광학 분석 시스템용 수.발광 소자 집적 모듈 개발

  • 본 발표에서는 광학적 분석 시스템에 적용 가능한 발광소자(광원)과 수광소자(광센서)를 집적화시키는 모듈(수 발광 집적모듈) 기술을 제시하고자 한다. 이러한 수-발광 집적모듈은 다양한 응용 분야에 적용 될 수 있다. 예를 들어, 광신호 감지를 위한 광통신용 송-수신 모듈(optical communication), 의료/진단 분야에서 단백질/DNA/박테리아 등의 검출 및 분석에 관한 바이오 센서(bio-sensor), 그리고 대기(가스)/수질 모니터링에 관한 환경센서 등 매우 광범위한 분야에 해당되는 요소 기술이라 할 수 있다. 특히, 이들 분야들 중 바이오 물질을 분석하고 검출하는 광학적 바이오 센서 기술은 높은 경제적 가치와 산업적 성장 잠재력으로 인해 오랫동안 활발한 연구가 진행되어 오고 있다. 이러한 광학적 바이오 센서에서 가장 범용적인 방법 중 하나가 항온-항체 면역반응을 기반으로 하는 형광 검출(fluorescence detection) 기법이다. 이러한 시스템은 전체적으로 광원, 광학계, 그리고 센서로 구성되는데 기존에 일반적으로 사용되고 있는 형광 현미경의 경우는 민감도가 우수하다는 장점은 있으나 상당히 고가이고 부피가 크며 복잡한 광학구성으로 이루어져 있다는 한계점을 가지고 있다. 이러한 맥락에서 고민감도를 확보하면서 휴대성, 고속처리, 저가 등의 특성을 가진 시스템에 대한 요구가 갈수록 증가하고 있다. 이를 해결하기 위한 핵심기술 중의 하나가 수-발광 부분을 집적화 시키는 기술이라 할 수 있다. 본 연구에서는 바이오 센서 기술의 하나로서 형광을 측정하여 혈액내의 진단 지표인자를 검출할 수 있는 휴대용 혈액진단기기에 적용되는 소형 수 발광 집적 모듈을 개발하였다. 혈액내의 검출 성분의 양에 따라 형광의 세기가 변화하게 됨으로써 정량적인 검출이 가능한 원리이다. 모듈의 구조는 크게 광원(발광소자), 광학계, 그리고 광센서(수광소자) 세 영역으로 나누어 진다. 광원은 635 nm 적색 레이저다이오드로서 형광체(Alexa Fluor 647/발광파장: 668 nm)를 여기 시키는 기능을 하며 장착된 볼렌즈 의해 샘플의 형광체 영역으로 집광된다. 광학계는 크게 시준렌즈(collimating lens)와 광학필터로 구성됨으로써 샘플로부터 발생되는 광을 적절하게 수광소자로 전달하는 기능을 하게 된다. 여기서 광학필터의 경우는 기본적으로 Distributed Bragg's Reflector(DBR) 구조로써 실리콘(Si) 포토다이오드 상부에 모노리식(monolithic)하게 형성되며 검출 샘플로부터 진행되는 레이저 광(잡음의 주원인)은 차단하고 형광(광신호)만 통과 시키는 기능을 하게 된다. 따라서 신호 대 잡음비(S/N ratio)를 향상시키기 위해서는 정밀한 광 필터링 기능이 요구됨으로써 박막의 세밀한 공정 조건과 구조적-광학적 특성 분석이 수행되었다. 마지막으로 포토다이오드 소자는 일반적인 구조 이외에 중앙에 원형 구멍이 형성된 특별한 구조가 적용된다. 이것은 포토다이오드 구조에 변화를 줌으로써 모듈 구조를 효율적으로 응용할 수 있다는 의미를 갖는다. 또한 포토다이오드의 전기적-광학적 측정 분석을 통해 잡음 및 감도 특성이 세부적으로 조사되며 형광신호를 효과적으로 측정할 수 있음을 확인하였다. 최종적으로 제작된 모듈은 약 $1{\times}1{\times}1cm^3$ 내외 정도의 크기를 갖는다. 요약하자면 본 발표에서는 광학적 바이오센서에 적용할 수 있는 소형 수-발광 소자 집적모듈을 소개한다. 전체 모듈 설계는 최소한의 부피를 가짐과 동시에 측정의 정밀성을 향상시키는데 초점을 맞추어 진행하였다. 세부요소인 광학필터와 포트다이오드의 경우 잡음 및 민감도에 미치는 중요성 때문에 세밀한 공정 및 특성분석이 수행되었다. 결론적으로 독자적인 설계 및 공정을 통해 휴대성 및 정밀성 등의 목적에 부합한 경쟁력 있는 수-발광 소자 집적모듈 제작 기술을 확보하였다.

Effects of TDN/CP Ratio on Nutrient Intake, Digestibility, Ruminal Fermentation and Blood Characteristics of Replacement Dairy Heifers (TDN/CP 비율에 의한 영양소 공급이 젖소 육성우의 영양소 섭취량, 소화율, 반추위 발효 및 혈액특성에 미치는 영향)

  • This study was conducted to investigate a proper management and nutrient supply for raising replacement heifers to improve lactating performance. In order to determine a proper TDN/CP ratio that would be suitable for domestic use, 13-month-old replacement heifers were fed in the ratios of 4.6:1(T1) and 4.3:1(T2), respectively, to examine the changes in nutritional intake, digestibility, characteristics of rumen fluid and blood compositions. While dry matter intake was same at 9.0kg/day, CP intake did not show any significant difference, though T2(1.45kg/day) was slightly higher than T1(1.34kg/day). In fiber intake and digestibility, two results showed no significant difference, though T1 was slightly higher than T2. The pH, NH3-N and VFA densities in rumen fluid examined at the fourth hour after feed intake did not show any significant difference either. Overall level of VFA was not influenced largely by the TDN/CP ratio, but the iso-butyrate and iso-valerate levels and the A/P ratio were significantly high in T1(p<0.05). In blood characteristics, the treatments showed no significant difference. In conclusion, the TDN/CP ratios of 4.3:1 and 4.6:1 did not significantly affect the characteristics of the ruminant stomach, but in comparison with digestibility of 13-month-old heifers, the ratio of 4.6:1 was more appropriate for increased intake of fiber and higher digestibility.

A Structural Equation Model on Quality of Life in Hemodialysis Patients (혈액투석환자의 삶의 질 구조모형)

  • This study was conducted to build a structural equation model explaining factors that affect hemodialysis patients' quality of life to test the goodness of fit of the model and identify directly and indirectly significant paths. To accomplish this, 230 patients receiving hemodialysis registered at study hospitals in G Metropolitan City and M City were randomly selected, and 201 were used in the final analysis. Data were collected using a self reporting questionnaire from February 1 to March 31, 2014. SPSS WIN 18.0 was used for descriptive statistics and correlation analysis of the general characteristics and for reliability analysis of the research tools. In addition, AMOS 18.0 was used for confirmatory factor analysis of latent variables, to determine the goodness of fit of the hypothetic model, and hypotheses testing. The results of this study were as follows: the fit of the modified model was 41.32, GFI = .96, AGFI = .92, RMR = .04, NFI = .96, CFI = .91 and TLI = .95. Twelve out of the 22 research hypotheses that were improved and improved were statistically significant. The results indicated that hemodialysis patients' quality of life was influenced directly by self-efficacy, depression, and self-care behavior, and indirectly by sleep disturbance, stress, fatigue, and family support. These variables explained 58.6% of the variation in quality of life. Overall, these findings suggest that nursing interventions for hemodialysis patients require strategies for managing patient depression and for increasing self-efficacy and self-care behavior to improve quality of life.

Effects of Dietary Resveratrol on Growth Performance, Blood Biochemical Parameter, Immunoglobulin, and Blood Antioxidant Activity in Broiler Chicks (Resveratrol의 첨가가 육계의 생산성, 혈액 생화학 특성, 혈액 내 면역글로불린과 혈액 내 항산화 인자에 미치는 영향)

  • This study was conducted to investigate the effects of dietary resveratrol on growth performance, blood biochemical parameters, immunoglobulin, and blood antioxidant activity in broiler chicks. Three hundred twenty one-day old broiler chicks were divided 8 treatments (C(-), basal diet; C(+), basal diet with antibiotics; DL-${\alpha}$-tocopherol 20 IU; DL-${\alpha}$-tocopherol 200 IU; resveratrol 20 ppm; resveratrol 200 ppm; methylated resveratrol 20 ppm; methylated resveratrol 200 ppm) with 4 replicates and 10 birds per replicate. Birds were reared for 35 days, and, at the age of 35 days, eight birds of average weight from each replicate were selected for blood samples collection. There were no significant differences on feed intake and feed conversion ratio. But final body weight and weight gain in antibiotics, resveratrol and methylated resveratrol treatments were significantly higher than no-antibiotics and ${\alpha}$-tocopherol treatments (P<0.05). There were no significant differences on carcass rate and relative organ weights among treatments, however, weights of liver and bursa of februcius in antibiotics, resveratrol and methylated resveratrol treatment were lower than other treatments. Weight of pancreas was high in resveratrol and methylated treatment. On the cecal microflora (total microbes, Coliform bacteria, Salmonella spp., and lactic acid bacteria), these in resveratrol and methylated resveratrol treatments didn't show the differences compared with those in no-antibiotics, antibiotics, and ${\alpha}$-tocopherol treatments. In the serum, there were no significant differences on creatinine, blood urea nitrogen (BUN), total protein, albumin, globulin, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) among treatments, though globulin contents of reseveratrol 200 ppm and methylated resveratrol 20 ppm treatments decreased compared to those of other treatments. Immunoglobulin (IgA, IgG and IgM) were significantly decreased in antibiotics and resveratrol treatments compared to that of no-antibiotics and ${\alpha}$-tocopherol treatments (P<0.05). Superoxide dismutase (SOD) like activity tended to increase in resveratrol groups (P<0.05), however, there was no significant difference on malondiakdehyde (MDA) content among treatments. In conclusion, these results showed that resveratrol derived from mulberry can be used as alternative of antibiotics through improvement of broiler's performance and maintain of health.

  • 투어멀린(Tourmaline)은 비대칭 쌍극자를 가진 유극성 결정체로 광물 중에서 영구적으로 전기분극의 특성을 띄고 있는 유일한 물질로써 일명 "전기석"이라고 알려져 있고, 자체의 미약전류(약 0,06mA)와 함께 음이온 및 원적외선의 발생으로 최근 들어 우리 주변에서 건강과 환경을 위한 관심고조의 대상인 육방정계의 압전성 및 초전성을 띄는 붕규산염(주요성분: Mg, Fe, B, Si, Ca)으로, 원석(면역지수 ~1000)에 비해 수백 배의 효력이 있는 투어말린 세라믹(면역지수 ~418,000)을 비롯하여 이것을 이용한 새로운 아이디어 신상품들이 계속 개발되고 있다. 뿐만 아니라, 물분자를 만나면 수소(H$^{+}$)와 수산기(OH$^{-}$)로 전기분해하여 친수기와 소수기 부분으로 구분하며, H$^{+}$와 OH$^{-}$는 각각 $H_2O$와 결합하여 활성이 강한 hydronium ion(H$_3$O$^{+}$ 과 계면활성 작용이 있는 hydroxyl ion(H$_3$O$_2$$^{-}$)을 생성하며, 물속에서 계속 불안정한 상태로 존재하는 수산기는 hydroxyl (-)ion을 형성하여 약 알카리성(pH~7.4)을 띄고 물의 클러스터 세분화와 미네랄을 공급해 줌으로 체내에 들어오면 살균, 항균능력이 있는 면역기능을 갖게 되며, 혈액을 정화하고 자율신경을 자극하여 교감신경의 흥분을 억제한. 이와 함께 전기분극을 띈 투어멀린 입자는 다른 물질에 비하여 경이적 수치의 원적외선을 발생하여 강력한 열 효과와 높은 침투력으로 인체의 혈액순환 촉진과 혈전용해 작용으로 건강과 활력에 도움을 주는 것으로 확인되어 많은 관심을 끌고 있다. 따라서 유익한 이용가치를 인정받고 있는 투어멀린을 유용한 타 물질과 혼합물을 구성하여 성형 또는 EPD 전착하여 소결한 뒤, 소결체의 특성을 분석하여 그 응용성을 개발, 확보하였다.

The Effects of Characteristics of Nurses on Knowledge and Nursing Performance Evaluation of Evidence Based Hemodialysis Nursing Practice in Hemodialysis Unit Nurses (혈액투석실에서 근무하는 간호사의 특성이 근거기반 혈액투석간호지식과 수행정도에 미치는 영향)

  • Purpose: The purpose of this study was to identify the effects of characteristics of nurses on knowledge and nursing performance of evidence based hemodialysis nursing practice among hemodialysis unit nurses. Methods: The participants were 180 nurses working in hemodialysis unit for more than 6 months in 23 private and general hospitals in Seoul and Gyeonggido. Data were collected from March 30th to April 15t in 2016 and were analyzed using stepwise regression analysis, descriptive statistics, t-test and ANOVA. Results: Nurses' knowledge on evidence based hemodialysis nursing practice was $15.87{\pm}4.52$ out of 23 points. Type of hospitals working in and adherence to evidence based hemodialysis guidelines were significant factors to knowledge and these two factors explained 30.0%. Nurses' nursing performance on evidence-based hemodialysis nursing practice was 4.52 out of 5 points. The performace level was significantly related to total nurisng career and necessity of hemodialysis nursing education and these two factors explained 8.0%. Conclusion: A development of guideline and continuing education is necessary for improving knowledge and performance of evidence based hemodialysis nursing practice.

Toxicity Evaluation of Effluent Gases from a Residental Fire by Rats (연립 주택 화재시 유독가스 방출 특성과 Rats를 이용한 독성평가)

  • It has long been recognized that exposure to fire-induced toxic gases is a fatal hazard confront-ing people in fires. In this study, an indoor fire experiment was conducted in an unoccupied residential building located in An-san city, Kyoung-gi province, and the composition of effluent gases, which include CO, $CO_2$, $O_2$, $SO_2$, NO and $NO_2$, were measured by a gas analyzer. A group of lab rats were exposed to the toxic gases released from fire, and the blood samples of the rats were gathered every 2 minutes. A toxicity evaluation was conducted by analyzing the concentrations of Glucose, AST(GOT), ALT(GPT), CBC Count and CO(carboxy)-Hb in the blood samples. Shown from the analysis is the does-response relationship between the CO concentration that rats were exposed to and the CO-Hb concentration in rat blood.

Fabrication and Medical Applications of Lotus-leaf-like Structured Superhydrophobic Surfaces (연잎 모사 구조로의 초소수성 표면 처리와 의료분야의 적용에 관한 연구)

  • Various biomaterials have been widely used for biomedical applications, including bio-organs, medical devices, and clinical devices like vessel, blood pumps, artificial kidneys and hearts, even in contact with blood. The issue of blood compatibility has been studied intensively to prevent negative effects such as thrombosis due to the implanted devices. The use of lotus-leaf-like structured surfaces has been extended to an increasing number of applications such as contamination prevention and anticorrosion applications. Various methods such as template, sol-gel transition, layer-by-layer, and other methods, developed for the fabrication of lotus-leaf-like surfaces have been reported for major industrial applications. Recently, the non-wettable character of these surfaces has been shown to be useful for biomedical applications ranging from blood-vessel replacement to antibacterial surface treatment. In this review, we provide a summary of current and future research efforts and opportunities in the development and medical applications of lotus-leaf-like structure surfaces.

Evaluating the Effects of Salicornia Extract on Performance, Egg Quality and Blood Profile of Laying Hens

  • The current experiment was conducted to evaluate the effects of adding Salicornia extract to the drinking water on the performance, egg quality, and blood profile of laying hens. A total of 216 Hy-Line Brown laying hens at 40 weeks of age were used in a 10-week experiment. The birds were allotted into three experimental treatments with three replications per treatment and 24 birds per replication. The treatments were CON (basal diet), T1 (1 cc of Salicornia extract per liter of drinking water), and T2 (5 cc of Salicornia extract per liter of drinking water). The collected data were analyzed using the SAS package program. The results indicated that addition of Salicornia extract to the drinking water of laying hens did not cause any negative effects on the performance, egg quality, or blood profile. Compared to the control treatment, the treatments with Salicornia extract remarkably increased egg production (P<0.05) in the last week of the study, improved egg shell thickness and significantly reduced the egg breaking rate (P<0.05). The results of this study showed that the addition of Salicornia extract improved egg shell quality; thus, Salicornia extract can decrease the egg breaking rate and increase production on commercial farms.

Flow Cytometric Characterization of Lymphocyte Subpopulations in the Korean Bovine (한우 혈액내 백혈구아군의 특성 및 말초혈액상)

  • Characterization of immune cell subpopulations in the bovine was performed using a direct immunofluorescence technique adaptable for routine and repeated monitoring. This whole blood procedure is faster and requires less volume than conventional density gradient isolation methods. Low inter-and intra-animal variations were seen in hematology parameters and in CD4, CD8 and CD2 lymphocyte subtypes. CD4 values were 30% of lymphocytes in male and 32% in females. Thriteen percent were CD8 in males and 13% in females. CD4: CD8 ratios were approximately two in both sexes. Fiffty three percent were CD2 in males and 54% in females. The mean RBC counts of peripheral blood were 7.20$\times10^6/{mm}^3$ for male cattle and 6.36$\times10^6/{mm}^3$ for females. The mean WBC counts were 8.09$\times10^3/{mm}^3$ for males and 7.09$\times10^3/{mm}^3$ for females. The percent of lymphocytes(63-65%) was higher than the percent of neutrophils(17-18%), the percent of eosinophils(11-15%), te percent of monocytes(4-5%), and percent of basophiles(1%).
