Contact Sensitivity to Dinitrochlorobenzene as a Marker Trait in the Indirect Selection for Body Mange and Coccidiosis Resistance in Broiler Rabbits

  • To determine the effects of genetic and environmental influences on cell mediated immune (CMI) responses in broiler rabbits, contact sensitivity to 2,4-Dinitrochlorobenzene (DNCB) was assessed in three temperate broiler breeds of rabbits, namely Soviet Chinchilla, New Zealand White and Grey Giant. The feasibility of using the contact sensitivity to DNCB as a marker trait in selection for disease resistance was examined. There were highly significant differences between breeds (p<0.01) in initial skin thickness and contact sensitivities to DNCB at 24, 48 and 72 hours. Initial skin thickness was greatest in the Soviet Chinchilla breed (mean 2.2484 mm), and was significantly greater (p<0.01) in males (2.4963 mm) than in females (1.7846 mm) (p<0.01). Highest contact sensitivity to DNCB was in the New Zealand White breed with mean increase in skin thickness of 1.1884, 0.9072 and 0.5879 mm at 24, 48 and 72 hours post challenge respectively. Delayed type hypersensitivity (DTH) reaction to DNCB at 24 hours post challenge had a highly significant association (p<0.01) with the incidence of body mange in rabbits. The results indicated a lowered contact sensitivity to DNCB at 24 hours post challenge was associated significantly (p<0.01) with an increase in incidence and severity of body mange, suggesting its potential value as a marker. The correlation s among contact sensitivities at 24, 48 and 72 hours were positive and highly significant (p<0.01); correlations between initial skin thickness and contact sensitivities were negative and highly significant (p<0.01). Another notable significant correlation was between body weight and delayed type hypersensitivity at 24 hours indicating that an enhanced CMI might be associated with better growth rate and general wellbeing.

Effects of Slurry Composting and Biofiltration Liquid Fertilizer on Growth Characteristic of Poplar Clones in a Reclaimed Land Mounding Soil (간척지 성토지 식재 포플러의 SCB액비 처리에 따른 클론별 생장특성)

  • We studied on the effects of slurry composting and biofiltration liquid fertilizer (SCBLF) on growth of two-year-old poplar clones planted in a reclaimed land mounding soil. The soil on the experimental site had lower concentrations of both exchangeable cations and salinity than before reclamation. However, the content of organic matter was low compared with the most soils. We applied SCBLF to the poplars six times and 5 L in each time for 80 days. Ten clones of six poplar species or hybrids were tested in this study: Populus alba ${\times}$ P. glandulosa(Clivus, 72-30, 72-31, Bongwha1), P. deltoides ${\times}$ P. nigra(Dorskamp), P. deltoides(Lux) ${\times}$ P. deltoides(Harvard)(97-19), P. euramericana(Eco28, I-476), P. nigra ${\times}$ P. maximowiczii(62-2) and P. Koreana ${\times}$ P. nigra var. italica(Suwon). Growth performance varied more among clones than among species. Average height growth of treated plots was 18% greater than control, and clones Clivus, 97-19, Eco28 and Dorskamp were more vigorous than other clones. Diameter at breast height in treated plots was 41% greater than control, and 97-19, Dorskamp, Eco28 and Clivus were the four best clones in this respect. Mean leaf area of treated trees was 26% greater for control trees. Chlorophyll content was similar between treated and controlled trees. Total nitrogen values in leaves were much higher in treated trees 18%. SCBLF treatment on poplars planted in reclaimed land helped tree growth.

A Study on the Main Diagnostic Code according to the Analysis of the Frequency of Fall Patients by Case-Centered Damage External Code (사례 중심의 손상외인코드 별 낙상환자 빈도수 분석에 따른 주진단코드 연구)

  • This study aimed to analyze patients hospitalized for injuries who fell using the data from 2020 to 2021 at institution A located in Gangneung-si, Gangwon-do, using codes for causes of injury. After classifying 20 codes from W00 to W19, which are external cause codes for fall patients, the most frequently occurring W18, W01, W10, and W13 were analyzed. The external cause of injury code W18 was other falls on the same plane, with the highest frequency of S72 and Z47, S72 being a fracture of the femur, and Z47 being orthopedic follow-up treatment. The external injury code W01 was determined to be a fall on the same plane due to slipping, tripping, and tripping, and like W18, S72, a fracture of the femur, and Z47, orthopedic follow-up treatment, were frequently reported. In W10, intracranial injuries such as concussion and epidural hemorrhage due to a fall on the stairs, S06, were common. Lastly, in W13, 91% of cases occurred in people in their 40s to 70s due to falls from buildings or structures, confirming that they occur frequently in middle-aged people, Z47 had the most frequent orthopedic follow-up treatment, and S72 had a fracture of the femur. It was found to be the second most common. In this way, the frequency of falling patients was analyzed, and the age and main diagnosis code at which most falls occurred were analyzed.

Clinical Significance of Combined Detection of Serum Tumor Markers in Diagnosis of Patients with Ovarian Cancer

  • Objective: To explore the predictive value of tumor markers, including cancer antigen 72-4 (CA72-4), cancer antigen 15-3 (CA15-3) and cancer antigen 125 (CA125), in single or combined detection, for the diagnosis of ovarian cancer. Methods: 120 patients diagnosed with ovarian cancer from August 2011 to March 2013 and 80 patients diagnosed with benign ovarian tumors were enrolled in this test, along with 50 health examination women randomly selected from the database as controls. Serum levels of CA72-4, CA15-3 and CA125 in this study were determined by electrochemiluminescence (ECL). Results: Serum levels of CA72-4, CA15-3 and CA125 in ovarian cancer were higher than those in healthy group and benign group (P<0.01).The sensitivity of combined detection of those three tumor markers for diagnosis of ovarian cancer was obviously higher than with single detection with each marker (P<0.01). Conclusions: CA72-4, CA15-3 and CA125 could be a good combination in the diagnosis of ovarian cancer. Patients whose tumor markers continue to increase should be highly suspected of malignancy.

The Effect of Escherichia coli Lipopolysaccharide on the Hormones in Rabbits

  • With a rabbit model, the present study was performed to examine the effect of Escherichia coli lipopolysaccharide (E. coli LPS) on hormones. Cortisol, epinephrine, and norepinephrine concentrations in LPS-treated groups were high at all sampling periods (from 3 hrs to 72 hrs) as compared to control group (p<0.05 or p<0.01). The each peak time was at respectively 24 hrs, 3 hrs, and 12 hrs. Insulin and glucagon concentrations in LPS-treated groups elevated up to 12 hrs (p<0.01 or p<0.05) with a each peak point at 12 hrs or 6 ks, while those of the rest sampling points (from 24 hrs to 72 hrs) were lower than that of control (p<0.05). Increase of cortisol concentration was generally dose-dependent, whereas the changes of the other hormones were irregular patterns. These observations show that E. coli LPS lead to releases of stress hormones such as cortisol, epinephrine, and norepinephrine and disturbances of endocrine systems. These LPS-induced hormonal disorders may cause physiologically fatal results.

Emulsion Stability of Cosmetic Facial Cream O/W Emulsions Prepared by Brij Type Non-ionic Emulsifie (Brij계 비이온성 혼합유화제를 이용하여 제조된 화장용크림 O/W 유화액의 유화안정성)

  • In this study, the stability of O/W cosmetic facial cream emulsions according to HLB values was evaluated by mixing nonionic surfactants, such as Brij 78&72 and Brij 98&92. Brij 78&72 (steareth-20&steareth-2, EMS-01), saturated fatty acid, and Brij 98&92 (oleth-20&oleth-2, EMS-02), unsaturated fatty acid, were used as mixed surfactants. The stability of the O/W emulsion was evaluated by using the emulsion viscosity, particle size, particle size distribution, and zeta-potential. The viscosity of the emulsion increased with the increase of time for EMS-01 while that of EMS-02 decreased with the increase of HLB value. The particle size of both EMS-01 and EMS-02 increased with time. The emulsifier with a HLB value of 10.8, which is the most similar to the required HLB value of mineral oil, 10.5, had the smallest particle size and highest density and also showed the highest emulsion stability. The zeta-potential of both emulsions tended to increase with the HLB value. No significant changes were observed in emulsions of the HLB value of 10.8 or more. The saturated fatty acid system, EMS-01, exhibited a higher zeta-potential value than that of the unsaturated fatty acid EMS-02 and also was superior in the stability.

Apoptotic effect of Pseudomonas aeruginosa exotoxin A in human tongue squamous cell carcinoma(SCC) 25 cells (Pseudomonas aeruginosa exotoxin A(PEA)가 사람혀 편평암종세포에서 나타나는 세포자멸사 작용)

  • Objectives : The purpose of the study is to examine the apoptotic effects of Pseudomonas aeruginosa exotoxin A(PEA) in squamous cell carcinoma(SCC) 25 cells. Methods : Cell growth reduction and apoptosis induced by PEA were confirmed by WST-1 assay, Hoechst 33258 staining, flow cytometry analysis, and Western blot assay. Results : The PEA treatment decreased the cell viability in a dose and time dependent manner: control; $100{\pm}0^e$(p<0.01), 0.1875 nM; $87{\pm}4.36^d$(p<0.01), 0.375 nM; $82{\pm}0.58^d$(p<0.01), 0.75 nM; $72{\pm}1.67^c$(p<0.01), 1.5 nM; $51{\pm}1.53^{bc}$(p<0.01), 7.5 nM; $31{\pm}1.20^{ab}$(p<0.01), 15 nM; $26{\pm}0.67^a$(p<0.01), control; $100{\pm}0^a$(p<0.05), 24 h; $51{\pm}1.53^b$(p<0.05), 48 h; $16{\pm}0.5^c$(p<0.05), 72 h; $12{\pm}1.67^d$%(p<0.05). The PEA was observed on SCC 25 cells with the half maximal inhibitory concentration(IC50) value of 1.5 nM at 24 hours. The PEA treated SCC 25 cells demonstrated several types of apoptotic indications, such as nuclear condensation, the increase of sub G1, and the cleavage of PARP-1 and DFF 45. Conclusions : PEA showed anti-cancer activity against SCC 25 cells via apoptosis. PEA may potentially contribute to human oral cancer treatment.

Relationship between Intersequence Pauses, Laying Persistency and Concentration of Prolactin during the Productive Period in White Leghorn Hens

  • Prolactin is considered to influence the taking of pauses in between ovulatory sequences in White Leghorn hens. Therefore modulating concentrations of prolactin using bromocriptine - a dopamine agonist during early life (17 to 36 weeks of age) could overcome the inhibitory effects of high concentration of prolactin on ovarian activity. The effect of modulation of prolactin concentration on egg production, sequence length and inter sequence pauses were studied by analyzing the oviposition records from 19 to 72 weeks were studied and compared with untreated controls. Bromocriptine administered subcutaneously (100 $\mu$g kg$^{-1}$ body weight or orally through feed (640 $\mu$g day$^{-1}$ bird$^{-1}$) resulted in a steady and sustained decrease in prolactin levels (p<0.01) during and after the withdrawal of treatment up to one reproductive cycle (72 weeks of age). The treated birds had comparatively longer sequences (p<0.01) and fewer pauses (p<0.01). Egg production increased (p<0.01) by fourteen per cent through subcutaneous administration and eleven per cent through oral feeding, over the control birds. It is concluded that the physiological pauses that occur during ovulatory sequences can be disrupted effectively using bromocriptine. Prolactin levels are modulated which may interfere with the follicular recruitment and subsequent oviposition thereby improve egg laying potential of the bird.

The efficiency of different types of extenders for semen cooling in stallions

  • Objective: This study was conducted to examine influence of skimmed milk-based extender (SM), INRA 96 extender and BotuSemen Gold extender on parameters of stallions' ejaculate during storage. Methods: In this study, 14 stallions between 4 and 20 years of age were monitored. Total and progressive motility, viability and morphology of sperm were evaluated at time intervals of 24, 48, and 72 hours after collection. Results: The total motility, progressive motility, and values of sperm with normal morphology were significantly higher in the INRA 96 and BotuSemen Gold extenders than in the SM (p<0.01). The sperm viability differed significantly in all extenders (p<0.01). The highest value of sperm viability was in INRA 96 (64.69%±0.67%) and lowest in SM (59.70%±0.81%). The highest differences occurred at 72 hours of storage. Values of total motility, progressive motility and sperm viability decreased over time (p<0.01). In case of sperm morphology there was no statistically significant decrease between 48- and 72-hour time intervals. Conclusion: It can be concluded that the extenders with a chemically defined composition have shown better indicators of insemination capabilities in ejaculates than the SM. The BotuSemen Gold extender is a suitable alternative to the INRA 96, when used within 48 hours; after 72 hours of storage, however, the INRA 96 showed a higher share of viable spermatozoa.

In vitro Fermentation of Rumen Microorganisms Cultured in Medium Supplemented with Bacterio-mineral Water (BMW) Produced from Bio-reacted Swine Manure

  • Bacterio-mineral water (BMW) produced from manure has been known to exert a number of positive effects on animal production and odor control. An experiment was conducted to examine the effects of BMW produced from bio-reacted swine manure on in vitro gas production, cellulose degradation, microbial growth and fibrolytic enzyme activities of mixed rumen microorganisms. The five levels of 0, 0.001, 0.005, 0.01 and 1.0% BMW were supplemented into serum vials containing mixed rumen microorganisms. Incubations were carried out anaerobically at $39^{\circ}C$ without shaking for 0, 12, 24, 48, 72 and 96 h. There were no significant (p>0.05) differences among the treatments for the initial rate of gas production. At 72 h incubation, the gas production tended (p<0.1) to be increased by the 0.01 and 1.0% BMW treatments compared with control and the 0.001% BMW treatment. At the end of incubation (96 h), the sample supplemented with 0.01% BMW was higher (p<0.05) than control (0% BMW) in the gas production. The microbial growth rate was increased by all the BMW treatments, while 0.01% BMW was most effective in stimulating the growth rate. Although the addition of BMW on the filter paper DM degradation was not significantly influenced throughout the incubation period except the 48 h incubation, DM degradation tended to be increased by all BMW treatments compared with control. The addition of both 0.005 and 0.01% BMW highly increased (p<0.05) CMCase activity compared with control after 24 h and 48 h incubation, while at the 72 h incubation the 0.01% BMW addition only significantly increased (p<0.05). After 72 h incubation, the xylanase activity was significantly (p<0.05) increased with the addition of 1.0% BMW compared with the addition of 0.001 and 0.005% BMW, while at the other incubation times, the xylanase activity was not different among the treatments. In conclusion, the 0.01% BMW of supplementation level would be the suitable addition level to stimulate rumen fermentation increasing microbial growth and cellulose degradation.
