Overall Survival and Clinicopathological Characteristics of Patients with Breast Cancer in Relation to the Expression Pattern of HER-2, IL-6, TNF-α and TGF-β1

  • The present study was conducted to investigate the prognostic significance of co-expression patterna of HER-2, IL-6, TNF-a and TGF-${\beta}1$ in breast cancer, by correlating the number of markers with positive expression with clinicopathological characteristics indicative of tumor progression and overall survival. One hundred thirty consecutive patients with primary breast cancer were prospectively included and evaluated. Serum concentrations of the above markers were measured by ELISA. Median split was used to subdivide patients with marker positive or negative expression. The presence of ${\geq}3$ positive markers was independently associated with extended lymph node (>3) involvement (aOR, 11.94, p=0.001) and lymphovascular invasion (aOR, 12.04, p=0.018), increasing the prognostic significance of each marker considered separately. Additional prognostic information regarding survival was also provided; as the number of positive markers increased, a gradually reduction of survival time was observed. In addition, patients with 4 positive markers had significantly shorter survival (25 vs 39 months, p=0.006) and a more than 4 fold increased risk of death (aHR, 4.35, p=0.003) compared to patients with 3 positive markers. Our findings suggest that the coexpression pattern of these four markers could be used clinically as a useful marker for tumor extension and outcome of breast cancer.

Predictors of Drug Calculation Competence of Nursing Students (간호 대학생의 약물계산역량에 영향을 미치는 요인)

  • Purpose: The objective of this study was to identify predictors of drug calculation competence of nursing students. Methods: A total of 120 students were recruited from 3 universities from November 10 to 20, 2011. The instruments for this study were drug calculation competence, self-efficacy for drug dosage calculation, anxiety for drug dosage calculation, and the academic self-efficacy scale. The data were analyzed by descriptive analysis, chi-square test, t-test, Scheffe test, partial correlation coefficients, and stepwise multiple regression using the SPSS 18.0 program. Results: The mean score of good competence group was $0.67{\pm}0.08$ and the mean score of no-good competence group was $0.42{\pm}0.10$. The drug calculation competence was positively related to self-efficacy for drug dosage calculation and academic self-efficacy scale, but negatively related to anxiety for drug dosage calculation after controlling personal attributes. The main predictors of drug calculation competence in nursing students were identified as anxiety for drug dosage calculation (${\beta}$=-.25, p=.046), academic self-efficacy (${\beta}$=.19, p=.035). These two factors explained about 10% of variance in drug calculation competence. Conclusion: Based on the results, the strategies reducing the anxiety for drug dosage calculation and improving the academic self-efficacy should be developed and implemented.

Volatile Oil Composition of Boxthorn (Lycium chinense M.) Leaves

  • Volatile components were extracted from leaves of two Boxthorn (Lycium chinense M.) cultivars by using simultaneous distillation and extraction, analyzed by gas chromatography-mass spectrometry. Seventy components were identified : 13 acids, 15 alcohols, 18 hydrocarbons, 13 carbonyls, three esters, three ionones, and five others. The principal volatile components (and their peak area percentage) were n-pentanol (11.2~30.2%), phytol (14.5~28.3%), hexadecanoic acid (13.5~17.1%) 2,3-dihydrobenzofuran (1.5~4.2%), benzyl alcohol (1.9-4.8%), phenylacetaldehyde (1.8~3.2%), and octadecadienoic acid (1.7~10.7%). Fresh leaves showed much higher peak area than that of dried leaf in n-pentanol, n-hexanol, cis-2-penten-l-ol, cis-3-hexen-l-ol, benzyl alcohol, and $\beta$-phenylethyl alcohol, while dried leaves showed much higher content than that of fresh leaves in 9-hydroxytheaspran A, octadecanoic acid and octadecadienic acid.

Cytotoxic Terpene Hydroperoxides from the Aerial Parts of Aster spathulifolius

  • Three new sesquiterpene hydroperoxides, 1-[3-(2-hydroperoxy-3-methylbut-3-en)-4-hydroxyphenyl]ethanone (2), $7{\beta}-hydroperoxy-eudesma-11-en-4-ol$ (3), and $7{\alpha}-hydroperoxymanool$ (4), together with three known compounds, germacrone (1), ent-germacra-4(15),5,10(14)-trien-$1{\alpha}$-ol (5) and teucdiol A (6) were isolated from the aerial parts of Aster spathulifolius (Compositae). Their structures were characterized using chemical and spectroscopic methods. The isolated compounds were tested for their cytotoxicity against five human tumor cell lines in vitro using a SRB method. The two new compounds, 3 and 4, showed moderate cytotoxicity against human cancer cells with $ED_{50}$ values ranging from 0.24 to $13.27\;{\mu}g/mL$.

  • Purpose: Lingual nerve (LN) damage may be caused by either tumor resection or injury such as wisdom tooth extraction, Although autologous nerve graft is sometimes used to repair the damaged nerve, it has the disadvantage of necessity of another operation for nerve harvesting. Moreover, the results of nerve grafting is not satisfactory. The nerve growth factor (NGF) is well-known to play a critical role in peripheral nerve regeneration and its local delivery to the injured nerve has been continuously tried to enhance nerve regeneration. However, its application has limitations like repeated administration due to short half life of 30 minutes and an in vivo delivery model must allow for direct and local delivery. The aim of this study was to construct a well-functioning $rhNGF-{\beta}$ adenovirus for the ultimate development of improved method to promote peripheral nerve regeneration with enhanced and extended secretion of hNGF from the injured nerve by injecting $rhNGF-{\beta}$ gene directly into crush-injured LN in rat model. Materials and Methods: $hNGF-{\beta}$ gene was prepared from fetal brain cDNA library and cloned into E1/E3 deleted adenoviral vector which contains green fluorescence protein (GFP) gene as a reporter. After large scale production and purification of $rhNGF-{\beta}$ adenovirus, transfection efficiency and its expression at various cells (primary cultured Schwann cells, HEK293 cells, Schwann cell lines, NIH3T3 and CRH cells) were evaluated by fluorescent microscopy, RT-PCR, ELISA, immunocytochemistry. Furthermore, the function of rhNGF-beta, which was secreted from various cells infected with $rhNGF-{\beta}$ adenovirus, was evaluated using neuritogenesis of PC-12 cells. For in vivo evaluation of efficacy of $rhNGF-{\beta}$ adenovirus, the LNs of 8-week old rats were exposed and crush-injured with a small hemostat for 10 seconds. After the injury, $rhNGF-{\beta}$ adenovirus($2{\mu}l,\;1.5{\times}10^{11}pfu$) or saline was administered into the crushed site in the experimental (n=24) and the control group (n=24), respectively. Sham operation of another group of rats (n=9) was performed without administration of either saline or adenovirus. The taste recovery and the change of fungiform papilla were studied at 1, 2, 3 and 4 weeks. Each of the 6 animals was tested with different solutions (0.1M NaCl, 0.1M sucrose, 0.01M QHCl, or 0.01M HCl) by two-bottle test paradigm and the number of papilla was counted using SEM picture of tongue dorsum. LN was explored at the same interval as taste study and evaluated electro-physiologically (peak voltage and nerve conduction velocity) and histomorphometrically (axon count, myelin thickness). Results: The recombinant adenovirus vector carrying $rhNGF-{\beta}$ was constructed and confirmed by restriction endonuclease analysis and DNA sequence analysis. GFP expression was observed in 90% of $rhNGF-{\beta}$ adenovirus infected cells compared with uninfected cells. Total mRNA isolated from $rhNGF-{\beta}$ adenovirus infected cells showed strong RT-PCR band, however uninfected or LacZ recombinant adenovirus infected cells did not. NGF quantification by ELISA showed a maximal release of $18865.4{\pm}310.9pg/ml$ NGF at the 4th day and stably continued till 14 days by $rhNGF-{\beta}$ adenovirus infected Schwann cells. PC-12 cells exposed to media with $rhNGF-{\beta}$ adenovirus infected Schwann cell revealed at the same level of neurite-extension as the commercial NGF did. $rhNGF-{\beta}$ adenovirus injected experimental groups in comparison to the control group exhibited different taste preference ratio. Salty, sweet and sour taste preference ratio were significantly different after 2 weeks from the beginning of the experiment, which were similar to the sham group, but not to the control group.

  • Concern for the effects of toxic chemicals on the environment leads the search for better bioassay test organisms and test procedures. Photobacterium phosphoreum was used successfully as a test organism and the luminometer detection technique was an effective and simple method for determining the concentration of toxic chemicals. With EC50 a total of 14 chlorine substituted phenols benzenes and ethanes were used for the experiments. The test results showed that the toxicity to P. phosphoreum increased in the order of phenol > benzene > ethane and the toxicity also increased with the number of chlorine substitution. Quantitative structure activity relationship (QSARO) model can be used to predict EC50 to save time and endeavor. Correlation was well established with the QSAR parameters such as log P, log S and solvatochromic parameter(Vi/100 $\pi$, ${\beta}$m and am). The QSAR modeling was used with multi-regression analysis and mono-regression analysis. These analyses resulted in the following QSAR : $log EC_{50} =2.48 + 0.914 log S(n=9 R2=85.5% RE=0.378) log EC_{50}=0.35 - 4.48 Vi/100 + 2.84 \pi^* +9.46{\beta}m-4.48am (n =14 R2=98.2% RE=0.012) log EC_{50} =2.64 -1.66 log P(n=5, R2=98.8% RE=0.16) log EC_{50}=3.44 -1.09 log P(n=9 R2= 80.8% Re=0.207)$

Atheroprotective nasal immunization with a heat shock protein 60 peptide from Porphyromonas gingivalis

  • Purpose: Immunization with Porphyromonas gingivalis heat shock protein 60 (PgHSP60) may have an immunoregulatory effect on atherogenesis. The aim of this study was to determine whether nasal immunization with a PgHSP60 peptide could reduce atherosclerotic plaque formation in apolipoprotein E knockout (ApoE KO) mice. Methods: Seven-week-old male ApoE KO mice were assigned to receive a normal diet, a Western diet, a Western diet and challenge with PgHSP60-derived peptide 14 (Pep14) or peptide 19 (Pep19), or a Western diet and immunization with Pep14 or Pep19 before challenge with Pep14 or Pep19. Results: Atherosclerotic plaques were significantly smaller in mice that received a Western diet with Pep14 nasal immunization than in mice that received a Western diet and no Pep14 immunization with or without Pep14 challenge. An immunoblot profile failed to detect serum reactivity to Pep14 in any of the study groups. Stimulation by either Pep14 or Pep19 strongly promoted the induction of CD4+CD25+ forkhead box P3 (FoxP3)+ human regulatory T cells (Tregs) in vitro. However, the expression of mouse splenic CD4+CD25+FoxP3+ Tregs was lower in the Pep14-immunized mice than in the Pep14-challenged or Pep19-immunized mice. Levels of serum interferon gamma (IFN-γ) and transforming growth factor beta were higher and levels of interleukin (IL) 10 were lower in the Pep14-immunized mice than in the other groups. Induction of CD25- IL-17+ T helper 17 (Th17) cells was attenuated in the Pep14-immunized mice. Conclusions: Nasal immunization with Pep14 may be a mechanism for attenuating atherogenesis by promoting the secretion of IFN-γ and/or suppressing Th17-mediated immunity.

Effect of Sterols on Phytophthora infestans and Oospore Production on detached Potato Plants (감자 역병균에 대한 스테롤류의 영향 및 감자절편에서의 난포자 형성)

  • The effects of media, cholesterol, $\beta$-sitosterol and lecithin on the growth and oospore production of the isolates KM10, U6, CDB6, MHB6, JD1 (A$^2$type) of Phytophthora infestans isolated in Korea and F8l7, DNC303 (A$^1$type), IB908, DN107 (A$^2$type) obtained from Japan were investigated. Mycelium of P. infestans grew better on V-8 juice agar and rye meal agar than on the other media. Oospores were produced most abundantly on V-8 juice agar. Mycelium extended more 16.6, 8.3, and 5.2% on V-8 juice agar supplemented with 5 $\mu\textrm{g}$/$m\ell$ of cholesterol, $\beta$-sitosterol and lecithin, respectively, and oospores are produced 76.0, 58.0, and 34.6 % on V-8juice agar supplemented with 5 $\mu\textrm{g}$/$m\ell$ of cholesterol, $\beta$-sitosterol and lecithin, respectively. Oospores more produced on detached potato plant disks when $A^1$ and $A^2$ type exist simultaneously which indicating that variation of population can occur in the field, but the rate of oospore formation and the number of oospores produced was low and small quantity.

Relationship of Physicochemical Characteristics and Ethanol Yield of Korean Barley (Hordeum vulgare L.) Cultivars

  • The grain and agronomic characteristics of Korean barley cultivars were investigated with respect to ethanol yield. Test weight, grain yield, and starch yield showed noticeable variation among the cultivars. Grain yields were higher in covered barley and non-waxy barley. Starch yield was higher in non-waxy barley than waxy barley. Protein, ${\beta}$-glucan, and starch content of tested cultivars ranged in 10.0-12.9%, 4.4-7.5% and 49.7-65.3%, respectively. Naked barley cultivar had higher starch content than covered barley cultivar. However, covered barley had high starch yield because it has higher grain yield than naked barley. Covered barley cultivar had higher husk content, ranging 7.6-14.0%, than that of naked barley cultivar, ranging 5.3-8.0%. Starch content was positively correlated with amylose content, test weight, ethanol yield and negatively correlated with protein, husk, ${\beta}$-glucan content. Ethanol yield per ton was positively correlated with starch content, but negatively correlated with husk content. Ethanol yield per hectare was positively correlated with starch yield, grain yield, grain weight and negatively correlated with protein, test weight. From this research, the important characteristics of barley cultivar as a bioethanol producing material were starch content and grain yield. Optimum barley genotype was non-waxy naked barley that had low protein, ${\beta}$-glucan, husk content, and high starch content and grain yield.

Interpersonal Communication Competence and Social Anxiety in Nursing Students (간호대학생의 의사소통 능력과 대인불안)

  • Purpose: This study aimed to identify a relationship between the communication competence and social anxiety in nursing students. Methods: With a cross-sectional survey, a convenience sampling of 303 nursing students was taken from P-university in Y-city. The instrument for the study was a questionnaire that is consisted of the general characteristics, an Interaction Anxiousness Scale (IAS) and a Global Interpersonal Communication Competence Scale (GICC). Data was analyzed using descriptive statistics, t-test, one-way ANOVA, Kruskal-Wallis, Pearson's correlation coefficients and stepwise multiple regression with SPSS version 21.0. Results: The mean score of the IAS was $41.89{\pm}8.74$ and the GICC was $3.51{\pm}0.34$. Communication competence significantly differed by the satisfaction of major (F=10.86, p<.001). In addition, the satisfaction of interpersonal relationships was significantly different in both communication competence (F=17.52, p<.001) and social anxiety (F=23.85, p<.001). Social anxiety had a negative correlation with communication competence (r=-.48, p<.001). Social anxiety accounted for 55.3% of the variance -social relaxation (${\beta}$=-.62), assertiveness (${\beta}$=-.18), interaction management (${\beta}$=.14) and efficiency (${\beta}$=-.13)- of communication competence's subcategories. Conclusion: It was found that communication competence has an influence on social anxiety. Therefore, education programs are needed to promote communication competence for a decline of social anxiety in nursing students.
