Anti-obesity Effect of Aster Yomena Ethanol Extract in High Fat Diet-induced Obese Mice (고지방 식이로 유도된 비만 생쥐에서 쑥부쟁이 에탄올 추출물의 항비만 효과)

  • Aster yomena (AY) have been used as a traditional medicine to treat cough, bronchial asthma, and insect bites in Korea. In this study, we evaluated the inhibition of adipogenesis in 3T3-L1 cells and in high-fat diet (HFD)-induced obese mice by AY ethanol extract. Lipid accumulation measurement indicates that AY markedly inhibited adipogenesis in a dose-dependent manner. qRT-PCR results demonstrated that the mRNA expression of adipogenic transcription factors such as peroxisome proliferator-activated receptor-${\gamma}$ ($PPAR-{\gamma}$) in 3T3-L1 cells were significantly down-regulated by AY treatment. And inhibited the expression of FAS, a protein responsible for lipid synthesis, transport and storage. Oral administration of AY (100, 250, and 500 mg/kg, P.O/daily for 4 weeks) was conducted in high-fat diet induced obese mice and C57BL/6 mice. AY was orally administered for 4 weeks to extract liver and epididymal fat, and hematoxylin and eosin staining(H&E staining) was observed. Observation showed that the fat concentration of liver tissue tended to decrease dose-dependently and decreased significantly at 500 mg/kg concentration. The AY-administered group of HFD-induced mice had a lower body weight gain, along with decreased triglycerides and total cholesterol compared with the control mice, however, the HDL-cholesterol/total cholesterol ratio was increased. These results indicate that AY exhibits anti-obesity effects in obese mice by decreasing in serum lipid levels and lipogenesis related gene.

Effects of Dykellic Acid Derived from Microorganism on the Cell Growth and Superoxide Dismutase Activity in Tobacco Photomixotrophic Cultured Cells (미생물 유래 Dykellic Acid가 담배 녹색배양세포의 생장 및 Superoxide Dismutase 활성에 미치는 영향)

  • To evaluate the biological effects of dykellic acid, a novel apoptosis inhibitor, isolated from microorganism on the plant cells, the cell growth, protein contents, and superoxide dismutase (SOD) activity were investigated in suspension cultures of tobacco photomixotrophic cultured (PM) cells on 12 days after different concentration of chemical treatment. The cells were cultured in MS medium containing 0.7 mg/L 2,4-D, 0.3 mg/L kinetin, 30 g/L sucrose and 200 mM NaCl at $25^{\circ}C$ in the light (100 rpm). Dykellic acid strongly inhibited the cell growth by evaluating the cell fresh wt and the ion conductivity in the medium ($IC_{50}$/, about 20 $\mu$M). The results as inhibition of cell growth and cell wall damage were same. The compound significantly increased the protein contents and the SOD specific activity in proportion with the dosage. The results suggested that dykellic acid may have biological activity in plant cells and tobacco PM cells may be suitable biomaterials for in vitro evaluation of the biological activity of natural products.

A Study on Performance Estimation and Operation Strategy of Biological Aerated Filter Using Semi-Empirical Biofilm Model (준 경험 생물막 모델(Semi-Empirical Biofilm Model)을 이용한 BAF 운전평가 및 적정 운전방안 도출)

  • The objective of this study is to find out whether the developed semi-empirical biofilm model can be applicable to real BAF pilot-scale wastewater treatment. In addition, the optimum operating conditions of BAF as a function of process variables such as organic loading change can be drawn based on the simulation results of model. The results will provide the economic and efficient BAF process design and operating control. As a result, developed semi-empirical biofilm model which is relatively simple compared to mathematical model can simulate three BAF processes consisted of 25 layers within 1 seconds. When this model was used for simulating real pilot scale BAF process and the simulated water quality values were compared to experimental ones, simulated TCOD, SCOD, TN, $NH_4{^+}$-N, $NO_x{^-}$-N, alkalinity values were different to experimental ones within 21%, 20%, 8.1%, 48%, 10%, and 23%, respectively. Therefore, if the BAF system was equipped with automatic control, the BAF process can be better efficiently adapted under the condition of significant change of influent loading.

Overnight Information E ects on Intra-Day Stoc Market Volatility (비거래시간대 주식시장정보가 장중 주가변동성에 미치는 영향)

  • Stock markets perpetually accumulate information. During trading hours the price instantaneously reacts to new information, but accumulated overnight information reacts simultaneously on the opening price. This can create opening price uctuations. This study explores the overnight information e ects on intra-da stock market volatility. GARCH models and the VKOSPI model are provided. Empirical data includes daily opening and closing prices of the KOSPI 200 index and the VKOSPI from March $3^{rd}$ 2008 to June $22^{th}$ 2010. Empirical results show that the VKOSPI signi cantly decrease during trading time when positiv overnight information moves the Korean stock upward. This study provides useful information to investors since the Korea Exchange plans to introduce a futures market for the VKOSPI soon.

  • TGARCH models characterized by asymmetric volatilities have been useful for analyzing various time series in financial econometrics. We are concerned with persistent volatility in the TGARCH context. Park et al. (2009) introduced I-TGARCH process exhibiting a certain persistency in volatility. This article applies I-TGARCH model to various financial time series in Korea and it is obtained that I-TGARCH provides a better fit than competing models.

Bacterial Adhesion And Penetration To e-PTFE Membrane Used For The Guided Tissue Regeneration (치주조직재생유도술용 비흡수성 차폐막 (e-PTFE membrane)에의 세균부착 및 침투)

  • This study was performed to observe the bacterial adhesion and penetration to e-PTFE membrane following guided tissue regeneration(GTR) procedure and to evaluate the association of the membrane exposure and bacterial contamination with the clinical outcome. For the study, ten infrabony defects in 9 patient were treated by mucoperiosteal flap operation including placement of the e-PTFE membrane. The treated teeth were monitored weekly for the membrane exposure, gingival recession and gingival inflammation. The membranes were retrieved after 4 to 6 weeks, examined by SEM for bacterial contamination and adherent connective tisue elements, and observed under LM for the bacterial penetration into membrane. Three months postsurgery, the defect sites were clinically reexamined for the changes in attachment level and probing depth. Comparison of the ultrastuctural findings and clinical outcome revealed that extent of membrane exposure and bacterial contamination of the membrane was inversely associated with clinical attachment gain. From this finding, the extent of membrane exposure and the bacterial contamination on the apical portion of the e-PTFE membrane at the time of removal seemed to be a critical determinant on the clinical outcome of GTR and the membrane exposure needs to be controlled for optimal results.

  • Despite the importance of chloride binding, it is very difficult to measure the binding capacity, in particular, for the concrete body in an existing structure: in fact, the measurement procedure for chloride binding is much influenced by the environmental condition such as temperature, fineness of sample and pore water extraction techniques. The present study concerns the quantification of the binding capacity of chloride ions in concrete using the X-ray diffraction (XRD) technique. Once the binding isotherm of chlorides was determined by the Langmuir isotherm, as a function of the W/C, curing age and binder type, the generation of bound chlorides (i.e. Friedel's salt) was simultaneously ensured by the XRD technique. The amount of bound chloride was then determined by analyzing the peak intensity for the bound chlorides in the XRD curve. It was found that an increase in the curing age and a decrease in the W/C resulted in an increase in the binding capacity.

  • This paper addresses EMC test results for the EQM Communications & Broadcasting Satellite(CBS) Ka-band Payload System. The aim of the EMC test is to verify that the payload system has no effect on the other system and is not affected by the external field. The compliance of EMC system requirements was verified by the analysis and measurement of the emission level from the payload system. The variation of performance by noise injection was measured to inspect the susceptibility of the payload system. The analysis and test results also showed that the electromagnetic interference has no serious effect on the overall payload system performance. It is confirmed that the EQM CBS Ka-band payload system can be operable in the space environment from the EMC performance's viewpoint. It is also obtained that better performance can be acquired by improving waveguide design and integration method.

  • Matrix metalloproteinases (MMPs) are a family of structurally and functionally related zinc-dependent enzymes responsible for proteolytic degradation of extracellular matrix components such as base membrane or interstitial stroma. MMPs play an important role in a variety of physiological and pathological tissue remodeling processes, including wound healing, embryo implantation, tumor invasion and metastasis. Since MMP-9 (gelatinase B) has unique ability to cleave type IV collagen, gene expression of MMP-9 has been focused on as a pharmacological target. Flavonoids are a class of compounds that are widely spread in plants. In the coures of screening for the suppressors of MMP-9 gene expression from natural products, Metasequoia glyptostroboides was selected. Six flavonoids, sciadopitysin, isoginkgetin, bilobetin, 2,3-dihydrohinokiflavone, luteolin and apigenin were purified as suppressors of MMP-9 gene expression from M. glyptostroboides. The suppressing activity of the isolated flavinoids on the MMP-9 gene expression was measured by gelatin zymography and Nothern blot analysis.

Production and identification of antisera against mu-opioid receptor using synthetic peptide epitope (Synthetic peptide를 이용한 mu-opioid receptor에 대한 항혈청의 생산과 검정)

  • In the present study we have analyzed the characteristics and distribution of the mu-opioid receptor(MOR) by raising anti-peptide antisera to the C-terminal peptide of MOR. The antisera against MOR was produced in New Zealand White rabbit against 15 residue corresponding to amino acids, 384-398 of the cloned rat MOR. The antigenic peptide was synthesized using an Applied Biosystems 432 solid-phase peptide synthesizer. The specificity and identification of the antisera were tested by analysis of transfected cells, epitope mapping and immunohistochemical method. COS-7 cells electroporated with MOR cDNA were used to evaluate the characteristics and subcellular distribution of MOR. MOR immunoreactivity was prodominent in the plasmalemma and subcellular compartments such as endoplasmic reticulum, Golgi apparatus and vesicle like structure. Furthermore, both tissue sections and transfected cell lines could be immunostained with these antisera and the immunoreactivity was abolished when anti-MOR sera were preincubated with the peptide against which they were raised. Based on epitope mapping analysis, all antisera appeared to have a similar epitope, which was determined to be within the last amino acid, 391-398. Moreover, immunohistochemistry showed that MOR immunoreactivity was observed in many brain areas including cerebral cortex, striatum, hippocampus, locus coeruleus and the superficial laminae of the dorsal horn. These stained spinal cord and brain areas showed the mirrored pattern observed in auto radiographic studies of mu-opioid binding as well as a pattern similar to that seen by is situ hybridization for MOR. Thus, several lines of evidence support the conclusion that the antisera produced in the present study most likely recognize mu-opioid receptor. These results suggest that MOR antisera may be utilized as useful tool to analyze the physiological and pharmacological studies for mu-opioid receptor in the future.
