Plasmid profiling of multi-drug resistant Vibrio sp. isolated from influent and effluent water samples of fish farms in Jeju, South Korea (제주 양식장 유입수과 방출수에서 분리한 다제내성 Vibrio균 플라스미드 프로파일링)

  • The objective of this study was to investigate the plasmid profiling of multi-drug resistant (MDR) Vibrio in influent (inflow) and effluent (discharged) water samples of fish farms in Jeju, South Korea. MDR isolates identified through disc diffusion susceptibility tests, were subjected to plasmid profiling. One hundred fifty Vibrio isolates were obtained from each influent and effluent water sample. All MDR isolates were subjected to plasmid profiling. Greater number of bacteria were enumerated from effluents (61%) comparing to influents (39%). High incidence of neomycin, sulfamethoxazole, amoxicillin and oxytetracycline resistance was observed among the isolates, which was higher in effluent samples. In contrast, Vibrio isolates were more susceptible to florfenicol, chloramphenicol, ciprofloxacin, and nalidixic acid. Among 99 (influent 39 and effluent 60) MDR isolates, a total of 58 (influent 38 and effluent 20) were found to bear plasmids ranging from 1.7 kb to >10 kb and showed 19 different antibiograms according to the size of plasmids. MDR isolates showed six and four distinct plasmid profiles in influent and effluent, respectively. Effluent samples contained more plasmid-carrying MDR Vibrio isolates with more diverse plasmid profiles and antibiograms, suggesting that fish farm tanks may serve as a reservoir of antibiotic resistance genes. The presence of plasmid-carrying MDR Vibrio isolates in fish farm effluent water may contribute to the dissemination of antibiotic resistance genes to the environments, which ultimately poses threat to human health.

A Study on Fire Spread and Evacuation Risk of Conduit Combustion in Ceiling Hiding Place (천장 은폐장소 전선관 연소에 따른 화재확산 및 피난 위험도에 관한 연구)

  • In this study, the ISO 5660 and ISO 5659 combustion tests were conducted with synthetic resin conduits (CD, VE) and metal conduit (ST) used for wiring work in electrical facilities, which can be installed in ceiling concealed places. Then, fire spreading and evacuation risks were analyzed based on the measured data. In the ISO 5660 test, CD of 120.5 MJ/㎡, VE of 81.9 MJ/㎡, and ST of 4.9 MJ/㎡ were measured. In the ISO 5659 test, the CD 1320, VE 731, and ST 102 were measured, and then the maximum smoke densities were measured for CD 605 s, VE 740 s, and ST 1,200 s. In terms of fire spreading and evacuation risk, the CD conduit, VE conduit, and ST conduit were in order. In the fire spreading risk analysis, total heat emission was calculated as 4,820 MJ/㎡, 4,267 MJ/㎡, and 196 MJ/㎡ for CD, VE, and ST, respectively. Evacuation risk analysis shows at transmittance of 89%, CD is 127 s, VE is 35 s, and ST is 969 s. At transmittance of 79%, representing almost invisible concentration, CD is 157 s and VE is 50 s. The CD and VE conduits had a high fire spreading and evacuation risks, while the ST conduit had little effect on fire spreading and evacuation risk.

Preparation and Release Profile of N8f-loaded Polylactide Scaffolds for Tissue Engineered Nerve Regeneration (조직공학적 신경재생을 위한 NGF를 함유한 PLA 담체의 제조 및 방출)

  • We developed the nerve growth factor (NGF) loaded poly (L - lactide) (PLA) scaffolds by means of emulsion freeze drying method to the possibility for the application of the nerve regeneration of spinal cord disease and the degeneration in Alzheimer's disease. The release amount of NGF from NGF loaded PLA scaffold were analyzed over a 4 week period in vitro at phosphate buffered saline (PBS), pH 7.4, at $37^{\circ}C$. It can be observed the open cell pore structure of porous scaffolds and can be easily controlled the pore structure by the controlling of formulation factors resulting in the controlling of the release rate and the release period. The stability of NGF during the preparation of PLA scaffold was evaluated by comparing the released amounts of total NGF, assayed NGF enzyme - linked immunosorbent assay (ELISA). Released NGF has been found to enhance the neurite sprouting and outgrowth from pheochromocytoma (PC-12) cells. These results suggest that the released NGF from NGF loaded PLA scaffold such as conduit type can be very useful for the nerve regeneration in the neural tissue engineering area.

  • Direct solid analysis of various kinds of metal samples has been conducted by glow discharge. In this laboratory, the gas-jet assisted glow discharge(GJGD) device has been developed and characterized. The effect of changes in applied current, cell pressure and flow rate on atomic emission signals obtained from a jet-assisted cathodic sputtering was investigate. The emission intensities of Cu, Zn, and Ar were measured. They were increased with the current. But the intensities were decreased by increasing the flow rate of argon due to the diffusion and transportation of particles into plasma. By increasing the pressure of the cell, the intensities were greatly decreased because of enhancement of redeposition onto the surface of the sample.

Cesium Release Behavior during the Thermal Treatment of High Bum-up Spent PWR Fuel (고연소도 경수로 사용후핵연료의 열처리에 따른 세슘 방출거동)

  • The dynamic release behavior of Cs from high burn-up spent PWR fuel was experimentally performed under the conditions of a thermal treatment process such as voloxidation and sintering conditions. In voloxidation process, influence of the oxidation and reduction atmosphere on the Cs release characteristic using fragment type of spent fuel heated up to $1,500^{\circ}C$ was compared. In sintering process, temperature history effect on Cs release behavior was evaluated using green pellet under 4% $H_2/Ar$ environment. Temperature range for complete Cs release from spent fuel fragment under voloxidation condition was about $800^{\circ}C{\sim}1,200^{\circ}C$, but that of green pellet under the reduction atmosphere was $1,100^{\circ}C{\sim}1,400^{\circ}C$. Key parameters on Cs release behavior from spent fuel was powder formation as well as the diffusion rate of Cs compound to grain boundary and fuel surface.

  • A two-dimensional turbulence model for the surface discharge of heated water into cross-flow field has been developed. The depth-averaged continuity, momentum and temperature equations, are solved by an efficient finite-difference procedure known as SIMPLE. Turbulent stresses and heat fluxes are determined from a depth-averaged version of the $textsc{k}$-$\varepsilon$ equation. Results of test run clearly demonstrate its effectiveness in handling strong turbulent phenomena in very shallow near-field region.

Prediction of VOCs Emissions from Multi-layers Materials (복합자재에서의 VOCs 방출량 예측에 관한 연구)

  • The purpose of this study is to predict VOCs emission rates from multi-layers materials, which are composed of single-layer materials having various VOCs emission rates, by using effective diffusion coefficients of the single-layer materials. The study was consisted of two parts; the one is the prediction of VOCs emission rates from multi-layer materials through numerical methods. The other is the measurement of VOCs emissions rates of wall composite and floor composite in Mock-up rooms for comparing the prediction and the experiments' values. The results of the study show that the short-term VOCs emission rates of multi-layers materials can be predicted from the effective diffusion coefficients of single materials in odor accuracy.

Dispersion of High Temperature and High Salinity Water Discharged from Offshore Desalination Plant (해상 담수화 공장에서 배출되는 고온고염 해수의 확산예측)

  • Dispersion of high temperature and high salinity water discharged from a desalination plant is numerically estimated to investigate its impact on marine environment. The plant is installed on a floating barge located in Jinhae Bay and takes 200 tons of seawater per day. Fifty tons of intake are changed into fresh water, while 150 tons of those are discharged as the water of 15℃ warmer and 1.33 times saltier than surrounding seawater. In this dispersion model, advection is described by two-dimensional tidal currents and turbulent diffusion is simulated by Monte Carlo technique. Decay of water temperature is modelled by heat exchange between the atmosphere and the ocean, while decay of water salinity is ignored. The distributions of temperature and salinity come to equilibrium when the dispersion model is run for 100 days for temperature and for 365 days for salinity, respectively. At equilibrium state the water temperature and salinity rise 0.01℃ and 0.001‰ higher than ambient seawater, respectively.

  • Three-dimensional wind field and atmospheric dispersion models have been developed for estimating the concentration distributions of radioactive materials released into atmosphere. The field tracer experiment near the Kori nuclear power plant located over complex terrain was carried out for validating the atmospheric dispersion model. The wind fields were one of the most important factors for calculating the concentration. Therefore several numerical simulations using the measured wind data were performed to get more accurate concentration distributions compared with the analyzed values of the tracer gas. The calculated concentration distributions agreed well in the case of the usage of the more measured wind data in wind field model.

Development of Three-Dimensional Trajectory Model for Detecting Source Region of the Radioactive Materials Released into the Atmosphere (대기 누출 방사성물질 선원 위치 추적을 위한 3차원 궤적모델 개발)

  • Background: It is necessary to consider the overall countermeasure for analysis of nuclear activities according to the increase of the nuclear facilities like nuclear power and reprocessing plants in the neighboring countries including China, Taiwan, North Korea, Japan and South Korea. South Korea and comprehensive nuclear-test-ban treaty organization (CTBTO) are now operating the monitoring instruments to detect radionuclides released into the air. It is important to estimate the origin of radionuclides measured using the detection technology as well as the monitoring analysis in aspects of investigation and security of the nuclear activities in neighboring countries. Materials and methods: A three-dimensional forward/backward trajectory model has been developed to estimate the origin of radionuclides for a covert nuclear activity. The developed trajectory model was composed of forward and backward modules to track the particle positions using finite difference method. Results and discussion: A three-dimensional trajectory model was validated using the measured data at Chernobyl accident. The calculated results showed a good agreement by using the high concentration measurements and the locations where was near a release point. The three-dimensional trajectory model had some uncertainty according to the release time, release height and time interval of the trajectory at each release points. An atmospheric dispersion model called long-range accident dose assessment system (LADAS), based on the fields of regards (FOR) technique, was applied to reduce the uncertainties of the trajectory model and to improve the detective technology for estimating the radioisotopes emission area. Conclusion: The detective technology developed in this study can evaluate in release area and origin for covert nuclear activities based on measured radioisotopes at monitoring stations, and it might play critical tool to improve the ability of the nuclear safety field.
