recall method. Antropometric assessments of the subjects were investigated by using the SBIA Method (Segmental Bioimpedance Assay, Inbody 3.0). The results were as follows: their mean body weight was 59.3 kg, their mean body height was 161.7 cm and their mean Basal Metabolic Indices (BMI) was 22.7. The status of their dietary fiber intake significantly increased (p < 0.05) after use of the supplements. Their body fat significantly decreased following the $3^{rd}$ stage of supplementation (p < 0.05), and their percent of body fat signiflcantly decreased after supplementation (p < 0.05). Their triceps and suprailiac circumference significantly decreased following the $3^{rd}$ stage of supplementation (p < 0.05). Their waist circumferences and waist-hip ratios (WHR) significantly decreased after both the $2^{nd}$and $3^{rd}$ stage of supplementation (p < 0.05). Their total cholesterol significantly decreased following the $3^{rd}$ stage of supplementation, as compared to their baseline results and those obtained following the 1$^{st}$ stage of supplementation (p < 0.05). Their LDL-cholesterol and LDL / HDL ratios significantly decreased following the $3^{rd}$ stage of supplementation, as compared to their baseline results and those obtained following the 1$^{st}$ stage of supplementation (p < 0.05). Their 4-HNE and total antioxidant statuses significantly decreased following the $2^{nd}$ and $3^{rd}$stage of supplementation, as compared to their baseline results and those obtained following the $1^{st}$ stage of supplementation (p <0.05). Their $2^{nd}$ stage supplementation results were significantly negatively correlated with their triceps measurements and WHR (p < 0.05). Their $3^{rd}$stage supplementation results were significantly negatively correlated with their anterior thigh measurements (p < 0.05). The above results show that prune, raspberry, silk peptide and dietary fiber supplementation to a routine diet improves body fat distribution, total cholesterol, and triglyceride levels. The decrease of abdominal fat and WHR were especially noteworthy. This implies a decrease of risk factors. (Korean J Community Nutrition 8(3) : 356~367, 2003)003)

Recovery Effects from Oxidative Cell Damage by So-Hap-Hyang-Won on Bovine Aortic Endothelial Cells (BAEC)

  • So-Hap-Hyang-Won, a traditional oriental medicine used in the treatment of stroke patients, was examined for its ability to reverse the cell damage caused by lipid peroxidation products and oxidative stress in bovine aortic endothelial cells (BAEC). The effects of herbal medicine on cell proliferation and recovery of oxidative damaged situation were studied in BAEC, which was considered an appropriate in vitro model for stroke resulting from various vascular diseases prevalent in advanced age. In a clinical study of stroke patients, So-Hap-Hyang-Won appeared to improve considerably arm and leg movements as well as consciousness disturbance condition, compared with other traditional medicines used for stroke. When BAEC were treated with extracts of the lyophilized herbal medicines, only that of So-Hap-Hyang-Won stimulated cell proliferation and showed no toxicity even at high concentrations. In studies of BAEC treated with extracts of the lyophilized material of the 14 components of So-Hap-Hyang-Won, only the extract of Foeniculi Fructus stimulated cell growth at all concentrations tested. Moreover, when cells were treated with Foeniculi Fructus (10 and 100 mg/ml) extract after prior exposure to t-BHP ($l0\mu\textrm{M}$) or HNE ($0.2\mu\textrm{M}$), lipid peroxidation products which are known to be involved in aging and vascular diseases, or after the exposure to SIN-l ($500\mu\textrm{M}$), which generates nitric oxide (NO) and other reactive oxygen species, there was substantial recovery from the oxidative damage, presumably due to the radical-scavenging effect of Foeniculi Fructus extract. Foeniculi Fructus not only showed stimulatory effects on cell growth and cell damage repair in BAEC, but also appeared to show the most anti-aging activity among all the herbal components of So-Hap-Hyang-Won.

소나무 천연집단(天然集團)의 변이(變異)에 관(關)한 연구(硏究)(I) (The Variation of Natural Population of Pinus densiflora S. et Z. in Korea(I) -Characteristics of Needle and Wood of Chuwang-san, An-Myeon-do and Odae-san Populations-)

  • 소나무 천연집단(天然集團)의 변이(變異)를 연구(硏究)하기 위(爲)해 주왕산(경북), 안면도(충남), 오대산(강원)의 3개 집단(集團)을 선발(選拔)하고 각 집단(集團) 20개체(個體)를 대상으로 하여 외형적(外形的) 특성(特性), 침엽(針葉)의 특성(特性) 및 재질특성(材質特性)에 대한 변이(變異)를 조사(調査)하였다. 그 결과(結果) 다음과 같은 성적(成績)을 얻었다. 1. 외형적(外形的) 특성(特性)중 수간(樹幹)의 통직성(通直性), 가지의 굵기, 분지각(分枝角)등에서 오대산 집단(集團)이 타 집단에 비하서 바람직한 형태(形態)를 가지고 있다. 2. 침엽(針葉)의 거치밀도(鋸齒密度), 기공열수(氣孔列數), 수지도수(樹脂道數) 등의 형질(形質)에서 집단간(集團間) 개체간(個體間)의 차이(差異)를 인정할 수 있다. 3. 수지도지수(樹脂道脂數)(RDI), 하표피층수(下表皮層數), 유관속(維管束)의 접근도(接近度)등의 관찰결과(結果) 조사된 3개 집 단 모두가 순수한 소나무라는 기준에서 약간의 편의偏倚)가 있다. 4. 추재율(秋材率)과 비중(比重)은 집단간(集團間)의 차이(差異)를 인정할 수 있으나 연륜폭(年輪福)과 가도관(假導管)의 길이에서는 그 차이를 인정할 수 없었다. 개체간(個體間)에는 비중(比重)과 가도관(假導管) 길이에서 유의차(有意差)가 인정된다. 5. 추재율(秋材率)은 11~20년에서 최고치(最高値)를 보이고 21~30년 사이에 감소하여 그 후로는 큰 변화가 없었다. 비중(比重)은 연륜증가에 따라 증가하는 경향을 보였으나 안면도 집단(集團)과 오대산 집단(集團)이 21~30년사이에 한번 감소하였고 주왕산 집단(集團)은 조금씩 계속 증가하였다. 가도관(假導管)은 3개 집단(集團) 모두 연륜의 증가에 따라 증가하는 경향을 보인다. 6. 이곳에 취급한 몇 형질(形質)로써 소나무 3개 집단간(集團間)의 차이가 안정될 수 있다.
