Pretreatment of Fish for the determination of polycyclic aromatic hydrocarbons using alkali digestion (알칼리분해를 이용한 어류 중 다환방향족탄화수소의 전처리방법)

  • Polycyclic Aromatic Hydrocarbons(PAHs) contamination arises from several sources including processing of food(smoking, direct drying, cooking) and environmental contamination of air, water, or soil, the later being considered as the most important. In this study, to establish the analytical method for some PAHs[benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in fish, alkali digestion time, extraction solvents, elution volume of florisil cartridge for clean-up have been optimized. The methodology involved saponification and extraction with n-hexane, clean-up on Sep-Pak florisil cartridges and determination by HPLC/FLD(High Performance Liquid Chromatography/Fluorescence Detector). Overall method recoveries for 8 PAHs spiked into these products ranged from 90 to 106%.

  • Aralia elata Seemann (AE) has long been used as a folk medicine for the treatment of various diseases including diabetes mellitus, anti-arthritic, and anti-gastric ulcer agent in Korea, Japan, and China. This study was performed to establish a simple and reliable HPLC/UV analytical method for determination of most active anti-hypertensive compound, a 3-O-${\alpha}$-L-rhamnopyranosyl($1{\rightarrow}$2)-${\alpha}$-L-arabinopyranosyl hederagenin 28-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}6$)-${\beta}$-D-glucopyranosylester (HE) for the standardization of the shoot extract of AE as a health functional food ingredient. The quantitative analytical method of HE was optimized by HPLC analysis using reverse-phase C18 column at $40^{\circ}C$ with $H_2O$ and acetonitrile (70:30, v/v) as an isocratic mobile phase at a flow rate of 1.0 mL/min and detection wavelength of UV 205 nm. This HPLC/UV analytical method showed good specificity and high linearity in the tested range of 0.03125-2.0mg/ml with excellent coefficient of determination ($R^2$) of 0.9999. The limit of detection and limit of quantification were $12.0{\mu}g/mL$ and $36.5{\mu}g/mL$, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 0.2% and 0.1%, respectively. These results indicate that the established HPLC/UV analytical method is very simple, specific, precise, accurate, and reproducible and thus can be useful for the quantitative analysis of HE as a functional anti-hypertensive compound in AE extract.

  • The rotor overhang is used to enhance air-gap flux and improve power density. Due to asymmetry in the axial direction caused by the overhang, the time consuming 3D analysis is necessary to design the motor with overhang. To solve this problem, this paper proposes the equivalent magnetic circuit model (EMCM) that can consider overhang effects without the 3D analysis by using effective air-gap length. The analysis time can be reduced significantly via the proposed EMCM. The reduction of the analysis time is essential for the preliminary design of the motor. In order to verify the proposed model, the 3-D finite-element method (FEM) analysis is adopted. 3-D FEM results confirm the validity of the proposed EMCM.

  • In this study, a convenient, fast and accurate inductively coupled plasma atomic emission spectrometry (ICP-AES) method has been optimized for the determination of osmium in aqueous solutions. The method makes use of the reaction of sulfurous acid with osmium to quantitative conversion of volatile Os(VIII) to non-volatile Os(IV) in the pH range 2-10. The response was found to be stabilized immediately after sulfurous acid reacted with osmium. The precision was calculated to be 0.5-4.5 % (RSD) under various ICP-AES conditions. The detection limit was 2.5-57.7 ng/g based on $3{\sigma}$ of the blank response (n=3) using a concentric flow nebulization.

Qualitative and quantitative assessment of process related impurities in Brigatinib raw material and formulations using HPLC

  • The presence of process related impurities in any drug or the drug product was associated with its safety, stability and efficacy. The overall literature survey proved that there is no method published on the assessment of process related impurities in brigatinib. In this study, a simple, reliable and stable HPLC qualitative method was reported for quantification of process related impurities with easy and quick extraction procedure. The impurities along with standard brigatinib was resolved on Lichrospher® C18 (250 mm × 4.6 mm; 5 ㎛ particle size) column in room temperature using methanol, acetonitrile, pH 4.5 phosphate buffer in 55:25:20 (v/v) at 1.0 mL/min as mobile phase and UV detection at 261 nm. The method produces well resolved peaks at retention time of 4.60 min, 12.28 min, 3.37 min, 7.34 min and 8.39 min respectively for brigatinib, impurity A, B, C and D. The method produces a very sensitive detection limit of 0.0065 ㎍/mL, 0.0068 ㎍/mL, 0.0053 ㎍/mL and 0.0058 ㎍/mL for impurity A, B, C and D respectively with calibration curve linear in the concentration range of 22.5-135 ㎍/mL for brigatinib and 0.0225-0.135 ㎍/mL for impurities. The method produces all the validation parameters under the acceptable level and doesn't produces any considerable changes in peak area response while minor changes in the developed method conditions. The method can effectively resolve the unknown stress degradation products along with known impurities with less % degradation. The method can efficiently resolve and quantify the impurities in formulation and hence can suitable for the routine quality analysis of brigatinib in raw material and formulation.

Comparison of Two- and Three-dimensional Approaches for the Numerical Determination of Equivalent Mechanical Properties of Fractured Rock Masses (균열암반의 역학적 등가물성의 수치해석적 결정을 위한 2차원 및 3차원 해석의 비교)

  • This paper compares the two- and three-dimensional (2D and 3D) approaches for the numerical determination of the equivalent mechanical properties of fractured rock masses. Both orthogonally-fractured model and discrete fracture networks (DFN) were used for the geometry and 2D models were cut in various directions from 3D model to compare their mechanical properties. Geological data were loosely based on the data available from Sellafield, UK. Analytical method based on compliance tensor transformation was used for investigation in orthogonally fractured rock and numerical experiments were conducted on fractured rock mass with DFN geometry. It is shown that 2D approach always overestimates the elastic modulus of fractured rock masses by a factor of up to around two because fractures are assumed to be perpendicular to the model plane in 2D problems. Poisson ratios tend to have larger values in 2D analysis while there is opposite trend in some sections. The study quantitatively demonstrates the limitation of the 2D approach that uses the simplified model from true 3D geometry.

Application and Validation of an Optimal Analytical Method using QuEChERS for the determination of Tolpyralate in Agricultural Products (QuEChERS법을 활용한 농산물 중 제초제 Tolpyralate의 최적 분석법 선발 및 검증)

  • BACKGROUND: Pesticides are broadly used to control weeds and pests, and the residues remaining in crops are managed in accordance with the MRLs (maximum residue limits). Therefore, an analytical method is required to quantify the residues, and we conducted a series of analyses to select and validate the quick and simple analytical method for tolpyralate in five agricultural products using QuEChERS (quick, easy, cheap, effective, rugged and safe) method and LC-MS/MS (liquid chromatography-tandem mass spectrometry). METHODS AND RESULTS: The agricultural samples were extracted with acetonitrile followed by addition of anhydrous magnesium sulfate, sodium chloride, disodium hydrogencitrate sesquihydrate and trisodium citrate dihydrate. After shaking and centrifugation, purification was performed with d-SPE (dispersive-solid phase extraction) sorbents. To validate the optimized method, its selectivity, linearity, LOD (limit of detection), LOQ (limit of quantitation), accuracy, repeatability, and reproducibility from the inter-laboratory analyses were considered. LOQ of the analytical method was 0.01 mg/kg at five agricultural products and the linearity of matrix-matched calibration were good at seven concentration levels, from 0.0025 to 0.25 mg/L (R2≥0.9980). Mean recoveries at three spiking levels (n=5) were in the range of 85.2~112.4% with associated relative standard deviation values less than 6.2%, and the coefficient of variation between the two laboratories was also below 13%. All optimized results were validated according to the criteria ranges requested in the Codex Alimentarius Commission (CAC) and Ministry of Food and Drug Safety (MFDS) guidelines. CONCLUSION: In conclusion, we suggest that the selected and validated method could serve as a basic data for detecting tolpyralate residue in imported and domestic agricultural products.

Numerical Investigation of an Unconditionally Stable Compact 2D FDTD Based on the Alternating-Direction Implicit Scheme

  • An unconditionally stable compact 2D Alternating-Direction Implicit (ADI) FDTD method for calculating dispersion characteristics of waveguide structures is proposed. The numerical stability and numerical dispersion relation of the proposed method are also presented and discussed. Numerical wavelengths for the dominant and higher order modes in a hollow waveguide are obtained from numerical simulations and compared with those from the analytical dispersion relation. The numerical results show that the proposed scheme has the potential to successfully analyze a class of waveguides having locally fine geometry with reduced numerical costs.

Virtual Experimental Characterization of 3D Orthogonal Woven Composite Materials (직교 직물 복합재료 물성치 예측을 위한 가상 수치 실험)

  • In this work, virtual material characterization of 3D orthogonal woven composites is performed to predict the elastic properties by a full scale FEA. To model the complex geometry of 3D orthogonal woven composites, an accurate unit structure is first prepared. The unit structure includes warp yarns, filler yarns, stuffer yams and resin regions and reveals the geometrical characteristics. For this virtual experiments by using finite element analysis, parallel multifrontal solver is utilized and the computed elastic properties are compared to available experimental results and the other analytical results. It is founded that a good agreement between material properties obtained from virtual characterization and experimental results. Using the method of this virtual material characterization, the effects of inconsistent filler yarn distribution on the in-plane shear modulus and filler yarn waviness on the transverse Young's modulus are investigated. Especially, the stiffness knockdown of 3D woven composite structures is simulated by virtual characterization. Considering these results, the virtual material characterization of composite materials can be used for designing the 3D complex composite structures and may supplement the actual experiments.

3D stress-fractional plasticity model for granular soil

  • The present fractional-order plasticity models for granular soil are mainly established under the triaxial compression condition, due to its difficult in analytically solving the fractional differentiation of the third stress invariant, e.g., Lode's angle. To solve this problem, a three dimensional fractional-order elastoplastic model based on the transformed stress method, which does not rely on the analytical solution of the Lode's angle, is proposed. A nonassociated plastic flow rule is derived by conducting the fractional derivative of the yielding function with respect to the stress tensor in the transformed stress space. All the model parameters can be easily determined by using laboratory test. The performance of this 3D model is then verified by simulating multi series of true triaxial test results of rockfill.
