The biological removal of 2,4,6-trinitrotoluene (TNT) was studied in a bench-scale bioreactor using a bacterial culture of strain OK-5 originally Isolated from soil samples contaminated with TNT. The TNT was completely removed within 4 days of incubation in a 2.5 L bench-scale bioreactor containing a newly developed medium. The TNT was catabolized in the presence of different supplemented carbons. Only minimal growth was observed in the killed controls and cultures that only received TNT during the incubation period. This catabolism was affected by the concentration ratio of the substrate to the biomass. The addition of various nitrogen sources produced a delayed effect for the TNT degradation. Tween 80 enhanced the degradation of TNT under these conditions. Two metabolic intermediates were detected and identified as 2-amino-4, 6-dinitrotoluene and 4-amino-2, 6-dinitrotoluene based on HPLC and GC-MS analyses, respectively. Strain OK-5 was characterized using the BIOLOG system and fatty acid profile produced by a microbial identification system equipped with a Hewlett Packard HP 5890 II gas chromatograph. As such, the bacterium was identified as a Stenotrophomonas species and designated as Stenotrophomonas sp. OK-5.
Several 2,4,6-trinitrotoluene (TNT) degrading bacteria were isolated from an activated sludge by an enrichment culture technique, and their TNT removal activities were examined. Among the isolates, strain C1 showed the highest degrading capability, and completely removed 100 or 200 mg I$\^$-1/ of TNT within 6 hours of incubation. This bacterium was identified as Klebsiella sp. The effects of different carbon sources on the removal of the parent TNT by Klebsiella sp. C1 were negligible, but the transformation rates of TNT metabolites such as amino-dinitrotoluenes and diamino-nitrotoluenes were higher with fructose addition compared to glucose addition. When nitrate was used as the nitrogen source, the degradation rates of TNT and hydroxylamino-dinitrotoluenes were higher than those with the ammonium addition. Although the TNT removal rate of Klebsiella sp. C1 was slightly higher in anaerobic conditions, the further transformations of TNT metabolites were more favorable in aerobic conditions.
2,4,6-Trinitrotoluene(TNT) was reduced into intermediate products by mixed culture incubated in anaerobic condition. To test the effects of extracellular material to electron transfer between sulfide and TNT, filtered medium of mixed culture was loaded in the test tubes with TNT and sulfide. The transformation rate was measured under four different conditions. The rate under microbial activity was the fastest among under different conditions. With sulfide or filtrate alone and TNT, the reactions were measured as the slowest reactions or no reactions occured, respectively. The reaction rate coefficient were calculated by linear regression and the first order kinetic was fitted best. Also, the plot of rate coefficients (K$_{f}$) showed linear relationships when at time zero TNT and sulfide concentration were 20 mg/1 and 6.0 mM, respectively. By extrapolation, reaction rate coefficient of 100% filtrate could be calculated as 0.0054/minute. However, reaction rate was affected by different concentration of sulfide, so it is a dependent of sulfide concentration. The results of this test showed TNT reduction rate can be limited more by microbial reaction than by mediation of filtrate or sulfide and filtrate alone.
본 연구에서는 폐 2,4,6-트리나이트로톨루엔(2, 4, 6-trinitrotoluene, TNT)의 친환경 연소처리 공정을 개발하고, 이를 이용한 연소조건 최적화를 통한 연소 잔유물 중 유기물 최소화에 대한 기초연구의 데이터를 제시하였다. TNT는 자체적으로 완전연소가 어려운 물질로 외부에서 열을 가해주는 조건인 버너의 가열시간 변화, 추진제(가연물)와 혼합하여 소각, 연소 후 잔유물에 대한 2차 고온처리의 방법으로 실시하였다. 실험결과 잔유물 내 유기물 함량 감소를 확인하였으며, 각 방법별 최소 7 ~ 10%의 유기물 함량을 나타내었다. 최적의 연소시간 조건에서 폐TNT의 연소 잔유물 중 유기물 함량은 1차 소각로만 사용한 경우 9% 수준을 보였으며, 동시에 폐가스의 환경친화성도 폐가스 실시간 분석에서 확인 되었다. 1차 소각이후 소각 잔유물을 외부의 전기로를 이용하여 추가적으로 고온처리 처리할 경우 유기물 함량을 2% 정도 추가적으로 감소시킬 수 있었다. 추진제가 포함된 연소공정에서는 추진제 함량에 따른 잔유물 중 유기물의 함량 감소가 가능함이 나타나 다양한 TNT 폐기물을 처리할 수 있음을 알 수 있었다. 폐기물관리법에서 소각 후 잔유물 내 유기물 함량 15% 미만을 충족하는 수치이다.
Various methods to degrade explosives efficiently in natural soil and water that include trinitrotoluene (TNT) have been studied. In this study, TNT in water was degraded by reduction with palladium (Pd) catalyst impregnated onto alumina (henceforth Pd-Al catalyst) and formic acid. The degradation of TNT was faster when the temperature of water was high, and the initial TNT concentration, pH, and ion concentration in water were low. The amounts of Pd-Al catalyst and formic acid were also important for TNT degradation in water. According to the experimental results, the degradation constant of TNT with unit mass of Pd-Al catalyst was $8.37min^{-1}g^{-1}$. The degradation constant of TNT was higher than the results of previous studies which used zero valent iron. 2,6-diamino-4-nitrotoluene and 2-amino-4,6-dinitrotoluene were detected as by-products of TNT degradation showing that TNT was reduced. The by-products of TNT were also completely degraded after reaction when both Pd-Al catalyst and formic acid existed. Even though there are several challenges of Pd-Al catalyst (e.g., deactivation, poisoning, leaching, etc.), the results of this study show that TNT degradation by Pd-Al catalyst and formic acid is a promising technique to remediate explosive contaminated water and soil.
본 연구에서는 염기성 수산화이온을 이용한 TNT의 분해 특성을 조사하였다. 이를 위해 TNT 처리 시 분광학적인 변화 특성을 관찰하고, pH 영향 및 반응생성물에 대해 정량적으로 조사하였다. 실험결과, pH=12에서 가수분해에 의해 TNT 수용액이 갈색을 띄는 파장 400-600nm 범위 내에서 흡광도가 증가함을 관찰하였다. 수용액 상의 pH=12에서 TNT 가수분해 시 pseudofirst-order 속도상수는 $0.0022min^{-1}$으로 나타났으며, 그 반응속도는 매우 느린 것으로 초기 TNT 농도인 $44{\mu}M$이 약 90% 정도 분해되려면 약 1,047min(17.44hrs)이 소요될 것으로 예상되었다. 반응 생성물로는 아질산이온과 포름산이 주로 생성되며, 기타 미량 성분으로 질산이온, 옥살산 등이 확인되었다.
The photocatalytic treatment of water contaminated with 2,4,6-trinitrotoluene (TNT) was explored in bench-scale experiments in batch mode using a Pyrex tube coated with a thin film of $TiO_2$ located inside a photoreactor. The reactor was aerated by purging it with compressed air before initiating the photocatalytic reaction. The rate of TNT degradation approximated first-order kinetics. The reaction rate constant decreased as the TNT concentration increased from 25 to 100 mg/L, while the first-order kinetics could be modeled using a Langmuir adsorption isotherm. The addition of the organic reductants methanol and EDTA significantly enhanced the rate of TNT degradation, with optimum results in the presence of 20% methanol by volume. EDTA increased the rate of TNT removal by enhancing the role of the reductants.
폭약 2,4,6-trinitrotoluene (TNT)스트레스 조건하에서 토양세균 Pseudomonas sp. HK-6의 세포반응에 대하여 조사하였다. 다양한 농도의 TNT에 노출됨으로서 약 70-kDa DnaK와 60-kDa GroEL의 스트레스 충격단백질 (stress shock proteins, SSPs)이 단떠질이 유도되었다. 이들 SSPs의 존재는 SDS-PAGE과 anti-DnaK와 anti-GroEL monoclonal antibodies를 이용한 Western bolt을 통하여 확인되었다. SSPs은 0.5 mM TNT로 6-12 시간 처리된 세포에서 나타났으며, TNT에 노출 후8시간대 에서 최대의 단백질 유도가 관찰되었다. $30^{\circ}C$에서 $42^{\circ}C$로 열변환충격을 주었을 때의 SSPs는 TNT노출에서와 유사한 유도양상을 보여주었다. TNT에 노출된 Pseudomonas sp. HK-6세포에서 유도된 SSPs의 존재는 배양된 세포의 수용성 단백질 분획에 대하여 2-D PAGE를 통하여 확인되었다. Coomassie brilliant blue R25O로 염색된 젤로부터 pH 3-10 범위에서 약 450 개의 spots이 탐침되었으며, 이들 가운데 12 개의 spots이 TNT 스트레스에 대하여 현저하게 유도되었다. Gel상에서 가장 짙게 나타난 대표적 인 spot에 대한 N-말단 아미노산 서열을 분석한 결과, $^1XXAKDVKFGDSARKKML^17$로서, Pseudomonas putida의 GroEL의 N-말단 아미노산 염기서열인 $^1XXAKDVKFGDSARKKML^17$과 동일한 것으로 분석되었다.
The decomposition of 2,4,6-trinitrotoluene (TNT) and the mass balance of nitrogen (N) species as products were investigated in a UV/H2O2system by varying pH, concentrations of $H_2O_2$, and $O_2$. All experiments were conducted in a semi-batch system employing a 50 mL reaction vessel and a coil-type quartz-tube reactor. In contrast with previous studies employing batch mode, TNT decomposition in the semi-batch mode was proportionally enhanced by increasing $H_2O_2$ concentration to 10 mM (0.034%), indicatingthat an inhibitory effect of excess $H_2O_2$on hydroxyl radical (${\cdot}OH$) can be negligible. N compounds are released as $NO_2^-$ in the early stages of the reaction, but $NO_2^-$ is rapidly oxidized to $NO_3^-$ by means of ${\cdot}OH$. $NH_4^+$ was also detected in this study and showed gradually the increase with increasing reaction time. In this study, $NH_4^+$ production can involve the reduction of nitro group of TNT concurrent with the production of $NO_3^-$. Of the N species originating from TNT decomposition, 12 ~ 72% were inorganic forms (i.e. [$NO_3^-$] + [$NO_2^-$] + [$NH_4^+$]). This result suggests that the large remaining N portions indicate that unidentified N compounds can exist.
금속 철(Fe$^{0}$ )에 의한 TNT의 환원실험을 회분식 반응조를 사용하여 환원 상태에서 연구하였다. 실험 결과, 환원에 의한 TNT의 농도감소는 유사 1차 반응으로 나타났고 반응상수는 반응조에 첨가한 철의 표면적에 선형적으로 비례하였으며, 교반 속도 60 rpm에서의 반응상수는 0.0981$min^{-1}$m$^{-2}$ L로 측정되었다. Triaminotoluene로 추정되는 최종산물이 용액내에 축적되었다. 철 표면에 흡착된 물질을 용출하기 위하여 acetonitrile혹은 물(인산염 완충액, pH=7.0)로 추출을 시도하였으나 용출되는 물질은 극히 미량이었다. 또한, 용액 내에서 생물학적 분해에서의 주요 중간산물인 aminodinitrotoluenes은 검출되지 않았다. 그러므로, 철에 의한 TNT의 환원시 니트로기의 환원이 순차적으로 발생하여 아미노기를 형성하지 않고, 3개의 니트로기에 대하여 동시에 발생하는 것으로 사료된다. 이에 TNT의 환원 모델을 제시하였다.
