Serovars of Xanthomonas campetris pv. oryzae Collected from Korea and Serological Diagnosis of Bacterial leaf Blight (우리나라 벼 흰빛잎마름병균 (Xanthomonas campestris pv. oryzae)의 혈청학적 분류 및 진단)

  • Seventy-one strains collected from Korea were classified into three serovars (designated A, B-I and B-II) by using agar gel diffusion test with the antisera produced against Xanthomonas campestris pv. oryzae isolates Q7472 and Q7502. Of 71 isolates tested, 65 isolates belonged to serovar A, 5 isolates were serovar B-I, and one isolate was serovar B-II. The isolates of serovar B-I and B-II could be distinguished clearly from those of serovar A showing marked autoagglutination. Xanthomonas campestris pv. oryzae was serologically diagnosed in rice leaves by agar gel diffusion tests, possibly being distinguished from Xanthomonas campestris pv. olyzicola and E. herbicola. The pathogen could be also serologically detected from the extracts of diseased leaves, squeezed immediately, heated at $100^{\circ}C$ or incubated in PSA. Serological detection of Xanthomonas campestris pv. oryzae is a more reliable and less time-comsuming method.

  • Infectious Pancreatic Necrosis Virus (IPNV) is one of the most important pathogens for inland fish farming and had been first reported in Korea from returning adult chum salmon (Oncorhynchus keta) at hatcheries on the east coast. During the past years, several viruses identified as IPNV were isolated not only from chum salmon, but also from gold fish (Carassius auratus), eel(Anguilla japonica), and rainbow trout (Salmo gairdneri). An isolate, coded DRT, from fingerlings of pan-cultured rainbow trout in Daechung Dam showed different serotype from three known reference serogroups of IPNV such as VR-299, Sp, and Ab. Antisera to three of these serotypes, however, partially neutralized the infectivity of this isolate. Anti-Sp type was rather effective than either anti-VR-299 or anti-Ab, implying DRT could be more closely related to Sp. DRT has been purified and its RNA genome segments were compated showing that the isolate does not belong to any of known serogroups even with some common antigenicity.

  • Generally, carrier chickens contaminate the processing plant equipment to such an extent that negative chickens procell afterwards result in contaminated. meat. This study was performed to investigate the prevalence of Complliobacter jejuni in two chicken procelling plants. Altogether two hundred samples were collected from cloaca, carcasses, chilling water, and evis-cerationknives at different processing stages during the period of June to September 1987. The isolated organisms were tested for distribution of biotype, serotype. The results obtained were summarized as follows: 1. C. jejuni was isolated from 41(34.2%) of 120 chicken feces, 9(45.0%) of 20 carC888eS before chilling, 11(55.0%) of 20 carcasses after chilling, 12(60.0%) of 20 eviscerationlmives. The evilceration knives and chilling water were considered as major means of croll contamination. 2. In biotyping 82 isolates of C. jejuni, 64(78.1%) were cl888ified as biotype I, and 18(21.9%) belonged to biotype II. 3. In serotyping 82 isolates of C. jejuni, 64(78.1%) were identified as serotype LIO 37, and 18(21.9%) were untypable.

젖소의 환경성 유방염 원인체인 그람음성균 분포 및 E. coli의 혈청형 조사

  • 본 연구는 젖소 유방염의 환경성 원인체인 그람음성균의 분포와 E. coli의 항생제 내성 및 사람 식중독과의 연관성을 조사하기 위해 E. coli 혈청형에 대해 조사한 바, 2001년과 2003년 그람음성균의 주요 균 중 분포에서 많은 차이가 났으며, 2003년에는 E. coli가 31.8%로 가장 높은 것으로 나타났다. 체세포수에 따른 그람음성균의 분포율에서는 1백만 cells/ml 이하가 65.3%로 가장 높았으며, 체세포수 3백만 이상의 유즙에는 10.8%만이 분포하고 있는 것으로 나타났다. 또한, E. coli의 항생제 내성율 양상에서는 페니실린, 클록싸신, 세팔로틴등이 70%이상의 높은 내성율을 나타내었으며, E. coli 혈청형에서는 O23, O111, O157과 같은 주요 장관출혈성 E. coli 식중독균은 분리되지 않았다.

Serotype Variations of Agglutinogen and Fimbriae in the Korean Isolates of Bordetella pertussis (국내 Bordetella pertussis 분리균주에서 Agglutinogen과 Fimbriae 혈청형 변이 분석)

  • Bordetella pertussis is pathogenic bacteria causing pertussis, a infectious respiratory disease for the infants. The incidence rate of pertussis was significantly decreased after introduction of vaccine. However, increased pertussis cases are recently reported in several countries with high vaccine coverage. One of the inferred reasons is genotype or serotype variation between circulating strains and vaccine strains. Therefore, it is required to confirm the variation status of the isolates by genotype or serotype analysis and the possibility of pertussis outbreak in Korea should be estimated. For this, the serotype variations of the isolates from $1999\sim2006$ were investigated in agglutinogen and fimbriae. As the result, the most frequent serotype in the isolated strains was agglutinogen 1 and fimbriae 2 serotypes. Moreover, serotype transition from vaccine serotypes to non-vaccine serotypes was observed. Especially, the transition pattern of agglutinogen serotype was directed to increase a different type (agg 1) from the vaccine type (agg 1,2). However, in case of fimbriae, the same type (fim 2) with vaccine strain was increased. These results were also observed in other countries with increasing incidence of pertussis. For more predictable results to know increasing possibility of pertussis incidence in Korea, the studies on genetic variations of antigenic determinant genes and prevalence of antibody titer in normal population should be performed in the further.

Characterization of Verotoxin-producing Escherichia coli Isolated from Domestic Foods (국내 유통식품에서 분리된 Verotoxin 생성 Escherichia coli의 특성)

  • The incidence of verotoxin-producing Escherichia coli(VTEC) was surveyed in domestic foods including hamburger, raw meats and vegetables from 1997 to 1999. The molecular biological characteristics of the isolates were analyzed. Three VTEC strain were isolated from 1,700 samples. Serotypes of those isolates were 0157 : H7, 026 H4, and 056 : Hl2, respectively. Serotype O26 : H4 produced VT I and VT II, and 055 Hl2 isolate produced VT I, however the 60 MDa plasmid DNA and eae gene were not found from both strains. One 0157 : H7 isolate produced VT II and harbour 60 MDa plasmid DNA, however eae gene was not found in the strain. Although they produced VT, it seemed that the virulence of two strains were relatively weak because of the lack of the eae gene. In addition, the serotype O157 : H7 isolate resistant to ampicillin and streptomycin, while isolates of serotype O26 : H4 and O55 : Hl2 were multi-resistant to antibiotics including ampicillin, carbenicillin , cephalothin, trimethoprim/sulfamethoxazole and tetracycline. 육계농장에서의 살모넬라 위생대책

  • 육계농장에서는 SE(Salmonella Enteritidis)감염도 있지만, 오히려 SE이외의 여러 가지 요인에 의한 혈청형 살모넬라에 오염되는 경우가 많다. 현재 육계농장에서는 계군을 살모넬라로부터 완전히 차단하는 것은 어렵다고 생각하고 있고, WHO에서도 살모넬라 오염을 가능한 한 감소시키는 쪽으로 살모넬라 대책의 지침으로 정하고 있다. 그러나, SE 및 ST(Salmonella Typhimurium)등 사람에게 병원성을 나타내는 혈청형에 의한 계군의 살모넬라 감염오염은 공중위생의 견지에서 완전히 제거하지 않으면 안되다. 여기에서 출하 및 수송시의 대책, 사료의 살모넬라 오염의 중요성 등 산란계와는 다른 점을 지적하면서 기하고자 한다. 일상의 위생관리에 관한 양계장에서의 대책과 살모넬라 검사법에 대하여는 제외하였다.

Host Affinities and Serological Distribution of Bradyrhizobium japonicum Indigenous to Korean Upland Soils (한국 밭토양에 분포한 Bradyrhizobium japonicum 의 숙주친화성과 혈청형)

  • As a basic experiment to enhance the symbiotic utility of atmospheric nitrogen on soybean, host affinities and serological distribution of Bradyrhizobium japonicum indigenous to five Korean upland soils were measured. Based on nodulation, the symbiotic affinities between indigenous B, japonicum and eight soybean cultivars were remarkably different among soil inocula. On the whole, the averaged affinities of B. japonicum populations to soybeans were favorable in order of Goseong > Milyang > Suweon soils, but those in Iksan and Namjeju soils were not, especially for Danweonkong, Jangkyeongkong, and Eunhakong soybean cultivars. Regression analyses between nodules mass and shoot dry weight of soybean yielded model with $R^2=0.51$ for Goseong, $R^2=0.45$ for Milyang, $R^2=0.38$ for Iksan, $R^2=0.28$ for Namjeju, and $R^2=0.24$ for Suweon soils. B. japonicum from sampled soils were serologically fell into more than seven serogroups such as YCK 117(34.1%), YCK 141(6.5%), YCK 321(6.5%), YCK 445(4.7%), YCK 338(2.9%), YCK 150(1.2%), YCK 258(0.6%). The dominant serogroup YCK 117 was distributed 51.9 for Namjeju, 45.8 for Goseong, 41.7 for Iksan. 34.2 for Sueveon, and 11.1% for Milyang soils.

Serological Distribution and Properties of Antibiotic Resistance of Escherichia coli from Patients with Diarrhea (설사환자로부터 분리한 대장균의 혈청형 분포 및 항생제 내성유형)

  • As a part of investigation for basic epidemiology of diarrheogenic disease, we attempted isolation of Escherichia coli from patients with diarrhea. Seven hundred and twenty-one strains of E. coli were isolated from 1,239 patients with diarrhea. Seasonal distribution of patient with diarrhea was shown the most high at August (18.2%). Age group distribution of patient was shown the most high at children (54.6%, 2 to 10 years old). The serotypes of 721 E. coli isolates were in order of serotype O44 (16.8%), O153 (8.6%), O1 (7.5%), O166(5.7%), O8 and O86a (4.7%), and O125 (4.6%). The supernates cultured 36 strains among 721 E. coli isolates were indicated cytotoxicity against monolayered Vero cells. All of the isolates were susceptible to amikacin. The isolates were resistant in order of novobiocin (99.0%), moxalactam (97.1%), carbenicillin (96.1%), tetracycline (90.4%), ampicillin (85.9%), gentamicin (84.0%), streptomycin (78.4%), cephalothin (46.6%) and polymyxin B (4.2%). In the antibiotic resistant patterns, 125 kinds of multiple resistance patterns of E. coli isolates were detected. The highest resistant pattern was ampicillin-carbenicillin-chloramphenicol-cephalothin-erythro-mycin- gentamicin-moxalactam-novobiocin-penicillin G-streptomycin-tobramycin-tetracycline-tri methoprim type (24.3%).

Pulsed-Field Gel Electrophoresis and Monoclonal Antibody Analysis of Leptospira interrogans Isolated in Korea (국내 분리 렙토스피라균의 단클론 항체 및 Genomic DNA의 Pulsed-Field Gel Electrophoresis 분석)

  • A total of 22 Leptospiua inlermgans field isolates from the ~ a t s captured in 5 provinces of Korea in 1996, and 6 antigenically closely related relerence serovars of lai, yeonchon, birkini. gem, mwogolo. and canicola were analysed. When the antigenic characteristics were analysed by reactivity with 7 monoclonal antibodies prepared with sh.ains belongng to serogroup Icterohaemorrhagiae. all 22 isolates showed the same reaction pattern with that of serovar lai. Large restriction fragment patterns obtained after cleavage of geno~nic DNAs with infrequently cuttimg restriction enzymes were analyzed by pulsed-field pel electrophoresis(PFGE). Identification of leptospira strains by PFGE with Nor I, Asc I or Iise I digests correlated with their antigenically typed serovars, silh a few exceptions. PFGE of isolates, except for JR89, digested wjth Nor I showed identical pattern w~th serovar lai, showing 13 Cragments between 940 kb and 63 kb. When PFGE pallerns of JR89 were compared with those of serovar lai, Not I digest showed additional two hands of 1000 kb and 460 kb, while Asc I digest showed 650 kb fragment and Fse I digest did not show the fragment of 280 kb. Whereas serovar yeonchon. which was isolated in Korea and identified as a new serovar previously. could be differentiated from serovar lai in antigenic reactivities with monoclonal antibodres. it showed the similar PFGE pattern with serovar lai includin~ reference and field isolates. It was suggested that Korean leptospiral field isolates are closely related in DNA level.
