[Retracted]Anti-inflammatory activities of octapeptides derived from tertomotide ([논문철회]Tertomotide 유래 옥타펩타이드의 항염증 활성)

  • Tertomotide is a peptide fragment of hTert and developed as a vaccine targeting cancer. It has been reportedly known to ameliorate inflammatory symptoms in clinical tests and in animal studies. However, the therapeutic potential of tertomotide is not supposed to be comparable to conventional anti-inflammatory agents due to low druglikeness In order to treat inflammations present in varous lesion, the structure of tertomotide is required to be modified. In this context, 12 octapeptides were designed based on tertomotide and screened for the anti-inflammatory activity in activated monocyte by measuring TNF-α secretion. As a result, some octapeptides has been exerted anti-inflammatory activity, comparable to or better than tertomotide and estradiol, known anti-inflammatory agents. This result is supposed to be helpful for developing therapeutic purpose exploiting other tertomotide-derived peptides and would be an example for designing novel drug based on active biomolecules with undesirable structure by convergence study of biology and computer-aided medicinal chemistry.

수종 생약의 항염증 작용

  • 1. 천연 항염증 물질의 분리 천연 항염증 약물 개발을 목표로 하여 한방과 민간에서 소염, 진통의 목적으로 이용되는 식물들을 대상으로 Carrageenin에 의해 유도된 백서 후족 부종의 억제를 지표로 Screening을 행한 결과, 2종의 식물의 부탄을 엑스가 항염증 활성을 나타내었다. 즉, 호랑가시나무(flex cornuta L. ) 잎 및 쑥부쟁이 (Aster yomena Makinc)의 부탄을 엑스가 Aspirin(300mg/kg)에 필적하는 항염증작용을 나타내었다. 이 식물들의 부탄을 엑스로 부터 각각 2종의 화합물을 분리하였다. 호랑가시나무로 부터는 01eanane type의 monodesmoside와 Pomolic acid bisdesmoside가, 쑥부쟁이로 부터는 Hederagenin bisdesmoside및 구조 미확정의 물질이 각각 분리되었다. 2.천연 항염증 물질의 구조 변환 항염증 약물로 많이 이용되고 있는 시호(Bupleurum falcatum L.)의 유효성분인 Saikosaponin-a의 용해도 개선을 위하여 그 유도체를 합성하였다. 합성된 Saikosaponin-a hemisuccinate의 K염은, 용해도 시험 결과 현저한 용해도 증가를 보였다.

A study on anti-inflammatory activity of fermented rice bran of Saccharomycopsis fibuligera isolated from Nuruk (누룩에서 분리한 Saccharomycopsis fibuligera 미강 발효물의 항염활성에 대한 연구)

  • This paper aims to increase functionality and competitiveness through fermentation of rice bran, which is used as a raw material for alternative foods and cosmetics. Cytotoxicity and anti-inflammatory effects were confirmed using rice bran extract fermented with Saccharomycopsis fibuligera A8 isolated from Nuruk. In the case of cytotoxicity, cytotoxicity was shown at 100 ㎍/mL for rice bran extracts, but cytotoxicity was not shown for fermented rice bran extracts. Meanwhile, as a result of confirming anti-inflammatory effects through inflammatory indicators such as nitric oxide, TNF-α, IL-1β, and IL-6, rice bran extracts showed anti-inflammatory effects at concentrations of 100 ㎍/mL or higher only nitric oxide and TNF-α. And fermented rice bran extracts exhibited anti-inflammatory effects at concentrations of 25 ㎍/mL, 25 ㎍/mL, 50 ㎍/mL, and 50 ㎍/mL, respectively, exhibiting anti-inflammatory effects in lower concentration. Meanwhile, as a result of testing the cytotoxicity and anti-inflammatory effects of heat-killed S. fibuligera, it was found that the heat-killed S. fibuligera showed anti-inflammatory effect in fermented products.

Preparation of Hot Water Extracts of Dandelion Leaves to Increase Anti-inflammatory Activity (항염증 활성이 증진된 민들레 잎의 열수 추출물 제조조건 설정)

  • To materialize anti-inflammatory compounds from dandelion leaves, we studied optimal conditions of hot water extraction and concentration. We investigated the properties of extracts on various temperatures between 60 and $90^{\circ}C$. Extraction yields at $60^{\circ}C$ were the highest at $39.2{\pm}1.3%$ and flavonoid contents were increased by heating. However, anti-inflammatory activities were remarkably decreased by increasing temperature. According to the results that the highest anti-inflammatory activities of water extract were demonstrated at $60^{\circ}C$, cell wall degrading enzyme treatment was carried out to increase extraction yield of anti-inflammatory compounds. The enzyme treatment resulted in increased yields and flavonoids from dandelion leaves of 1.5-fold and anti-inflammatory activities were not decreased. Also, column chromatography was used for concentration of anti-inflammatory compounds from extract of dandelion leaves treated with enzyme. Anti-inflammatory activities of adsorbed compounds on Amberlite XAD-2 and Sepabeads SP-850 were significantly increased. However, yields of adsorbed compounds were increased over two-folds on Sepabeads SP-850 compared to Amberlite XAD-2. Thus Sepabeads SP-850 was more effective than Amberlite XAD-2 to concentrate the anti-inflammatory compounds of dandelion leaves.

  • Antiinflammatory and antioxidative effects of mulberry fruit were investigated by using bioassay screening system. The extract of muberry fruits showed inhibitory effect in phospholipase $A_2$ activity and cyclooxygenase II activity. Among nine varieties of mulberry tree, the antiinflammatory activities of the Shingwangppong, Ficus, Cheongilppong and Keomseolppong were higher than that of the other varieties. Also, antioxidative activity of mulberry fruit was examined by DPPH free radical scavenging method. The radical scavenging activity of the mulberry fruit decreased as following order : Shingwangppong > Keomseolppong > tetraploid Ficus> diploid Ficus.

  • 염증 (Inflammation)은 병원체, 손상된 세포, 자극물질 등으로 인한 손상에 대해 작용하여 조직이나 장기의 손상을 재생하는 작용으로써 신체 방어 기전들 가운데 매우 중요한 역할을 하는 것으로 알려져 있다. 이러한 염증반응이 과다할 경우 각종 염증성 질환 혹은 암 등을 유발하는 원인으로 발전할 수 있어 항염증제의 개발은 전 세계적으로 중요시되고 있다. 선천적 면역을 담당하는 대식세포는 lipopolysaccharide(LPS), 활성산소 (ROS) 등에 의해 자극되어 염증인자 생성에 관여한다. 따라서 본 연구에서는 항염증 소재 개발을 목표로 하며, 국내 천연소재를 활용한 기능성 항염증 소재로 전북 순창군에서 재배된 초석잠(Stachys Sieboldii MIQ.)과 돼지감자 (Helianthus tuberosus L.)를 이용하여 항산화 활성을 평가하고자 하였으며, 대식세포주를 활용한 세포독성 및 항염증 활성에 대한 효능을 확인하고자 하였다. 본 연구는, 전북 순창 지역에서 재배 된 초석잠과 돼지감자를 사용하여 각 조건의 추출 용매, 온도, 시간별 추출물의 Total polyphenol 함량 평가를 통한 최적 추출조건 선정을 진행하고, 선정된 추출조건의 추출물의 항산화 활성을 측정하기 위해 DPPH & ABTS radical scavenging activity 분석 및 Total flavonoids 함량 분석을 통해 항산화 효능 평가를 진행하였다. 또한 항염증 소재로의 활용을 위해 대식세포인 Raw 264.7을 사용하여 농도별 MTT assay를 진행하여 세포독성 평가를 진행하였고, Nitric oxide (NO) 생성억제 효능을 확인하여 항염증 활성을 평가하였다. 실험 결과, Total polyphenol 함량 분석을 통해 최적 추출조건이 선정된 초석잠 (25℃, 주정 60%, 3 h), 돼지감자 (25℃, 주정 40%, 1 h)은 최적 조건에서 약 58 mg GAE/g 및 158 mg GAE/g의 총 폴리페놀 함량을 보였으며, DPPH & ABTS radical scavenging activity 및 Total flavonoids 함량 분석한 결과, 초석잠이 돼지감자보다 더 높은 항산화 활성을 나타내었다. 대식세포 실험에서의 추출물 처리군의 세포독성 측정 결과, 100 ug/mL 이내의 농도에서 독성활성이 나타나지 않음을 확인하였고, Nitric oxide (NO) 생성 억제활성 측정을 통해 LPS 처리군 대비 접종량 100 ug/mL 기준 각각 초석잠 약 47%, 돼지감자 약 49% 수준의 항염증 활성을 확인하였다.

Anti-inflammatory and Wrinkle Improvement Effects of Peptides from Ginseng Berry Amino Acidic Complex (진생베리 아미노산 복합체로부터 분리한 펩타이드의 항염, 주름개선 효과)

  • Ginseng berry (GB) contains Ginsenoside Re and has anti-inflammatory and anti-wrinkle properties. In this study, TLC fractions 1, 2, and 4 of the ginseng berry amino acid complex were identified and analyzed by HPLC. And identified a peptide (AP-1) by LC/MASS analysis of fraction 1. The anti-inflammatory activity was confirmed by investigating the inhibitory effect of AP-1 on NO production. In addition, collagen synthesis using procollagen type I C-peptide (PIP) ELISA kit was 50% higher effective than that of the control group. From these results, the peptide isolated from ginseng berry amino acid complex is considered to have anti-inflammatory and anti-wrinkle effect, and may be useful as an anti-inflammatory and anti-aging cosmetic raw material.

Antioxidant and Antiinflammation Activities of Prunus persica Tree Extracts (야생 복숭아나무 추출물의 항산화 및 항염증 효과)

  • Reactive oxygen species (ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Several lines of evidence provided that ROS appears to cause to develop aging and various diseases. High level of hyaluronic acid with decreased molecular weight has been detected in patients with inflammatory diseases including rheumatoid arthritis. In this study, we have conducted to investigate the antioxidant and hyaluronidase inhibitory activities of Prunus persica Batsch var. davidiana Maximowicz in order to screen the bioactive substances which can be developed as possible anti-inflammatory agents. Among the extracts of Prunus persica Batsch var. davidiana Maximowicz, EtOAc extract exhibited the strongest effect on antioxidant experiment such as DPPH, Ferric- Thiocyanate and Rancimat. Also, EtOAc extract showed a potent hyaluronidase inhibitory activity.

Effect of Soluble EPCR on the Anti-Inflammatory Effects by Activated Protein C (수용성 EPCR에 의한 활성화된 단백질 C의 항염증 작용에 관한 연구)

  • In this study, we evaluated the effect of soluble EPCR(Soluble Endothelial Protein C Receptor, sEPCR) on the anti-inflammatory activities by activated protein C(APC) in endothelium. We demonstrated that sEPCR inhibited the barrier protective activity, the inhibition of neutrophils adhesion toward endothelial cells and the inhibition of transendothelial migration by APC in endothelial cells. Interestingly, sEPCR also blocked the mechanism by which APC inhibited the expression of cell adhesion molecules(CAM) by TNF-alpha in endothelial cells. These results suggested that the anti-inflammatory activities of APC was inhibited by sEPCR which blocked the binding motifs of Gla domain of APC to membrane bound EPCR. This finding will provide the important evidence in the development of new medicine for the treatment of severe sepsis and inflammatory diseases and good clue for understanding unknown mechanisms by which APC showed the anti-inflammatory activities in endothelium.

Antioxidant and Anti-inflammatory activity of Saccharomycopsis fibuligera fermented Lindera obtusiloba flower extract (Saccharomycopsis fibuligera 발효 생강나무 꽃 추출물의 항산화 및 항염효과)

  • In this study, the antioxidant activity and anti-inflammatory activity of Lindera obtusiloba flower fermentation extracts were identified. To evulate antioxidant activity and antioxidant concentration, polyphenol concentration measurements, flavonoid concentrations measurements, DPPH experiments, and ABTS experiments were conducted. In Flavonoids, DPPH and ABTS assay, antioxidant activity were increased after fermentation. At this time, the flavonoid concentration was 5.0%, the DPPH experiment showed 33.27% and the ABTS experiment showed 29.82% antioxidant increase. In the anti-inflammatory experiment, we conducted a cytotoxicity experiment and an anti-inflammatory experiment for LPS-induced inflammation. Cytotoxicity showed low cytotoxicity in both control and fermentation groups, but lower cytotoxicity in fermentation groups. In the case of NO production inhibition, fermented Lindera obtusiloba flowers showed an increase in anti-inflammatory activity by more than 50% compared to the control group, showing that they can be used as a cosmetic ingredient with anti-inflammatory function.
