Compositional Changes of Functional Oil from Algae Oil during the Lipase-Catalyzed Production (Algae유로부터 디글리세롤 함유 기능성 유지의 효소적 합성 중 구성성분 변화에 관한 연구)

  • In the absence of organic solvent, di- and mono-acylglycerol enriched functional oil was produced by lipase-catalyzed glycerolysis in a stirred tank batch reactor. After glycerolysis for 48 hr, functional oil consisted of $47.1\%\;TAG,\;33.6\%\;DAG,\;and\;18.7\%$ MAG. In MAG and DAG, docosahexaenoic acid was found as $10.7\%\;and\;8.1\%$, respectively. During the reaction, total tocopherol content decreased, and $0.065\%$ tocopherols were existed in the functional oil produced after 48hr reaction.

Synthesis and Characterization of Mono- and Diacylglycerol Enriched Functional Oil by Enzymatic Glycerolysis of Corn Oil (옥수수유로부터의 효소적 glycerolysis에 의한 monoacylglycerol과 diacylglycerol 함유 기능성 유지 합성 및 특성연구)

  • Mono-and diacylglycerol-enriched oil was produced from corn oil through enzymatic glycerolysis using 1,3-specific immobilized lipase in solvent-free system and stirred-tank batch reactor. HPLC analysis revealed enriched oil was respectively composed of: 45.05, 16.27, 23.05, and 14.98% triacylglycerol, 1,3-diacylglycerol, 1,2-diacylglycerol, and monoacylglycerol; 13.21, 0.15, 2.02, 34.36, 49.12, and 1.14 mol% palmitic, palmitoleic, stearic, oleic, linoleic, and linolenic acids; and 0.014, 0.029, 0.010 and 0.053% ${\alpha},\;{\gamma},\;{\delta}-$, and total tocopherols. Physiochemical and melting properties of enriched oil were evaluated. Oxidative stability study revealed enriched oil showed higher peroxide and p-anisidine values than corn oil. Rosemary extracts (100 to 300 ppm) reduce oxidation.

A Study on Alkyl Glucoside Synthesis by Amphiphilic Phase Enzyme Reaction (양친매상 효소반응에 의한 알킬글루코시드의 합성연구)

  • An amphiphilic phase enzyme reaction was used to synthesize alkyl glucosides from glucose and alkyl alcohol with immobilized ${\beta}$-glucosidase using four glycol ether cosolvents(monoglyme, diglyme, 2-methoxyethanol, and 1,4-dioxane). Monoglyme was shown to be the best cosolvent for the amphiphilic phase medium composed of water/cosolvent/alkyl alcohol admixture. To obtain high yield of alkyl glucoside by amphiphilic phase enzyme reaction, hydrophilicity-hydrophobicity of amphiphilic media and enzyme microenvironment was optimized from the viewpoints of substrate solubility, enzyme activity, water activity, and dynamic equilibrium between glucose alcoholysis and glucoside hydrolysis. Under optimum reaction conditions, the highest concentrations of hexyl, octyl, decyl, and dodecyl glucosides were 18.2, 9.68, 7.27, and 6.03g/L, respectively.

A Study on Glycoside Synthesis Using Alginate-enclosed Microspheres (Alginate-enclosed Microspheres를 이용한 배당체 합성에 관한 연구)

  • Latex microspheres of styrene/acryl copolymer with acrylamide functional group were used for the stable covalent immobilization of an enzyme applicable for enzymatic synthesis of glycoside. The latex microspheres were coated with polyethyleneimine to establish structural and functional properties relevant to the covalent Immobilization with a high retention of activity. Polythyleneimine-coated microspheres satisfactorily immobilized the invertase for methyl fructoside synthesis, and model reaction were formed into alginate-enclosed microspheres biocatalyst. Using the alginate-enclosed microspheres biocatalyst, the yield of model glycoside was obtained as high as 52.2% at concentration of aqueous 30%(v/v) methanol and 0.291mo1/1 sucrose solution with 2U/ml of activity. The present study showed that the latex microspheres were successfully applied to enzymatic synthesis of glycoside.

  • 효소는 생체내의 합성, 분해, 산화, 환원 등 복잡한 화학반응이 상온, 상압, 중성부근에서 효율적으로 진행되게 하는 단백질이 주성분인 유기촉매이다. 현재 알려져 있는 효소의 종류는 수백만종 이상으로 추정되며 그 중 100여종 이상은 순수한 결정상태이며 약 600종 정도는 어느 정도 순수하게 징제되고 있다. 이들 효소의 분자량은 Ribonuclease의 12,700에서 부터 L-Glutamate dehydrogenase나 Carboxylase의 1,000,000 이상으로 광범위하다.

  • 1980년대 초반에 'Neurosteroid'라는 개념이 소개 되면서, 지난 20년간 이 분야에 많은 연구결과들이 쌓이게 되었다. 그리고, neurosteroid는 과거에 알고 있었던 스테로이드들과는 다른 방법으로 생물체에 생리적 작용을 하는 것으로 알려졌으며, 특히 신경계 내에서는 신경전달물질과 같은 작용을 한다는 것이 알려지면서 신경계 내에서의 자체적인 스테로이드 호르몬 합성에 관한 연구들이 본격화되기 시작했다. 지난 10년 사이에 많은 review 논문들이 발표되고 있지만, neurosteroid와 신경계의 특성상스테로이드 호르몬의 신경계에 미치는 기능에 대한 연구가 쉽지 않으며, 정밀하게 조절되는 신경계의 스테로이드 함량변화를 조절하는 합성 효소에 대한 연구는 아직도 초보적인 단계에 머물러 있다. 이에 본 논단에서는 neurosteroid에 대한 소개와 함께 최근 우리실험실의 연구결과를 소개하고자 한다.

Inhibitory Effects of Bile Acids on the Cholesterol Biosynthesis in Cultured Hepatocytes (배양 간세포내에서의 콜레스테롤 합성에 대한 담즙산의 저해효과)

  • The present work tested the inhibitory effects of bile acids on the cholesterol biosynthesis and the activity of HMG-CoA reductase in cultured rat hepatocytes. The uptake of bile acids in hepatocytes were increased in according to the different bile acid concentrations and culture times. The rate of cholesterol synthesis in cells were inversely decreased to the bile acid concentrations and culture times. As expected, insulin injection (4 units/100g body weight) showed an enhancing effect of the cholesterol synthesis and the HMG-CoA reductase activity. The addition of bile acids in medium of insulin-treated hepatocytes also showed the suppressing effect. This effect was directly confirmed in isolated hepatic icrosomes by the test of HMG-CoA reductase activity. In the test of $Na^+$,$K^+$-ATPase activity in the isolated hepatocyte membrane, only the cholic acid did not stimulate the enzyme system. The reason of such difference is not obvious, but this result indicates that the cholic acid could be absorbed by simple diffusion.

Enzymatic Synthesis of Low Trans Fats Using Rice Bran Oil, Palm Stearin and High Oleic Sunflower Seed Oil (미강유, 팜스테아린 및 고올레인산 해바라기씨유를 이용한 저트랜스 유지의 효소적 합성)

  • Low trans fats were synthesized by interesterification of rice bran oil (RBO), palm stearin (PS) and high oleic sunflower seed oil (HO) using TLIM from Thermomyces lanuginosa. After 24-h reaction, physicochemical characteristics such as fatty acid and triacylglycerol composition, solid fat content, melting point, tocopherol, oryzanol and phytosterol contents were evaluated. Trans fatty acid contents of the produced low-trans fats showed less than 0.5 wt%. Mostly, triacylglycerol species in the products were palmitoyl-linoleoyl-oleoyl-glycerol (PLO), palmitoyl-oleoyl-oleoyl-glycerol (POO) and palmitoyl-oleoyl-palmitoyl-glycerol (POP). Total tocopherol contents ranged from 6.94 to 11.83 mg/100 g while $0.18{\sim}0.49$ mg/100 g of $\gamma$-oryzanol and $182.47{\sim}269.08$ mg/100 g of phytosterols were observed depending on the substrates ratios. When the content of PS in the reaction substrate was increased, solid fat content and slip melting points were increased.
