Purification and Characterization of Recombinant Monellin Produced from Yeast Culture Medium (효모 배양액으로부터 재조합 모넬린의 정제와 특성 연구)

  • The monellin, a sweet-taste protein, was expressed and secreted in Saccharomyces cerevisiae. The secreted menellin was concentrated using an ultrafiltration membrane with a nominal molecular weight cut off of 3,000 or by ammonium sulfate precipitation. The monellin was purified by G-25 gel filtration chromatography, followed by CM-Sepharose ion exchange chromatography. The purified monellin was characterized by SDS-PAGE (SDS-Polyacrylamide Gel Electrophoresis) and PHLC. The molecular weight of monellin was found to be 10,700 dalton, and its purity was over 95%.

  • 플루란은 글루코스 삼량체인 말토트리오스(maltotiose)를 반복단위로 하고, 그 삼량체와는 다른 결합인 $\alpha$-1,6결합에 의해 반복결합한 고분자 선상 중합체이다(Fig. 1) 플루란은 검정색 효모인 aureobasidium pullulans에 의해 배양 생산된 천연 다당류의 일종이다. 플루란은 원료가 저렴하기도 하지만 플루란으로 만들어진 막, 필름(1), 섬유(2) 등의 일반적인 성형물은 투명성, 강인성, 산소 불투과성이 있으며 땅속 수중에서 자연 분해되며 소각에 의해 유독가스와 고엽이 발생하지 않으므로 공해를 동반하지 않는다.(중략)

  • 어류나 갑각류 등의 종묘 생산을 위해 아직은 살아 있는 먹이 생물이 필수적이며, 대체 사료가 일부 개발되었지만 실제로 종묘생산시에 가장 많이 사용되고 있는 먹이 생물은 외국산 Artemia이다. 특히 몇 년 전부터는 Artemia 대량 수출국인 미국 유타주에서 Artemia 채취를 일부 금지시켰으며, 동남아시아에서도 자국의 종묘생산용을 제외하고는 수출용 Artemia 채취를 금지할 예정이므로 이로 인한 외화 낭비를 막기 위해 이에 대한 대체먹이 생물의 개발이 시급한 실정이다. (중략)

  • The present study was attempted to obtain the basic data concerning a reasonable preparing method and the optimum fermentation conditions of Kochujang (Red pepper paste). To establish the standard qualify of Kochujang, changes of the chemical composition and the numbers of bacteria and feasts in Kochujang during fermentation were observed. Moisture, salts and crude ash contents of Kochujang were not changed significantly during fermentation. Titrable acidity and amino nitrogen gradually increased with the time-passed, whereas crude fat gradually decreased with the time-elapsed. And reducing sugar and total nitrogen increased until 40 days, but slightly decreased after this period. The numbers of bacteria and yeasts in the ingrients for the preparation of Kochujang were $3.9{\times}10^7/g$, $1.5{\times}10^3/g$ in red pepper powder, $7.6{\times}10^4/g$, $2.8{\times}10^2/g$ in salts. respectively, but those of sugar and malt were not more than 100/g. Microbial counts in Kochujang during fermentation increased until 40 days, but those are gradually decreased after that.

  • In order to investigate the characteristics for RNA accumulation, continuous cultures of S. cerevisiae MTY62 under the carbon-limitation or carbon-phosphate-limitation were performed. In the carbon-limited chemostat, the highest RNA content of 144 mg-RNA/g-dry cell was observed at the dilution rate of $0.30\;hr^{-1}$. In the carbon and phosphate-limited chemostat, the culture condition with the dilution rate of $0.25\;hr^{-1}$ gave the maximal RNA content of 209 mg-RNA/g-dry cell. The accumulation rate of RNA $(mg-RNA/L\;{\cdot}\;hr)$ was decreased at higher dilution rate in the carbon and phosphate-limited chemostat.

  • For the purpose of obtaining microorganisms producing high fructosyl transferase, the screening test was carried out. Among more than three hundred isolates, an isolate (C23-isolate) was selected for high fructosyl transferase producer from the dirts at the coffee vendig machine. The morphological and cultural characteristics of the isolate C23 on various culture media were studied and identified as Aureobasidium pullulans var. melanigenum.

  • Effects of various elictitors were investigated to enhance the production of berberine in plant cell suspension cultures of Thalicrtrum rugosum. Treatments of yeast elicitor, 15 different types of abiotic elicitors, 16 kinds of fungal elicitors from three species of fungi were performed. Cell growth and berberine production were examined and compared for both normal and elicitor treated cultures. No distinguished increases in berberine yield by the addition of elicitors could be attained.

  • Automatic addition of glucose, ammonium and phosphate to alcohol distillery wastewater and their control at low concentrations have been carried increase the cell concentration of a baker's yeast cultivated in the wastewater. Glucose was automatically added using dissolved oxygen as the control parameter, and maintained below 300 mg/L. Ammonium was automatically added by a pH-stat method and maintained in the low range of 12.6~17.4 mM. An automated FIA system, which used an ascorbic acid-based method was developed for the automatic analysis nad addition of phosphate. With this system, the phosphate concentration was succesfully analysed and controlled afrer 19.4 hr in the range 23.3~43.4 mg/L. The cell concentration was increased by 33.0-fold by the addition of these three nutrients. The overall specific growth rate of the yeast was 0.19 $hr^{-1}$.
