Comparative Molecular Similarity Indices Analysis (CoMSIA) on the Melanogenesis Inhibitory Activities of Alkyl-3,4-dihydroxy- benzoate and N-alkyl-3,4-dihydroxybenzamide Derivatives.

  • To find a new substance with superior melanogenesis inhibitory activity, the bioactivities of alkyl-3,4-dihydroxy-5-substituted benzoate (A) and N-alkyl-3,4-dihydroxy-5-substituted benzamide (B) derivatives as substrate of tyrosinase were measured in mouse melanoma cells. And the bioactivities analyzed using comparative molecular similarity indices analysis (CoMSIA). From the CoMSIA model, when cross-validation value (q$^2$) is 0.713 at four components, the pearson correlation coefficient ($r^2$) is 0.900. Unknown compounds were predicted, using QSAR analyzed results from the CoMSIA methods. Excellent agreement was obtained between the measured and the predicted bioactivities of unknown compounds. As the results of prediction from CoMSIA, we could conclude that the bioactivities were increased from pl$_{50}$=3.18-4.80 to above 5.17 by creation of 6-methylheptyl, n-pentylphenyl and 2-hydroxypentylphenyl group etc,.,.

  • 새로운 2-(4-(6-chloro-2-benzoxazolyloxy)phenoxy)-N-phenylpropionamide 유도체들의 구조 변화에 따른 발아 전, 논피(Echinochloa crus-galli)에 대한 제초활성과의 3D-QSAR 관계를 상이한 정렬방법에 따라 비교 분자장 분석(CoMFA)과 비교분자 유사성 지수분석(CoMSIA) 방법으로 연구하였다. 가장 양호한 3D-QSAR 모델은 atom based fit 정렬과 CoMFA장과 CoMSIA장의 조합 조건에서 유도된 CoMFA 모델(AI-2)과 CoMSIA 모델(AII-4)이었다. CoMFA 및 CoMSIA 등고도로부터 제초활성은 N-phenyl 고리 상 치환기의 구조변화로 개선될 수 있었다.

  • In order to search new inhibitors against farnesyl protein transferase (FPTase), a series of 2,3-bis-benzylidenesuccinaldehyde derivatives (1-29) were synthesized and their inhibition activities ($pI_{50}$) against FPTase were measured. From based on the reported results that the inhibitory activities of dimers 2,3-bis-benzylidenesuccinaldehydes were higher than those of monomers cinnamaldehydes, 3D-QSARs on FPTase inhibitory activities of the dimers (1-29) were studied quantitatively using comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) methods. The statistical qualities of the optimized CoMFA model II ($r^2{_{cv.}}$= 0.693 and $r^2{_{ncv.}}$= 0.974) was higher than those of the CoMSIA model II ($r^2{_{cv.}}$ = 0.484 and $r^2{_{ncv.}}$ = 0.928). The dependence of CoMFA models on chance correlations was evaluated with progressive scrambling analyses. And the inhibitory activity exhibited a strong correlation with steric factors of the substrate molecules. Therefore, from the results of graphical analyses on the contour maps and of predicted higher inhibitory active compounds, it is suggested that the structural distinctions and descriptors that contribute to inhibitory activities ($pI_{50}$) against FPTase will be able to applied new inhibitor design.

Structural Characteristics that Influence on the Insecticidal Activity of 2-(n-Octyl)pseudothiourea Analogues against the Diamondback Moth (Plutella xylostella, L.)

  • Structural characteristics that influence on the insecticidal activity ($pI_{50}$) of 2-(n-octyl)isothiourea analogues (1-45) against the diamondback moth (Plutella xylostella, L.) based on three dimensional quantitative structure activity relationships (3D-QSARs) were discussed quantitatively using a comparative molecular field analysis (CoMFA) and a comparative molecular similarity indeces analysis (CoMSIA) methods. The statistical values of the CoMFA 2 model were better than those of the CoMSIA 1 model. The CoMFA 2 model was the optimized model with the correlativity (the training set: Ave. = 0.104 & PRESS = 0.613) and the predictability (the test set: Ave. = 0.086 & PRESS = 0.096). Insecticidal activities with the optimized CoMFA 2 model were dependent upon steric factors (79.4%) of $R_1-R_3$ substituents. From the analytical results of CoMFA contour maps, it is predicted that the R1 substituent of 1-45 which has a steric favor in a broad space, $R_2\;and\;R_3$ groups with a steric favor in a narrow space and a H-bond donor favor would have better the insecticidal activity.

  • 감수성(SBC) 및 저항성(RBC) 잿빛곰팡이병균(Botrytis cinerea)에 대한 N-phenyl-O-phenylthionocarbamate 유도체들의 선택적인 살균활성에 대한 3차원적인 구조와 활성과의 관계(3D-QSAR)를 CoMFA와 CoMSIA 방법으로 검토하였다 그 결과, 통계적으로 CoMFA 모델(M5)보다 CoMSIA 모델(M7)이 양호하였으며 살균활성의 선택성에 미치는 요소는 CoMSIA 모델(M7)의 정전기장에 의존적이었다. 그러므로 CoMSIA 모델(M7)의 등고도로부터 N-phenyl 고리의 meta-위치에 음하전을 띄는 수소결합 주게에 의하여 선택성이 개선될 것으로 예상되었다.

  • 새로운 25종의 Imazethapyr 유도체, (2-(4-isopropyl-4-methyl-5-oxo-2-imidazolin-2-yl)-3-(N-methyl-N-(X)치환-phenylaminooxoacetyl)-5-methylpyridine)들을 합성하여 치환기(X) 변화에 따른 발아 전 후, 피(Echinochla crus-galli.)의 제초활성에 미치는 3-(N-mothy-N-(X)치환-phenylaminoozoacetyl) group의 영향을 검토한 바, 발아 전보다 발아 후의 제초활성에 더 큰 영향을 미침을 알 수 있었다. 발아 후의 제초활성은 X-치환기의 전자밀게 효과와 입체상수 $(E_s)$에 의존적이었으며 가장 큰 제초활성을 나타내는 화합물로는 $bulky(E_s<0)$하고 전자밀게$(\sigma<0)$가 치환된 화합물, 15(4-t-butyl group)와 20(3,5-dimethyl group)이었다. 그리고 높은 제초활성을 나타낼 것으로 예상되는 화합물의 조건들이 검토되었다.

  • 새로운 17종의 imazapyr 유도체인 3-(N-methyl-N(X)-치환-phenylaminooxoacetyl)-2-(4-isopropyl-5-oxo-2-imidazoline-2-yl)pyridine, 6을 합성하고 3-(N-methyl-N(X)-치환-phenylaminooxoacetyl group중의 X-치환기 변화에 따른 구조와 옥수수(Zea mays L.)와 참비듬(Amaranthus viridis L.)에 대한 발아 전의 제초활성에 미치는 관계(SAR)를 검토 한 바, 옥수수의 경우에는 MR상수의 적정값(optimal value)이 $5.56\;cm^3/mol$이었으므로 이 값보다 크거나 작을수록 제초활성이 작았다. 참비듬의 경우에는 $L_1$상수의 적정값이 3.34 A이므로 이 값에 가까울수록 제초활성이 클 것임을 알 수 있었다. 특히, 2,5-difluoro group 치환체, 6i는 참비듬에 가장 큰 제초활성을 나타내었으나 옥수수에는 가장 작은 제초활성을 미치므로써 현저하게 두 초종간에 선택성을 나타내는 화합물이었다.

두 점박이 응애(Tetranychus urticae)에 대한 N'-phenyl-N-methylformamidine 유도체의 살충활성과 새로운 고활성 화합물들의 설계 (Insecticidal Activity of N'-phenvl-N-Methylformamidine Analogues against Two Spotted Spider Mite (Tetranychus urticae) and Design of New Potent Compounds)

  • 새로운 고활성의 살충성 화합물을 예측하고 설계하기 위하여 N'-phenyl-N-methylformamidine 유도체들의 두점박이 응애(Tetranychus urticae)에 대한 살충활성과 물리-화합적인 설명인자들 사이의 2차원적인 정량적 구조-활성관계(2D-QSAR)와 분자 홀로그래피적인 정량적 구조-활성관계(HQSAR)를 구체적으로 검토하였다. 2D-QSAR 모델(1 및 3)로부터 입체적인 요소로서 $R_3$-치환기의 폭($B_2$)과 $R_4$-치환기의 적정한 전체 쌍극자능율값(TDM=2.025 D)이 살충활성에 주로 영향을 미쳤으며 살충활성은 $R_3$- 및 $R_4$-치환기들에 의존적이었다. 특히, 2D-QSAR 모델(3)과 HQSAR 모델 F2에 의하여 설계된 새로운 유력한 활성화합물(P1; $EC_{50}$=0.516 ppm)의 살충활성은 Amitraz에 비하여 약 34.3배 높을 것으로 예측되었다.

Pharmacophore Models of Paclitaxel- and Epothilone-Based Microtubule Stabilizing Agents

  • Microtubules play an important role in intracellular transport, mobility, and particularly mitosis. Paclitaxel (Taxol$^{TM}$) and paclitaxel-like compounds have been shown to be anti-tumor agents useful for various human tumors. Paclitaxel-like compounds operate by stabilizing microtubules through interface binding at the interface between two ${\beta}$-tubulin monomers in adjacent protofilaments. In this paper we present the elucidation of the structural features of paclitaxel and paclitaxel-like compounds (e.g., epothilones) with microtubule stabilizing activities, and relate their activities to spatial and chemical features of the molecules. CATALYST program was used to generate three-dimensional quantitative structure activity relationships (3D-QSARs) resulting in 3D pharmacophore models of epothilone- and paclitaxel-derivatives. Pharmacophore models were generated from diverse conformers of these compounds resulting in a high correlation between experimental and predicted biological activities (r = 0.83 and 0.91 for epothilone and paclitaxel derivatives, respectively). On the basis of biological activities of the training sets, five- and four-feature pharmacophore hypotheses were generated in the epothilone and paclitaxel series. The validation of generated hypotheses was achieved by using twelve epothilones and ten paclitaxels, respectively, which are not in the training sets. The clustering (grouping) and merging techniques were used in order to supplement spatial restrictions of each of hypothesis and to develop more comprehensive models. This approach may be of use in developing novel inhibitor candidates as well as contributing a better understanding of structural characters of many compounds useful as anticancer agents targeting microtubules.

6-Bromobenzo[4,5]imidazo[$1,2{\alpha}$pyridin-8,9-dione 유도체들의 Photosystem II 저해활성에 관한 3D-QSAR 분석 (3D-QSAR Analysis on the Photosystem II Inhibition Activity of 6-Bromobenzo[4,5]imidazo[$1,2{\alpha}$]pyridin-8,9-dione Analogues)

  • 기질 분자로서 6-bromobenzo-[4,5]imidazo[$1,2{\alpha}$]pyridin-8,9-diones 유도체의 photosystem II의 저해활성에 관한 3차원적인 정량적 구조와 활성과의 관계(3D-QSARs)들을 비교 분자장분석(CoMFA)과 비교분자 유사성 지수분석(CoMSIA) 방법으로 각각 검토하였다. CoMFA 모델은 CoMSIA 모델보다 높은 예측성과 상관성을 갖는 모델이었다. 또한 최적화된 CoMFA 2 모델의 기여도는 입체장(90.4%)에 가장 의존적이었다. CoMFA 등고도 분석으로부터, 치환기(R)로서 모체로 연결되는 탄소원자(ipso carbon)에 곁가지를 가진 치환체가 도입될수록 저해활성이 증가할 것으로 보인다. 또한, imidazol 고리와 pyridine 고리 사이의 위치에 양하전을 가질수록 활성이 높을 것으로 예상되며 친수성을 띄는 선 형태의 치환기가 도입되는 경우에 활성이 개선될 것으로 판단되었다.
