Association of Endocrine Factors (Insulin-Like Growth Factor-II and Binding Protein-3) with Litter Size in Pigs

  • Litter size has been one of the important economic traits in porcine reproduction. The insulin-like growth factor (IGF) system has been shown to mediate actions of the steroid hormone or to synergize with other endocrine factors so that it consequently plays roles in reproductive processes, including ovulation, implantation, maintenance of pregnancy, and fetal development. However, the effect of the serum IGF system on porcine litter size has not been deeply studied. Therefore, this study was conducted to relate serum IFG-II concentration and IGF binding protein-3 (IGFBP-3) expression with porcine litter size. Moreover, the possible association of those with estrogen receptor (ER) as a candidate gene for litter size was investigated. Swine were separated into two groups showing high and low litter sizes, and sera were collected from sows in the estrous cycle to postnatal growth of their female progeny. Serum IFG-II concentration was measured by radioimmunoassay and IGFBP-3 expression was detected by Western ligand blotting. During the estrous cycle, IGFBP-3 expression in both groups decreased moderately from metestrus to estrus, but IFG-II concentration showed a reverse pattern. Also, IFG-II concentration and IGFBP-3 expression decreased gradually as pregnancy proceeded. Unlike IGFBP-3, IFG-II decreased moderately as newborn pigs grew. Significant differences in serum IFG-II amount between the two groups were detected at 60 (p<0.01), 75, 90, and 105 d (p<0.05) of pregnancy and at 60 (p<0.01), 45, and 105 d (p<0.05) of postnatal growth. Furthermore, based on ER genotypes, a high litter size group with genotypes AB and BB showed lower IFG-II concentration than a low litter size group with a genotype AA during pregnancy. Taken together, the results indicate that the serum IFG-II and IGFBP-3 are correlated with the litter size in pigs.

In vitro Methanogenesis and Fermentation of Feeds Containing Oil Seed Cakes with Rumen Liquor of Buffalo

  • Eight feeds (mixture of wheat straw and oil seed cakes in 3:1 ratio) were evaluated for methane emission and fermentation pattern with buffalo rumen liquor as inoculum in an in vitro gas production test. The cakes tested were groundnut cake (GNC), soybean cake (SBC), mustard seed cake (MSC), cotton seed cake (CSC), karanj seed cake expeller extracted (KCEE), karanj seed cake solvent extracted (KCSE), caster bean cake expeller extracted (CBCEE) and caster bean cake solvent extracted (CBCSE). The gas production (ml/g dry matter) was significantly higher with SBC and MSC followed by CSC, GNC, KCSE, KCEE, CBCSE and was the lowest with CBCEE. Methane emission was significantly lower with KCEE, KCSE, CBCEE, CBCSE (20.32- 22.43 ml/g DM) than that with SBC, GNC, CSC (27.34-31.14 ml/g DM). Mustard seed cake was in-between the two groups of oil cakes in methane production. In vitro true digestibility was highest with SBC followed by GNC, CSC, MSC, KCSE, KCEE, CBCSE and CECEE. Ammonia nitrogen level was positively correlated with the amount of protein present in the cake. Total holotrich protozoa were significantly higher with SBC, whereas, large spirotrich protozoa tended to be lower than with other cakes. The counts of small spirotrich and total protozoa were similar with all the cakes. Total volatile fatty acid production and acetate to propionate ratio were significantly higher with SBC and significantly lower with KCEE as compared to the other cakes. Among the conventional oil cakes tested in the present experiment (GNC, SBC, MSC and CSC), mustard seed cake-based feed produced the minimum methane without affecting other fermentation characteristics adversely.

  • A. rhizogenes A4 균주를 접종하여 청피홍심무우(Raphanus sativus cv. Chungpihongsim)의 모상근 배양을 확립하였다. 형질전환된 뿌리는 MS 배지의 기본염을 1/2로 희석하고, pH는 5.2, sucrose는 3%로 조정한 배양조건에서 최적성장을 보였다. 형질전환된 조직에서 합성되는 물질인 opine, 즉 agropine과 mannopine이 모상근의 추출액에서 검출되었다. 배지에 2, 4-D와 Kinetin이 첨가될 경우 모상근의 탈분화와 더불어 세포내에 안토시아닌의 합성이 유도되었으며, 그 중 2, 4-D $0.45\;\mu\textrm{M}$과 kinetin $2.3\;\mu\textrm{M}$의 첨가에서 최대의 색소 축적능을 보였다. 모상근의 탈분화와 함께 유도되는 안토시아닌의 paper chromatography 전개양상은 경작뿌리에서 추출한 색소에 비하여 다소 차이를 보였지만 모든 안토시아닌의 aglycone은 pelargonidin으로 확인되었다. 이러한 시료의 총 안토시아닌 함량은 0.49 mg/g(f.w.)로 계산되었다.

조숙 3면잠의 유발과 실용형질 (Induction and Characterization of Precocious Trimolting Silkworm, Bombyx mori)

  • 세직도견사의 생산을 최종목적으로 해서, imiazole 계항유약 호르몬 활성물질의 1종인 "KK-42"의 투여와 고온처리에 의해서, 효과적인 조숙 3면잠의 유발조건을 탐색하고, 유발된 3면잠의 성장, 사료 효율 및 견사질을 조사한 결과는 다음과 같다. 1. KK-42의 투여에 의하여 고율로 조숙 3면잠이 유발되었다. 효과적인 KK-42의 농도는 10$\mu\textrm{g}$/마리이었다. 한편, 고온처리에 의한 조숙 3면잠의 유발은 KK-42의 투여에 비하여 적었고, 3령누에를 38$^{\circ}C$에서 48시간 처리한 것이 36시간처리한 것보다 효과적이었다. 2. KK-42에 의한 조숙 3면잠의 최대체중은 약 1.8g로서, 대조의 4면잠에 비하여 상당히 가벼웠고, 4령기의 성장은 4면잠의 5령기에 가까운 종령기적 패턴을 나타내었다. 3. KK-42의 투여와 고온처리에 의한 조열 3면잠의 경과일수는 대조의 4면잠에 비하여 상당히 단축되었다. 특히, 뽕잎육에 있어서 KK-42에 의한 조숙 3면잠의 경과일수는 4면잠에 비하여 5.17일 단축되었다. 4. 조숙 3면잠의 견과 견사직도는 대조의 4면잠에 비하여 상당히 작고 가늘었다. 즉, 조숙 3면잠견의 견사직도는 KK-42를 처리한 경우 1.39d이고, 고온처리한 경우 1.88d이었다. 5. 조숙 3면잠견의 견사크기는 KK-42의 투여나 고온처리나 다같이 대조의 4면잠에 비하여 상당히 작았다. 6. KK-42에 의한 한숙 3면잠의 사료효율은 대조의 4면잠에 비하여 낮은 편이었다. 즉, 조숙 3면잠(대성잠)의 견층중 1g당 식하건물뽕잎량은 7.97cg인데 비하여 대조의 4면잠은 9.20cg이었다.

The change of sleeping and lying posture of Japanese black cows after moving into new environment

  • Objective: Environmental change is one of the stressful events in livestock production. Change in environment disturbs cow behavior and cows require several days to regain a stable behavioral pattern. Sleeping posture (SP) and lying posture (LP) have been used as indicators for animal that are relaxed and well-acclimated to their environment. The aim of this study was to examine the time required by Japanese black cows for stabilization of SP and LP after moving into new environment. Methods: Seven pregnant Japanese black cows were used. Cows were moved into new tie-stall shed and their sleeping and lying posture measured 17 times during 35 experimental days. Both SP and LP were detected by accelerometer fixed on middle occipital and hip-cross, respectively. Daily total time, frequency, and average bout of both SP and LP were calculated. Results: Daily SP time was the shortest on day 1 and increased to the highest on day 3. It then decreased until day 9, after that stabilized about 65 min/d till the end of experiment. Daily LP time changed in same manner as daily SP time. The average SP bout was the longest on day 1, and then decreased to stable level on day 7. On the other hand, the average LP bout was the shortest on day 1, and it increased to stable level on day 7. Conclusion: These results showed that pregnant Japanese black cows needed 1 week to stabilize their SP. However, there were different change patterns between the average SP and LP bout, even though the change pattern of daily SP and LP time were similar.

수경재배에 의한 고랭지 시금치의 여름철 안정생산 기술 개발 (Development of Stable Production Technique of Summer Spinach (Spinacia oleracea L.) in Soilless Culture in the Highlands)

  • 고랭지 시금치의 여름철 수경재배에 적합한 품종과 육묘 배지의 선발, 적정 양액조건의 구명 및 시설비가 적게 들고 생산성이 높은 수경재배 방식의 개발을 위하여 본 연구를 수행하였다. 고랭지에서 여름철 수경재배에 적합한 시금치 품종은 '퀸토'로 다른 품종에 비하여 생육이 좋고 수량도 많았다. 시금치의 생육에 적합한 양액의 조건은 pH 6.0, EC $2.0dS{\cdot}m^{-1}$$NH_4-N$의 비율은 30%이었다. 육묘용으로 피트모스와 펄라이트를 7:3의 용적비로 혼합한 상토를 200공 트레이에 채우고 시금치 종자를 파종하여 육묘한 경우 우레탄스펀지나 암면 플러그에서 육모한 것보다 입묘율이 높았다. 이렇게 육묘한 묘로 수경재배를 한 결과 혼합상토로 육묘한 경우의 수량은 암면 및 우레탄스펀지에 비하여 18% 및 24%가 증가하였다. 토양 위에 비닐을 깔아 만든 박막재배 베드에 lcm 두께로 혼합배지(피트 : 펄라이트 = 7 : 3)를 채운 후 스티로폼 정식판을 올려놓고 시금치를 재배할 경우 담액재배에 비하여 초장은 18% 길었고 수량은 9% 증가하였으며, 근권 온도 pH 및 EC는 담액재배와 비슷한 양상으로 변화하였다.

이트리아 소결체의 특성에 글라스프릿 첨가가 미치는 영향 (Effect of Glass Frit Addition on Characteristics of Yttria Ceramics)

  • The semiconductor and display industries require the development of plasma resistant materials for use in high density plasma etching process equipment. Yttria (Y2O3) is a ceramic material mainly used to ensure good plasma resistance properties, which requires a dense microstructure. In commercial production, a sintering process is applied to reduce the sintering temperature of Y2O3. In this study, the effect of the addition of glass frit to the sintered specimen was examined when manufacturing yttria sintered specimens for semiconductor process equipment parts. The Y2O3 specimen was shaped into a Ø50 mm size and then sintered at 1,600 ℃ for 1~8 h. The characteristics, X-ray diffraction pattern, densities, contraction rate of the specimen, and swelling of the surface of the Y2O3 specimens were investigated as a function of the sintering time and glass frit addition. The Y2O3 specimen exhibited a density of over 4.9 g/cm3 as the sintering time increased, and the swelling phenomenon characteristics were improved by glass frit, by controlling particle size.

폐콘크리트의 탄산화 특성 분석 (Analysis of carbonation characteristics on waste concrete)

  • 본 연구에서는 폐콘크리트 분쇄물의 탄산화 가능성 검토를 위해 폐콘크리트 분쇄물의 기초물성 측정 및 탄산화를 실시하였으며, 탄산화 시료의 CaCO3 생성량, 결정성 분석 및 미세구조를 관찰하였다. 폐콘크리트 분쇄물의 탄산화 전 CaCO3 함량은 14.51 %이었으며, 24시간 탄산화 시 CaCO3 함량은 28.52 %, 미분쇄 후 24시간 탄산화 시의 CaCO3 함량은 32.73 %이었다. 탄산화 시간에 따른 폐콘크리트 분쇄물의 탄산화는 초기 6시간까지 급격히 이루어졌으나, 12시간 이후 24시간까지의 탄산화는 서서히 진행되었다. 특히 12시간 이후 24시간까지의 CaCO3 생성량은 2.32 % 수준에 불과하였다. 폐콘크리트 탄산화 후 미세구조 관찰 시 calcite 형상의 CaCO3 결정을 발견할 수 있었으며, 또한 XRD 패턴에서도 동일 결정 피크를 검출할 수 있었다. 따라서 본 연구에서 생성된 CaCO3 결정은 calcite라는 것을 확인할 수 있었다.

Comparison of Indigenous Browses and Sunflower Seed Cake Supplementation on Intake and Growth Performance of Dual-purpose Goats Fed Buffel Grass (Cenchrus ciliaris) Hay

  • A study to compare the effects of supplementing Delonix elata, Grewia similis, Tamarindus indica and sunflower seed cake on intake and growth rate of dual-purpose goats fed low quality Buffel grass (Cenchrus ciliaris) hay was carried out. Twenty-eight male goats aged five to seven months (mean weight 12.93${\pm}$3.94 kg) were randomly allocated to four dietary groups in a completely randomised design. The diets were hay plus Grewia similis, hay plus Delonix elata, hay plus Tamarindus indica and hay plus sunflower seed cake. All diets were supplemented with maize bran. The experimental period was 90 days. Voluntary dry matter intake of the supplements was higher for Tamarindus indica (275.5 g/day) and Grewia similis (201.8 g/day) and lowest for sunflower seed cake (81g/day). Goats supplemented with Grewia similis had the highest hay intake (183.8 g/day) while those supplemented with sunflower seed cake had the lowest hay intake (98.9 g/day). Animals fed browse supplements gained significantly more weight (p<0.001) than those with sunflower seed cake. There were no significant differences in live weight change between goats fed the different browses. However, those fed Tamarindus indica gained an average of 20.79 g/d which was slightly higher than the gains for those on Grewia similis and Delonix elata while those fed sunflower seed cake lost weight. Correspondingly, goats supplemented with browse leaf meals had higher feed conversion ratios than those supplemented with sunflower seed cake and required 23.91 to 35.06 g DM of feed to produce one g of weight gain per day. In a separate study, the DM disappearance pattern indicated that Grewia similis and Delonix elata were highly degradable compared to Tamarindus indica. At 24 h of incubation, DM degradability was 627, 588 and 345 g/kg DM for Grewia similis, Delonix elata and Tamarindus indica, respectively. In another study in vivo DM digestibility ranged from 46.1% (for hay alone) to 56.2% (for hay plus Grewia similis). It was concluded that the addition of Tamarindus indica, Grewia similis and Delonix elata leaf meals to Cenchrus ciliaris hay resulted in increased total DM intake, in vivo digestibility and growth rate. Therefore, leaf meals of indigenous browses particularly Tamarindus indica and Grewia similis could be used as supplementary feeds for small ruminants grazing on poor quality roughages during the dry season rather than use of expensive, less effective and intermittently available sunflower seed cake.

Temporal Pattern of cAMP Concentrations and α-Actin mRNA Expression in Skeletal Muscle of Cimaterol-Fed Rats

  • Twenty four female Sprague-Dawley rats weighing about 190 g were used to examine changes in muscle cAMP concentrations and steady-state levels of skeletal muscle ${\alpha}$-actin mRNA during chronic administration of cimaterol, a ${\beta}$-adrenergic agonist. Cimaterol was mixed in a powdered rat diet at 10 mg/kg diet. At 3 and 21 days after the start of treatment, skeletal muscle and heart samples were collected for the measurement of cAMP concentrations and skeletal muscle ${\alpha}$-actin mRNA levels. Cimaterol increased (p < 0.01) body weight gain gradually during the first seven days of the trial period, but not thereafter. Most skeletal muscle weights and the ratio of muscle weight to body weight were increased (p < 0.05) by cimaterol treatment both at 3 and 21 days. Heart weight was also increased (p < 0.05) by cimaterol treatment at 3 and 21 days, but the ratio of heart weight to body weight was increased (p < 0.05) only at 3 day. Cimaterol decreased (p < 0.05) cAMP concentration of gastrocnemius muscle at both 3 and 21 days after treatment. However, cimaterol tended (p = 0.07) to increase cAMP concentration at 3 days in the heart. Cimaterol tended (p = 0.08) to increase the steady-state level of ${\alpha}$-actin mRNA by 60% in gastrocnemius muscle at 3 days but had no effect at 21 days. The results indicate that the pattern of hypertrophic response to chronic dietary administration of cimaterol is different between cardiac and skeletal muscle. In skeletal muscles it appears that the hypertrophy induced by cimaterol is partly due to stimulated myofibrillar protein synthesis at a pre-translational level.
