홍화자 추출물의 치주 경조직 재생 촉진 효과

  • 최근 치의학 영역에서 사용되는 천연물 특히 생약제에 대한 연구가 활발히 진행되고 있다. Scutellariae Radix, Centella asiatica 등이 치주인대세포의 활성을 증가시키고 홍삼사포닌이 배양한 치주인대세포의 성장, 분화에 관계된다는 보고 등이 이에 해당된다. 본 연구에서는 홍화자 메탄을 추출물과 키토산의 치주인대 세포의 증식, 분화, 석회물 결정 생성 촉진작용을 검토하여 치주경조직 재생 약물로서의 사용여부를 보고자 하였다. 치주인대 세포는 교정치료목적으로 경희의료원에 내원한 환자의 제1 소구치를 발거하고 치근시작점에서 중앙으로 1/3되는 지점에서 치주인대 조직을 절취하여 1차 배양하였다. 실험에는 계대배양하여 5-7 세대의 세포를 사용하였다.

Effects of Safflower (Carthamus tinctorius L.) Seed Powder on Fracture Healing in Rats (홍화(Carthamus tinctorius L.)씨 분말의 랫드 골절에 대한 치유 효과)

  • Safflower (Carthamus tinctorius L.) is a thistle-like annual plant mainly grown in dry hot climates as an oilseed or birdseed. Traditionally, the oil has mainly been sold in the health food market because it is unsaturated having high linoleic and oleic acid levels. With increased health consciousness in recent years, the oil quality has become a more general health issue. This study was designed to understand whether safflower seed powder has positive effects on the fracture healing in rats. Simple transverse fracture of rat fibula was made with a rotating diamond disc saw. The histologic changes of rats were observed with a scanning electron microscope. The fractured fibulae showed a complete fusion at the fracture site in the 4th to 5th week after a simple transverse fracture. Administration of safflower seed powder facilitated the speed of histologic changes without affecting qualitative changes. These results suggest that safflower seed powder nay have substances that help the fracture healing process.

  • Mt. Jiri, where is located in southwestern area of Korea, can be a treasury of herbal medicine plants grown wild. More than 1200 species currently classified as herbs are grown or cultivated. Recently, the studies on the safflower which attracts the interests in the inhibition of skin aging in the modern people have been actively investigated, and this study aims to confirm that polyphenol compound, the main ingredient of safflower, can be a natural material to cause the delayed effect on the skin aging, inhibiting the generation of wrinkle in the 30 to 40s middle aged females. Primarily, this study is to confirm the anti-oxidizing effect by DPPH assay in vitro, and to evaluate the efficacy of inhibition or improvement on the wrinkle generation in the face skin by clinical trials, additionally. In addition, we are confident to contribute on the improvement of functional cosmetic industry, developing the sample product of anti-aging mask pack which has the main ingredient with the extract of safflower.

Analysis on Red-colored Dyeing by using Non-destructive UV-visible and Fluorescence Spectrophotometry (비파괴 자외-가시 및 형광 분광 분석법을 이용한 적색계 전통 염료 분석)

  • This study examined non-destructive UV-Vis spectrophotometry as well as 3-D fluorescence spectrophotometry of textile that made use of red dye such as Sappan wood, madder, Safflower, Gromwell. The authors produced two textile specimen that were dyed by not only two kinds of textile (cotton and silk) but also three kinds of mordanting (no-mordanting, alumen and iron), and they investigated effects of each dye material upon investigation results. At analysis with UV-Vis spectrophotometry of dyed textile specimen, dyeing made by sappan wood, madder and gromwell had significant difference depending upon mardant regardless of kinds of textile, and safflower had no significant difference depending upon textile and mordant. At analysis with 3D-fluorescence spectrophotometry, specimen dyed with sappan wood had difference with mordants, and with madder, there were difference with textiles, and safflower had inherent fluorescence spectrum regardless of textiles and mordants, while gromwell had no fluorescence spectrum.

Major Morphological Characteristics of Safflower(Carthamus tinctorius L.) Accessions (홍화 수집종의 주요 형태적 특성)

  • Morphological characteristics of 16 collected safflower (Carthamus tinctorius L.)accessions were studied at Gyongbuk A.T.A. Uisong Medicinal Plant Experiment Station in 1998. As for plant height, 5 lines were tall(more than 131cm), which were mainly foreign safflower. Middle and small were 5 and 6 accessions, respectively. Number of branch per plant was 11∼13 in 2 accessions of domestic safflowers while foreign safflowers have more than 50 branches per plant. Leaf shapes were oblong(3 lines), lanceolate(10 lines) and broadly linear(3 lines). There were three types of leaf margin shape which were repand(7 lines), serrate(4 lines) and parted serrate(5 lines). Flower color was white(1 line), yellow(13 lines) and red orange(2 lines). As for flower head shape, 14 lines were conical and 2 lines were flattened. There were thorns on bract of flower head in 11 lines but in 5 lines there were not. Domestic safflower lines had seeds of conical shape. Contrastively, seed shapes of foreign saffower lines were oval.

Effects of Dietary Safflower Seed Powder and Chitosan on Hatching Egg Production and Eggshell Quality in Aged Egg-type Breeder Hens (홍화씨 분말 및 키토산의 사료 내 첨가가 종란 생산성 및 후기 난각질에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary safflower seed (SFS) powder and chitosan on hatching egg production and eggshell qualify in aged egg-type breeder hens. A total of four hundred 54-week-old Hy-Line Brown breeder hens were divided into five the groups and fed experimental diets either with addition of 0.25 and 0.50% SFS powder or 0.10 and 0.20% chitosan or devoid of all for 6 weeks. There were no significant differences in feed intake and laying performances among the groups. The rate of sellable egg in the groups fed diets containing SFS powder or chitosan was significantly increased compared to that of control (P<0.05), irrespective of dose-dependent manner. The addition of SFS powder or chitosan significantly improved the eggshell strength (P<0.01). Eggshell thickness was also significantly increased in the groups 134 diets containing chitosan as compared with that of control (P<0.01). Fertility and hatchability of egg set were significantly improved by dietary chitosan that those of control (P<0.05). ike contents of tibial Ca and P were significantly increased in the groups fed diets containing 0.5% SFS powder, 0.1 and 0.2% chitosan as compared with those of control. The levels of blood estrogen, calcitonin and parathyroid hormone were not affected by the dietary treatments. The overall results indicated that the proper use of some feed additives such as safflower seed powder and chitosan might provide means of improving eggshell quality and reproductive performances in aged egg-type breeder hens.

Effect of Carthami Tinctorii Fructus Herbal-acupuncture Solution(CTF-HAS) on Gene Expression in HepG2 carcinomar cells (Oligonucleotide chip를 이용한 홍화자약침액(紅花子藥鍼液)이 간암세포주(肝癌細胞柱)의 유전자(遺傳子) 발현(發顯)에 미치는 영향(影響))

  • Objective : It has long been known about the osteogenic effect of CTF-HAS on bone tissues. However, it has not been determined the effect of CTF-HAS on cancer cells. The purpose of this study is to screen the CTF-HAS mediated differentially expressed genes in cancer cells such as HepG2 hepatoma cells lines. Oligonucleotide microarray approach were employed to screen the differential expression genes. Methods : CTF-HAS was prepared by boiling and stored at $-70^{\circ}C$ until use. Cells were treated with various concentrations of CTF-HAS(0.1, 0.5, 1.5, 10, $20mg/m{\ell}$) for 24 h. Cytotoxicity was tested by MTT assay. To screen the differentially expressed genes in cancer cells, cells were treated with $1.5mg/m{\ell}$ of CTF-HAS. For oligonucleotide microarray assay, total RNA was used for gene expression analysis using oligonucleotide genechip (Human genome U133 Plus 2.0., Affimatrix Co.). ResuIts : It has no cytotoxic effects on HepG2 cells in all concentrations (0.1, 0.5, 1.5, 10, $20mg/m{\ell}$). More than twofold up-regulated genes were 19 genes. The number of more than twofold down-regulated genes was 13. Discussion : This study showed the screening of CTF-HAS mediated differentially regulated genes using combined approaches of oligonucleotide microarray. The screened genes will be used for the better understanding in therapeutic effect of CTF-HAS on cancer field.

Efficiency of Dyes Extraction and Dyeing of Safflower according to pH Condition (pH에 따른 홍화(紅花)의 색소(色素) 추출(抽出)과 염색(染色)의 효율성(效率性)``)

  • Effect of the pH condition was examined on extraction and dyeing of the yellow and red dyes of safflower. Absorption of dye solution extracted from safflower was changed by pH of solvent. In the case of yellow dyes, cotton and ramie weren't dyed regardless of pH of dye bath, but silks were significantly dyed at pH4. Raw silk showed better dyeability than refined silk. In the case of red dyes, cotton and ramie absorbed red dyes selectively, but silks absorbed yellow dyes more significantly than red dyes. Efficiency of extraction and dyeing of yellow dyes in acidic range were find out higher than that in neutral range. Red dyes was effectively extracted at pH11 and dyed at pH6.

Effect of Various Natural Antioxidants on the Safflower Oil (홍화유에 대한 천연 항산화제의 항산화력 비교)

  • To elucidate the effect of various natural antioxidants on the safflower oil, rosemary extract, green tea extract, isoflavon, Phaffia rhodozyma extract, tocopherol, sesamol, and BRA as a control were added to the safflower oil and stored at 60$^{\circ}C$ for 4 weeks. During storage, its viscosity and antioxidative activity were determined. Viscosity of the oil increased with increasing storage period and was related with its antioxidative acvtivity. Antioxidative avtivity among antioxidants used in this study were in order of green tea extract>BHA>tocopherol>rosemary extract>isoflavon>sesamol>Phaffia rhodozyma extract. Green tea extract was the best in terms of antioxidative acvtivity. After 4 weeks, its peroxide value decreased up to 80.4% of the control, and 42.1% for acid value and 47.4% for TBA value decreased.
