It was investigated whether or not such helpers as worker bee, bee-cocoon and egg-cup etc, have any effects on oviposition and colony foundation of the bumblebee queen, Bombus ignitus. Among the helpers tested, the callow workers of B. ignitus and B. terrestris showed the most remarkable effects on the oviposition rates to 92% and 88%, respectively. The live cocoon as a helper improved oviposition rate over 60%. A narcotized old worker 10-20 days-aged after emergence, showed similar effects to a callow worker on the colony development such as oviposition rate, colony foundation and progeny-queen production. On the other hand, dried cocoon, callow honeybee worker or egg-cup did not show a positive effect as a helper. In the number of workers recruited to a foundation queen, two workers showed better effect than one worker on the colony development, with no difference between two and more.

A study was conducted on three pollination methods on oriental melon(sageageol-ggul) grafting with pumpkin(seongjutozoa) for the labor-saving and to improve fruit set. Fruit weight, flesh thickness and fruit setting rate of oriental melon were greater in growth regulators treatment than those of pollinated by bees. Sugar content and hardness of fruits pollinated by bees were higher than those of by growth regulators. From the last ten days of the February to the first ten days of the March, fruit setting rate was $95\%$ in fruit setting growth regulators, whereas it was $46\%$ and $45\%$ in pollinated by honey and bumble bee, respectively. After the middle of March, the percentage of fruit setting was >$98\%$ in all the pollination methods. The cultivation under plastic houses of oriental melon, suitable fruiting time far the pollination by bees was decided after middle days of the March. Chromaticity and especially the value of 'a' of fruit of oriental melon pollinated by bees were higher than those of growth regulators. The percentage of fermented fruits of bee pollinated and growth regulators treated was $6.7\~9.1\%\;and\;28.1\%$, respectively. The weight of 100 seeds of bees pollinated were higher than that of growth regulators. The more increased the weight of 100 seeds the less appeared the rate of fermented fruits. The percentage of marketable fruits of the honey and bumble bee pollinated and that of growth regulators treated was $82\%,\;80.3\%\;and\;62.5\%$, respectively. The decreasing rate of fruit weight during storage of bees pollinated was less than those of growth regulators. In these results, the introduction of honey bee and bumble bee for the pollination of oriental melon was able to labor-saving of fruit set and increase of fruit quality.
Chilling temperature and colony development in Korean native bumblebee, Bombus ignitus and introduced B. terrestris were firstly investigated. Among four chilling temperatures of -2.5, 0, 2.5 and 5$^{\circ}C$, 2.5$^{\circ}C$ showed the best result at the survival rate after artificial hibernation in both species. The survival rate after chilling was somewhat higher in B. ignitus until three months of cold treatment, but it was higher in B. terrestris at four months of cold treatment. In colony development of queens broken diapause by CO$_2$ treatment, oviposition rate and preoviposition period of B. ignitus were 72.5% and 17.4 days, respectively. These values were 6.1% higher and 7.4 days shorter than those of B. terrestris. The period up to colony foundation, the first male and queen emergence of B. ignitus were 62.8,66.4 and 63.0 days, respectively, and these values were 2.1-29.5 days shorter than those of B. terrestris. But, the duration up to first worker emergence of B. ignitus and B. terrexrris did not differ as 28.6 and 28.4 days, respectively. On the other hand, the rate of colony foundation and progeny-queen production of B. terrestris, which are the important indication in evaluating the quality of colony, were 14.2 and 13.5%, respectively and these values are 3.8 to 5.7 fold higher than those of B. ignitus. Besides, the numbers of progenies of B. terrestris reached 104.2 workers, 317.9 males and 21.1 queens, which corresponded to 1.1-1.8 fold those at B. ignitus. Therefore, above results showed that colony development of B. terrestris is superior to that of B. ignitus.
This experiment was executed to investigate the effect of the carpenter bee as a pollinator on the fruit set increment of sweet pepper in summer hydroponics in the alpine area of Gangwon province, Korea. Where the number of fruit sets were increasing during the treatment of pollination by the carpenter bee, total yields during the whole growing period were not different. The number of seeds per fruit in the treatment of the carpenter bee increased more than that in the control by 7.3 percent increment, but the percentage of blossom-end rot increased. As fruit weight range of SS size (below 120g) decreased and that of L size (from 181g to 210g) increased by the pollination treatment, so the percentage of standardized products for the export to Japan improved. Therefore this result was effective for the fruit production of high quality for the export to Japan and it will be necessary for further study on the method of reducing blossom-end rot to be done.
Bumblebees are widely used to pollinate crops in greenhouses and fields. Here we firstly developed an apparatus for the oviposition induction of the bumblebee Bombus ignitus using electricity. The apparatus consists of boxes for colony initiation, part of temperature control, part of heat transfer, and moving shelf. The result shows that the rates of oviposition and colony foundation in the newly developed apparatus are respectively 3.9% and 5.2% higher than in the existing apparatus using hot water. More importantly, the newly developed apparatus is 75% cheaper in costs and can more save energy than existing apparatus. These results indicate that the newly developed apparatus could serve as an effective apparatus for the oviposition induction of B. ignitus.
The effect of $CO_2$-treatment on interrupting diapause of Bombus ignitus was examined to provide a means for year-round rearing of the bumblebee. When mated young queens were exposed to 65% or 99% $CO_2$ for 30 min daily during two consecutive days, oviposition rate increased to 75% and 77%, respectively, comparing 50% in $CO_2$-untreated queens. At the same time, the days needed to first oviposition shortened to 17-18 days in $CO_2$-treated queens, comparing to 30 days in $CO_2$-untreated queens. $CO_2$-treatment at the second day after mating was appropriate to the oviposition and colony development. $CO_2$-treatment showed a positive effect on the oviposition and colony development, but less than them of overwintered queen in numbers of produced progeny. It can be concluded that $CO_2$-treatment to B. ignitus is insufficient to produce commercial grade bumblebee colony in spite of its capability for promoting oviposition, because the treatment failed to form a big colony.
The pollinating activity of bumblebee, Bombus ignitus was investigated at the blossom of Peach [Prunus persica (L.) Batsch] planted in vinyl house. The major time of foraging activity by B. ignitus was between 08:00 and 7:00, and the peak time of pollinating activity by B. ignitus was 11 :00 in the peach vinyl house from February 28 to March 5 in 2002. The rate of fruit set and the rate of marketable fruit by foraging activity of B. ignitus were much better than artificial pollination, and the effect of labor reduction by the pollinating activities of B. ignitus released against th ε artificial pollination was took remarkable profits over 93%. Therefore, the foraging activity of B. ignitus in peach vinyl house brought satisfactory results among the rate of fruit set, the rate of marketable fruit and the effect of labor reduction against the artificial pollination.
To increase the mating rate of Bombus ignitus used as insect pollinator, we investigated the sexual maturity time of B. ignitus. In investigating ovary development such as the number of eggs per ovariole and spermatheca size, the time of sexual maturity of queen was 10 days after eclosion. In case of male, the number of sperm was 246 thousand at immediately after eclosion, and was highest as 480 thousand at 9 days, and tended to show a dramatic decline at 35 days (87 thousand). The more mating time, the less the number of sperm. In consideration of number of sperms, the time of sexual maturity of male was 3-15 days after eclosion. In the sexual maturity time of queen in mating, the queen was not mated at immediately after eclosion, and showed a decrease in 20 days. In terms of the rate of mating and oviposition, the favorable time for mating of queen was 9-20 days. On the other hand, the male showed 3.3% of mating at immediately after ecolosion, showed the highest of 43.3% at 6 days after eclosion, and tended to decrease in 25 days. The sexual maturity time for mating of male was 6-20 days. In summary, our results indicate that sexual maturity time of B. ignitus in reproductive organs and mating is most favorable in 9-12 days after eclosion for queen and 6-9 days for male.
Pure Bombus ignitus venom samples were submitted to two-dimensional gel electrophoresis. A total of 64 excised spots were analyzed by mass spectrometry. Three main proteins resulted in the identification have not been described in other bee venoms before. Dose-dependence against human carcinoma (Hep3B, BT-20, A549 and AGS) were observed from 1ng/ml to 100ng/ml. Expecially, the treatment of 100ng/ml B. ignitus venoms showed the highest cytotoxicity with 55% against hepatocellular carcinoma (Hep3B). The B. ignitus venoms showed strong antimicrobial activities against Enterococcus faecium and Shigella sonnei, and practically antimicrobial activity against the other microorganisms tested. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of E. faecium and S. sonnei, were 0.256ug/ml, respectively.
