Studies on the Chemical Constituents Biological Activities of Mulberry Extracts (오디(Mulberry) 추출물의 성분분석 및 생리활성에 관한 연구)

  • The freezed Mulberry (10 kg) was extracted with 80% EtOH, concentrated, and fractionated with a series of n-hexane, ethyl acetate, and water on a separatory funnel. A portion of ethyl acetate soluble (22 g) was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents. The isolated compounds were identified by cellulose TLC, $^1H-$,$^{13}C$-NMR, FAB, and EI-MS. Quercetin-3-O-${\beta}$-D-glucopyranoside (Compound I), protocatechuic acid (Compound II), p-hydroxybenzoic acid (Compound III) were isolated from the ethyl acetate soluble fraction. In antioxidative activities of the fractionated extractives using DPPH radical scavenging test, EtOAc and water soluble fractions indicated better than BHT as contro and in in vitro tests using MTT assay, there was no cytotoxicity. Also, tyrosinase inhibition and anticancer activities were not so good, but there may be a potential as a cosmetic raw material because the cell extension effect was excellent.

Phytochemical Study of Adenophora stricta Roots (당잔대(Adenophora stricta) 뿌리의 성분연구)

  • Adenophora stricta Miq. (Campanulaceae) is an annual herb, which has been used as a traditional medicine in Korea, Japan and China to treat bronchial asthma, tonsillitis, and hypertension. In this study, 12 compounds were isolated from the roots of A. stricta and isolates were identified to be methyl adenophorate (1), decursidin (2), L-tryptophan (3), D-1,2,3,4-tetrahydronorharmane-3-carboxylic acid (4), vanillic acid 4-O-${\beta}$-D-glucopyranoside (5), syringic acid 4-O-${\beta}$-D-glucopyranoside (6), vanillin (7), vanillic acid (8), p-hydroxybenzaldehyde (9), p-hydroxybenzoic acid (10), p-hydroxyacetophenone (11) and linoleic acid (12). Decursidin (2) and D-1,2,3,4-tetrahydronorharmane-3-carboxylic acid (4) is firstly reported from A. stricta in current study.

Development of PCR-Based Screening Methods for Macrolide Type Polyketides in Actinomycetes

  • About two thirds of the naturally occurring antibiotics have been discovered from actinomycetes. Therefore, the probability of discovering further new antibiotics from actinomycetes is declining as many known metabolites are isolated repeatedly. However, various efforts leave been made in order to enhance the probability of discovering novel compounds. In the present study, we have developed new screening strategies based on the antibiotic biosynthetic pathway, and the genetic information, utilizing polymerase chain reaction. We have selected macrolide type polyketides. In order to divide the ansamycin group antibotic of macrolide type polyketides, we have selected 3-amino-5-hydroxybenzoic acid (AHBA) moiety which contains a biosynthetically unique structural element in the group as a target molecules. Oligonucleotide primers were designed to amplify DNA fragments of macrolide type polyketide synthase and AHBA synthase genes from fourteen actinomycetes species. This method was successfully applied to all three of the known macrolide type polyketide produccing actinomycetes tested. In addition, it also identified the presence of potential macrolide type polyketide producing genes from seven actinomycetes that were known to produce none of macrolide type polyketides, and AHBA biosynthetic genes in one actinomycetes. This technique is potentially useful for the screening of new antibiotices and cloning of their biosynthetic genes.

Antimicrobial and Antioxidative Activities from Moutan Cortex Extract (목단피 추출물의 항균 및 항산화 작용)

  • Methanol extract and various solvent fractions from Moutan Cortex were tested for their antimicrobial activities, free radical scavenging activities and antioxidative activities, and phenolic compounds in ethylacetate fraction were analyzed by GC and HPLC. In the antimicrobial activities test, the ethylacetate fraction of methanol extract showed stronger than other fractions. The antimirobial activities were more effective against Gram positive bacteria than Gram negative bacteria. Minimum inhibitory concentration(MIC) of ethylacetate fraction showed 156-1250$\mu\textrm{g}$/ml against Cram positive bacteria and 2500-5000$\mu\textrm{g}$/mg against Gram negative bacteria. The free radical scavenging activities and antioxidative activities using linoleic acid were higher in ethylacetate fraction. The antioxidative activity of ethylacetate fraction was similar to ${\alpha}$-tocopherol. The 3 major phenolic compounds were analyzed by GC and HPLC and these content were determined. The content of p-hydroxybenzoic acid, methyl gallate and gallic acid were 1.35%, 14.61% and 4.01%, respectively.

Chemical Constituents of Nelumbo nucifera Seeds

  • The phytochemical study for the extract of Nelumbo nucifera (Nymphaceae) seeds has led to the isolation of ten compounds including five simple phenolic compounds, two indole derivatives, a flavonoid glycoside, two abscisic acid derivatives. The interpretation of 1D and 2D NMR and ESI-Q-TOF-MS spectroscopic data revealed the chemical structures of isolates to be p-hydroxybenzoic acid (1), protocatechuic acid (2), (E)-p-coumaric acid (3), (E)-ferulic acid (4), (E)-sinapate-4-O-${\beta}$-$\text\tiny{D}$-glucopyranoside (5), tryptophan (6), 3-indoleacetic acid (7), isoschaftoside (8), dihydrophaseic acid (9), dihydrophaseic acid 3'-O-${\beta}$-$\text\tiny{D}$-glucopyranoside (10). To the best of our knowledge, 1 - 5 and 7 were identified for the first time from N. nucifera seeds, and the presence of dihydrophaseic acid (9) and its glucoside (10) were demonstrated secondly in this plant.

Graft Copolymerization of Acrylic Monomer Containing Aromatic Carboxylic Acid Group onto EPDM and Their Mechanical Properties (EPDM에 방향족 카르복시산을 함유하는 아크릴 단량체의 그라프트 공중합과 기계적 특성)

  • In this study, p-acryloyloxybenzoic acid(ABA) was synthesized with p-hydroxybenzoic acid(HBA) and acryloyl chloride(AC). The synthesized ABA monomer was grafted onto ethylene-propylene-diene rubber(EPDM) in toluene using benzoyl peroxide(BPO) as an initiator. The structures of ABA and EPDM-g-ABA were characterized by FT-IR, $^1H$-NMR, and $^{13}C$-NMR spectrometer. The graft ratio of EPDM-g-ABA increased with increasing the concentration of the initiator and the monomer. Mechanical properties such as tensile strength and compression set of the EPDM-g-ABA were improved with increasing the graft ratio. The $T_g$ and initial decomposition temperature were also increased with increasing the graft ratio.

Characterization of Biological Treatment by an Isolated Phenol-Degrading Bacterium (페놀분해세균의 분리 및 생물학적 처리 특성)

  • 20 bacterial strains capable of growing on phenol minimal medium were isolated from soil and wastewater by the enrichment culture technique, and among them, one isolate which was the best in the cell growth was selected and identified as Bacillus sp. SH3 by its characteristics. Strain SH3 could grow with phenol as the sole carbon source up to 15 mM, but did not grow in minimal medium containing above 20 mM of phenol. The optimal conditions of temperature and initial pH for growth and phenol degradation were 30$^{\circ}$C and 7.5, respectively. This strain could grow on various aromatic compounds such as catechol, protocatechuic acid, gentisic acid, o-, m-, p-cresol, benzoic acid, p-hydroxybenzoic acid, anthranilic acid, phenyl acetate and pentachlorophenol, and the growth-limiting log P value of strain SH3 on organic solvents was 3.1. In batch culture, strain SH3 degraded 97% of 10 mM phenol in 48 hours. In continuous culture under the conditions of 20 mM of influent phenol concentration and 0.050 hr$^{-1}$ of dilution rate, the treatment rate of phenol was 94%.

  • From the 70% EtOH extract of Paeonia lactiflora roots (Paeoniaceae), fourteen phenolic and related compounds were isolated. They were identified as ${\alpha}-tocopherol$ (1), dioctylphthalate (2), ${\alpha}-tocospiro$ B (3), paeonol (4), 3,3'-di-O-methylellagic acid(5), 3,4'-di-O-methylellagic acid (6), benzoic acid (7), aromadendrin (8), p-hydroxybenzoic acid (9), (+)-catechin (10), gallic acid (11), nicotinamide (12), methyl gallate (13) and $1,2,3,4,6-penta-O-galloyl-{\beta}-D-glucose$ (14) by spectroscopic methods. Among these compounds, 1-3, 5, 6, 8 and 12 were isolated for the first time from this plant.

Identification and Growth Inhibition of Phytotoxic Substances from Tomato Plant (토마토식물의 독성물질 확인과 생장억제작용)

  • Phenolic compounds such as gallic acid, ferulic acid, p-hydroxybenzoic acid, vanillic acid, salicylic acid, tannic acid, and hydroquinone were identified from the aqueous extracts and volatile substances of tomato plant by paper chromatography, high performance liquid chromatography and gas chromatography. The seed germination and seedling growth of the experimental species, lettuce and egg plant, were severely inhibited in 5$\times$10-3M of phenolic reagents identical to those identified from tomato plant. Germination and growth rate of test species in 5$\times$10-4M and 5$\times$10-5M were higher than that of 5$\times$10-3M. Therefore, 5$\times$10-3M of phenolic compounds would be assumed to be threshold concentration for allelopathic effects.

Comparative Analysis of Antioxidant, Anti Aging and Phenolic Compounds of Different Solvent Extracts from Saccharina japonica and Costaria costata (추출용매에 따른 쇠미역과 다시마의 항산화, 항노화 활성과 페놀화합물 비교분석)

  • This study analyzed the polyphenol, flavonoid contents, antioxidant activity, anti-aging activity and phenol component contents of Saccharina japonica (SJ), Costaria costata (CC) extracts with hot water, 95% methanol, 95% prethanol for investigating possible utilization of SJ and CC extracts. The result revealed that the SJ and CC methanol extracts showed the highest polyphenol and flavonoid contents, 4.63 mg TAN/g, and 4.19 mg QUE/g respectively. Also, the SJ and CC methanol extracts showed higher antioxidant activity than prethanol and hot water extracts, whereas the ABTS radical scavenging activities were the highest in prethanol extracts (IC50 = 15.4, 10.3 ㎍/µL). In anti-aging activity for evaluating the anti-wrinkle activity and skin whitening activity, the CC methanol extracts had high collagenase inhibitory activity (88.3%), and the SJ prethanol extracts showed higher elastase inhibitory activity (19.0%) compared to other extracts. Then the tyrosinase inhibitory activity was significantly higher in the SJ and CC methanol extracts (41.8, 30.3%, respectively), whereas prethanol extracts were the lowest. To identify the phenol component contents of SJ and CC extracts, 4-hydroxybenzoic acid, naringenin, naringin and nicotinic acid were measured using LC-MS/MS. As a result, the phenol contents were the highest in SJ methanol extract (4-hydroxybenzoic acid), SJ and CC prethanol extract (naringin and naringenin) and CC prethanol extract (nicotinic acid). Lastly, the antioxidant activity of SJ and CC showed high correlations with polyphenol and flavonoid contents (R = -0.946~0.883). These results suggest that prethanol or methanol extracts of SJ and CC have higher antioxidant activities, anti-aging activity and the potential to be used as material for health functional food and cosmetics.
