Effect of Lysozyme, Glycine, Lysine and EDTA on the Growth of Lactobacillus plantarum (Lysozyme과 Glycine, Lysine 및 EDTA의 첨가가 Lactobacillus plantarum의 성장에 미치는 영향)

  • Effect of 200 ppm lysozyme. 0.1 glycine and 0.1% lysine on inhibition growth of Lactobacillus plantarum was investigated in MRS broth. All samples except control were effective in inhibiting the growth and especially the combination of lysozyme and glycine was observed to be highly effective. The mixture effect against microbial growth was increased as concentration of lysozyme with glycine or lysozyme with EDTA was respectively increased. Lactobacillus plantarum almost didn't grow in MRS broth containing lysozyme(>200 ppm) with glycine(>0.5%), or lysozyme(>200 ppm) with EDTA(> 0.8 mM). It was found that the growth of L. plantarum could be extremely inhibited in 120 ppm lysozyme with at least 0.8 mM EDTA compared with control.

Effect of Premixing of TDEAT and $\textrm{NH}_3$ on TiN Formation (TDEAT와 $\textrm{NH}_3$ 예비혼합 처리가 MOCVD TiN형성에 미치는 영향)

  • TDEAT(TI[N(C$_{2}$H$_{5}$)$_{2}$]$_{4}$)와 NH$_{3}$반응기체를 이용하여 MOCVD(Metal Organic Chemical Vapor Deposition)TiN 박막을 형성하였다. 반응기체들은 chamber내에 주입하기 전에 적절한 기상반응을 유도시켜 주었으며, TiN박막 형성에 미치는 예비혼합 효과를 관찰하였다. 두 반응기체의 예비혼합을 이용하여 낮은 탄소의 함유와 함께 -800$\mu$Ωㆍcm의 비교적 낮은 비저항을 나타내었다. 또한 NH$_{3}$의 유량 증가에 따라 도포성이 상당히 증가되고 있는데 이같은 도포성 향상 효과는 기상반응에 의하여 형성되는 중간상의 낮은 흡\ulcorner계수에 기인하는 것으로 여겨진다. QMS(Quadruple Mass Spectrometer)분석을 이용하여 두가지 경쟁적 반응을 포함한 전체 반응식을 제시하였다. TDEAT/NH$_{3}$혼합증착원의 경우 particle이 관찰\ulcorner지 않았으며 이것은 기상반응의 정도를 효과적으로 조절한데 기인하는 것으로 여겨진다. 결과적으로 반응기체의 예비혼합은 막질 및 도포성 개선과 함께 particle생성억제에 매우 효율적인 방법임을 알 수 있었다.다.

Robust ridge regression for nonlinear mixed effects models with applications to quantitative high throughput screening assay data (비선형 혼합효과모형에서의 로버스트 능형회귀 방법과 정량적 고속 대량 스크리닝 자료에의 응용)

  • A nonlinear mixed effects model is mainly used to analyze repeated measurement data in various fields. A nonlinear mixed effects model consists of two stages: the first-stage individual-level model considers intra-individual variation and the second-stage population model considers inter-individual variation. The individual-level model, which is the first stage of the nonlinear mixed effects model, estimates the parameters of the nonlinear regression model. It is the same as the general nonlinear regression model, and usually estimates parameters using the least squares estimation method. However, the least squares estimation method may have a problem that the estimated value of the parameters and standard errors become extremely large if the assumed nonlinear function is not explicitly revealed by the data. In this paper, a new estimation method is proposed to solve this problem by introducing the ridge regression method recently proposed in the nonlinear regression model into the first-stage individual-level model of the nonlinear mixed effects model. The performance of the proposed estimator is compared with the performance with the standard estimator through a simulation study. The proposed methodology is also illustrated using quantitative high throughput screening data obtained from the US National Toxicology Program.

Inhibition of Nitric Oxide Production and Hyaluronidase Activities from the Combined Extracts of Platycodon grandiflorum, Astragalus membranaceus, and Schisandra chinensis (길경, 황기와 오미자 혼합추출물의 NO 억제활성과 Hyaluronidase 억제활성 효과)

  • In this study, the optimal extraction conditions for three medicinal herbs as functional sources against inflammatory and arthritic diseases were developed. Traditional medicinal herbs were screened for their inhibition of hyaluronidase (HAse) activity and nitric oxide (NO) synthesis. For the screening of anti-inflammatory properties, ethanolic extracts of 53 species of traditional medicinal herb were examined. We confirmed that Astragalus membranaceus (A.R.), Schisandra chinensis (S.F.), and Platycodon grandiflorum (P.G.) inhibit NO production. For extraction from all three herbs simultaneously, an ethanol concentration of 95%, a 1:2:1 mixture ratio, and at 50 rpm mixing speed, for over 12 h and at $30^{\circ}C$ was the best condition for optimal extract yield and NO inhibition effects. HAse inhibition from the three herb extraction was three fold higher than single samples. The ethanol extracts were fractionated with various solvents (n-hexane, chloroform, ethyl acetate, n-butanol, and water). The ethyl acetate-soluble fraction of the herb mixture showed the highest extract yield (13%) and NO inhibition effects (73%). In conclusion, this study provides experimental evidence that a mixture of P.G., A.R., and S.F. could be used as a source of antioxidant ingredients in the food industry.

Effect of Aeration Mechanism on Livestock Manure Liquid Fertilization (폭기형태가 돈분뇨 액비 부숙특성에 미치는 영향)

  • Three types of aeration system were installed in experimental liquid fertilization tanks to investigate the change of characteristics of pig slurry used as a raw material for making livestock liquid fertilizer. The aeration systems of the reaction tanks were composed of three major part: the air suppling part (blower), the air pipe part, and the air diffuser part. In the first tank (reactor A), the air was supplied from the bottom of the reaction tank through air pipe system connecting air diffuser with commercial ordinary blower. In the second tank (reactor B), the air diffuser was located 10cm above the bottom of the reactor. In the third tank (reactor C), the pure air was supplied with circulating pjg slurry. The oxygen content of pure air was about 90%. The pure air was mixed with pig slurry by mechanically in the air suppling part (blower) and the air pipe part. The agitation effect was highest in the reactor C than other reactors. The contents of SS, COD, T-N and T-P of each samples collected at middle part of all reactors were 8,500, 4,188, 694 and 422mg/L; 9,000, 4,247, 813 and 356mg/L; 8,667, 6,910, 973 and 269mg/L, respectively.

Synergistic effects of grape branch and Pleurotus eryngii extract combination against inflammation on activated mast cells and atopic dermatitis-like skin lesions in mice (포도가지와 새송이버섯 혼합 추출물의 항염증과 아토피 피부염 개선 상승효과)

  • The aim of this study was to investigate the synergistic anti-inflammatory and anti-dermatitis effects of grape branch extract (GBE) and Pleurotus eryngii (PEE) combinations on the active immune cells and atopic dermatitis-like skin lesions in mice. The results showed that the combination of GBE ($12.5{\mu}g/mL$) and PEE ($500{\mu}g/mL$) led to much stronger inhibitory effects on the production of inflammatory mediators, such as NO and $PGE_2$ than that exhibited by GBE ($25{\mu}g/mL$) and PEE ($1000{\mu}g/mL$) alone, even at higher concentrations, in LPS-stimulated RAW 264.7 macrophages. The combination of GBE and PEE synergistically inhibited the production of TNF- and IL-6 in LPS-stimulated RAW 264.7 macrophages and PMA plus A23187-activated HMC-1 cells. Furthermore, combined GBE and PEE had a stronger ameliorative effect than GBE and PEE alone by inhibiting the clinical sores, IgE, and IL-4 on atopic dermatitis-like skin lesions in mice. Collectively, these results suggested that the combination of GBE and PEE produced a synergistic anti-inflammatory and anti-atopic dermatitis effect on immune cells and atopic dermatitis-like skin lesions in mice.

Soot Formation Characteristics of Concentric Diffusion Flames with Mixture Fuels (이중동축류 화염을 이용한 혼합연료의 매연생성 특성에 관한 연구)

  • The synergistic effect of ethylene/propane and ethylene/methane mixtures on soot formation is studied experimentally with a concentric co-flow burner. The integrated soot volume fractions, laser light scattering signal and PAH concentrations are measured for different fuel supply configurations. The synergistic effect in ethylene/propane diffusion flames is found to be affected not only by the composition of mixture but also by the way of mixing. Comparing to the homogeneously mixed ethylene/propane case, the increase of soot formation is observed when propane is supplied through the inner nozzle, while the decrease is observed when propane is supplied through the outer nozzle. However, the measured PAH concentration distributions are inconsistent with the current view of the synergistic effect of ethylene./propane mixture on soot formation. Virtually no synergistic effect is observed in ethylene-methane flames regardless of the fuel supply configuration, which suggests the important role of $C_3$ species produced during the propane pyrolysis process for the synergistic effect.

Structure Identification and Cage Occupancy Analysis of the Mixed Gas Hydrates Containing 1-Propanol and 2-Propanol (propanol계 혼합 하이드레이트의 구조 및 동공 점유 분석)

  • 일반적으로 알콜계 물질은 가스 하이드레이트 형성에 저해제로 사용된다고 알려져 있으나, 최근의 연구를 통해 2-propanol이 일부 조건에서 촉진효과가 나타난다고 보고되고 있다. 본 연구에서는 알콜계 물질인 1-propanol과 2-propanol의 가스 하이드레이트 격자내로의 포집여부와 그로 인한 촉진 혹은 저해 작용 그리고 구조적 특성에 대해 알아보았다. $CO_2$$CH_4$ 기체에 대하여 1-propanol 혹은 2-propanol을 첨가하여 형성된 혼합 하이드레이트의 3상평형 기상(V)-물(Lw)-하이드레이트(H))을 측정하였다. 그 결과 $CO_2$의 경우 1-propanol과 2-propanol이 저해 작용을 함을 확인하였으며 농도가 높을수록 저해작용이 커짐을 알 수 있었다. 반면, $CH_4$의 경우 1-propanol에서는 저해 및 촉진효과가 거의 나타나지 않았지만, 2-propanol에서는 촉진효과가 나타났으며 5.6 mol%에서 촉진효과가 가장 크게 나타났다. 혼합 하이드레이트의 구조규명 및 동공 점유 분석을 위해 $^{13}C$ NMR과 XRD분석을 하였으며, 그 결과 2-propanol과 1-propanol을 포함하는 혼합 하이드레이트는 구조 II를 형성하며, 2-propanol과 1-propanol은 큰 동공에 포집되고, 기체는 작은 동공에 포집됨을 확인할 수 있었다. 본 연구의 결과는 알콜계 물질을 첨가제로 사용하는 가스 하이드레이트 공정에서 매우 유용한 기초자료가 될 것으로 사료된다.

Antioxidant Activity of Ginseng in a Chicken Model System (닭고기 model system에서의 인삼의 항산화 효과)

  • Antioxidant activity of ginseng was studied in a chicken model system containing sodium nitrite, sorbic acid and ascorbate with or without ginseng. Results obtained are as follows. 1. The antioxidant effect of ginseng increased with increasing concentration but the increment decreased with increasing concentration. 2. In the sample containing sodium nitrite, sorbic acid and ascorbate, the addition of ginseng enhanced the antioxidant effect. Treatment with ginseng, nitrite, sorbic acid and asrorbate( G-NaSoAs) showed the highest antioxidant activity whereas that with nitrite and sorbic acid(NaSo) did the lowest.
