Phenolic Components from the Leaves of Cornus controversa H. (층층나무 잎의 Phenol성 성분 (I))

  • Twelve phenolic components were isolated from the aqueous acetone extract of the leaf of Cornus controversa H. (Cornaceae). On the basis of chemical and spectroscopic evidence, the structures of these components were established as gallic acid, $1-O-galloyl-{\beta}-{_D}-glucose$, $1,6-di-O-galloyl-{\beta}-{_D}-glucose$, $1,2,3-tri-O-galloyl-{\beta}-{_D}-glucose$, $1,2,6-tri-O-galloyl-{\beta}-{_D}-glucose$, 3,4,6-tri-O-galloyl ${_D}-glucose$, eugeniin, gemine D, quercetin, quercitrin, hyperoside and rutin.

Inhibitory Effects of Hydrolysable Tannins on Tyrosinase Activities in B16 Mouse Melanoma Cells

  • To investigate skin whitening natural substances, the effects on melanogenesis by measuring the tyrosinase activity and the melanin contents of three hydrolysable tannins, $1,2,6-tri-O-galloyl-{\beta}-D-glucose$ (1), 2,3-(S)-HHDP-D-glucose (2) and pedunculagin (3) in B16 melanoma cells were examined. $1,2,6-Tri-O-galloyl-{\beta}-D-glucose$ (1), 2,3-(S)-HHDP-D-glucose (2) and pedunculagin (3) inhibited tyrosinase activity in B16 melanoma cells in a dose-dependent manner.

  • Seventy three hydrolysable tannins and related compounds were isolated from seven Euphorbia plants. Among them, 28 compounds including nine gallotannins, eleven ellagitannins and eight related compounds were selected according to the structural similarity. Cytotoxicity of them on the human tumor cell lines including A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT-15 were evaluated by the SRB method in vitro. 3,4,6-Tri-O-galloyl-D-glucose was shown to exhibit most potent cytotoxic effect($4.4{\mu}g/ml).

  • Phytochemical study of the constitutive polyphenolics of the flower aqueous alcohol extract of Tamarix tetragyna was carried out. The new sulphated flavonol, quercetin 3',4'-dimethyl ether $3-O-KSO_3$ as well as the new natural galloyl glucose, $2-O-galloyl-({\alpha}/{\beta})-^4C_1-glucopyranose$ were isolated and characterized. The known sulphated flavonols, kaempferol 7,4-dimethyl ether $3,5-di-O-KSO_3$, quercetin 7-methyl ether $3,3',4'-tri-O-KSO_3$, quercetin 7,4'-dimethyl ether $3-O-KSO_3$ and quercetin $3-O-KSO_3$ and the known sulphated phenolics, isoferulic acid $3-O-KSO_3$ and ellagic acid 4,4'-dimethyl ether $3-O-KSO_3$ were also separated and identified. The structures were established by conventional methods of analysis and confirmed by $^1H-,\;^{13}C-NMR$ and negative ESI-mass spectrometry. 2D-homonuclear chemical shift correlation NMR experiment was applied for the new natural galloylglucose.

  • For the use of tannins in the whitening-effect cosmetics, inhibitory effect against tyrosinase activity was determined. Three condensed tannins including gallocatechin, gallocatechin 3',4'-di-O-gallate and epicatechin 3-O-gallate and three hydrolyzable tannins, 1,2,6-tri-O-galloyl-${\beta}$-D-glucose, 2,3-(S)-HHDP-D-glucose and pedunculagin showed 15-29% mild inhibitory effects against tyrosinase activity.

Anti-inflammatory Effect on RAW 264.7 Cells and Antibacterial Effect on Cutibacterium acnes of Compounds Isolated from Sedum takesimense (섬기린초(Sedum takesimense)에서 분리된 화합물들의 RAW 264.7 Cell에 대한 항염증 효과와 여드름 원인균에 대한 항균 효과)

  • This study investigated anti-inflammatory and antibacterial activities of Sedum takesimense ethanolic extract, and 3 isolated compounds. To confirm anti-inflammatory and anti-acne activities, a nitric oxide (NO) inhibition assay, pro-inflammatory factor (TNF-α, IL-1β, and IL-8) inhibition assays, and minimum inhibitory concentration (MIC) tests were performed. The 3 isolated compounds were identified as 4,6-di-O-galloylarbutin (OGA), 2,4,6-tri-O-galloyl-glucose (OGG), and 1,2,4,6-tetra-O-galloyl-β-glucose (TOGG). The ethanolic extract and isolated compounds (OGA, OGG, TOGG) effectively inhibited production of NO and pro-inflammatory cytokines (TNF-α, IL-1β, and IL-8). Furthermore, OGG and TOGG exhibited MIC values toward Cutibacterium acnes of 12.5 ㎍/mL and 3.2 ㎍/mL, respectively. These results suggest that S. takesimense extract exerts an anti-inflammatory effect on LPS-induced RAW264.7 cells, and an antibacterial efficacy against C. acnes.
