Effects of Fermented Diet Using Probiotics from Pine Needle Microbes on Growth Performance, Blood Characteristics, Carcass Traits and Economy of Pigs (솔잎 토착미생물을 이용한 발효사료 급여가 돼지의 성장, 혈액성상, 도체형질 및 경제성에 미치는 영향)

  • This study was conducted to investigate the effect of probiotic from pine needle microbes on growth performance, blood characteristics, carcass traits and economy of pigs. One hundred eighty pigs (L${\times}$Y${\times}$D, average body weight=75 kg) were grouped and housed with 15 animals per pen for 3 replications per each treatment. The ingredients and pine needle microbes mixed and fermented on anaerobic condition for 10 days to make the fermented diet. The basal diet was substituted with 0 (CON), 3 (T1), 5 (T2) and 10% (T3) of fermented diet, the pigs were fed for 42 days and slaughtered at the end of the trial to measure the carcass traits. Crude protein concentration of fermented diet on 10 day was higher (p<0.05) than that of on 0 day, while crude fiber concentration and pH were lower (p<0.05). The supplementation of fermented diet did not affect on growth performance and feed intake. Hemoglobin concentration of whole blood in T3 was higher (p<0.05) than that of CON, and hematocrit concentrations in CON and T1 were higher (p<0.05) than that of T2 and T3. The concentration of platelet was higher (p<0.05) in T1 than in T2 and T3. The CON had higher (p<0.05) total cholesterol, LDL-cholesterol and triglycerides concentrations than the treatments, whereas lower (p<0.05) HDL-cholesterol concentration. Carcass weight and dressing in T1 and T3 were higher (p<0.05) than those of CON and T2. The backfat thickness was higher (p<0.05) in CON and T2 than in T3. The carcass grade and economy improved by supplementation of fermented diet compare to CON. In conclusion, the supplementation of fermented diet using probiotics from pine needle microbes could improve health and meat quality and reduce the feed cost of pigs.

Hydrochemical and Isotopic Characteristics, and Origin of Noble Gas for Low-temperature Hot Spring Waters in the Honam Area (호남지역 저온형 온천수의 수리지화학적 및 안정동위원소 특성과 영족기체의 기원에 관한 연구)

  • Geochemical composition, stable isotopes $({\delta}^{18}O,\;{\delta}D,\;{\delta}^{34}S)$ and noble gases(He, Ne and Ar) of nine hot spring water and three groundwater for five hot springs(Jukam, Hwasun, Dokog, Jirisan, Beunsan) from the Honam area were analyzed to investigate the hydrogeochemical characteristics and the hydrogeochemical evolution of the hot spring waters, and to interpret the source of sulfur, helium and argon dissolved in the hot spring waters. The hot spring waters show low water temperature ranging from 23.0 to $30.5^{\circ}C$ and alkaline characteristics of pH 7.67 to 9.98. Electrical conductivity of hot spring waters is $153{\sim}746{\mu}S/cm$. Groundwaters in this area were characterized by the acidic to neutral pH range$(5.85{\sim}7.21)$, the wide electrical conductivity range $(44{\sim}165{\mu}S/cm)$. The geochemical compositions of hot spring and groundwaters can be divided into three water types: (1) $Na-HCO_3$ water type, (2) Na-Cl water type and (3) $Ca-HCO_3$ water type. The hot spring water of $Ca-HCO_3$ water type in early stage have been evolved through $Ca(Na)-HCO_3$ water type into $Na-HCO_3$ type in final stage. In particular, Jurim alkaline(pH 9.98) hot spring water plotted at the end point of $Na-HCO_3$ type in the Piper diagram is likely to arrive into the final stage in geochemical evolution process. Hydrogen and oxygen isotopic data of the hot spring water samples indicate that the hot spring waters originated from the local meteoric water showing latitude and altitude effects. The ${\delta}^{34}S$ value for sulfate of the hot spring waters varies widely from 0.5 to $25.9%o$. The sulfur source of most hot spring waters in this area is igneous origin. However, The ${\delta}^{34}S$ also indicates the sulfur of JR1 hot water is originated from marine sulfur which might be derived ken ancient seawater sulfates. The $^3He/^4He\;and\;^4He/^{20}Ne$ ratios of the hot spring waters range from $0.0143{\times}10^{-6}\;to\;0.407{\times}10^{-6}\;and\;6.49{\sim}584{\times}10^{-6}$, respectively. The hot spring waters are plotted on the mixing line between air and crustal components. It means that the He gas in the hot spring waters was mainly originated from crustal sources. However, the JR1 hot spring water show a little mixing ratio of the helium gas of mantle source. The $^{40}Ar/^{36}Ar$ ratios of hot spring water are in the range from $292.3{\times}10^{-6}\;to\;304.1{\times}10^{-6}$, implying the atmospheric argon source.

Effects of a Chelated Copper as Growth Promoter on Performance and Carcass Traits in Pigs

  • Three studies were conducted to investigate whether a chelated Cu can replace $CuSO_4$ as a growth promoter in pigs. In Exp. 1, a total of 240 piglets (Large White${\times}$Landrace, $7.36{\pm}0.10kg$) were randomly allocated to 1 of 3 treatments with 8 replicates and 10 piglets per pen. Treatments included a NRC control ($CuSO_4$, 6 mg/kg), two Cu supplementations from either $CuSO_4$ or $Cu(HMTBa)_2$ at 170 mg/kg. Pigs fed $Cu(HMTBa)_2$ were 6.0% heavier than pigs fed either the NRC control or 170 mg/kg $CuSO_4$ (p = 0.03) at the end of the experiment. During the 42 days of experimental period, pigs fed $Cu(HMTBa)_2$ gained 9.0% more (p = 0.01), tended to eat more feed (p = 0.09), and had better feed efficiency (p = 0.06) than those fed $CuSO_4$. Compared with the 6 mg/kg $CuSO_4$ NRC control, liver Cu was increased 2.7 times with 170 mg/kg $CuSO_4$ supplementation, and was further increased with $Cu(HMTBa)_2$ (4.5 times, p<0.05). In Exp. 2, a total of 616 crossbred piglets (PIC, $5.01{\pm}0.25kg$) were randomly allocated to 1 of 4 treatments with 7 replicates and 22 piglets per pen. Treatments included a NRC control (from $CuSO_4$), and three pharmaceutical levels of Cu (150 mg/kg) supplemented either from C$CuSO_4$, tri-basic copper chloride ($Cu_2[OH]_3C1$), or $Cu(HMTBa)_2$. Pigs fed $CuSO_4$ or $Cu(HMTBa)_2$ had better feed efficiency (p = 0.01) and tended to gain more (p = 0.08) compared with those fed the NRC control. Pigs fed $Cu_2[OH]_2C1$ were intermediate. Pigs fed $Cu(HMTBa)_2$ had the highest liver Cu, which was significantly higher than those fed ($Cu_2[OH]_3C1$) or the negative control (p = 0.01). In Exp. 3, a total of 1,048 pigs (PIC, $32.36{\pm}0.29kg$) were allotted to 6 treatments with 8 replicates per treatment and 20 to 22 pigs per pen. The treatments included a NRC control with 4 mg/kg Cu from $CuSO_4$, a positive control with 160 mg/kg Cu from $CuSO_4$, and incremental levels of $Cu(HMTBa)_2$ at 20, 40, 80, and 160 mg/kg. During the overall experimental period of 100 days, no benefit from 160 mg/kg $CuSO_4$ was observed. Pigs fed $Cu(HMTBa)_2$ had increased ADG (linear and quadratic, $p{\leq}0.05$) and feed efficiency (linear and quadratic, $p{\leq}0.05$) up to 80 mg/kg and no further improvement was observed at 160 mg/kg for the whole experimental period. Pigs fed 80 mg/kg $Cu(HMTBa)_2$ weighed 1.8 kg more (p = 0.07) and were 2.3 kg heavier in carcass (p<0.01) compared with pigs fed 160 mg/kg $CuSO_4$. In addition, loin depth was increased with increased $Cu(HMTBa)_2$ supplementation with pigs fed 80 mg/kg $Cu(HMTBa)_2$ had the greatest loin depth (p<0.05). In summary, $Cu(HMTBa)_2$ can be used to replace high $CuSO_4$ as a growth promoter in nursery and grower-finisher pigs.

Effect of Dietary Energy, Protein on Growth and Blood Composition of Cross Bred Chicks (유색육용계의 성장과 혈액성상에 사료단백질 및 에너지가 미치는 영향)

  • To acquire essentially necessary basic data to establish feeding system by verifying appropriate dietary energy and protein level for the growth of commercial slow growing broiler chicks within the country, two experiments were conducted for 5 weeks. One day old, 1,404 male and female broiler chicks were used for the experiments, and 26 chicks were placed at each pen. The energy level of feed was maintained about 3,000 or 3,100 kg/kcal for whole breeding period of 5 weeks, and protein content was adjusted about 20, 21, and 22% during the first two weeks and the content was adjusted to 18, 19, 20, 21, and 22% from the 3 to 5 weeks old of the experiment. The categories of body weight and feed intake amount were monitored to calculate the productivity and blood sampling was conducted for the analysis at the end of each experiment. Experiment 1:Although the productivity by the ME content difference during $0{\sim}2$ weeks did not have significant difference and the body weight increase by the difference of CP content and feed intake amount did not have much difference, the feed requirement rate was statistically improved in CP 21 and 22% treatment groups compared to the CP 20% group (P<0.05). The feed ME 3,100 kcal/kg treated group during $3{\sim}5$ weeks after starting the experiment revealed to show improved feed requirement rate (P<0.05). Within the period of experiment, the CP 22% treated group resulted to show significant body weight increase compared to the groups treated with low levels of CP (P<0.05) and the feed requirement rate was improved in high CP treated group compared to low CP treated groups, but the feed intake amount did not show significant difference between treated groups. During the experiment period, the body weight increase and feed requirement rate revealed to interact between ME and CP (P<0.05). During the whole experiment period of the 5 weeks, the feed requirement rate was improved in ME 3,100 kcal/kg treated group than the groups treated with ME 3,000 kcal/kg, and the CP (20) 18% treatment groups resulted to show higher values than other treatment groups (P<0.05). Body weight increase was high in CP (22) 22% treated groups than those of CP (21) 21% and (20) 18% treated groups, and the interaction between ME and CP was found at body weight increase and feed requirement rate (P<0.05). Although blood albumin and total cholesterol levels were elevated in ME 3,100 kcal/kg treated group than ME 3,000 kcal/kg treated group, but neutral fat content was reduced (P<0.05). On the other hand, the total cholesterol content was increased in CP (22) 21% treated group than CP (22) 20% and CP (20) 18% treated groups (P<0.05). Experiment 2: The body weight increase in 0-2 weeks was higher in ME 3,100 kcal/kg treated group than ME 3,000 kcal/kg treated group, and it was highly improved in CP 22% treated group than CP 20% treated group by showing the interaction between CP and ME (P<0.05). The significant improvement of feed requirement rate was observed in CP 21% and 22% treated groups compared to CP 20% treated group (P<0.05). The productivity between the growth period from 3 to 5 weeks of age and whole growth period resulted to show no significant difference. Although no difference was observed in blood composition between treated groups, the interaction of ME and CP on cholesterol content was accepted at the range of P<0.05). Therefore, it is considered that the appropriate dietary protein level within feed for the physiology of growing broiler chicks was 22% or more for the first two weeks and protein level of 21% or 20% from 3 to 5 weeks old for the maximization of productivity. Even if the energy level within feed had some partial effects on the productivity, but did not show consistency. So, further experiments needto be conducted by differentiating the energy level.

Effects of Dissolved Oxygen and Depth on the Survival and Filtering Rate and Pseudofeces Production of a Filter-feeding Bivalve (Unio douglasiae) in the Cyanobacterial Bloom (남조류 대발생 환경에서 수심과 용존산소 변화에 따른 담수산 이매패(말조개)의 생존율, 여과율 및 배설물 생산)

  • We performed the experiment to evaluate the effect of different DO concentrations (0.5, 4.5 and 9.0 $mgO_2L^{-1}$) and water depths (20, 50 and 80 cm) on the filtering rate, mortality, and pseudifeces production of Unio douglasiae against the cyanobacterial bloom (mainly Microcystis aeruginosa). A solitary-living bivalve U. douglasiae was collected in the upstream region of the North Han River (Korea). The harvested mussels were carefully transferred to the laboratory artificial management system, which was controlled temperature $(18{\pm}2^{\circ}C)$, flow rate (10L $h^{-1}$), food $(Chlorella^{TM})$, sediment (pebble and clay), light intensity (ca. $20{\mu}mol$ photons), and photocycle (12 L : 12 D). In the field observation, the mussel mortality was significantly correlated with water temperature, pH and DO concentration (P<0.05). The mortality was decreased with water depth; 65, 90, 80% of mortality at 20, 50, 80 cm water-depth, respectively. Filtering rate (FR) showed the highest value at 50 cm water depth, and thereby the concentration of chlorophyll-${\alpha}$ decreased continuously by 94% of the control at the end of the experiment. In contrast, FR decreased by 34% of the initial concentration at 20 cm water depth. Over the given water-depth range, the mussel FR ranged from $0.15{\sim}0.20L\;gAFDW^{-1}hr^{-1}$ during the 18hrs of experiment, and thereafter, they appeared to be approximately 0.11, 0.26 and 0.30 L $gAFDW^{-1}hr^{-1}$ at 20, 50 and 80cm water depth, respectively. FR was highest with the value of 0.46L $gAFDW^{-1}hr^{-1}\;at\;0.5mgO_2 L^{-1}$ at the early stage of the experiment, while it increased with DO concentration. Maximum pseudofaeces production was 11.2 mg $gAFDW^{-1}hr^{-1}\;at\;9.0mgO_2L^{-1}$. Our results conclude that U. douglasiae has a potential to enhance water quality in eutrophic lake by removing dominant cyanobacteria, but their effects vary with environmental parameters and the water depth at which they are located.

  • Factors affecting the survival of out planted Cryptomena and Japaneses Cypress seedlings. (I) - Especially on the drying and water soaking duration ­ There are often the cases of poor survival percentage for the economic forest establishments of Cryplomeria japonica D. Don. and Chamaecyparis obtusa End. This study was carried out to investigate the survival factors of both 1-1 seedlings and Cryptomena cutting Seedlings by room drying conditions, day drying conditions, OED green water soaking treatment and drying duration at the forest nursery of Chonnam National University from February to September. 1979. The results are as follows: 1. From 11 a.m. to 3 p.m. fluctuation in then moisture contents(MC) rapidly decreased and M.C. fluctuation in indoor until 5 p.m. showed the same results with the amount of an hour fluctuation in outdoor. 2. M.C. fluctuations between OED green and non-treatment of these seedlings were silghtly different. 3. The survival percentage of OED green and non-treatment of seedlings were highly significant in the room drying conditions and the day drying conditions respectively The effects of water soaking treatment on rooting was negligible. The survival percentage of seedlings of OED green was higher than that of non-treatment. 4. Although survival percentage of Japanese Cypress was almost in accord with Cryptomena, the power of resistance to drying was lower m Japanese Cypress than m Cryptomeria. 5. Compared with Cryptomena seedlings the survival percentage of Cryptomeria cutting seedlings was higher and not influenced at all in case of being drying for two hours; but it was highly significant in interactions between the drying time and the water soaking time. 6. When Cryptomena and Japanese Cypress seedling become drying in indoor for four days, the survival percentage remarkably reduced after two days and also was highly significant among numbers of drying days.

Hsp90 Inhibitor Induces Cell Cycle Arrest and Apoptosis of Early Embryos and Primary Cells in Pigs

  • Heat shock protein 90 (Hsp90) is ATPase-directed molecular chaperon and affects survival of cancer cell. Inhibitory effect of Hsp90 by inducing cell cycle arrest and apoptosis in the cancer cell was reported. However, its role during oocyte maturation and early embryo development is very insufficient. In this study, we traced the effects of Hsp90 inhibitor, 17-allylamino-17-demethoxygeldanamycin (17-AAG), on meiotic maturation and early embryonic development in pigs. We also investigated several indicators of developmental potential, including structural integrity, gene expression (Hsp90-, cell cycle-, and apoptosis-related genes), and apoptosis, which are affected by 17-AAG. Then, we examined the roles of Hsp90 inhibitor on viability of primary cells in pigs. Porcine oocytes were cultured in the NCSU-23 medium with or without 17-AAG for 44 h. The proportion of GV arrested oocytes was significantly different between the 17-AAG treated and untreated group (78.2 vs 34.8%, p<0.05). After completion of meiotic maturation, the proportion of MII oocytes was lower in the 17-AAG treated group than in the control group (27.9 vs 71.0%, p<0.05). After IVF, the percentage of penetrated oocytes was significantly lower in the 17-AAG treated group (25.2%), resulting in lower normal pronucleus formation (2PN of 14.6%). Therefore, the inhibition of meiotic progression by Hsp90 inhibitor played a critical role in fertilization status. Porcine embryo were cultured in the PZM-3 medium with or without 17-AAG for 6 days. In result, significant differences in developmental potential were detected between the embryos that were cultured with or without 17-AAG. Terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) showed that the number of containing fragmented DNA at the blastocyst stage increased in the 17-AAG treated group compared with control (7.5 vs 4.4, respectively). Blastocysts that developed in the 17-AAG treated group had low structural integrity and high apoptotic nuclei than those of the untreated control, resulting in decrease the embryonic qualities of preimplantation porcine blastocysts. The mRNA expressions of cell cycle-related genes were down-regulated in the 17-AAG treated group compared with control. Also, the expression of the pro-apoptotic gene Bax increased in 17-AAG treated group, whereas expression of the anti-apoptotic gene Bel-XL decreased. However, the expression of ER stress-related genes did not changed by 17-AAG. Cultured pESF cells were treated with or without 17-AAG and used for MIT assay. The results showed that viability of pESF cells were decreased by treatment of 17-AAG ($2{\mu}M$) for 24 hr. These results indicated that 17-AAG decreased cell proliferation and increased cell death. Expression patterns Hsp90 complex genes (Hsp70 and p23), cell cycle-related genes (cdc2 and cdc25c) and apoptosis-related genes (Bax and Bcl-XL) were significantly changed by using RT-PCR analysis. The spliced form of pXbp-1 product (pXbp-1s) was detected in the tunicamycin (TM) treated cells, but it is not detected in 17-AAG treated cells. In conclusion, Hsp90 appears to play a direct role in porcine early embryo developmental competence including structural integrity of blastocysts. Also, these results indicate that Hsp90 is closely associated with cell cycle- and apoptosis-related genes expression in developing porcine embryos.

Combustion Characteristic Study of LNG Flame in an Oxygen Enriched Environment (산소부화 조건에 따른 LNG 연소특성 연구)

  • The ultimate objective of this study is to develop oxygen-enriched combustion techniques applicable to the system of practical industrial boiler. To this end the combustion characteristics of lab-scale LNG combustor were investigated as a first step using the method of numerical simulation by analyzing the flame characteristics and pollutant emission behaviour as a function of oxygen enrichment level. Several useful conclusions could be drawn based on this study. First of all, the increase of oxygen enrichment level instead of air caused long and thin flame called laminar flame feature. This was in good agreement with experimental results appeared in open literature and explained by the effect of the decrease of turbulent mixing due to the decrease of absolute amount of oxidizer flow rate by the absence of the nitrogen species. Further, as expected, oxygen enrichment increased the flame temperatures to a significant level together with concentrations of $CO_2$ and $H_2O$ species because of the elimination of the heat sink and dilution effects by the presence of $N_2$ inert gas. However, the increased flame temperature with $O_2$ enriched air showed the high possibility of the generation of thermal $NO_x$ if nitrogen species were present. In order to remedy the problem caused by the oxygen-enriched combustion, the appropriate amount of recirculation $CO_2$ gas was desirable to enhance the turbulent mixing and thereby flame stability and further optimum determination of operational conditions were necessary. For example, the adjustment of burner with swirl angle of $30\sim45^{\circ}$ increased the combustion efficiency of LNG fuel and simultaneously dropped the $NO_x$ formation.

The Effects of Nitrogen Type and Fertilized Depth on Leaching and Absorption of Nitrogen in Paddy Soil and Growth and Yields of Rice (질소비료(窒素肥料)의 형태(形態)와 시용심도(施用深度)가 질소(窒素)의 용탈흡수(溶脫吸收) 및 수도(水滔) 생육수량(生育收量)에 미치는 영향(影響))

  • In order to find the effects of nitrogen type (ammonium sulfate and urea fertilizer) and fertilized depth, (0~10cm, 0cm, 5~10cm, 10~15cm, 15~20cm, and 20cm below) on leaching and absorption of nitrogen in paddy soil, and growth and yields of rice, the pot culture experiment was carried out, using the variety Jaekun, one of the Korean leading variety. Experimental results were Summarized as follows: 1. No variations of the pH of percolating water were induced by the differences of nitrogen types and their fertilized depth (Table. 2). 2. The leaching of nitrogen was less in ammonium sulfate and top soil fertilizing plots than in urea and subsoil fertilizing plot, and the growth of rice in early stage was more promoted in ammonium sulfate and topsoil fertilizing plots (Table. 1, 7 and 8). 3. Leachng of nitrogen through the percolating water almost came to an end at the most numerous tiller stage (Table 1). 4. The absorption of nitrogen of each part of the rice plant in the harvesting stage correlated closely with the yields of each part (Table 5, 6, 9 and 10) and the leaching of nitrogen in the early stage was inversely proportion to the absorption of nitrogen of rice plant in the harvesting time (Table 1, 5, 6, 9 and 10). 5. The number of spikes was more numerous in ammonium sulfate plots than in urea plots on an average, so that the yields were higher in the ammonium sulfate plots than in urea plots although no differences in the grain number per spike were found in above two plots. The number of spikes was more numerous in topsoil fertilizing plots than in subsoil fertilizing plots, but the grain number per spike was less in former than in latter, so that no difference in yields was found. The absorption of nitrogen correlated closely with the yields in complete paddy grains (Table 5, 9, and 10). 6. At the ammonium sulfate fertilizing plots, the number of spikes was more numerous in topsoil fertilizing plots than in subsoil fertilizing plots, (among the each of the topsoil plots, 0~10cm and 5~10cm fertilizing plots kept more spikes than the 0cm fertilizing plots), but the grain number per spike was less in former than in latter (among the each of topsoil plots, no differences were found), so that no significant difference in yields was showed between the topsoil and subsoil fertilizing plots, but the results showed the tendency that the yields were highest in 0~10cm plots and the lowest in 20cm below plots. At the urea fertilizing plots, the number of spikes decreased in proportion to the increasing of fertilized depth, but no variations were found in the grain number per spike, so that the yields decreased in proportion to the increasing of fertilized depth. The absorption of nitrogen correlated closely with the yields in complete paddy grains (Table 5, 6, 9, and 10). 7. When fertilized in topsoil, the number of spikes was more numerous in ammonium sulfate plot than in urea plot, but the grain number per spike variated reversely, so that no differences were found in the yields between the ammonium sulfate and the urea plots, when fertilized in subsoil, both the number of spikes and the grain number per spike were larger in ammonium sulfate than in urea plot, so that the yields were also higher in ammonium sulfate plots (Table 5, 6, 9 and 10). 8. The weight of straw and its nitrogen absorption were higher in ammonium sulfate plot than in urea plot and decreased in proportion to the increasing of fertilized depth. Among the each of topsoil fertilizing plots, the 0~10cm and the 5~10cm fertilizing plots excelled the 0cm plot (Table 5, 6, 9 and 10). 9. No significant variations in the fertilizer treatments were found in the characters of heading date, maturing date, length of culm, length of spike, weight of empty grain, 1,000 grain weight, and one liter weight.
