지황 (地黃)의 성분연구 (Phytochemical Studies on Rehmanniae Radix)

  • Twenty-four compounds were isolated from the 70% ethanolic extract of Rehmanniae Radix (Scrophulariaceae) cultivated in Ubo-myeon, Gunwi-gun, Gyeongbuk province, Korea and their structures were identified as four iridoids [6-O-(4''-O-${\alpha}$-L-rhamnopyranosyl) vanilloyl ajugol (17), ajugol (18), aucubin (19), and catalpol (20)], three phenethyl alcohol glycosides [decaffeoyl acteoside (15), isoacteoside (16), and acteoside (21)], five sugar derivatives [ethyl ${\beta}$-D-fructofuranoside (7), eleutheroside C (14), mannitol (22), raffinose (23), and stachyose (24)], two terpenoids [remophilanetriol (4) and glutinolic acid (11)], a lignan, paulownin (2), and eight others [${\beta}$-sitosterol (1), daucosterol (6), monopalmitin (3), pinellic acid (9), uracil (5), adenosine (12), jio-cerebroside (10), aeginetic acid 5-O-${\beta}$-D-quinovoside (8), aeginetoyl ajugol 5''-O-${\beta}$-D-quinovoside (13)]. The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. Among these compounds, paulownin (2), monopalmitin (3), uracil (5), daucosterol (6), ethyl ${\beta}$-D-fructo-furanoside (7), and eleutheroside C (14) were isolated from this plant for the first time.

Fundamental approach to development of plastic scintillator system for in situ groundwater beta monitoring

  • The performance of a plastic scintillator for use in an in situ measurement system was analyzed using simulation and experimental methods. The experimental results of four major pure beta-emitting radionuclides, namely $^3H$, $^{14}C$, $^{32}P$, and $^{90}Sr/^{90}Y$, were compared with those obtained using a Monte Carlo N-particle (MCNP) code simulation. The MCNP simulation and experimental results demonstrated good agreement for $^{32}P$ and $^{90}Sr/^{90}Y$, with a relative difference of 1.95% and 0.43% between experimental and simulation efficiencies for $^{32}P$ and $^{90}Sr/^{90}Y$, respectively. However, owing to the short range of beta particles in water, the efficiency for $^{14}C$ was extremely low, and $^3H$ could not be detected. To directly measure the low-energy beta radionuclides considering their short range, a system where the source could flow directly to the scintillator was developed. The optimal thickness of the plastic scintillator was determined based on the suggested diameter. Results showed that the detection efficiency decreases with an increase in the depth of the water. The detection efficiency decreased drastically to approximately 10 cm, and the tendency was gradually constant.

  • Electrical properties of the $\alpha$-In$_2$Se$_3$ single crystals grown by use of the Bridgman technique were examined in the transition temperature range between $\alpha$-phase and $\beta$-phase. $\alpha$-In$_2$Se$_3$ single crystal has ' the rhombohedral structure and lattice constants are a=4.025 $\AA$, c=28.771 $\AA$ in c-axis. The transition temperatures of the stoichiometric $\alpha$-In$_2$Se$_3$ single crystal is 198.8$^{\circ}C$ according to the specimens. The temperature of $\alpha$longrightarrow$\beta$ phase transition decreased but the temperature of $\beta$longrightarrow$\alpha$ phase transition increased as the number of heating-cooling cycle increased.

  • 본 연구는 구급대원의 스트레스대응과 현장충격스트레스가 신체적 증상에 어떤 영향을 주는지를 증명하고자 하였다. 소방공무원 970명을 대상으로 설문 조사를 하였고, 자료수집은 2007년 1월부터 12월까지 실시하였으며, 자료분석은 SPSS 14.0과 AMOS 7.0 통계패키지를 사용하였다. 스트레스 대응이 신체적 증상에 미치는 영향을 분석한 결과, 스트레스 대응 1(p<.001), 4(p<.01), 2(p<.05) 순서로 신체적 증상 1에 유의한 영향을 주는 것으로 나타났다. 스트레스 대응 1(${\beta}$=.302), 4(${\beta}$=121)이 높을수록, 스트레스 대응 2(${\beta}$=-.070)이 낮을수록 신체적 증상1이 높아지는 것으로 나타났으며, 스트레스 대응이 신체적 증상 1을 설명하는 설명력의 정도는 16.5%이었다. 스트레스 대응 1(p<.001), 3(p<.001), 4(p<.001) 순서로 신체적 증상 2에 유의한 영향을 주는 것으로 나타났다. 스트레스 대응 1(${\beta}$=.261), 3(${\beta}$=129), 4(${\beta}$=129)가 높을수록 신체적 증상2가 높아지는 것으로 나타났으며, 스트레스 대응이 신체적 증상2를 설명하는 설명력의 정도는 16.3%이었다. 스트레스 대응 1(p<.001), 4(p<.001), 2(p<.05) 순서로 신체적 증상3에 유의한 영향을 주는 것으로 나타났다. 스트레스 대응 1(${\beta}$=.238), 4(${\beta}$=156)이 높을수록, 스트레스 대응 2(${\beta}$=-.081)이 낮을수록 신체적 증상 3이 높아지는 것으로 나타났으며, 스트레스 대응이 신체적 증상 3을 설명하는 설명력의 정도는 13.1%이었다.

  • Column chromatographic separation of the MeOH extract from the roots of Dicentra spectabilis yielded fourteen compounds, menisdaurin (1), menisdaurilide (2), trans-N-p-coumaroyltyramine (3), trans-N-p-feruloyltyramine (4), 4-O-feruloylquinicacid (5), chlorogenic acid (6), 3-O-feruloylquinicacid (7), ferulic acid (8), protopine (9), Kaempferol 3,7-di-O-${\beta}-{\text\tiny{D}}$-glucopyranoside (10), kaempferol 3-O-${\beta}-{\text\tiny{D}}$-glucopyranosyl-7-O-${\alpha}-{\text\tiny{L}}$-rhamnopyranoside (11), ${\alpha}-rhamnoisorobin$ (12), astragalin (13), and nicotiflorin (14). Their structures were determined on the basis of NMR spectroscopic data. Among them, compound 1, 3-8, and 10-14 isolated from this plant were reported for the first time. The isolated compounds (1-14) were tested for nitric oxide (NO) inhibitory activity on lipopolysaccharide-stimulated RAW 264.7 cells. Compound 3, 4 and 12 significantly inhibited NO production. Moreover, Compound 3 suppressed pro-inflammatory cytokines ($TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) in a dose- dependent manner. These data suggest that compound 3 possess anti-inflammatory activity and might be useful natural materials for development of anti-inflammatory agent.

  • 식용식물자원으로부터 활성소재를 찾기 위하여 흰씀바귀 뿌리를 80% MeOH로 추출하고, 얻어진 추출물을 EtOAc, n-BuOH및 $H_2O$로 용매 분획하였다. EtOAc와 n-BuOH 분획에 대하여 column chromatography를 반복하여 4종의 화합물을 분리하였다. 각각에 대하여 2D-NMR을 포함한 스펙트럼 데이터의 해석과 문헌 자료를 조사하여 zaluzanin C (1), $9{\alpha}-hydroxyguaian-4(l5),10(14),11(13)-triene-6,12-olide$ (2), $3{\beta}-O-{\beta}-D-glucopyranosyl-8{\beta}-hydroxyguaian-4(15),\;10(14 )-diene-6,12-olide$ (3), $3-O-{\beta}- D-glucopyranosyl-8{\beta}hydroxyguaian-10(14)-ene-6,12-olide$ (4)로 구조를 결정하였다. 이들 화합물에 대하여 ACAT(Acyl-CoA: cholesterol acyltransferase), DGAT (diacylglycerol acyltransferase) 및 FPTase(farnesyl-protein transferase)의 활성에 미치는 억제효과를 측정하였다. Compound 1과 Compound 2는 DGAT에 대한 활성억제효과에 있어서 $IC_{50}$ 값이 각각 0.13 mM, 0.10 mM로 나타났고, FPTase에 대하여는 각각 0.15 mM, 0.18 mM로 나타났으며, ACAT에 대하여는 약한 억제 활성을 나타냈다. 따라서 흰씀바귀는 항암 및 항고혈압의 소재 개발에 있어서 유용한 자원으로 활용될 수 있을 것이다.

화랑곡나방(Plodia interpunctella Hubner) Carboxylesterase-III의 정제 및 생화학적 특성 (Purification and Biochemical Characterization of Carboxylesterase-III from Plodia interpunctella Hubner)

  • Purification and biochemical experiments on the carboxylesterases-III (CE-III) from the indian meal moth, Plodia interpunctella (Hubner) were carried out to understand their enzymemological characteristics. The CE-III from the fifth instar larvae was purified by means of ammonium sulfate fractionation, gel permeation choromatography and ion exchange choromatography. The optimal temperature for the reaction of the CE-III on the 4 substrates ($\alpha$-Na, $\alpha$-Nb, $\beta$-Na and $\beta$-Nb) was confirmed at 4$0^{\circ}C$. The optimal pH for the reactions on the substrates $\alpha$-Na and $\alpha$-Nb was 7.5. But the optimal pH on the substrate $\beta$-Na and $\beta$-Nb was 8.0. The optimal substrate concentration for the reactions of the CE-III was 3.16 X 10$^{-3}$ M in $\alpha$-Na and $\beta$-Nb. On the substrate $\beta$-Na and $\alpha$-Nb, the optimal substrate concentration was 1.0 X 10$^{-3}$ M for CE-III. The $V_{max}$ and $K_{m}$ values of the carboxylesterases were varied by the substrates as followings: the $V_{max}$ of CE-III was 45.9 for $\alpha$-Na, 52.6 for $\beta$-Na, 36.4 for $\alpha$-Nb, and 83.3 ($\mu$ mol/min/mg protein) for $\beta$-Nb. The $K_{m}$ of CE-III was 1.43 X 10$^{-4}$ M for $\alpha$-Na, 3.57 x 10$^{-5}$ M for $\beta$-Na, 9.17 X 10$^{-5}$ M for $\alpha$-Nb, and 7.14 X 10$^{-5}$ M for $\beta$ -Nb, respectively. The CE-III seemed to have somewhat high thermostability considering that the temperature for effective denaturation on activity was about 5$0^{\circ}C$ ~ 6$0^{\circ}C$.EX>.EX>.

착색시간에 따른 칼라렌즈 색좌표 이동 (Color lens CIE L*a*b* coordination transfer by tinted time)

  • 5분 간격으로 착색시간을 증가시켜 R, G, Y, B칼라렌즈를 제작하였다. 색 좌표 분석은 분광측색계와 CIE $L^*a^*b^*$ color system를 사용하였다. 착색 시간에 따른 색좌표 이동 형태는 $b^*(Y-B)={\beta}a^*+{\alpha}$이며, 거의 선형적으로 변하였다. 적색의 경우 착색시간이 길수록 $G{\rightarrow}R$, $B{\rightarrow}Y$ 형태로 이동하고, Parameter ${\alpha}$, ${\beta}$ 값은 각각 -3.49, 0.90을 얻었다. 녹색의 경우 색좌표 이동은 $R{\rightarrow}G$, $B{\rightarrow}Y$ 형태이다. 10분의 착색 시간에서 가장 순수한 녹색 칼라를 얻었다. Parameter ${\alpha}$, ${\beta}$값은 각각 -0.72, -0.55을 얻었다. 황색과 청색의 경우 모두 착색 시간이 길수록 순수한 색이 된다. 황색에서 Parameter ${\alpha}$, ${\beta}$값은 각각 14.11, 1.58이고, 청색에서 Parameter ${\alpha}$, ${\beta}$값은 각각 -11.62, 1.30을 얻었다.

Anti-oxidative and Antibacterial Constituents from Sedum hybridum

  • Phytochemical studies on the whole extract of Sedum hybridum L., a Mongolian medicinal plant, has been undertaken to isolate active principles responsible for its anti-oxidative and antibacterial activities. Eighteen known compounds, i.e. (1) quercetin, (2) kaempferol, (3) herbacetin-8-O-${\beta}$-D-xylopyranoside, (4) myricetin, (5) gossypetin-8-O-${\beta}$-D-xylopyranoside, (6) gallic acid, (7) 2,4,6-tri-O-galloyl-D-glucopyranose, (8) 6-O-galloylarbutin, (9) myricetin-3-O-${\alpha}$-L-arabinofuranoside, (10) quercetin-3-O-${\alpha}$-L-arabinofuranoside, (11) caffeic acid, (12) ethylgallate, (13) (-) epigallocatechin-3-O-gallate, (14) palmitic acid, (15) stearic acid, (16) stearic acid ethyl ether, (17) ${\beta}$-sitosterol and (18) ${\beta}$-sitosteryl-O-${\beta}$-D-glucopyranose have been isolated and their molecular structures identified by spectroscopic analysis. Thirteen substances including seven flavonol components (1, 2, 3, 4, 5, 9 and 10), five gallic acid derivatives (6, 7, 8, 12 and 13) and caffeic acid (11) exhibited significant, dose-dependent, DPPH radical scavenging activity. Galloyl esters 12 and 13 were revealed to be main active principles for the antibacterial property of the extract of Sedum hybridum L.
