Purification and Production Conditions of Antimicrobial Compound from Methylotrophic Actinomycetes MO-16 (Methanol 자화방선균 MO-16으로부터 항균성 물질의 정제 및 생산조건)

  • A methylotrophic actinomycetes strain MO-16, which produce the antimicrobial compound, was isolated from soil and supposed as Amycolatopsis sp. based on taxonomic studies. The cell-free extract of methanol-grown strain MO-16 showed dehydrogenase activity for methanol and formaldehyde when various electron acceptors were added for oxidation. On the other hand, methanol did not affect the production of antimicrobial compounds, and organic nitrogen sources such as corn steep liquor and peptone were better than inorganic nitrogen sources. These compounds showed broad antimicrobial spectrum to the tested strains such as bacteria and yeast. The antimicrobial comounds were very stable under heat(121$^{\circ}C$), acid(pH2.0), alkali(pH11.0) treatments. These compounds were isolated by ethylacetate extract, silica gel column chromatography and reverse phase HPLC. Two compounds(peak 1 and 2) were detected as antimicrobial compounds through the HPLC analysis. The peak 2 was purified as a single compound and revealed a 98% purity.

Expression of cytokeratin 10, 16 and 17 as biomarkers differentiating odontogenic keratocysts from dentigerous cysts

  • Objectives: Odontogenic keratocysts (OKCs) have a tendency to recur and possess an aggressive nature. the aim of the present study was to evaluate cytokeratin (CK) expression patterns as a method for the differentiation between dentigerous cysts (DCs) and OKCs, as their histomorphologic appearance are often indistinguishable. Materials and Methods: Formalin-fixed, paraffin-embedded tissue sections of 43 OKCs and 38 DCs were immunohistochemically analyzed with i-solution in a quantitative manner in order to evaluate the immunoreactivity of CK 10, 16 and 17. Results: CK 10 expression was evident in 79.1% of OKCs but found in only 18.4% of DCs (P<0.05), and CK 10 expression was observed to occur more frequently in OKCs (mean 25.45%) than in DCs (2.19%) (P<0.05). The expression of CK 16 was evident in 79.1% of OKCs but found in only 7.9% of the DCs (P<0.05) and CK 16 expression was observed to occur more frequently in OKCs (mean 4.33%) than in the DCs (0.61%) (P<0.05). The expression of CK 17 was evident in 88.4% of OKCs but seen in only 15.7% of the DCs (P<0.05) and CK 17 expression was observed to occur more frequently in OKCs (mean 31.11%) than in the DCs (2.37%) (P<0.05). Conclusion: The immunohistochemical detection of CK 10, 16 and 17 can be utilized as a valuable biomarker for use in distinguishing between OKCs and DCs, which have clinically significant differential diagnoses.

The Effects of College Life Adaptation Program on Nursing Students College Life Adaptation and Nursing professional attitude (대학신입생의 대학생활적응 프로그램이 간호대학생의 대학생활적응, 간호전문직 태도에 미치는 효과)

  • The purpose of this study was to investigate the effects of college life adaptation program on nursing students' college life adaptation and nursing professional attitude. College life adaptation program of a freshman at college which consists of two area : theory-practice and liberal-cultivation of character and college freshman for 70 hours, 5 a week in 10 session for 2 weeks. The collected data were analyzed using paired t-test, ANOVA. As a result of the study, the score for college life adaptation(t=11.42, p=<.000) and nursing professional attitude(t=12.45, p=<.000) increased significantly after college life adaptation program. The conclusion, this study showed that college life adaptation program was effective in improving nursing students' college life adaptation and nursing professional attitude.

Correlation between Microvascular Density and Matrix Metalloproteinase 11 Expression in Prostate Cancer Tissues: a Preliminary Study in Thailand

  • Background: Prostate cancer is a major concern of public health. Microvascular density (MVD) is one of the prognostic markers for various solid cancers. Matrix metalloproteinase 11 (MMP11) plays an important role in angiogenesis and changes in its expression level are known to be associated with tumor progression and clinical outcome. Aim: To investigate the relationship between MVD and MMP11 expression in prostatic adenocarcinoma tissues. Materials and Methods: The expression levels of MMP11 and MVD were analyzed immunohistochemically for 50 specimens of prostatic adenocarcinoma. Results: MMP11 was mainly expressed in stromal cells but rarely seen in epithelial cells. Mean MVD was $36/mm^2$, and it was correlated significantly only with bone metastases. MVD was also significantly correlated with MMP11 expression (r=0.29, p=0.044). Conclusions: MMP11 may alter the stromal microenvironment of prostate cancer to stimulate tumor angiogenesis.

  • The stability constant of the complex between salicylic acid($H_2A$) and Fe(III) ion has been determined using UV-Vis absorption spectrometry and pH titration method in O.1M $KNO_3$ aqueous solution at $20^{\circ}C$ and compared with reference value, $logK_f=16.48$. The $pK_{a1}$ and $pK_{a2}$ of $H_2A$ are $2.92{\pm}0.08$ and $12.90{\pm}0.13$, respectively and the $logK_f$ of 1:1 complex Fe(III)-$H_2A$ system is $11.88{\pm}0.12$ at $20^{\circ}C$.

Production of Biopolymer Flocculant by Bacillus subtilis TB11

  • A microbial flocculant-producing gram-positive bacterium, strain TE11, was isolated from soil samples, and was identified as Bacillus subtilis by using the Midi system, the Biolog system, 16S rDNA sequence analysis, and some physiological and morphological characteristics. The maximum flocculant capsular biopolymer of TE11 strain (BCP, 4.9mg/ml) was obtained when it was grown in GA broth medium containing 3% glutamic acid, 2% glycerol, 0.5% citric acid, 0.5% $NH_4$Cl, 0.05% $MgSO_4.7H_2O,\; 0.05%\;K_2HPO_4\;,\; and\; 0.004%\; FeC1_3. 6H_2O,\; pH 7.2,\; at\; 30^{\circ}C$ for 70 h with shaking. When glycerol was used as an additional carbon source in the GA medium, TE11 produced only flocculant BCP without any by-product. The flocculant (BCP) was found to aggregate suspended kaolin and activated charcoal powder without cations, and its flocculating activity was significantly enhanced by the addition of bivalent cations such as $Ca^{2+}.Zn^{2},\; and\; Mn^{2+}$. The flocculation activity by addition of $Ca^{2+}$ was high in an acidic pH 4.0. In the case of $Zn^{2+}$, high flocculating activity remained without significant loss in the broad range of pH 4.0 to 9.0.

  • Infection of the uterine cervix by human papilloma viruses (HPV) may be associated with cervical pre-cancer and invasive cervical carcinoma if left untreated. With advance in molecular techniques, it has become easier to detect the resence of HPV DNA long before the appearance of any lesion. This study concerned cervical scrape samples of 310 married non-pregnant women attending a gynecology outpatient department for both Pap and PCR testing to detect HPV DNA. Nested PCR using primers for L1 consensus gene with My9/My11 and GP6+/GP5+followed by multiplex PCR were carried out to detect HPV 16 and HPV18. Result: HPV prevalence was 11.9% out of which 3.67% cases of negative for intra-epithelial lesion or malignancy (NILM) and in 71.1% (27/38) of atypical cervical smears were HPV positive. There was increasing trend of high-risk-HPV positivity (HR HPV 16 and 18), from 20% in benign cytology (NILM) to 42.9 % in LSIL, 71.41% in HSIL and 100% in SCC. There was highly significant association of HPV infection with cervical lesion ($x^2=144.0$, p<0.01) and also with type specific HPV prevalence ($x^2=7.761^*$, p<0.05).

Enhanced Biomass Productivity of Freshwater microalga, Parachlorella kessleri for Fixation of Atmospheric CO2 Using Optimal Culture Conditions (최적 배양 조건을 이용한 CO2 제거 목적의 담수 미세조류 Parachlorella kessleri의 바이오매스 생산성 향상)

  • This study attempted to improve the growth of the freshwater microalgae, Parachlorella kessleri, through the sequential optimization of culture conditions. This attempt aimed to enhance the microalgae's ability to fixate atmospheric CO2. Culture temperature and light intensity appropriate for microalgal growth were scanned using a high-throughput photobioreactor system. The supplied air flow rate varied from 0.05 to 0.3 vvm, and its effect on the growth rate of P. kessleri was determined. Next, sodium phosphate buffer was added to the culture medium (BG11) to enhance CO2 fixation by increasing the availability of CO2(HCO3-) in the culture medium. The results indicated that optimal culture temperature and light intensity were 20℃-25℃ and 300 μE/m2/s, respectively. Growth rates of P. kessleri under various air flow rates highly depended on the increase of the culture's flow rate and pH which determines CO2 availability. Adding sodium phosphate buffer to BG11 to maintain a constant neutral pH (7.0) improved microalgal growth compared to control conditions (BG11 without sodium phosphate). These results indicate that the CO2 fixation rate in the air could be enhanced via the sequential optimization of microalgal culture conditions.

Growth characteristics and distribution pattern of a brackish water clam, Corbicula japonica along an estuarine salinity gradient in Seomjin River (섬진강 하구역에서 염분구배에 따른 일본재첩의 분포와 성장특성)

  • The purpose of this study was to determine the growth characteristics and distribution pattern of a brackish water clam Corbicula japonica in Seomjin River. Field samples were taken from 14 stations with salinity gradients during spring. Salinity at the bottom layer ranged from 1.0 psu to 32.9 psu, with low salinities in the upper area of the river. In particular, salinity at St.11 was decreased drastically to be ca. 15.0 psu, indicating an intermediate salinity zone. The distribution pattern of C. japonica was related to the salinity gradient, with the highest densities of $2,102ind.m^{-2}$ at Station 13, followed by $1,507ind.m^{-2}$ at Station 11. Here, we focused on the growth characteristics of collected C. japonica collected at two stations with different salinity values. The relationship between shell length and total weight was highly correlated ($R^2=0.91$, P<0.001) at Station 13 compared to that at Station 11 ($R^2=0.72$, P<0.001). On the other hands, the degree of correlation between shell length and shell height (SH) or shell width (SW) at Station 11 (SH: $R^2=0.91$, P<0.001; SW: $R^2=0.69$, P<0.001) was higher than that at Station 13 (SH: $R^2=0.64$, P<0.001; SW: $R^2=0.48$, P<0.001). In addition, fatness index of C. japonica at Station 13 was significantly (P < 0.001) higher than that at St. 11 (t-test value=-22.8, p<0.001). This implies that C. japonica at Station 13 might have enhanced their somatic growth, whereas C. japonica at Station 11 might have this kind of defense mechanism their internal organization against the salinity stress. Ecologically, this kind of defense mechanism of C. japonica against salinity flucuation may play an important role in their survival strategy.

Correlation Between the Prolongation of P300 Latency during Sleep Deprivation and Personality Variables (수면박탈에 의한 P300 잠복기 연장과 성격요소와의 연관성)

  • Objectives: The purpose of this study is to investigate the relationship between the extent of prolongation in P300 latency by sleep deprivation and personality variables. Methods: Eighteen healthy male college students participated in this study. Subjects remained awake for 37 hours under continuous surveillance. In the morning and evening of two consecutive study days, P300 was checked four times. MMPI and STAI-T were checked in the morning of the first day. The 18 subjects were divided into two groups according to their extent of P300 prolongation by sleep deprivation: Group A consisted of short P300 latency prolonged subjects, and group B of long latency prolonged subjects. The MMPI profiles and STAI-T scores of these two groups were compared. Results: Group B showed significantly higher scores in the Mf subscale (t=-2.16, df=16, p=0.046) and Pa subscale (t=-2.61, df=16, p=0.019) than group A. Group B also showed higher F subscale scores at a marginally significant level (t=-2.11, df=16, p=0.052). Conclusion: These results suggest that subjects with higher scores in F, Mf, and Pa subscales tend to have delayed cognitive process and decreased efficiency of mental process by sleep deprivation. It can be hypothesized that individuals who are sensitive, passive, dependent, and easily projecting are susceptible to the deterioration of cognitive function by total sleep deprivation.
