  • Induction of triploid cherry salmon, Oncorhynchus ma.sou was performed by the heat shock procedure. The triploids were induced by 15 and 20 min heat shock treatments after 10 min fertilization at $28^{\circ}C$. Incidences of the triploidy were $91.4\%\;and\;89.8\%$, respectively. Nucleus and erythrocyte from the induced triploids larger than those of the diploids in major axis, minor axis, major axis/minor axis, surface area and volume. The chromosome number of diploid was 2n= 66 (17 pairs of metacentrics or submetacentrics, 16 pairs of acrocentrics or telocentrics), and that of triploid was 3n=99. Satellites were observed in the short arm of the largest telocentrics, and these may be useful marker to determine the polyploidization. Diploid have outgrown to triploid, and female have also outgrown to male in the triploid group during the 22 months of observation period after hatching. During the spawning season, 22 to 26 months after hatching, diploid were observed the growth retardation because of their sexual maturation, however the triploid showed continuous growth.

  • Growth of triploid abalone, Haliotis discus hannai induced by cody (3$^{\circ}C$) shock and its feed efficiency were investigated from larva to adult for 51 months. After 51 months from triploidy induction, the triploid abalones have outgrown to diploid abalones in shell length and total weight. Triploid abalones with inhibition of extrusion of first polar body (3n-1pb) were outgrown to diploid abalones, however, triploid abalones with inhibition of extrusion of second polar body (3n-2pb) were not significantly different from diploid controls in shell length and total weight through the whole rearing period (P<0.05), because of their heterozygosity differences. Daily feeding rates and feed conversion rates decreased with the growth of abalones and both rates had no differnce between two experimental groups. After 51 months from inducing triploid, conditin index of triploid abalone (64.1%) was higher than that of diploid control (59.4%) (P<0.05). Survival rate was 63.0% in triploid group (3n-1pb 62.0%, 3n-2pb 64.0%) and 62.0% in diploid group during the experimental period.

Maturity and Spawning of the Triploid Pacific Abalone, Haliotis discus hannai (북방전복, Haliotis discus hannai 3배체의 생식능력)

  • The gonadal development of triploid and diploid Pacific abalones, Haliotis discus hannai was histologically investigated in spawning season. Diploid abalones had matured oocytes and spermatozoa, but most triploid had spermatocytes or developing oocytes that was slightly retarded in gonadal development compared to diploid abalones. In spawning experiment of triploid and diploid abalones, spawning rates of diploid male and female were 100%, but those of triploid female was 50% and male was 25% respectively. Investigation of spawned abalone eggs and spermatozoa revealed that length of diploid sperms head were 17.47 ${\mu}m$, breadth of head were 10.31 ${\mu}m$, length of spermatozoa were 130.72 ${\mu}m$, but those of triploid spermatozoa were 11.83 ${\mu}m$, 7.89 ${\mu}m$ and 103.36 ${\mu}m$ respectively. Triploid spermatozoa were significantly small to diploid spermatozoa (p < 0.05). The eggs of diploid and triploid were not different in size. The cross experiment between oocytes produced by triploids and spermatozoa by diploids ($3n{\times}2n$ cross) revealed that no fertilization were occurred, and $2n{\times}3n$ cross also revealed same result.

  • A Study was conducted by using a histological method to examine gonadal development in diploid and triploid mud loach, Misgurnus mizolepis. Shape of ovaries from both groups looked like a single mass however, size of triploid ovary was significantly smaller than that of diploid ovary. Testis of triploid was also similar that of diploid in shape, but it was smaller than that of diploid. Ovarian development and oogenesis in diploid were significantly more rapid than those process in triploid, and first matured eggs were observed 100-days after hatching (SL, 5.68cm). Triploid ovary from hatching to 9-month-old had external appearance of undeveloped gonad and a few of oocytes of perinucleolus stage. However, normal matured eggs like those of female diploid were observed in one 5year-old triploid fish ovary examined. Although testicular development and sperrnatogenesis of diploid were prosperous sexually, testicular development of triploid were rather retarded compare to their male diploid counterpart in spermatogenesis.

  • Morphometrical data for diploid and induced triploid cherry salmon, Oncorhynchus masou were analyzed prior to the spawning season to compare their commercial values. The following traits were measured with the diploids and induced triploids : ungutted and gutted body weight; body length; dressing percentage; condition factor; gonad weight and gonad index; belly thickness; viscera weight; viscera index; body circumference at the pelvic, dorsal and anal fins; area of the cross sections at the pelvic and dorsal fins; two belly thickness traits measured on each cross section; total height, height and width of each cross section; three body shape traits; and nine cross section shape traits. Body length and body weight of diploids were larger than those of induced triploids and dressing percentage, gonad weight, gonad index, viscera index, and liver index were also larger in diploids. However, induced triploids showed higher values in one belly thickness trait and some section shapes than diploids. Differences in body traits were due to the sterility of induced triploids. Therefore, induced triploid cherry salmon appears to have greater potentials for the commercial values than their diploids.

  • In this study, we invesgated a cytogenetic analysis of the Pacific triploid abalone, Haliotis discus hannai induced by low temperature treatment. We got a lot of mitotic metaphase chromosome spreads from the triploid and diploid Pacific abalones' hatched larvae (trochophores). The chromosome number of diploid abalone was 2n = 36 and that of triploid abalone was 3n = 54, so the chromosome number of triploid abalone was 1.5 times higher than that of diploid abalone. We developed a modified flow cytometric method for Pacific abalone from the existing methods. We uesd 51 months aged triploid and diploid Pacific abalones' hemolymph to get the DNA contents by flow cytometry. The DNA content of diploid abalone was 1.743 pg/cell and the DNA content of triploid abalone was 1.49 times higher than that of diploid one. It proved that triploid abalone was consisted with two sets of maternal diploid and one set of paternal genome.

  • Triploid cherry salmon (Oncorhynchus masou) were induced by the heat shocking treatment of fertilized eggs 10 minutes post fertilization at $28^{\circ}C$ for 15 minutes. Haematological and physiological characteristics of diploid and the induced triploid fish were examined just before the spawning period. Triploids had significantly lower concentrations of circulating red blood cells than that of diploids (P<0.01). However, oxygen consumption, hematocrit, hemoglobin, RBC, and mean concentration of hemoglobin within the RBC of triploids were higher than those of diploids. The concentrations of total protein, albumin and cholesterol from triploids were also higher than those from diploids.

  • This study was conducted to examine early gonadogenesis by using a histological method for the appearance of primordial germ cells (PGCs), protrude of genital ridge, and formation of primitive gonads in diploid and triploid mud loach, Misgurnus mizolepis. The pattern of early gonadogenesis including appearance of PGCs, formation of genital ridge, and development of primitive gonad in both diploid and triploid were not different histologically. Characteristics of PGCs of triploid were also the same as those of diploid. However, gonadal length of diploid was significantly longer than that of triploid (P<0.05).

A comparative analysis of cell cycles in diploid and induced triploid tissues in marine medaka(Oryzias dancena) (해산송사리(Oryzias dancena) 2배체와 유도 3배체 조직의 세포주기 비교)

  • The aim of this study was to conduct a comparative analysis of the diploid and induced triploid cell cycles in marine medaka, Oryzias dancena, tissues. The mean fraction of cells in the G1, S, and G2+M phases was 85.5%, 7.6%, and 6.9%, respectively, in the tail fin tissues of diploid fish and 91.2%, 3.6%, and 5.2%, respectively, in those of induced triploid fish. The mean fraction of cells in the G1, S, and G2+M phases were 78.4%, 10.6%, and 11.0%, respectively, in the liver tissues of diploid fish; 86.2%, 5.9%, and 7.9%, respectively, in those of induced triploid; 79.3%, 9.4%, and 11.3%, respectively, in the gill tissues of diploid fish; and 85.7%, 5.4%, and 8.9%, respectively, in the induced triploid fish. The differences among the tissues were statistically significant within both the diploid and induced triploid fish (p<0.05). Mitosis was more active in each tissue of the diploid fish than in the corresponding tissues of the induced triploid fish and mitosis was more active in the liver and gill tissues than in the tail fin tissues in both the diploid and induced triploid marine medaka.

  • Experiments were carried out to examine early life history, growth and gonadal development of triploid rainbow trout to evaluate their suitability for commercial aquaculture. A high degree ($96.5\%$) of triploid fish was induced when the fertilized eggs were treated with heat shock. Mean embryonic development time of triploids was slightly shorter than diploids and survival rate of triploid at swim-up was $80.2\%$ relative to control. Erythrocytes and their nuclei of triploid were larger in both axes than those of diploid. Triploid rainbow trout showed improved growth relative of diploid after 1 year of age. Histological analysis of the gonads revealed sterility in triploid fish.
