We have previously shown that the addition of silkworm hemolymph to a culture medium increases the longevity of insect and mammalian cells by inhibiting apoptosis. This indicates that the component which inhibits apoptosis is contained in the silkworm hemolymph, The apoptosis-inhibiting component was isolated from silkwonn hemolymph and characterized in our previous study. A database search using the N-terminal amino acid sequence of this component as a template resulted in a 95% homology with a low molecular weight lipoprotein, the so called ’30K protein' of unknown function. In this study, the 30K protein gene was expressed in mammalian and insect cells to confirm the apoptosis-inhibiting effect. The overexpression of 30K protein in mammalian cell inhibited the staurosporin-induced apoptosis by the prevention of the activation of caspase 3. Using an Autographa californicanuclear polyhedrosis virus (AcNPV) system, the 30K protein was overexpressed also in insect cells. The expression of the 30K protein increased the longevity of baculovirus-infected insect cells by inhibiting apoptosis. These results suggest that the 30K protein is a novel anti-apoptotic protein.
We investigated the induction of resistance to Haemaphysalis longicornis infestation in rabbits that had been immunized with recombinant H. longicornis P27/30 protein. The success of immunological control methods is dependent upon the use of potential key antigens as tick vaccine candidates. Previously, we cloned a gene encoding 27 kDa and 30 kDa proteins (P27/30) of H. longicornis, and identified P27/30 as a troponin I-like protein. In this study, rabbits that were immunized with recombinant P27/30 expressed in Escherichia coli showed the statistically significant longer feeding duration for larval and adult ticks (P<0.05), low engorgement rates in larval ticks (64.4%), and an apparent reduction in egg weights, which suggest that H. longicornis P27/30 protein is a potential candidate antigen for a tick vaccine. These results demonstrated that the recombinant P27/30 protein might be a useful vaccine candidate antigen for biological control of H. longicornis.
An antiviral protein (CAP30) with ribosome-inactivating activity was purified from the leaves of Chenopodium album L. through ammonium sulfate precipitation and column chromatography using S-Sepharose, Blue-Sepharose, FPLC Suprose12 HR, and FPLC Mono-S. The molecular wight of CAP30 was estimated to be 30kD. CAP30 was thermostable, maintaing its activity even after incubation at $70^{\circ}C$ for 30 min, and was stable in the pH range of 6 to 9. In a cell-free in vitro translation system using rabbit reticulocyte lysate, protein synthesis was inhibited by the addition of CAP30 with an $IC_{50}$ of 2.26pM. The comparison of N-terminal amino acid sequences of this protein with known ribosome-inactivating proteins (RIPs) revealed that it had some sequence homology with PAP-S and PAP-R from pokeweed (Phytolacca americana)and dodecandrin from P. dodecandra, but had no sequence homology with RIPs from other plants belonging to different orders. The mosaic symptoms on tobacco leaves caused by cucumber mosaic virus infection was completely inhibited by 100 ng/ml of the pure CAP30 protein.
BACKGROUND/OBJECTIVE: The incidence of colorectal cancer (CRC) has been attributed to higher intake of fat and protein. However, reports on the relationship between protein intake and CRC are inconsistent, possibly due to the complexity of diet composition. In this study, we addressed a question whether alteration of protein intake is independently associated with colonic inflammation and colon carcinogenesis. MATERIALS/METHODS: Balb/c mice were randomly divided into 4 experimental groups: 20% protein (control, 20P, 20% casein/kg diet), 10% protein (10P, 10% casein/kg diet), 30% protein (30P, 30% casein/kg diet), and 50% protein (50P, 50% casein/kg diet) diet groups and were subjected to azoxymethane-dextran sodium sulfate induced colon carcinogenesis. RESULTS: As the protein content of the diet increased, clinical signs of colitis including loss of body weight, rectal bleeding, change in stool consistency, and shortening of the colon were worsened. This was associated with a significant decrease in the survival rate of the mice, an increase in proinflammatory protein expression in the colon, and an increase in mucosal cell proliferation. Further, colon tumor multiplicity was dramatically increased in the 30P (318%) and 50P (438%) groups compared with the control (20P) group. CONCLUSIONS: These results suggest that a high protein diet stimulates colon tumor formation by increasing colonic inflammation and proliferation.
The effect of various dietary protein levels on growth and body composition of young common carp raised from 56 g to 170 g in recirculating system was investigated for 15 weeks when they were fed to visual satiety three times daily. Five experiemtal diets were formulated to contain 40, 35, 30, 25 and 21% protein levels and 3.56, 3.59. 3.63, 3.66 and 3.69 kcal/g diet GE levels respectively. Mean survival rates of the fish fed the 40, 35, 30 and 25% protein diets were not different but sig-nificantly higher than that of the fish fed the 40, 35, 30 and 25% protein diets were not different but sig-nificantly higher than that of o the fish fed the 21% protein diet(P<0.05) Weight gain (g/tank) of common carp fed the 30% protein diet was the best. However weight gain of the fish fed the 25, 30, 35 and 40% protein diets were not different but significantly better than that of the fish fed the 21% protein diet. Feed efficiency ratio of the 21% protein diet was significantly lower (P<0.05) than for other groups of diets which were not different among them. Protein efficiency ratio for the 21% protein diet was significantly lower (P<0.05) than for other groups of diets which were not different among them(P>0.05) Dietary protein level had no effect on hemoglobin content in the fish(P>0.05) Crude protein contents of whole body of the fish fed the 35 and 40% protein diets were significantly higher than that of the fish fed the 21$$\mid$% or 25% protein diet(P<0.05) Body crude lipid contents of the fish fed the 21 and 25% protein diets were significantly higher than that of the fish fed the 30% or 35% protein diet. Crude ash contents of the fish fed the 35 and 40% protein diets were significantly higher than that of the fish fed the 21% or 25% protein diet(P<0.05) Moisture content of the fish fed the 35% protein diet was significantly higher than that of the fish fed the 21% protein diet(P<0.05) In considering growth performance of common carp and efficiency of diet dietary protein level could be lowered up to 25% without the reduction of young common carp production in recirculating system.
An experiment was conducted on rainbow trout(Oncorhynclus mykiss) for eight weeks to investigate the growth performance of the fish fed with different dietary protein with constant diet energy of $20kJg^{-1}$. Four diets containing 25, 30, 35 and 40% crude protein were used. The highest mean final weight was obtained for the fish fed with diet having 35% protein. Growth performance in terms of Specific Growth Rate (SGR), Food Conversion Ratio (FCR) and Protein Efficiency Ratio (PER) were calculated for each diet. There were no significant differences in SGR but the highest value was exhibited by fish fed with 35% protein diet. Significant differences were found among FCR of different diets. Diets with 35 and 40% crude protein gave better FCR value than that of 25 and 30% crude protein. Although significant differences were not found between PER of different diets but PER of diet with 35% protein was found to be better than PER of both high and low protein diets (diets of 40 and 30% crude protein). It is concluded that diet having 35% protein with protein energy ratio of $17.53mgkJ^{-1}$ was suitable for rainbow trout (O. mykiss) among the protein spectrum used.

Generally known psittacosis or ornithosis is a disease of birds caused by the bacterium Chlamydia psittaci. Humans are accidential hosts and are most commonly infected from avian sources. It raises hepatitis or neurosis. As major outer membrane protein (MOMP) of Chlamydia psittaci has been known to play a role in the avoidance of host immune defenses, research on developing a Chlamydia vaccine has focused on the MOMP. In this study, the gene encoding the major outer membrane protein (MOMP) of the Chlamydia psittaci strain 6BC was cloned and expressed in Escherichia coli strain M-15. The recombinant DNA was cloned by fusion prokaryotic expression vector pQE30-GFPII. Expression of the recombinant protein was performed in E. coli and was induced by IPTG. The size of expressed recombinant protein is 74.220 kDa (MOMP, 43.260 kDa; GFP expression region, 30 kDa; $6{\times}His$ tag, 960Da). This protein was purified by using his-tagging-inclusion body. Recombinant protein was reconfirmed through ELISA test and western blot with antibody against pQE30-GFPII. It will be useful antibody development.

In order to investigate the feasibility of 30K protein from silkworm (Bombyx mori) hemolymph (SH) on the proliferation of human cells, a simple separating procedure by anion-exchange chromatography system with Q-Sepharose fast flow gel was established. The 30K protein was eluted with an optimized condition of 0.16 M sodium chloride in 20 mM tris buffer (pH 9.0). The separated 30K protein at three concentrations of 0.04, 0.12, and 0.4 mg/ml was added to the culture medium with various human cells, such as chondrocytes, periosteum-derived cells, and MRC-5 cells, and their growth rates were measured. The cell growth rate at protein concentration of 0.4 mg/ml was always higher than that without 30K protein in all human cells tested, suggesting that the 30K protein has positive effect on the increase of the life span of human cells.

A feeding trial was conducted to determine the optimum dietary protein and lipid levels for the growth of juvenile muddy loach Misgurnus anguillicaudatus. Eight experimental diets (designated as P20L7, P20L14, P30L7, P30L14, P40L7, P40L14, P50L7 and P50L14) were formulated to contain 20%, 30%, 40% and 50% protein with 7% and 14% lipid. Three replicate groups of muddy loach (1.5 g/fish) were fed one of the experimental diets ad libitum for 8 weeks. Survival of fish fed the P20L14 diet was lower than in other groups (P<0.05). Specific growth rate (SGR) and feed efficiency (FE) of fish were affected by dietary protein (P<0.05) and lipid (P<0.005) levels. SGR and FE of fish tended to decrease with increasing dietary lipid level at the same protein level. There were no significant differences in SGR and FE among fish fed diets containing different protein levels ranging from 30% to 50% with the same lipid level. Whole body moisture and lipid contents were affected by dietary protein and lipid levels. Crude lipid content of fish fed the 14% lipid diets was higher than that of fish fed 7% lipid diets at each protein level. The results of this study indicate that a diet containing 30% protein with 7% lipid would be suitable for the optimum growth and effective feed utilization of juvenile muddy loach.

Eight experimental diets (P20L7, P20L14, P30L7, P30L14, P40L7, P40L14, P50L7 and P50L14) were formulated to contain 20%, 30%, 40% or 50% protein combined with either 7% or 14% lipid. Three replicate groups of fish (mean mass: 3.9 g/fish) were fed one of the experimental diets ad libitum for 8 weeks. Survival of fish fed the P20L14 diet was lower than that of fish fed the P40L14, P50L7 and P50L14 diets. Growth of fish fed diets containing 7% lipid increased with increasing protein level (up to 50% protein); growth of fish fed diets containing 14% lipid increased with increasing protein level (up to 30% protein). The feed efficiency of fish fed a diet with 50% protein and 7% lipid was higher than that of other groups. Whole body moisture and lipid contents were affected by dietary lipid level but not by dietary protein level. The crude lipid contents of fish fed 14% lipid diets were higher than those fed 7% lipid diets across all protein levels (other than the 50% level). Thus, under our experimental conditions, an increase in dietary protein level improved growth and feed efficiency of fish; a diet containing 50% protein with 7% lipid was optimal for growth and effective feed utilization in juvenile long snout bullhead.
