Synthesis of 1,3-Dioxolan-2-yliden Derivatives and Their Antifungal Activities (1,3-Dioxolan-2-yliden 유도체들의 합성과 항진균 활성)

  • (1H-1,2,4-Triazolyl) methy-4-(sub). phenyl-5-methyl-1,3-dioxolan-2-yliden (3) derivatives were synthesized and tested for their antifungal activities. The designed compounds with a 1,2,4-triazolylmethyl group at the 4-position of 1,3-dioxolan-2-yliden moiety were synthesized by reaction of difluorinated olefins(2) with (2R, 3R)-2-(2,4-dihalophenyl)-1-(1H-1,2,4-triazol-l-yl) butane-2,3-diol (1). These compounds were tested for in vitro antifungal activities against 16 fungi species. The MIC values were determined by the micro broth dilution method. In general, 1,3-dioxolan-2-yliden derivatives showed antifungal activities in vitro. Among them, (4R, 5R)-4-(2,4-difluorophenyl)-5-methyl-2-[1-(3,4-methylenedioxypheny)meth-ylidene)-1,3-dioxolon-4-yl(1H-1,2,4-triazollyl)methane showed superior antifungal activities to fluconazol and ketoconazol.

  • SKI 2053R and SKI 2053R-human serum albumin(HSA) mixture were examined for their antigenicity in Hartley guinea pigs as well as C57BL/6 mice in comparison with distilled water (DW), HSA and DW-HSA conjugate. Several antigenicity tests, including acitive systemic anaphylaxis(ASA), passive systemic anaphylaxis (PSA), passive cutaneous anaphylaxis (PCA) and indirect hamagglutination test (IHA), were performed according to the Established Regulations of National Institute of Safety Research. The results were as follows: 1. When guinea pigs were sensitized with SKI2053R or SKI2053R-HSA emulsified with complete Freund's adjuvant(CFA), these animals showed negative reactions in ASA and PSA. 2.No blue spot was observed on the back skin of guinea pigs in the PCA test. 3. Sera from guinea pigs revealed a negative reaction in IHA. 4.Guinea pigs were sensitized by HSA emulsified with CFA as a positive control, and these animals showed positive reactions in ASA, PSA, PCA, and IHA. As shown above, SKI2053R was considered to possess neither antigenic, nor haptenic properties, and confirmed not to have the haemagglutinating activity.

  • Synthesis of an optically active azabicyclo[3.2.1]octane skeleton as a backbone of the tropane alkaloids has been achieved by employing intramolecular Mannich reaction. Utilizat ion of (R)-${\alpha}$-methylbenzylamine as a chiral auxiliary provided an excellent cyclization of amino dioxolane precursor. However, this auxiliary did not afford high asymmertic induction for the preparation of the optically active cyclization precursor.

  • The Jig cells are fabricated in the drying room, and consisted of elemental sulfur used as a cathode active material, lithium metal used as a anode material and 1M $LiCF_{3}SO_{3}$ dissolved in TG (Tetraglyme)/DIOX (1,3-Dioxolane) used as a electrolyte. The four kinds of electrolytes with different content of TG and DIOX are prepared. The electrochemical properties of the foregoing electrolytes-based lithium sulfur batteries are analyzed by AC impedance experiments. The conductivity of four different electrolytes is investigated. The conductivity of electrolyte [1M $LiCF_3SO_3$ dissolved in TG/DIOX (50:50, vol.)] is higher than that of other three kinds of electrolytes with different volume ratio (70:30, 30:70) and single solvent (TG).

  • Racemic nuciferal(1) and nuciferol(2), the terpenic natural perfumeries, have been synthesized by a simple procedure. The benzylic halide 6; 1-(1-chloroethyl)-4-methylbenzene, was prepared by converting p-tolualdehyde(4) into 1-(p-tolyl)-1-ethanol(5), followed by convertion of 5 into corresponding chloride. The Grignard reagent of 6 was reacted with the bromoacetal 7, 2-(2-bromoethyl)-1,3-dioxolane, to give a crosscoupling product 8, which was hydrolysed to 4-(p-tolyl)-pentanal (9). The Wittig reaction of isopropylide 10 with 9 yielded arcurcumen(11). The stereospecific allylic oxidation of the gem-dimethyl olefin 11 with selenium dioxide afforded a trans-aldehyde, (${\pm}$)-1, which was reduced to corresponding alcohol, (${\pm}$)-2.

Cell Viability and Flow Cytometry Analysis of a Novel Antitumor Agent, Heptaplatin in Human Melanoma Cell Line, SK-MEL-28 (신규항암제인 Heptaplatin의 인체 흑색종세포(SK-MEL-28)에 대한 세포생존률 및 유세포 분석)

  • Heptaplatin, cis-Malonato[(4R,5R)-4,5-bis(aminomethyl)-2-isopropyl-1,3-dioxolane]platinum(II), is a novel platinum-based antitumor agent with clinical potential against human stomach cancer and the 3rd generation of the cisplatin. This study was performed to study how cisplain, heptaplatin and sunpla which is a mixture of heptaplatin and mannitol (w: w=l : 2) affect cell viability of SK-MEL-28 human melanoma cell line. Heptaplatin ($IC_{50}$/; 95.35 $\mu$M) and sunpla ($IC_{50}$/; 10.95 11M) were less effect than cisplatin (IC $_{50}$; 10.92 $\mu$M) on the SK-MEL-28 cells. By cell cycle analysis using flow cytometry, it was identified that the cells were arrested at G2/M phase by cisplatin, heptaplatin and sunpla, and percentage of cell death group was increased according to increasing of time and concentration. These results suggest that cisplatin, heptaplatin and sunpla are a novel anticancer agent against human melanoma cell.l.

  • Steric and electronic parameters of 4'-substituents play significant roles in steering the conformation of nucleoside analogues. In order to investigate the relationship of 4'-group with antiviral enhancement, novel 4'-hydroxymethyl-5'-norcarbocyclic adenosine phosphonic acid analogues were designed and synthesized from 2,2-dimethyl-1,3-dioxolane-4-ethanol (5) using reiterative Grignard addition and ring-closing metathesis (RCM) as key reactions. The synthesized adenosine phosphonic acids analogues (22) and (23) were subjected to antiviral screening against HIV-1. Compound (23) exhibited moderate anti-HIV activity ($EC_50$ = 8.61 ${\mu}M$) in the CEM cell line.

  • 티오요소류, 폴리에테르류, 염료와 같은 유기화합물이 첨가된 황산동 전기도금욕에서 석출된 구리의 광택, 평활력효과를 무첨가욕과 기존 광택제가 첨가된 도금욕에서와 각각 비교하여 이들 유기첨가제가 구리의 석출상태와 음극분극에 미치는 영향을 Hull cell 실험과 현미경관찰, 전기화학적 측정방법에 의해 고찰하였다. 황산동 전기도금욕에서 thiourea, 2-mercapto-2-imidazoline, 1-acetyl-2-thiourea와 같은 유기 황화합물은 비교적 고전류 밀도 범위에서 결정성장이 억제되고 입자가 미세한 구리석출을 일으켜 양호한 광택효과와 평활력을 가져오나, 특히 저전류밀도에서는 해로운 줄무늬가 발생한다는 점에서 많은 개선의 여지가 필요하다. 그러나 이들 유기 황화합물에 1,3-dioxolane polymer와 소량의 염소 이온을 혼합 첨가할 것 같으며 넓은 전류밀도 범위에서 양호한 광택과 평활력이 있는 동석출을 얻을 수 있었다.

Electrochemical Properties of Lithium Sulfur Battery with Silicon Anodes Lithiated by Direct Contact Method

  • It is hard to employ the carbon materials or the lithium metal foil for the anode of lithium sulfur batteries because of the poor passivation in ether-based electrolytes and the formation of lithium dendrites, respectively. Herein, we investigated the electrochemical characteristics of lithium sulfur batteries with lithiated silicon anode in the liquid electrolytes based on ether solvents. The silicon anodes were lithiated by direct contact with lithium foil in a 1M lithium bis(trifluoromethane sulfonyl) imide (LiTFSI) solution in 1,2-dimethoxyethane (DME) and 1,3-dioxolane (DOL) at a volume ratio of 1:1. They were readily lithiated up to ~40% of their theoretical capacity with a 30 min contact time. In particular, the carbon mesh reported in our previous work was employed in order to maximize the performance by capturing the dissolved polysulfide in sulfur cathode. The reversible specific capacity of the lithiated silicon-sulfur batteries with carbon mesh was 1,129 mAh/g during the first cycle, and was maintained at 297 mAh/g even after 50 cycles at 0.2 C, without any problems of poor passivation or lithium dendrite formation.

  • 가스 하이드레이트는 작은 고체 부피 내에 막대한 양의 가스를 저장할 수 있다는 특성으로 인하여, 최근 천연가스 혹은 메탄의 저장 매체로 활용하기 위한 연구가 활발히 진행중에 있다. 하지만 실제 응용을 위해서는 미세구조 분석이 수행되어 하이드레이트 형태로 저장할 수 있는 정확한 저장 용량을 파악할 필요가 있다. 본 연구에서는 여러가지의 고리형 에테르, 고리형 에스테르 및 고리형 케톤 화합물들을 테스트하여 메탄 가스와 반응하는 6가지의 새로운 sII 혹은 sH 하이드레이트 형성제를 파악하였다. 또한 새로이 발견된 형성제 모두에 대하여 하이드레이트 상평형도 측정하였다. 얻어진 상평형 데이터는 하이드레이트 안정영역과 게스트 분자 크기 간에 뚜렷한 상관관계가 있음을 입증하였다. 아울러 형성된 하이드레이트 샘플은 고체 분말 X-선 회절과 고체상 13C NMR 분석을 수행하여 하이드레이트 구조와 게스트 포집률을 조사하였다. 마지막으로, 비슷한 화학 구조식을 갖고 있음에도 2-methyltetrahydrofuran과 3-methyltetrahydrofuran, 혹은 4-methyl-1,3-dioxane과 4-methyl-1,3-dioxolane은 서로 다른 하이드레이트 결정 구조를 보여 주었는데, 이러한 차이는 하이드레이트 결정 구조를 결정짓는 게스트 분자 크기, 즉 임계 게스트 분자 크기를 파악하는 데에도 매우 유용한 정보를 제공할 수 있을 것이라 판단된다.
