Induction of Erythromycin by Virginiamycin Inducing Factor, Virginiae Butanolide C (Virginiamycin 생합성 유도인자 Virginiae Butanolide C에 이한 Erythromycin 생산 유도)

  • Virginiae butanolide C(VB-C) is one of the butyrolactone autoregulators, which triggers the production of virginiamycin in Streptomyces virginiae. In order to investigate the function of VB-C as inducer in other strains, Streptomyces erythraeus was used as a test strain(parent). VB-C binding receptor gene was introduced into S. erythraeus(transformant) and the production of VBs and specific VB-C binding protein were analysed in parent and transformant. When 300ng/ml of the synthetic VB-C was added at 0, 20, 44 h cultivation of the parent and at 44 h cultivation of the transformant, the initial production times a antibiotics were shortened by more than 8 and 6 h, respectively. The transformant showed strong antibiotic activity against B. subtilis. These results suggest that the VB-C might have an ability to induce the production of secondary metabolites in S. erythraeus.

  • With the increase of the use of antibiotics and invasive procedures, infections caused by multidrug-resistant Acinetobacter baumannii(MRAB) are increasing. We screened the antibiotic producing strain B-51 for antibacterial activity against MRAB from the soils and studied the effects of culture medium on the antibiotic production of B-51. The medium conditions for maximum antibiotic productivity of B-51 was 2% glycerol, 0.5% soybean meal, 0.01% $CaCl_2$, 0.01% $MgSO_4{\cdot}7H_2O$ and 0.01% $KH_2PO_4$ at an initial pH of 6.0, at $30^{\circ}C$ for 76 h.

Adsorption Characteristics of Sulfonamide Antibiotic Compounds in GAC Process (GAC 공정에서의 Sulfonamide계 항생물질 흡착특성)

  • Adsorption performance of sulfonamide antibiotic compounds such as sulfadimethoxine(SDM), sulfachloropyridazine(SCP), sulfamethazine(SMT), sulfathiazole(STZ) and sulfamethoxazole(SMX) on granular activated carbon(GAC) was evaluated in this study. The coal-based activated carbon was found to be more effective than other carbons in adsorption of sulfonamide antibiotic compounds. The wood-based activated carbon was less effective than coconut- and coal-based carbon in adsorption nevertheless having larger pore volume and specific surface area than others carbons. The maximum adsorption capacities(X/M) of coal-based activated carbon for the five sulfonamide species was 1.3$\sim$1.5 and 1.8$\sim$2.1 times larger than coconut- and wood-based activated carbon, respectively. Carbon usage rates (CUR) of coal-, coconut- and wood-based activated carbons for SCP were 3.55 g/day, 4.29 g/day and 6.47 g/day, respectively. Similar results were obtained in the adsorption of the rest four sulfonamide species. It is concluded that coal-based activated carbon could removed the sulfonamide antibiotic compounds better than other material-based activated carbons.

Occurrence and Distribution of Selected Veterinary Antibiotics in Soils, Sediments and Water Adjacent to a Cattle Manure Composting Facility in Korea (국내 우분 퇴비화 시설 인근 농경지 및 수계 중 Tetracycline 및 Sulfonamide 계열 항생물질의 분포특성)

  • There has been increased concern regarding the release of antibiotics to different environmental compartments due to the possibility of the development of antibiotic resistant bacteria. However, limited information is available regarding the occurrence, fate, and transport of antibiotics in Korea in both the aqueous phase and in solid phases such as sediment and soil. Therefore, this study was conducted to monitor the concentration of released antibiotics in surface water, sediment, and soil adjacent to a cattle manure composting facility in Korea. Specifically, the following six antibiotics were monitored: tetracycline (TC), chlortetracycline (CTC), oxytetracycline (OTC), sulfamethazine (SMT), sulfamethoxazole (SMX), and sulfathiazole (STZ). To extract and quantify the antibiotics from different environmental compartments, solid phase extraction (SPE) and high performance liquid chromatography mass spectrometry (HPLC/MS) techniques were adopted. The concentration of the six antibiotics ranged from below the detection limit (BDL) to 0.71 ${\mu}g$/L in surface water, from BDL to 27.61 ${\mu}g$/L in sediment, and from 0.12 to 157.33 ${\mu}g$/L in soil. In addition, higher concentrations of antibiotics were observed in surface water and sediment at locations closer to the composting facility indicating that composting is the source of the antibiotics found in the environment. Furthermore, higher concentrations of antibiotics were observed in the solid phase (sediment and soil) than the aqueous phase. These findings indicate that the possibility of antibiotic resistant bacteria is increased because such bacteria are more stable in the solid phase. Overall, longterm monitoring of the aqueous phase and solid phase is necessary to gain a better understanding of the impact of antibiotics from source on the environment in Korea.

  • An antifungal substance was purified from culture broth of Pseudomonas flulorescens 2112 that showed a broad-spectrum antagonistic activity against various phytopathogenic fungi including capsici. The substance was identified as 2,4-diacetylphloro-glucinol basd on NMR analysis. The 2,4-diacetylphloroglcinol showed antibiotic activity in broad acidic range from pH 1.0 to pH 9.0. About 83% of initial activity was remained after incubation for 30min ar $60^{\circ}C$, however, the activity was dropped up to 50% after 30 min incubation in $80^{\circ}C$. When the nucleotides of P. capsici treated with 2,4-diacetylphloroglucinol were labeled with[$^{3}$ H]-Adenin, the newly synthesized and radioactive-labeled RNA was significantly reduced than those of untreated P. capsici. indicating that the 2,4-diacetylphloroglucinol inhibits RNA synthesis.

An Antifungal Agent Produced by Bacillus thuringiensis BK4, an Antagonistic Bacterium against Fusarium Wilt Disease of Tomato (항진균성 항생물질을 생산하는 Bacillus thuringiensis BK4의 항생물질 정제와 토마토 시들음병의 효과적인 방제)

  • The optimum production condition for the antibiotic from Bacillus thuringiensis BK4 was determined, and the suppression rate of Fusarium-wilt by the butanol-extracted antibiotic was verified by employing tomatoes in vitro and in vivo pot tests. Cell growth and antifungal activity were the best when 0.5% xylose and 0.2% peptone No.3 were given as carbon and nitrogen sources, respectively, in the presence of 5mM $CaCl_2$. The partially purified antibiotic successfully prevented Fusarium oxysporum pathogen in pot experiments. When the pots were treated with both live cells and the partially purified antibiotic, an additive-effect was seen in the suppression of Fusarium-wilt, but synergistic effect was not detected. The antibiotic, denoted BK4, purified by Sephadex LH-20 column chromatography was eluted with a single peak at a retention time of 38 min. on prep-HPLC; Minimum inhibition concentration of the homogenous antibiotic was determined to be 50${\mu}$g/ml.
