  • 돼지 체세포 핵이식에 있어서 체외발달률을 재고하기 위하여 공여세포의 종류, 활성화 방법, 배양조건 등이 체외발달에 미치는 영향을 검토하였다. 도축장에서 채취한 난소에서 미성숙난자를 채취하여 NCSU-37 배양액에 10% pFF 와 0.6mM Cystein, 1mM dbcAMP, 0.1 IU/㎖ human menopausal gonadotrophin(hMG)에서 20시간 배양후 dbcAMP 와 hMG 가 없는 배양액에서 18-24 시간 추가배양 성숙 후 성숙된 난자를 핵이식의 수핵난자로 사용하였다. (중략)

  • 작약은 염증성 질환을 치료하기 위해 사용되어 온 전통 약용식물이다. 최근 작약은 대식세포에서 면역조절인자의 분비를 증가시키고 포식작용을 증가시킨다고 보고되었다. 그리하여 본 연구에서 추출조건별 작약의 대식세포 활성화 유도를 비교하였다. 온도조건 별 작약추출물은 4℃에서 60℃까지는 면역조절인자의 분비를 증가시켰지만, 80℃에서는 면역조절인자의 분비가 다소 감소하였다. 60℃에서 시간별 추출조건에서는 1시간에 24시간까지 면역조절인자의 분비가 유사하였다. 따라서 본 연구결과를 종합해 볼 때, 작약은 60℃에서 1시간 추출하는 것이 대식세포 활성화를 위한 최적 조건이라고 판단된다.

활성화 과정에 따른 돼지 단위발생란의 발달

  • 돼지 핵이식란의 발달은 다른 종의 것과 비교하여 효율이 낮다. 그 이유 중 하나가 난자활성화율이 낮기 때문이다. 따라서, 본 실험은 돼지 난자의 인위적 활성화처리에 따른 단위발생율을 조사함으로써, 이후 핵이식란 생산의 효율을 높이고자 수행되었다. 돼지 난포란을 10% pFF, 0.1mg/ml cysteine, 10IU/ml PMSG, 10IU/ml hCG, 10ng/ml EGF가 첨가된 TCM-199배양액에서 22시간 동안 배양한 후, 성선자극 호르몬이 배제된 배양액에서 추가로 22시간 동안 배양하여 성숙을 유도하였다. 체외성숙이 야기된 난자는 난구세포를 제거한 후 제2극체가 보이는 난자만을 선별하였다. 선별된 난자는 1)아무것도 처리하지 않은 대조군과 2)전기자극(2.0㎸/cm, 30$\mu\textrm{s}$), 2)7% ethanol에서 5분 배양 3)5$\mu$M ionomycin에서 4분 배양의 groups들로 나누어 활성화 처리 후 5분 동안 TCM-199에서 세정하고 다시 4시간 동안 6-DMAP에서 배양한 후 12시간에 난자를 염색하여 핵상을 분석하였고, 나머지는 4mg/ml BSA가 첨가된 NCSU-23에서 39$^{\circ}C$, 5%$CO_2$ 배양기에서 각각 6~7일 동안 배양을 실시하였다.

  • Bovine follicular oocytes were matured in vitro for 3D hr and exposed to Dulbecco's phosphate buffered saline containing 7% ethanol for 7 minutes for their activation, and incubated in TCM199 containing 10% fetal calf serum (FCS) and cytochalasin D (CD). When the oocytes were exposed to cytochalasin D for 0, 5, 10 and 15 hr, 5 hr showed highest developmental rate to blastocyst (16%), expanded blastocyst (13%) and hatched blastocyst (7%). Also when the oocytes were cultured for 7 hours in medium contai ning 0, 2.5, 5 and 7.5 ${\mu}\textrm{g}$/ml cytochalasin D, 2.5 ${\mu}\textrm{g}$/ml showed highest developmental rate to blastocyst (13%), expanded blastocyst (7%) and hatched blastocyst (4%). In conclusion, cytochalasin D played very important role for parthenogenetic development of follicular oocytes and 5 hr of exposure time to CD and 2.5 ${\mu}\textrm{g}$/ml CD in the medium was optimal for development of the follicular oocyte.

Effects of Maturation Duration and Activation Treatments on Activation and Development of Porcine Follicular Oocytes (돼지 난모세포의 단위발생에 있어서 성숙시간과 활성화 처리가 활성화와 발달에 미치는 영향)

  • This study is a part of research that development of effective genetic resources preservation system using the in vitro spermatogenesis, in vitro insemination and culture system. We aimed for establishment of in vitro culture system with in vitro activated porcine oocytes. The porcine oocytes were matured for 48 hours in $TCM199+10\%$ FCS and activated with $7\%$ ethanol. The activated oocytes were cultured for 7 days in $TCM199+10\%$ FCS or $NCSU23+0.4\%$ BSA medium. The activated oocytes were not developed to the blastocyst stage in $TCM199+10\%$ FCS medium. However in $NCSU23+0.4\%$ medium, those were developed to blastocyst with $3\%$ of treated oocytes. We extended maturation duration of porcine follicular oocytes fur 48, 52, 56, 60, 64, 68, and 72 hours and activated with $7\%$ ethanol and cultured using $NCSU23+0.4\%$ BSA medium. The six percents of activated oocytes were developed to blastocyst in 48 hours and $10\%$ in 52 hours with comparatively low rates suggested to be not fully activated by regenerated MPF. Maturation durations from 56 hours to 68 hours supported to develop upto $11.9\~18.3\%$ of blastocysts. However the developmental rate was declined to $7.2\%$ at 72 hours of maturation duration because of cytoplasmic deterioration. The assumed time window for activation will be $56\~68$ hours of maturation duration. When the matured oocytes were activated with electric pulse of 1, 1.2, 1.4, 1.6, 1.8 and 2.0kV/cm for $80{\mu}s$, although appling the electric current once was not enough for activation, appling twice with 1.6kV/cm for $80{\mu}s$ was shown the highest developmental rate with $11.3\%$. When those were compared with activating methods, $15.7%$ of blastocyst rate was obtained in the $7\%$ ethanol. That was higher than those in electric pulse with $9.5\%$ and calcium ionophore method with $5.8\%$. In this experimental condition, the $7\%$ ethanol treatment was the most effective method for activating porcine oocytes.

  • 최근 고해상도 영상이 필요하게 되었으며, 저해상도 영상을 고해상도 영상으로 변환하는 딥러닝 기반의 초해상도 알고리즘에 대한 연구가 활발히 진행되고 있다. 그럼에도 불구하고 딥러닝 기반의 초해상도 알고리즘은 하드웨어의 한계로 인해 임베디드 시스템에서 실행시간이 느린 단점이 있다. 본 논문에서는 심층신경망 기반의 초해상도 알고리즘의 네트워크 구조를 제시하고 다양한 활성화 함수에 따른 화질 및 실행시간 성능을 분석한다. 실험 결과, 젯슨 나노보드의 다양한 활성화 함수 중 화질과 실행 시간의 관계에서 도출한 최적의 활성화 함수가 PReLU 함수임을 확인하였다.

Manufacture of Activated Carbon based on Solid Residue after Lignin Pyrolysis (리그닌 열분해 잔류고형물을 원료로 한 활성탄의 제조)

  • In this study, activated carbon was prepared from solid residue after lignin pyrolysis by using zinc chloride as an activation agent. The steam activation method was adopted to manufacture activated carbon from solid residue after lignin pyrolysis. The effect of process operation variables such as activation temperature, activation time and mass of activation agent added to char on the pore structure and specific surface area of the activated carbon was investigated. Activated carbon with high surface area and well-developed pore structure could be prepared, when solid residue after lignin pyrolysis was mixed with zinc chloride of 300 wt% and then the mixture was activated for 1 hour at $1000^{\circ}C$ in a stream of nitrogen.

  • 본 연구는 색 조명 시각 자극이 정상인의 기억작용에 어떠한 영향을 미치는가를 평가해보고자 하였다. 피험자는 대학생 성인 남, 여 20대 (20-30세) 각각 10명을 대상으로 실시하였으며 자체 제작한 기억 테스트(Memory test)를 활용하였다. 색채 환경은 방음 암실에서 백색광원을 파장대별로 필터링한 색 조명을 사용하여 제시하였으며 실험 측정 파라미터로서는 정확도(accuracy), 반응시간(Response time) 그리고 학습시 주로 활성화 되는 뇌파인 알파와 세타파의 주파수 대역을 측정하여 기억기능을 수행하는 대뇌의 기억 활성화 상태를 색 조명에 따라 비교 분석 하였다. 측정결과 각각의 색조명에 대한 정확도는 파랑색에서 증가하고 반응시간을 노랑과 파랑에서 감소했다. 뇌파의 활성화에서도 흥분이나 긴장을 상징하는 빨강의 색보다는 파랑색과 보라색과 같은 단파장의 색이 알파파나 세타파를 활성화 시키는데 영향을 줌을 알 수 있었으며 이러한 연구가 감성적 색 조명 디자인의 응용이나 색 관련 제품 계발에 적극 활용될 수 있을 것이라 기대된다.

Production of Activated Carbon from Waste Walnut Shell Using Phosphoric Acid and Its Adsorption Characteristics for Heavy Metal Ion (인산활성화제에 의한 폐호도껍질을 원료로 한 활성탄제조 및 이의 중금속 이온 흡착특성)

  • The production characteristics of activated carbon from waste walnut shell have been investigated by taking activation temperature, activation time, amount of activating agent, and kind of activating agent as the major influential factors. The adsorption capacity of the activated carbon which was produced using phosphoric acid as the activating agent increased with activation temperature and showed its greatest value at about $550^{\circ}C$. Yield for activated carbon was observed to decrease continuously as the activation temperature was raised. The optimal activation time for the highest adsorption capacity was found to be about 2 hr, and as the activation time increased the yield for activated carbon was showed to decrease continuously. The increase in the amount of activating agent resulted in the increase of the yield for activated carbon, however, excessive amount of activating agent deteriorated its adsorption capacity reversely. The variations of the microstructure of activated carbon observed by SEM with several influential factors, correlated very well with its changes in the adsorbability with the same factors and the kind of activating agent was found to play an important role in the determination of the adsorption capacity of activated carbon. To investigate the adsorption characteristics of the produced activated carbon, the adsorption reactions of $Cu^{2+}$ ion were examined using the produced activated carbon as the adsorbent. In general, the kinetics of the adsorption of $Cu^{2+}$ ion was observed to follow a 2nd-order reaction and the rate constant for adsorption reaction increased as the initial concentration of adsorbate was diminished. The equilibrium adsorption of $Cu^{2+}$ was explained well with Freundlich model and its adsorption reaction was found to be endothermic. The activation energy for adsorption was calculated to be 13.07 kcal/mol, which implied that the adsorption reaction was very irreversible, and several thermodynamic parameters of adsorption reaction were estimated using van't. Hoff equation and thermodynamic relationships.

Application of CV Cycling to the Activation of the Polymer Electrolyte Membrane Fuel Cell (고분자 전해질막 연료전지의 활성화를 위한 CV 활성화법)

  • The activation process of the membrane-electrode assembly (MEA) is important for the mass production of the polymer electrolyte membrane fuel cell. The conventional activation process for the MEA requires excessive time and hydrogen gas and it might become the barrier for the commercialization of the fuel cell. The conventional activation process is based on hydrolysis of ion conducting membrane. In the study, we suggest the cyclic voltammetry (CV) technique as an on-line activation process and the CV activation process consists of two steps : 1) the humidification of the polymer electrolyte membrane and the electrode with 100% RH humidified nitrogen ($N_{2}$) gas, and 2) the removal step of the oxide layer on the surface of the Pt catalyst with CV cycling. The cycling reduces the activation time of the MEA by 2.5 h and use of hydrogen gas by 1/4.
