해양심층수 소금을 이용한 민어 염건품 제조 및 저장 중 품질 변화 (Changes in Quality of Salted and Dried Brown-Croaker Product Prepared with Deep Seawater Salt)

  • 해양심층수 및 해양심층수 소금을 이용하여 민어 염건품 제조 조건을 확인한 결과, 민어 어체의 전처리 조건이 염건품 품질에 매우 큰 영향을 주는 것으로 나타났으며, 특히 내장만을 제거하고 부레와 복부지방층을 제거하지 않으면 염의 침투도 원활하지 않으며, 가공 중 지방의 산패나 미생물의 성장에 의한 품질 저하가 일어나는 것을 확인하였다. 아울러 어체 중량에 대해 20% 염 농도로 염장하는 것이 적절하였고, 특히 이 염장 농도에서 염장 시간에 따른 제품의 염도나 품질에 영향을 주는 것이 확인되었는데, 염장시간은 상온($20\sim25^{\circ}C$)에서 12시간이 적절하였다. 한편, 건조온도는 $30^{\circ}C$ 조건에서 24시간 건조하는 것이 제품의 품질에 가장 바람직한 조건이었다. 한편, 해양심층수 소금과 시판 소금을 사용하여 동일한 조건에서 제조한 민어 염장품의 저장 중 품질 변화를 관찰한 결과, 시판 소금의 경우 소금에서 유래하는 미생물 등에 의해 가공 중 품질의 저하는 물론이고 저장 중에도 POV나 AV의 증가, 생균수의 증가 정도가 해양심 층수 소금을 사용한 것에 비해 높은 것으로 나타났다. 또한 지방산 변화는 관능검사 결과, 시판 소금에 비해 해양심층수 소금이 민어 염건품 제조에 있어서 가공 공정 중이나 저장 중에도 품질을 유지하는데 훨씬 효과적이라는 것을 알 수 있었다. 결론적으로 해양심층수는 염장 후 2차 수세 시에 해양심층수를 사용하고, 염장 시에는 어체 중량에 대해 20% 농도로 상온에서 12시간 정도 염장을 행한 다음 $30^{\circ}C$ 온도 조건에서 24시간 건조하는 조건이 적절하다는 것을 알 수 있었다. 아울러 민어 염건품의 경우 냉장 유통 과정에서 2주 이상을 경과하지 않는 것이 바람직하다는 것을 알 수 있었다.

하수슬러지의 혐기적 소화효율 향상을 위한 전처리 효과 (The Effect of Pre-treatment on the Anaerobic Digestion of waste Activated Sludge)

  • 혐기성소화공정에서 슬러지의 용해가 전체 반응속도를 제한하고 있다. 용해율 향상을 위해 산(pH1.5, 3, 4, 5) 및 알칼리처리(pH9, 10, 13), 열처리(50, 100, 150, $200^{\circ}C$) 및 초음파처리(400W, 20kHz, 15, 20, 25, 30. 35, 40, 50, 60min)가 각 조건에서 실시되어 아래의 결과를 얻었다. 용해효과는 SCOD기준으로 무처리에 비해 산처리는 효과가 매우 낮았고, 알칼리처리는 pH13에서 986%, 열처리는 $200^{\circ}C$에서 959%, 초음파처리는 처리시간 35분에서 온도고정의 경우 802%, 온도 무고정의 경우 1123%의 높은 효과를 나타내어, 초음파처리가 가장 효과적이었다. VS/SS비율은 전처리에 따른 용해율과 정의 상관성을 가져, 용해율 효과의 지표로써 이용 가능했다. 총가스발생량은 열처리 $200^{\circ}C$가 무처리에 비해 1.8배, 열처리 $150^{\circ}C$가 1.4배, pH9의 알칼리처리가 1.2배, 온도 무고정 초음파 90분처리가 1.3배의 가스증산 효과를 나타내어, 열처리의 경우가 가장 효과적이었다. 또한 초음파처리의 경우, 온도고정보다 무고정의 경우가 저온 열처리의 동시효과가 있어 보다 효과적이었다.

SCM435 구조용 합금강의 플라즈마 질화에 미치는 전처리(Q/T)의 영향 (The Effect of Pretreatment(Q/T) on the Plasma Nitriding of SCM435 Structural Steel)

  • The effects of pre-heat treatment(Q/T) on microstructure and hardness of SCM435 structural steel nitrided by micro-pulse plasma was investigated. The quenching and tempering temperatures for obtaining matrix hardness of SCM435 steel on range of HRC30 to HRC40 desired for machine parts were about $860^{\circ}C$ and $500^{\circ}C$ respectively. The case depth of SCM435 nitrided at $480^{\circ}C$ for 5 hours was independent of pre-heat treatment condition and was approximately $150{\mu}m$. However, hardness and compactness of nitrified layer on Q/T treated specimen were more heigher than annealed specimen. The case depth increased linearly with the increase of nitriding temperature, however, the hardness of nitrified layer decreased with the temperature. Phase mixture of ${\gamma}^{\prime}$-phase($Fe_4N$) and ${\varepsilon}$-phase($Fe_3N$) were detected by XRD analysis in the nitrified layer formed at optimum nitriding condition, and only single ${\gamma}^{\prime}$-phase was detected in the nitrified layer formed at higher nitriding temperature such as $540^{\circ}C$. The optimum nitriding temperature was approximately $480^{\circ}C$ which is lower than tempering temperature for preventing softening behavior of SCM435 matrix during nitriding process and the surface hardness of nitrified layer obtained by optimum preheat treatment condition was about Hv930.

한국산 유색미 속겨의 안토시아닌 색소에 의한 견직물염색 (Silk Dyeing Method in Natural Pigments - In Case of Korean Colored Rice Bran -)

  • This research was conducted to establish the efficient use of the colored rice bran fur dyeing textiles. To investigate the fabric dyeability of the colored rice bran extract, the anthocyanin pigments were extracted with water of different temperature ranges of 40 ~ 8$0^{\circ}C$ and were dyed on silk fabrics with different dyeing temperature $25^{\circ}C$~6$0^{\circ}C$, at acidic pH and neutral pH, respectively. Aluminum chloride was preheated with mordant K/S value and dyeing fastness of dyed silk fabrics were examined. The anthocyanins of the colored rice bran were stable and red color at acidic pH, red purple or purple blue at neutral pH, but unstable, blue color at alkaline pH. If extracting temperature and dyeing temperature of dyeing solution were higher, the dyeability was high, but the color of dyed fabric showed red tone. When extracting temperature was 8$0^{\circ}C$ and dyeing temperature of dyeing solution was 6$0^{\circ}C$, the dyeability was best. Without mordant, the dyeability of silk fabrics was higher in acidic pH than in neutral pH solution. With mordant, the dyeability was higher than without mordant, and also higher in acidic pH. Pretreatment of aluminum chloride resulted in the increase of color intensity and stability. The laundering fastness of dyed fabrics was good from grade 5 to grade 3-4. Because of the anthocyanins sensitivity on light radiation, the light fastness of dyed fabrics was poor from grade 3 to grade 1-2.

Photocatalytic Cell Disruption of Giardia lamblia in a $UV/TiO_2$ Immobilized Optical-Fiber Reactor

  • Disinfection of a waterborne pathogenic protozoa, Giardia lamblia, by the conventional chlorine method has been known to be difficult. An alternative disinfection has been carried out by using a UV -light illuminating optical­fiber photoreactor. Light intensity diffused from one piece of a clad-removed optical-fiber was $1- 1.5{\mu}Em^{-2}s^{-1}$. Disinfection capability in a UV -light irradiated optical-fiber reactor suspended with 0.01 g $TiO_{2}\;dm^{-3}$ was 1.4 times that in the same reactor without $TiO_{2}$ photocatalysts. To resolve the absorption and scattering of UV light by the particles themselves as well as the difficulty of recycling particles in the slurry­type reactor, $TiO_{2}$ which was obtained by a hydrothermal method, was immobilized on clad-removed optical fibers. Such pretreatment of fiber surface resulted in an excellent transparency, which enhanced the UV light to diffuse laterally from a fiber surface. Coating time of the prepared solution by the hydrothermal method was not effective after more than two times. Disinfection capability in the $TiO_{2}$-immobilized optical-fiber reactor was $83\%$ in 1 h at $40^{\circ}C$, which was slightly higher than $76\%$ at $22^{\circ}C$ and $68\%$ at $10^{\circ}C$. Disinfection capability at $22^{\circ}C$ increased from $74\%$ at an initial pH of 3.4, through $76\%$ at pH 6.5, to $87\%$ at an initial pH of 10. Oxygen supply with air-flow rate of 5 $cm^3\;min^{-1}$ did not seem to increase the disinfection capability with UV /immobilized $TiO_2$.

전처리 조건 및 황산화물에 대한 Mn-Cu계 촉매의 탈질특성 (NOx Removal of Mn Based Catalyst for the Pretreatment Condition and Sulfur Dioxide)

  • Mn-Cu계 촉매를 사용하여 암모니아 SCR(Selective catalytic Reduction)공정에서 질소산화물 제거능을 측정하였다. 반응온도를 3가지 형태로 변화시키는 조건에서 촉매에 대한 초기반응온도의 영향을 조사하였다. 200, 300 그리고 $340^{\circ}C$에서 소성한 촉매를 사용하여 온도변화에 따른 질소산화물 전환율, 그리고 $H_2$-TPR 시스템에서 온도변화에 따른 수소소모율을 측정하였다. 이산화황가스 공급유무를 조절함으로써 촉매에 대한 이산화황가스의 불활성화 효과를 파악하였다. $340^{\circ}C$ 이상의 온도에서 소성한 촉매는 열적쇼크에 의해 일부 불활성화되며, 이러한 불활성화 원인은 비표면적과 수소 전환율 결과로부터 추론할 수 있다.

돌산갓 전처리 추출물의 항균활성 및 열안정성 (Antimicrobial Activity and Heat Stability of Water-Pretreated Extract of Leaf Mustard Dolsan(Brassica juncea))

  • 돌산갓 중의 천연 항균활성 물질을 이용하여 식품의 보존 효과를 증진시킬 목적으로, 갓을 물로 전처리하여 항균활성과 allylisothiocyanate(AIT) 함량을 조사하였다. 갓을 마쇄하여 $30^{\circ}C$에서 72시간 까지 진탕하면서 전처리할 때, 휘발성 AIT 함량은 전처리 2시간에서 가장 많았고, 전처리한 농축물 추출물(WPE)의 항균활성은 21시간 이후 부터 나타나서 48~72시간에 가장 강하게 나타내었고, 농축하지 않은 것은 WPE와는 달리 2~3시간에서도 항균활성을 나타내었지만 20시간 이후 부터는 동일한 패턴의 항균활성을 나타내었다. Ethylacetate 분획물에서 항균활성이 높았으며, 갓의 WPE는 전처리하지 않은 것보다 1.2~1.4배 항균활성이 증가되었다. WPE는 S. typhimurium과 S. aureus에 강한 저해활성을 나타내었으며, $121^{\circ}C$, 30분간 열처리하여도 항균활성은 거의 그대로 유지되었고, 전처리하지 않은 것은 항균활성이 거의 없거나 약하게 나타났다.

효모의 원형질체 형성조건 (Factors Affecting Protoplast Formation of Yeast)

  • 이속 효모간 원형질체 융합에 의해 새로운 알콜발효 균주를 개발하기 위한 전단계로서, Arthro-bacter luteus에서 얻은 zymolyase 5,000을 세포벽 분해효소로 사용하여 전분분해 효모인 C. tropicalis, E. fibuligera 및 알콜발효 효모인 S. cerevisiae 등의 원형질체 형성조건에 대하여 검토하였다. 원형질체 형성의 최적 pH와 온도는 각각 pH 7.5와 35$^{\circ}C$였다. 또한 원형질체 형성에 대한 2-mercapto-ethanol의 효과는 50mM농도에서 가장 좋았으며 삼투압 안정제로는 0.6M KCl이 적당하였다.

Effects of Cold Pretreatment and Medium Composition on Anther Culture Initiation in Strawberry

  • Callus culture initiation of strawberry (Fragaria${\times}$ananassa Duch.) was investigated at different Murashige and Skoog (MS) medium strengths, types and concentrations of plant growth regulators, and incorporating a cold pretreatment period to determine the optimal nutritional and environmental conditions. No high quality callus was induced on MS media without auxin regardless of medium strength. When 6-benzylaminopurine (BA) was combined with indole acetic acid (IAA), naphthalene acetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D), high quality callus were highly induced compared to medium supplemented with auxin alone. When $0.5mg{\cdot}L^{-1}$ BA was combined with IAA, NAA, and 2,4-D, high quality callus induction was more effective than the medium supplemented with the other BA concentrations. The best combination of auxin and cytokinin for high quality callus induction was $1.0mg{\cdot}L^{-1}$ NAA and $0.5mg{\cdot}L^{-1}$ BA. Although the differences in callus induction were not significant, high quality callus induction at half strength MS medium was more effective than at full strength medium. When $30g{\cdot}L^{-1}$ sucrose was added to the half strength MS medium, the rate of high quality callus induction increased. The optimum cold pretreatment temperature and period for high quality callus induction were $4^{\circ}C$ and 72 h, respectively. Regeneration rate of high quality callus increased in MS medium supplemented with thidiazuron.

Catecholamines에 관(關)하여 -제4편(第四編) : 심실전동발생(心室顫動發生)에 있어서의 catecholamines의 의의(意義)- (Role of Catecholamines in Ventricular Fibrillation)

  • Although it has been well known that ventricular fibrillation is the most important complication during hypothermia, much investigation has failed to show the exact nature of the etiology of ventricular fibrillation. Recently, there has been considerable research on the relationship between sympathetic activity and ventricular fibrillation under hypothermia. Cardiac muscle normally contains a certain amount of norepinephrine and the dramatic effect of this catecholamines on the cardiac muscle is well documented. It is, therefore, conceivable that cardiac catecholamines might exert an influence on the susceptibility of heart muscle to tachycardia, ventricular fibrillation and arrhythmia, under hypothermia. Hypothermia itself is stress enough to increase tonus of sympatheticoadrenal system. The normal heart is supplied by an autonomic innervation and is subjected to action of circulating catecholamines which may be released from the heart. If the reaction of the heart associated with a variable amount of cardiac catecholamines is. permitted to occur in the induction of hypothermia, the action of this agent on the heart has not to be differentiated from the direct effects of cooling. The studies presented in this paper were designed to provide further information about the cardio-physiological effects of reduced body temperature, with special reference to the role of catecholamines in ventricular fibrillation. Healthy cats, weighing about 3 kg, were anesthetized with pentobarbital(30 mg/kg) intraperitoneally. The trachea was intubated and the endotracheal tube was connected to a C.F. Palmer type A.C. respirator. Hypothermia was induced by immersing the cat into a ice water tub and the rate of body temperature lowering was $1^{\circ}C$ per 5 to 8 min. Esophageal temperature and ECG (Lead II) were simultaneously monitored. In some cases the blood pH and serum sodium and potassium were estimated before the experiment. After the experiment the animals were killed and the hearts were excised. The catecholamines content of the cardiac muscle was measured by the method of Shore and Olin (1958). The results obtained are summarized as follows. 1) In control animal the heart rate was slowed as the temperature fell and the average pulse rates of eight animals were read 94/min at $31^{\circ}C$, 70/min at $27^{\circ}C$ and 43/min at $23^{\circ}C$ if esophageal temperature. Ventricular fibrillation was occurred with no exception at a mean temperature of $20.3^{\circ}C(21-l9^{\circ}C)$. The electrocardiogram revealed abnormal P waves in each progressive cooling of the heart. there was, ultimately, a marked delay in the P-R interval, QRS complex and Q-T interval. Inversion of the T waves was characteristic of all animals. The catecholamines content of the heart muscle excised immediately after the occurrence of ventricular fibrillation was about thirty percent lower than that of the pre-hypothermic heart, that is, $1.0\;{\mu}g/g$ wet weight compared to the prehypothermic value of $1.41\;{\mu}g/g$ wet weight. The changes of blood pH, serum sodium and potassium concentration were not remarkable. 2) By the adrenergic receptor blocking agent, DCI(2-3 mg/kg), given intramuscularly thirty minutes before hypothermia, ventricular fibrillation did not occur in one of five animals when their body temperature was reduced even to $16^{\circ}C$. These animals succumbed at that low temperature, and the changes of heart rate and loss of myocardial catecholamines after hypothermia were similar to those of normal animals. The actual effect of DCI preventing the ventricular fibrillation is not predictable. 3) Administration of reserpine(1 mg/kg, i.m.) 24 hours Prior to hypothermia disclosed reduced incidence of ventricular fibrillation, that is, six of the nine animals went into fibrillation at an average temperature of $19.6^{\circ}C$. By reserpine myocardial catecholamines content dropped to $0.045\;{\mu}g/g$ wet weight. 4) Bretylium pretreatment(20 mg/kg, i.m.), which blocks the release of catecholamines, Prevented the ventricular fibrillation under hypothermia in four of the eight cats. The pulse rate, however, was approximately the same as control and in some cases was rather slower. 5) Six cats treated with norepinephrine(2 mg/kg, i.m.) or DOPA(50 mg/kg) and tranylcypromine(10 mg/kg), which tab teen proved to cause significant increase in the catecholamines content of the heart muscle, showed ventricular fibrillation in all animals under hypothermia at average temperature of $21.6^{\circ}C$ and the pulse rate increased remarkably as compared with that of normal. Catecholamines content of cardiac muscle of these animals markedly decreased after hypothermia but higher than control animals. 6) The functional refractory periods of isolated rabbit atria, determined by the paired stimulus technique, was markedly shortened by administration of epinephrine, norepinephrine and isoproterenol. 7) Adrenergic beta-blocking agents, such as pronethalol, propranolol and sotalol(MJ-1999), inhibited completely the shortening of refractory period induced by norepinephrine. 8) Pretreatment with either phenoxftenbamine or phentolamine, an adrenergic alphatlocking agent, did not modify the decrease in refractory period induced by norepinephrine. From the above experiment it is possible to conclude that catecholamines play an important role in producing ventricular fibrillation under hypothermia. The shortening of the refractorf period of cardiac muscle induced by catecholamines mar be considered as a partial factor in producing ventriculr fibrillaton and to be mediated by beta-adrenergic receptor.
