This study was designed to examine the in vitro antioxidant and anti-inflammatory effects of kohlrabi (Brassica oleracea var. gonglodes) extract. Kohlrabi was extracted using 70% ethanol and then fractionated sequentially with n-hexane, ethyl acetate and butanol. Antioxidative ability was evaluated by bioassays using total polyphenol contents and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid diammonium salt) radical scavenging activity. Ethyl acetate fraction of kohlrabi was best on total polyphenol contents ($27.33{\pm}0.26mg\;GAE/g$) and ABTS radical scavenging effects ($IC_{50}\;172.9{\pm}1.6{\mu}g/mL$).For the anti-inflammatory activity in RAW 264.7 cells, the EtOAc fraction showed the highest inflammatory effect. Dose response studies were performed to determine the inhibitory effect of EtOAc fraction of kohlrabi on pro-inflammatory mediators in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. The EtOAc fraction of kohlrabi inhibited the NO and $PGE_2$ production and the protein level of iNOS and COX-2, and protein expression of pro-inflammatory cytokines (TNF-${\alpha}$, IL-6 and IL-$1{\beta}$), in a dose-dependent manner. These results suggest that kohlrabi has considerable potential as a ingredient with antioxidative and anti-inflammatory effects.

This study was intended to test anti-inflammation and cytoprotective effect against UVB after Sorghum nervosum was extracted with 70% ethanol. The efficacy of Sorghum nervosum was assessed regarding cell viability analysis, reactive oxygen species measurement, anti-inflammation, a change in COX-2 protein, and cytoprotective effect against UVB. According to the results of experiment, the cell viability of 97% or higher was shown at all concentrations of Sorghum nervosum in RAW264.7 macrophage, HaCaT cell. And in anti-inflammatory NO inhibitory activity, a concentration-dependent inhibitory effect was shown. And COX-2 protein expression was also significantly (p<.001) inhibited at 25, $50{\mu}g/mL$. With regard to cytoprotective effect against UVB, in the quantitative analysis results of reactive oxygen species within the cell, it was verified that Sorghum nervosum extract had an effect on an decrease in the total amount of ROS. When the results of study are considered comprehensively, it is thought that there is possibility of Sorghum nervosum development as raw materials for cosmetics showing an anti-inflammation and cytoprotective function against UVB.

Purpose: Nonsteroidal anti-inflammatory drugs that prohibit biosynthesis of arachidonic acid metabolites have been considered potent host modulation agents. The aim of this review was to determine the effect of nonsteroidal anti-inflammatory drugs adjunctive with nonsurgical periodontal treatment in patients with periodontal disease. Materials and Methods: Three electronic databases were searched to identify relevant studies. The methodological quality and mean differences of the change in clinical attachment level and probing depth were analyzed according to Cochrane review methods. Results: Twelve studies were included in the methodological assessment and nine studies were suitable for inclusion in the meta-analysis. The mean difference in the clinical attachment level gain did not differ significantly between the nonsteroidal anti-inflammatory drugs and control groups at any observation time. The highest mean difference in clinical attachment level gain was 0.30 mm at 4 weeks (95% confidence interval = -0.37 to 0.97). There was a significant mean difference in the probing depth reduction, of 0.34 mm (95% confidence interval = 0.29 to 0.40) at 6 weeks. Conclusion: Therefore, nonsteroidal anti-inflammatory drugs have additional therapeutic effect when administrated with nonsurgical periodontal treatment.

The effects of volatile flavor extracts of eight different herbal medicines, Juniperus rigida (JR), Saussurea lappa SL), Cnidium officinale (CO), Angelica gigas (AG), Eugenia caryophyllata (EC), Angelica tenuissima (AT), Mentha arvense (MA), and Artemisiae argyi (AA), were investigated on LPS-stimulated inflammation using Raw 264.7 cells. The volatile flavor extracts of CO and AG considerably inhibited LPS-stimulated NO, $PGE_2$, IL-6, and TNF-${\alpha}$ (except AG) production, as well as iNOS expression. Major volatile components of CO were identified as ligustilide and of ${\beta}$-eudesmol as AG by GC-MS analysis. Thus, these results suggest that the volatile extracts of CO and AG may be useful as potential therapeutic agents for inflammation-associated disorders.
In this study, we selected Gardeniae fructus (GF) as an anti-inflammatory functional material and improved the biological activity of GF through the treatment of ${\beta}$-glucosidase. For the simple evaluation of anti-inflammatory activity, the inhibitory activity of GF extract (GFE) on the production of NO by RAW264.7 cells in the presence of LPS was examined. ${\beta}$-glucosidase originating from Aspergillus niger or Aspergillus fumigatus has effectively improved the anti-inflammatory activity of GFE. The enzyme treatment raised the activity of GFE by more than 10 times. The optimum conditions for the enzyme reaction were at pH 4.6, $45^{\circ}C$, and 20 U/mL for 24 h with agitation. In addition, in vitro production of cytokines (IL-$1{\beta}$, IL-6, TNF-${\alpha}$), COX-2, and the NF-${\kappa}B$ activation of RAW264.7 cells decreased more in the presence of GFE treated with ${\beta}$-glucosidase originating from Aspergillus niger (GFAN) than in the presence of GFE. These results suggest that enzyme-treated GFE might be a potential candidate for natural anti-inflammatory food materials.

This work was carried out to investigate the anti-inflammatory effects of Paulownia coreana Uyeki on abirritant, atopy and acne skin. Paulownia coreana Uyeki has been used as a traditional medicine having anti-febrile, anti-inflammation effect in Korea, Paulownia coreana Uyeki loaves were extracted with 70% EtOH. Its superoxide anion radical scavenging activity and inhibitory effect on LPS-induced NO production were examined. The extract inhibitied the generation of NO and $PGE_2$ induced by LPS in the macrophage cell line (Raw 264.7). Consistent with the inhibitory effects on No and $PGE_2$ generation, the extract inhibited expression of iNOS and COX-2. In further study, it was found that the extract prevented $IkB-{\alpha}$ degradation, as demonstrated by western blot analysis of $IkB-{\alpha}$ protein level. However, the extract treatment did not affect cell viability at $100{\mu}g/mL$ concentration in both human skin fibroblast and Raw 264.7 cells in vitro. Thus, the present study suggests that Paulownia coreana Uyeki leaves extract have significant anti-inflammatory activity and potential as an anti-irritation material.

Abalone, a marine organism inhabiting the west coast of Korea, has antioxidant and anti-inflammatory effects, and is a resource with potential to be used in various industries such as antibiotic development and cosmetic raw materials. In this study, we chose abalone among various marine lives on the west coast. Antibacterial peptide (AMP) was separated from abalone and its derivative Ab4-7 was identified and its physiological activity was studied. The treatment of Ab4-7 in inflammatory RAW 264.7 to check the anti-inflammatory efficacy nhibited inflammatory cytokines, TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased mRNA manifestation of HO-1, genes related to antioxidants. Based on the strong antioxidant and anti-inflammatory effects of Ab4-7, the effects of Ab4-7 in the inflammatory RAW 264.7 cells were identified through RT-PCR, which regulates the gene Matrix Metalloproteinase (MMPs) that induces a variety of inflammatory skin diseases by engaging in the decomposition of the extrocellular matrix metalloproteinase (ECM). Taken together, it is concluded that Ab4-7 has a powerful anti-inflammatory and antioxidant effect and can be used as a functional material for various inflammatory skin disease treatments by controlling the genes associated with matrix metalloproteinase (MMPs).
In this study, we investigated the variation in free sugars, amino acids, antioxidant activity, and anti-inflammatory activity of Solanum nigrum Linne based on harvest time. Major amino acids identified by HPLC analysis were proline, histidine, and serine. The highest content of total amino acids were found in S. nigrum aerial parts and roots harvested on July 10th and August 10th. Four kinds of free sugars (fructose, glucose, sucrose, maltose) were detected in S. nigrum, and the free sugar content varied significantly with harvest time. The fructose content of S. nigrum decreased with as harvest time increased. The total polyphenol content of S. nigrum was highest in those harvested on August 30th. The antioxidant activity of ethanol extract from S. nigrum collected at different harvest times were measured by DPPH and ABTS radical scavenging assays. The anti-inflammatory activity of these extracts were assayed via nitric oxide suppression in C6 glioma cells with a lipopolysaccharide (LPS)-induced inflammatory response. The anti-inflammatory activity and antioxidant effects were the highest in the extract from S. nigrum collected on August 30th. Good correlations were observed between antioxidant and anti-inflammatory activities in ethanol extract of S. nigrum roots harvested on August 30th.

The purpose of this study is for checking anti-inflammatory effects of Syzygium aromaticum ethanol extract. For this, we investigated biological active evaluation about anti-inflammatory effects by Syzygium aromaticum ethanol extract. Syzygium aromaticum was extracted with 70% ethanol. This extract was tested for the cell viability on RAW 264.7 cell line by MTT assay, nitric oxide inhibitory activity and expression of inflammatory mediators. The extract showed low cytotoxicity as more than 90% cell viability in under 25 ㎍/ml concentration. On LPS-induced RAW 264.7 cell line, nitric oxide inhibition activity result showed that the extract reduced NO productions in a concentration-dependent manner. Expression of inflammatory cytokines as PGE2, TNF-α and IL-1β decreased in a concentration-dependent manner and iNOS and COX-2 proteins expression were decreased significantly in western blot analysis. We confirmed that the Syzygium aromaticum ethanol extract has excellent anti-inflammatory effect. This results suggested that extract of Syzygium aromaticum may have value as the cosmetic materials.

Red rose petals are usually disposed but they are an abundant source of phenolics and traditionally used as food supplement and as herbal medicine. Of the Various phenolics, they are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of red rose ethanolic extracts(GRP) on lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results demonstrated that pretreatment of GRP(500㎍/mL) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects byred rose petal were observed in the following. Red rose petal inhibited the translocation of NF-κB from the cytosol to the nucleus via the suppression of IκB-α phosphorylation and also inhibited LPS-stimulated NF-κB transcriptional activity. These findings suggest that red rose petal exert anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of red rose petal. Therefore, red rose petal could be regarded as a potential source of natural anti-inflammatory agents.
