본 연구는 선천선고혈압쥐(SHR)에서와 마찬가지로, 높은 정상혈압 및 중등 정도의 본태성고혈압 환자에서 Valyl-Tyrosine(VY)의 항고혈압 효과 여부를 결정하기 위해 수행되었다. 29명의 지원자에 대하여 무작위 이중맹검 위약검사를 실시하였으며, 3 mg의 VY를 포함하는 100-ml 음료와 100ml-위약 음료를 조제하여 사용하였다. 연구대상은 VY (남16/여1, 45.5${\pm}$3.2세, 146.4${\pm}$2.3/90.5${\pm}$1.8 mmHg) 집단과 위약(P) (남11/여1, 48.8${\pm}$3.0 세, 145.5${\pm}$2.4/92.3${\pm}$1.8 mmHg) 집단으로 나누었다. 대조(C)기간 3주 째에 VY-음료 또는 P-음료를 하루 2번씩 투여하기 시작하여 4주 간의 실험(E)기간(BP)은 매주 측정되었으며, 아침에 앉은 자세에서 측정하였다. 혈액 표본은 C기간과 E기간의 마지막 날 채취하였다. VY집단에서, 수축기(S) 및 이완기(D) 혈압의 감소는 첫째 주에 각각 9.7 및 5.3 mmHg (P < 0.001)이었으며, E기간의 시작에 따른 넷째 주에는 각각 9.3 및 5.2 mmHg (P < 0.001)이었다. P 집단에서는 SBP와 DBP 모두 변하지 않았다. VY 집단의 혈압은 회복기간의 끝까지 점진적으로 증가하였다. VY집단에서, 혈장 안지오텐신(Ang) I과 VY의 농도는 현저히 증가한 반면 Ang II와 알도스테론은 VY 투여 후에 현저히 감소하였다. VY는 SHR에서와 마찬가지로 중등 정도의 고혈압 환자에서도 Ang I-전환효소 억제를 통하여 현저한 항고혈압 효과를 가지는 것으로 보이며, 어떤 부작용도 검출되지 않았다.
Background: In korea the agricultural community widely uses organophosphorous, and organophosphorous poisonings are increasing every year. We compared change in activity of acetylcholinesterase and pseudocholinesterase by organophosphorous and by the interaction of ethanol and organophosphorous. We also compared the effect of reversible anticholinesterase drugs, physostigmine and neostigmine The object of this study is to investigate the effects of several anticholinesterase drugs and on how ethanol influences the activity of cholinesterase. Materials and Methods: Fifteen male university students were randomly selected, and blood samples were taken from the antecubital vein. The acetylcholinesterase in the RBC and the pseudocholinesterase in the serum were extracted and separated. The enzyme activity change was measured by the electrometric method. After adding acetylcholine, the pH change was measured with a pH meter. Results and Conclusion: Our results indicated that reversible anticholinesterase drugs decreased the cholinesterase activity more efficiently than organophosphorous. The acetyl cholinesterase and pseudocholinosterase activity were decreased by ethanol. When ethanol was added, oxime a cholinesterase activator, increased acetylcholinesterase activity but does not increased pseudocholinesterase activity.
Chronic alcoholism is considered a common cause of malnutrition. Especially, micronutrient deficiency may playa critical role in the incidence of alcoholic liver diseases. This study was conducted to investigate the effect of folate deficiency and ethanol consumption on cholesterol metabolism and the antioxidative system in rats. Plasma concentration of total cholesterol was increased by ethanol administration in folate-fed rats. HDL-cholesterol tended to be higher in the folate-fed group, but it was not significant. The plasma and hepatic levels of malondialdehyde were increased after chronic ethanol feeding, but dietary folate depressed the plasma malondialdehyde content of rats. Ethanol or folate feeding did not significantly change alcohol dehydrogenase activity. But folate feeding increased catalase activity in ethanol-fed rats. There was no significant change in superoxide dismutase activity among the experimental groups. Glutathione peroxidase activity tended to decrease by chronic ethanol feeding, but dietary folate did not affectthe glutathione peroxidase activity of chronic ethanol-fed rats. Glutathionine-S-transferase activity was not affected by ethanol feeding or folate deficiency. The plasma and hepatic levels of retinol decreased after chronic ethanol feeding. The hepatic level of retinol significantly decreased in ethanol-fed rats by folate deficiency. The plasma level of $\alpha$-tocopherol tended to be low in the folate deficient group with ethanol feeding, but there was no difference among the experimental groups in the hepatic level of $\alpha$-tocopherol. These results demonstrate that chronic ethanol consumption changes the plasma cholesterol metabolism and antioxidative system of rats, and optimal folate feeding in ethanol-fed rats exerts protective effects to some extent.

This study was undertaken to evaluate the antihyperglycemic, antilipid peroxidative, and antioxidant effects of the ethanol extracts of Artemisia iwayomogi (Ai) in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Sprague-Dawley rats with a single intravenous injection (45 mg/kg b.w.) of STZ. The diabetic rats were then randomized to the diabetic and Ai extract therapy groups which were treated with Ai extract at doses of 1, 2, and 3 g/kg b.w./day, respectively, for 14 days. Oral administration of Ai (2 g/kg b.w.) significantly decreased their intake of food. Dosage of 2 g/kg of the extract significantly decreased blood glucose levels in the glucose level in diabetic rats after 4 day, there was no significant difference observed at 1 and 3 g/kg. A dose of 2 or 3 g/kg of the Ai extract significantly reduced plasma glucose levels in STZ-induced hyperglycemic rats at 7 days. The hypoglycemic effect of Ai at a dose of 2 g/kg was significantly more effective than that of STZ-control. The effect was more pronounced in 2 g/kg than 1 g and 3 g/kg. A significant reduction in triglycerides (TG) and free fatty acids (FFA), and a significant increase in liver glycogen were observed in treated diabetic rats at doses of 2 g/kg after 14 days of treatment. Administration of Ai extracts to diabetic rats showed a significant decrease in liver malondialdehyde (MDA) levels. The activity of superoxide dismutase (SOD) was significantly increased in the 3 g extract-supplemented groups. The activities of glutathione peroxidase (GSH-px) and catalase (CAT) were significantly increased in the 1 g and 3 g extract-supplemented groups. Ai extract significantly increased glutathione-S transferase (GST) activity in a dose-dependent manner compared with treatment in STZ-control rats. Our result supports the fact that the administration of Ai extract is able to reduce hyperglycemia and hyperlipidemia risk, and also reduce the oxidative stress in diabetic rats.

This study investigated the beneficial effects of crude saponins from soybean cake on body weight and glucose tolerance in high-fat (37% calories from fat) diet fed C57BL/6 mice. The mice were supplemented with three doses of saponins (0.5%, 1.0%, and 1.5%, wt/wt) and 1.0% Garcinia cambogia (wt/wt), positive control for 9 weeks. The body weight, visceral fat weight and epididymal adipocyte area were significantly reduced in the saponin supplemented groups in a dose dependent manner compared to the high-fat group. Saponins did not significantly affect food intake; however, cambogia significantly lowered food intake compared to the high-fat fed control group. The crude saponins from soybean cake supplement significantly lowered plasma leptin, triglyceride and total cholesterol levels, whereas they significantly elevated the fecal excretion of triglyceide in a dose dependent manner compared to the high-fat group. Cambogia did not affect the fecal excretion of lipid in the diet-induced obese mice. Supplementation of 1.5% saponin reduced the hepatic triglyceride content compared to the high-fat group. High-fat induced glucose intolerance with the elevation of blood glucose levels compared to the normal group; however, the saponins supplement significantly improved postprandial glucose levels. After 9 weeks of being fed a high-fat diet, the mice presented with significantly increased activities of hepatic fatty acid synthase and fatty acid ${\beta}$-oxidation; however, saponins and cambogia normalized these activities. These results indicate that saponins from soybean cake exhibit a potential anti-obesity effect and may prevent glucose intolerance by reducing body weight and plasma lipids, increasing fecal lipid excretion and regulating hepatic lipid metabolism in high-fat fed mice.
In the present work we investigated the effect of regular physical exercise on the activities of erythrocyte antioxidant enzyme, plasma total radical-trapping antioxidant potential(TRAP) and plasma level of lipid peroxidation(malondialdehyde, MDA) in 64 healthy male, aged 34-67 years. The study population were divided in two groups: small amount of exerciser(exercise time less than 10min/d) and moderate amount of exerciser(exercise time more than 20min/d) according to their physical exercise habits measured by a questionnaire. Erythrocyte superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and catalase(CAT), plasma TRAP, as well as plasma MDA were determined. Erythrocyte GSH-Px and plasma TRAP were higher in moderate amount of exercisers than those in small amount of exercisers by 17% and 26%, respectively. No significant differences were observed in erythrocyte SOD, CAT and plasma MDA between the two groups. Mean exercise time was positively correlated with the erythrocyte GSH-Px activity and plasma TRAP significantly. The results would sugest that regular moderate exercise enhances antioxidant defences against reactive oxygen species and may increase the likelihood of a healthier life span.

An ISFET urea sensor was fabricated by immobilizing the urease using photosensitive polymer, poly(vinyl alcohol)-SbQ on the H$^+$ sensing $Si_3$$N_4$ thin film of pH-ISFET. The sensor could determine the urea concentration in the range of 1∼50 mg/dl with fast response and good repeatability. For its application to clinical analysis, the interferences of the various materials which cause inhibition in urease catalytic reactions in blood was investigated. The results of the urea measurements in blood plasma using the ISFET urea sensor were compared with these of conventional spectrophotometric method.

This study was to investigate the hypolipidemic effect of powdered mulberry leaves (PML) and water extract of powdered mulberry leaves (WML) on high-fat fed mice. Male C57BL/6 mice were divided into four groups; a normal group (N), a high-fat (HF) group, a high-fat group supplemented with PML (HF-PML) and a high-fat group supplemented with WML (HF-WML). The PML or WML was added to a standard diet based on 1% dried mulberry loaves (1g PML/100g diet and 0.22g WML/100g diet) for 6 weeks. Body weight and organ weights were not different among thle groups in high-fat fed mice, whereas food intake and daily energy intake were significantly (p<0.05) lowered in the HF-PML group. In plasma and liver, the supplementation of PML and WML significantly (p<0.05) lowered cholesterol and triglyceride concentrations compared to the HF group. The HDL-cholesterol/total cholesterol ratio was significantly (p<0.05) higher in the HF-PML and HF-WML groups than in the B:.w group. The fecal triglyceride and cholesterol concentrations were significantly (p<0.05) increased in the HF-PML and HF-WML groups compared to the HF group. Hepatic lipid regulating enzyme activities, fatty acid synthase, fatty acid ${\beta}-oxidation$ and carnitine palmitoyl transferase were significantly lower in the HF group than in the N group. However, the activities of these hepatic lipid regulating enzymes activities were significantly (p<0.05) elevated in the HF-PML and HF-WML groups compared to the HF group. Accordingly, these results suggest that PML and WML improve plasma and hepatic lipid levels partly by increasing fecal lipid excretion and enhancing hepatic lipid regulating enzymes activities.
This study examined the effect of Momordica charantia L. (bitter melon: BM) on lipid and hepatic antioxidative enzyme levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats by injection of streptozotocin (STZ), and rats were fed for 4 weeks with experimental groups divided into four groups: a normal control group, STZ-control and STZ-BM 5% & STZ-BM 10% treated groups. Levels of free fatty acids (FFA), high-density lipoprotein cholesterol (HDL-chol), triglycerides (TG) in plasma and malondialdehyde (MDA) & protein in liver, catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST), and xanthine oxidase (XOD) were measured in liver cytosol. Level of HDL-chol significantly increased in the STZ-BM 5% diabetic group. TG & FFA levels were significantly higher in all diabetic groups compared to the control group. MDA and protein levels were significantly higher in the STZ-BM 5% group compared to all other experimental group. CAT level was higher in the supplementary group with BM compared to the STZ-control group, although the difference was not significantly different. SOD level was not significant in any experimental groups. GST level was significantly higher in the BM-treated groups compared to the STZ-control group. XOD level was significantly lower in the BM 5% group and significantly decreased in all experimental groups. These results show that supplementation of BM fruit powder may have beneficial effects on diabetic complications and damage caused by oxidative stress.
