Effects of Low Dose Gamma Radiation on the Early Growth of Pepper and the Resistance to Subsequent High Dose of Radiation (저선량 방사선이 고추의 초기생육과 후속고선량 내성에 미치는 영향)

  • Pepper (Capsicum annuum L. cv. Jokwang and cv. Johong) seeds were irradiated with the doses of 0-50 Gy to investigate the effect of the low dose ${\gamma}$-radiation on the early growth and resistance to subsequent high dose of radiation. Following effect was observed in Johong cultivar but not in Jokwang cultivar Germination and early growth rate of Johong cultivar were noticeably increased at 4, 8 and 20 Gy irradiation group. Resistance to subsequent high dose of radiation of Johong cultivar increased at almost all of the low dose irradiation group. Especially it was highest at 4 Gy irradiation group. Plant groups of Johong cultivar showing the resistance to subsequent high dose of radiation, which had been pre-irradiated with 4 Gy and 8 Gy, were also higher in the carotenoid contents and enzyme activity than the plant group not irradiated previously.

Effects of low dose $\gamma$-ray on the early growth of tomato and the resistance to subsequent high doses of radiation (저선량 $\gamma$선 조사가 토마토의 초기생육과 후속고선량 $\gamma$선 저항성에 미치는 영향)

  • Tomato (Lycopericum esculentum $M_{ILL}$ cv. Seokwang and cv. Housemomotaro) seeds were irradiated with the doses of $1{\sim}20$ Gy from $^{60}Co$ $\gamma$-ray source to investigate the effect of the low dose $\gamma$-ray radiation on the early growth and resistance to subsequent high dose of radiation. Germination rate of seeds irradiated with low dose $\gamma$-ray was enhanced in Seokwang cultivar but not in Housemomotaro cultivar. Seedling height increased in 4 Gy and 8 Gy irradiation group of both cultivars. Plant height of Seokwang cultivar was depressed in low dose irradiation group but fresh weight was increased in 2 Gy and 4 Gy irradiation group. In Housemomotaro cultivar, plant height increased in 12 Gy and 20 Gy irradiation group and fresh weight increased in 4 Gy and 20 Gy irradiation group. Growth inhibition of tomato plants by high dose radiation was noticeably reduced by pre-irradiation of low dose radiation. Resistance to subsequent high dose of radiation was enhanced in 2 Gy and 8 Gy Irradiation group of Seokwang cultivar and in 2 Gy and 12 Gy irradiation group of Housemomotaro cultivar.

  • Soybean (Glycine max L. cv. Hwangkeum) seeds were irradiated with the dose of $0{\sim}20$ Gy to investigate the effect of the low dose ${\gamma}-ray$ radiation on the early growth and resistance to subsequent high dose of radiation. Germination rate was not enhanced in the seeds irradiated with low dos ${\gamma}-ray$ but plant height and fresh weight increased in the low dose irradiation group. The optimal radiation dose for the growth increasing was 8 Gy in soybean plant. Growth inhibition of soybean plants by high dose radiation was noticeably reduced by pre-irradiation of low dose radiation, Resistance to subsequent high dose of radiation was effective in 8 Gy and 20 Gy irradiation group.

Assessment of Nucleus-DNA Damage in Red Pepper Cells Treated with γ-Radiation through Comet Assay (Comet 분석을 통한 방사선처리 고추세포의 핵 DNA 손상평가)

  • We employed single cell gel electrophoresis method (comet assay) to analyze the degree of nucleus-DNA damage in the leaves of red pepper (Capsicum annuum L.) seedlings exposed to $^{60}$ CO v-radiation stress. Nucleus-DNA damage was measured as the ratio of tail length (T) to head length (H) in individual comet image isolated from pepper leaf cell. The T/H ratio of control-cells and treated-cells at 50 or 100 Gy were 1.28 and 3.54 or 3.39, respectively, suggesting that nuclei of pepper cells were severely damaged in the integrity of DNA strand by the treatment of enhanced v-radiation. The percentage of head-DNA in control-cells was 76.8%, whereas those of 50 and 100 Gy treated-cells were 55.9% and 59.9%, respectively. Pretreatment of low dose (4 to 20 Gy) radiation to seeds decreased DNA-damage in the leaves of seedlings treated with high dose radiation at 50 or 100 Gy. In this experiment, we developed a sensitive, reliable and rapid method for evaluating genotoxic effect in the nuclei of plant cells by employing comet assay.
