Protective Effect of Combinational Antidotes Composed of Physostigmine and Procyclidine Against Nerve-agent Poisoning

  • Antidotal efficacy of physostigmine plus procyclidine, the combinational prophylactics for organophosphate poisoning, was evaluated in rats and guinea pigs. To assess the dose-response relation-ship in rats, various doses (0.3-6.0mg/kg) of procyclidine in combination with a fixed dose (0.1mg/kg) of physostigmine were pretreated subcutaneously 30 min prior to subcutaneous exposure to nerve-agents. Physostigmine alone exerted protection ratios of 2.44, 1.20, 1.50, 1.50 and 2.20 folds for tabun, sarin, soman, cyclosarin and V-agent, respectively. Interestingly, coadmnistration of procyclidine with physostigmine exhibited remarkable synergistic effects in a dose-dependent manner, leading to 4.00~8.00 folds for tabun, 2.15-8.50 folds for sarin, 1.92~507 folds for so man, 2.15~2.90 folds for cyclosarin, and 2.71~10.50 folds for V-agent. On the contrary, a low effect (l.65 fold) was achieved with the traditional antidotes atropine (17.4 mg/kg) plus 2-pralidoxime (30 mg/kg) treated immediately after soman poisoning. Noteworthy, the combinational prophylactics markedly potentiated the effect of atropine plus 2-pralidoxime to 6.13 and 12.27 folds with 1.0 and 3.0 mg/kg of procyclidine, respectively, against soman poisoning. In guinea pigs, the physostigmine plus procyclidine prophylactics exerted protective effects of 3.00~4.70 folds against soman intoxcation, which were much higher at low doses (0.3~1.0 mg/kg) of procyclidine than those in rats. Taken together, it is proposed that the combinational prophylactics composed oj physostigmine and procyclidine could be a promising antidote regimen for the poisoning with organophosphates possessing diverse properties.

Trace Metal Contents in Cereals, Pulses and Potatoes and Their Safety Evaluations (우리나라 곡류, 두류 및 서류중 중금속 함량 및 안전성 평가)

  • 국내에서 생산된 곡류 9종 416건, 두류 5종 296건 및 서류 2종 156건에 대해 수은 함량은 Mercury Analyzer로, 납, 카드뮴, 비소, 구리, 망간, 아연 등은 습식분해 후 ICP로 분석하였다. 본 연구 결과, 곡류중 중금속 함량[최소~최대(평균),mg/kg]은 다음과 같았다. 수은 0.0001~0.051(0.007), 납 0.01~0.39(0.13), 카드뮴 0.001~0.098(0.023), 비소 0.01~0.38(0.09), 구리 0.06~11.85(1.73), 망간 0.91~39.15(7.47), 아연 1.35~24.15(9.32) mg/kg으로 나타났다. 또한 두류중 중금속 함량(mg/kg)은 수은 0.0002~0.031(0.005), 납 0.01~0.38(0.12), 카드뮴 0.005~0.098(0.030), 비소 0.01~0.37(0.10), 구리 0.03~6.56(2.44), 망간 0.85~22.97(8.16), 아연 2.40~40.18(11.25) mg/kg이었다 서류중 중금속 함량(mg/kg)은 수은 0.002~0.036(0.017), 비소0.01~0.20(0.08), 구리 0.02~2.91(0.84), 망간 0.26~9.48(2.54), 아연 0.35~6.11(2.23)mg/kg이었다. 본 연구에서 얻어진 분석치들은 국내외 다른 연구자들의 분석치와 비슷한 것으로 나타났으며, 이는 우리나라에서 생산된 곡류, 두류, 서류중의 중금속 함유량은 오염된 것이 아닌 자연함유량 수준인 것으로 파악되어 우리나라 사람이 이들 농산물에서 섭취하는 중금속량으로 인한 위해성은 없는 것으로 판단된다. 또한 우리나라 국민이 곡류, 두류, 서류 등을 통해 섭치하는 납, 수은, 카드뮴 등의 중금속 주간섭취량은 FAO/WHO에서 중금속 안전성 평가를 이해 정한 잠정주간섭취혀용량인 PTW1의 0.2~19%를 차지하고 있다.

Environmental Contamination and Bioavailability of Toxic Element around the Daduk Mine Area, Korea (다덕광산 주변지역에서의 독성원소들의 환경오염 및 인체흡수도)

  • In order to investigate the extent and degree of arsenic and heavy metal contamination and the bioavailability of toxic elements around the abandoned mine in Korea, an environmental geochemical survey was undertaken in the Daduk mine. After appropriate preparation, tailings, soil, stream sediment, crop plant and fingernail samples were analysed for As, Cd, Cu, Pb and Zn by ICP-AES and ICP-MS. Elevated levels of 8,782 mg/kg As, 8.3 mg/kg Cd, 489 mg/kg Cu, 3,638 mg/kg Pb and 919 mg/kg Zn were found in tailings from the Daduk mine. These significant concentrations can impact on soils and sediments around the tailing ponds. Mean concentrations of As, Cd, Pb, Cu and Zn in soils are significantly higher than those in world average soil, especially for As and Pb. Element concentrations in sediments decrease with distance from the tailing ponds due to a dilution effect by the mixing of uncontaminated sediments. Arsenic and Cd are elevated in rice grains and stalks, and Cu and Zn concentrations in chinese cabbage, sesame and bean leaves are higher than the upper limit values for normal plant. Arsenic concentration in fingernails of farmers are higher than the normal level with a maximum value of 1.5 mg/kg. The post-ingestion bioavailability of toxic heavy metals in some paddy and farmland soils has been also investigated using the SBET (simple bioavailability extract test) method. The method utilises synthetic leaching fluids closelyanalogous to those of the human stomach. The quantities of As, Cd, Cu, Pb and Zn extracted from paddy soils after 1 hour indicated 15.9, 65.4, 46.2, 39.4 and 29.4% bioavailability, respectively and for farmland soils, 12.4, 26.0, 31.2, 29.3 and 19.4% bioavailability, respectively. The results of the SBET indicate that regular ingestion of soils by the local population could pose a potential health threat due to long-term toxic element exposure.

3-week repeated dose oral toxicity study of Clausena excavata extract in ICR mice (ICR 마우스에서 핑크왐피 추출물의 3주간 반복 투여 독성 연구)

  • Medicinal herb in Asian countries has been traditionally used for a long time. However, the safety and adverse effect of medicinal herb have not been established yet. The aim of this study is to evaluate toxicity in the oral administration of Clausena excavata (C. excavata) in male ICR mice for 3 weeks and the noobserved adverse effect level (NOAEL). C. excavata has been used as a medicinal herb for the treatment of dermatopathy, malaria, abdominal pain, dysentery, and enteritis. C. excavata was orally administered daily for 21 days at a dose of 100, 250, 500, 1000, and 2000 mg/kg/day (MPK). There were no significant differences in mortalities, clinical signs, body weight changes, hematological, and serum biochemistry examination in all animals administrated with C. excavate. Consequently, these findings indicated that C. excavata did not affect the toxic effect in ICR mice and the NOAEL of C. excavata was considered as more than 2000 MPK.

Toxicity Study of Red Ginseng Acidic Polysaccharide (RGAP) : Single and 2-week Repeated Oral Dose Toxicity Study in Rats

  • The present study was conducted to investigate the single and 2-week repeated dose toxicity of red ginseng acidic polysaccharide (RGAP) in Sprague-Dawley rats. The test article was administered orally to rats at dose levels of 0, and 2000 mg/kg/day for single dose toxicity study and at dose levels of 0, 250, 500, and 1000 mg/kg/day for repeated dose toxicity study. In both studies, there were no treatment-related effects on mortality, clinical signs, food and water consumption, ophthalmoscopy, urinalysis, hematology, serum biochemistry, necropsy findings and organ weights of all animals treated RGAP. Based on these results, it was concluded that the 2-week repeated oral dose of RGAP may have no toxic effect in rats at a dose level of 1000 mg/kg/day. In the condition of this study, the no-observed-adverse-effect level (NOAEL) was considered to be 1000 mg/kg/day for both sexes.

A Study on the Distribution of Heavy Metals in Soils and Native Plants of Goggyochun (곡교천 주변의 토양과 자생식물의 중금속분포에 관한 연구)

  • This study was conducted to investigate the distribution pattern of plants on the streamside of Goggyochun, and to survey the distribution of heavy metal-contaminated soils and plants. A total of 11 survey sites were selected along the Goggyochun during the period of between June and September 1999. The wet land plants were composed of 44 species. The most frequently occurring species in the study sites were Rumex crispus, Persicaria hydropiper, Cyperus amuricus and Setaria viridis, and among them the Humulus japonicus, Erigeron canadensis and Phragnites communis species were dominants of the community. Analysis of soil properties showed that organic matter and clay contents were higher in the upstream region while sand content was higher in the downstream region. The pH and the organic matter contents of soils were at range of 6.70-9.94 and 2.39-15.16% respectively. The average contents of Pb and Cd in soils were 11.96mg/kg and 1.64mg/kg respectively, and these values were higher as compared with those of ordinary soil in Korea. The average contents of Zn and Pb in Persicaria hydropiper were 8.30mg/kg and 7.38mg/kg respectively, and these values were higher than other plants. The distribution of heavy metals in plants varied in accordance with the species regardless of the sites.

  • This study was investigated to compare the volatile flavor components in fuji, tsugaru, hongro and jonathan (Malus pumila M.). In the results of analyzing the volatie components of four varieties apples with GC-FID and GC/MS, 100, 68, 85 and 94 volatiles were identified in fuji, tsugaru, hongro and jonathan apples, respectively. Hexanal, butanol, (E)-2-hexenal, hexanol,3-methy-1-butanol, ethy1actate, ethyl butyrate, 2-methybutyl acetate and hexy1 acetate were major flavor components in the four varieties of apple, though there were several differences in composition of colatiles.

Effects of Ethylacetate Fraction of Persimmon Leaves on Experimentally-induced Gastric Mucosal Damage and Gastric Ulcers in Rats

  • The protective effects of the ethylacetate fraction of persimmon leaves(PEF) against experimentally induced gastric mucosal damage and gastric ulcers were evaluated in ratss. In prophylatic study, 100 mg/kg ethylacetate fraction of persimmon leaves (PEFH) exhibited a total protection of 73.8% and 65.7% against HCl-ethanol and 0.2N NaOH-induced gastric mucosal membrane lesions, respectively, which was superior to cimetidine 50 mg/kg, a commonly used anti-ulcer drug. PEFH showed excellent anti-ulcer effects against pylorus ligation induced gastric ulcers, compared to the control group, however, 50 mg/kg ethylacetate fraction of persimmon leaves (PEFL) and PEFH did not affect ulcers induced by water immersion stress, and that is inferior to cimetidine 50 mg/kg. In conclusion, the results suggest that the ethylacetate fraction of persimmon leaves can be used both in prevention and treatment of experimentally induced gastric mucosal damage and ulcers.

  • Previously, we have reported an antitumor efficacy of liposomal N-(phosphon-acetyl)-L-aspartic acid (or PALA) in C-26 tumor bearing Balb/c mice, where PALA in liposome was administered one day after tumor inoculation. In this report, we have investigated the therapeutic potency of liposomal formulation of PALA, which was administered eight days after tumor inoculation in the same C-26 tumor bearing mice. The C-26 murine colon tumor inoculated mice were randomized for the in vivo therapy and the survival was measured after a single intraperitoneal injection of the drug. When the therapy was initiated eight days after tumor inoculation, DSPC-PALA at 150 mg/kg resulted in a significant increase in median survival time (MST) of 56% over the control group which received MES/HEPES buffer alone. However, none of the free PALA and DSPG-PALA liposome doses caused a statistically significant increase in MST over control group at the 95% confidence level. At 750 mg/kg dose, free PALA caused a marginally significant improvement in MST by 34%, but both 375 mg/kg and 150 mg/kg doses of free PALA caused only a 2% and a 4% increase in MST, respectively. These results show that PALA in neutrally charged liposome can exhibit considerably greater potency than free PALA in established C-26 tumor bearing mice.

Single Dose Toxicity Test of Bojungikkeehapdaechilkitang water extract in Male and Female ICR Mice (보중익기합대칠기탕(補中益氣合大七氣湯) 추출물의 ICR마우스에서 경구 단회투여독성 평가)

  • An herbal water extract of Bojungikkeehapdaechilkitang(BDT) was prepared to test it for single-dose and repeated-dose toxicity, genotoxicity and reproductive toxicity, and to obtain a 50% lethal dose$(LD_{50})$, approximated lethal dose(ALD), and approximated target organs for BDT. The extract was tested on female and male ICR mice according to KFDA Guideline 1999-61 at doasge level of 2000, 1000, 500, 250 and 125mg/kg/10mL In this study, clinical signs, mortalities and gross findings of principal organs were observed for 14 days of single dosing, and afterwards in some cases. The ALD and $LD_{50}$ of BDT extract obtained in this study was>2000mg/kg for both male and female ICR mice. Also, any possible digestive toxicity of BDT extract was found to be above 1000mg/kg in both male and female ICR mice. The results of this study strongly suggest that BDT extract has no toxic effect at dosage level below 500mg/kg.
