1,4-Thiazine Carboxanilide의 합성: Pummerer 반응에서의 인접기 참여효과 (A Synthesis of 1,4-Thiazine Carboxanilide: Neighboring Group Participation in Pummerer Reaction)

  • 새로운 ${\alpha},{\beta}$-불포화Carboxanilide계열의 살균제를 개발을 목적으로 4-acetyl-3-methyl-N-phenyl-1,4-thiazine-2-Carboxamide(6)의 합성에 대하여 기술하였다. Dihydro-1,4-thiazine methyl ester 11을 산화하여 생성된 sulfoxide 7을 Pummerer 반응하여 $\alpha$-acetoxy dihydro-1,4-thiazine 10a를 얻었다. 반며에 Dihydro-1,4-thiazine Carboxanilide sulfoxide14는 동일한 반응조건 하에서 중간체 sulfonium ion의 carboxanilide기가 관여한 vinylogous Pummerer 반응의 전위에 의하여 중간체를 15를 통하여 acetoxymethyl dihydro-1,4-thiazine 18이 생성되었다. 1,4-Thiazine carboxanilide 6은 $\alpha$-acetoxy dihydro-1,4-thiazine 10a를 산촉매로 처리한 다음 생성된 21a를 가수분해하고 아닐린과 결합하여 합성하였다

  • Several new pyridine, pyridinethione, pyrazole [3, 4-b]-pyridine, pyrido [1, 2-a]-1, 3-thiazine and pyrido[1, 2-a] pyridine thione derivatives have be synthesised via the reactions of 2-methyl-3-ethoxycarbonyl-4-phenyl-5-cyano-1, 4, 5, 6-tetrahydro-pyridine-6-one 2 with different rfeagents. The structures of the newly synthesised derivatives were established on the basis of elemental analyses and spectral data studies.

  • Synthesis of $\alpha$-piperidino and $\alpha$-morphelino styryl quinoxalinone 2f, 2g respectively by facile one step method is reported. The Michael adducts (3a-d) obtained by the interaction of 2-styryl-2 (1H) quinoxalinone (2) and ethylacetoacetate have been treated with resorcino and hydroxylamine separately. With resorcinol the chromones (4) are obtained whereas with ydroxylamine isoxazoles (6) are the products. Michael addition of acetylacetone to 2 leads to 3-[1'-aryl-2'-(2'-hydroxy-3'-quinoxalinyl)ethyl]-2, 4-pentanediones (5) which undergo cyclisation with hydroxylamine to give isoxazoles (7). Addition of thiopenol and thioglycolic acid to 2 gives 3-$\alpha$[$\beta$-(phenyl)-$\beta$-(plenylthio)]ethyl-2(1H)-quinoxalinone (8) and 3-$\alpha$-[$\beta$-phenyl)-$\beta$-(hydroxycarbonylmethylithio)]-ethyl-2(1H)-qui noxalinone (9) respectively. 2-Bromomethyl-2(1H)-quinoxalinone (1b) reacts with thioglycolic acid to gives S-[2 (1H)-oxoquionoxaline-3-yl-methyl] mercaptoacetic acid (10) which on cyclisation with acetic anhydride/pyridine affords 1, 2, 5, 6-tetrahydro [1, 4]thiazino[4, 3-a] quinoxaline-1, 6-dione (11).

  • 새로운 cephalosporin 항생물질인 7-[(3,4-dihydro-6-methoxycarbonyl-2,2-dimethyl-2H-1,4-thiazin-3-yl)acetamido]-3-[(substituted pyrimidin-2-yl)thiomethyl]-3-cephem-4-carboxylic acid derivatives(2a∼2d)유도체들을 합성하였다. 7-ACA의 3-위치에는 pyridinylthiomethyl기를 도입시키고, 7-위치에는 thiazine를 치환시켜서 구조 변화에 대한 항균 활성을 측정하였다. 그 결과 이들 유도체들은 기준물질인 cefotaxim보다 낮은 항균력을 나타내었다.

1,3-티아졸리딘술폭시드의 디히드로-1,4-티아진 유도체로의 전위에 관한 연구 (A Study on the Rearrangement of 1,3-Thiazolidine Sulfoxides to Dihydro-1,4-thiazines)

  • 4-아세틸-5,6-디히드로-2-메틸-1,4-티아진-3-카르복실산 유도체 24를 상응하는 티아졸리딘 술폭시드의 고리확대에 의하여 제조하였다. 2-메틸-1,3-티아졸라딘-2-아세트산 유도체 12를 산화하여 시스와 트랜스 술폭시드, 14 및 15를 얻었으며 이들의 구조는 수소핵자기 공명스펙트럼, 적외선 흡수스펙트럼, 그리고 중수소 치환반응에 의한 regioselectivity에 근거하여 결정하였다. 산촉매 존재하에서 시스와트랜스 술폭시드, 14 및 15는 중간체 술페닌산 18을 거쳐 디히드로-1,4-티아진 24로 전환되었다. 그러나 중성조건($100^{\circ}C$의 DMF)에서 트랜스 술폭시드 15는 중간체 술페닌산 21을 거쳐 isomeric 디히드로-1,4-티아진 27로 전환하였다. 화합물 24와 27의 생성반응기전도 논의하였다.

  • The ring expansion of 1,3-thiazolidines 4 derived from ${\beta}$-ketoacid derivatives to the corresponding dihydro-1,4-thiazines 1 by using the action of chlorine on 4 has been achieved. In the chlorinolysis unisolable sulfenyl chlorides 5 may be formed from chlorosulfonium ions 11 by ${\beta}$-elimination involving carbonyl activated methylene hydrogens. Addition of sulfenyl chloride to the internal double bond appears to form probable thiiranium ions 14, which in turn gave 1 with loss of acidic proton. Imminium ions 15 could be hydrolyzed easily to give enol 8. As a side reaction, dihydrothiazine that was formed was further chlorinated to produce dichlorides 16 which were rearranged readily to the chloromethyl compounds 10.

1,3-티아졸리딘 술폭시드의 입체구조에 관한 연구 (A Study on the Stereochemistry of 1,3-Thiazolidine)

  • 한 분자 내에 3개의 chiral 중심이 있는 1,3-티아졸리딘 술록시드 1의 입체화학을 중수소치환반응 및 trapping 반응을 통해 밝혔다. 3-아세틸-1,3-티아졸리딘 5의 부분이성질체를 분리한 다음 6과 8을 각각 술록시드로 산화시켜 상응하는 $\alpha$-시스 10, $\alpha$-트랜스 11과 $\beta$ -시스 12, $\beta$ -트랜스이성체 13을 얻었다. 술폭시드 10은 중성 조건하의 환류하는 벤젠 또는 톨루엔 용액 중에서 열역학적으로 보다 안정한 13으로 이성질화하였다. 중수소치환반응에 의해서 술폭시드 13과 술폭시드 11의 2-메틸기의 수소원자는 중수소로 치환되었다. 술폭시드 10과 12로부터 sigmatropic 전위에 의해서 생성된 중간체 슬페닌산 25 및 26은 2-머캡토벤조티아졸(2-MBT)에 의하여 trap되어 디술피드 27 및 28로 전환되었으나 동일한 반응조건하에서 술폭시드 11 및 13은 환팽창 생성물 디히드로-1,4-티아진 29로 전환되었다. 산촉매 존재하에서 술폭시드 10, 11, 12는 술폭시드 13을 통해서 디히드로-1,4-티아진 29로 정량적으로 전환되었다. 술폭시드들의 이성질화 및 29의 생성메카니즘도 기술하였다.

  • 4-Arylazo-3-phenyl-5-aminopyrazoles (5a, b) and substituted hydroxythiazoles 8a, b were synthesized from the reaction of 4a, b with hydrazine hydrate and mercaptoacetic acid respectively. Compounds 5a, b and 8a, b were also obtained from coupling of 2a, b with 6 and 7, respectively. 4H-1, 4-Benzothiazine 11 was prepared from 1 and 10. The resaction of the diazonium salts 2a-c with diethyl 3-amino-2-cyanopenet-2-en-1, 5-dicarboxylate 12 was also reported.

  • New cephalosporin antibiotics, 7-[(3,4-dihydro-6-methoxycarbonyl-2,2-dimethyl-2H-1,4-thiazin-3-yl)acetamido]-3-substituted-3-cephem-4-carboxylic acid derivatives 2a-2d, 7 -[(3,4-dihydro-6-ethoxycarbonyl-2,2-dimethyl-2H-1,4-thiazin-3-yl)acetamido] -3-substituted-3-cephem-4-carboxylic acid derivatives 3a-3d and 7-[(3,4-dihydro-6-methoxycarbonyl-2,2-dimethyl-2H-1,4-thiazin-3-yl-1-(S)-oxide)acetamido]-3-substituted-3-cephem-4-carboxylic acid derivatives 4a-4d were synthesized. Antibacterial activities of these new cephalosporin derivatives and the relationship between their structures and their activities were examined. Among them, 7-[(3,4-dihydro-6-methoxycaronyl-2,2-dimethyl-2H-1,4-thiazin-3-yl-1-(S)oxide)-acetamido]-3-[(1,2,3-triazol-5-yl)thiomethyl]-3-cephem-4-carboxylic acid 4d exhibited the antibacterial activities against Gram(+)and Gram(-) bacteria.

피록시캄의 용매 비의존 결정구조 (The Solvent-Independent Structure of Piroxicam)

  • The three-dimensional structures of piroxicam crystallized from two different solvents, toluene and toluene/hexane mixture respectively, are proved identical: $C_{15}H_{13}N_3O_4S,\;M\;=\;331.35$, monoclinic, a = 7.128(1), b = 15.146(2), c = 13.956(2) ${\AA},\;{\beta}=\;97.33(1)^{\circ},\;V\;=\;1494.37{\AA}^{3},\;Dx\;=\;1.472\;g/cm^{3},\;Z\;=\;4,\;space\;group\;P2_{1}/c,\;Mo\;K{\alpha}(\lambda=\;0.71073\;{\AA})$, F(000) = 688, T = 295 K, R = 0.0611 for 1993 unique observed reflections. The thiazine ring exhibits a half chair conformation. An amide group is involved in an intramolecular hydrogen bond to the hydroxy group, O(17)-H(17)${\cdots}O(15){\AA}$. The molecule is planar within 2 ${\AA}$ with the interplanar angle $127.9(4)^{\circ}$ between pyridine and benzene rings. A molecular chain parallel to [011] is formed by two intermolecular hydrogen bonds N(16)-H(6)${\cdots}O(11)$ and C(6)-H(6)${\cdots}O(11)$, and the molecular chains are held together by van der Waals forces.
