Antioxidative Activity of Solvent Fraction and Isolation of ANtioxidative Compound from Chestnut Husk (밤 귀피의 용매분획별 항산화 활성과 항산화 물질의 분리)

  • To enhance the utilization of chestnut husk discarded in the processing company antioxidative activities and compounds were investigated. Antioxidative activities of solvent fractions from chestnut husk were examined by benzoic acid hydroxylation method ferric thiocyanate method and DPPH test. Ethyl acetate fraction showed strong antioxidative activities comparable to BHA. Active compounds were isolated and purified from ethyl acetate fraction by Sephadex LH-20 column chromatography and preparative HPLC. A major active compound gallic acid was identified by $^{1}$H and $^{13}$ C-NMR. The phenolic acid contents was analyzed by GC and ellagic acid (172.22 mg%) and gallic acid (107.39 mg%) were major phenolic acid of chestnut husk.

  • Lactobacillus spp.의 항산화 효과를 측정한 결과 L. bulgaricus LB 207의 cell lysate와 intact cell 모두에서 항산화 효과가 높게 측정되었다. 또한 이들 균주들은 높은 ferrous iron 흡착능을 나타내었으나 항산화 효과 사이의 연관성을 설명하기는 어려웠다. 유산균의항산화 메커니즘과 in vivo 실험을 통한 더욱 구체적인 항산화 효과에 대한 연구가 필요 하며, 다양한 항산화 메커니즘을 통하여 항산화 유산균 인간의 활성산소 축적의 위험으로부터 보호해 줄 수 있을 것으로 생각된다.

Evaluation of Antioxidant and Antimicrobial Activities of Solvent Extracts from Coriolus versicolor (구름버섯(Coriolus versicolor)의 용매 추출물에 대한 항균 및 항산화 활성 조사)

  • Antioxidant activities of various solvent extracts from Coriolus versicolor were investigated for their total polyphenol content, total flavonoid content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical scavenging activity. C. versicolor extracts were produced by organic solvents such as ethanol, ethyl acetate, and methanol. Total polyphenol and flavonoid contents in the ethanol and ethyl acetate extracts were higher than those in the methanol extract. DPPH radical scavenging activity of methanol extract showed the highest value of 80.3%, which was similar to that of ascorbic acid (85.5%). All extracts showed good (>90.0%) ABTS cation radical scavenging activity. The antimicrobial activities of these extracts from C. versicolor were evaluated against six strains of bacteria using the disc diffusion method. All extracts showed antimicrobial activities against all tested bacteria except Staphylococcus aureus. These results indicate that various extracts from C. versicolor could be useful as natural antioxidant and antimicrobial agents.

Effects of Turmeric (Curcuma longa L.) Bioactivity Compounds and Lipid Peroxidation Inhibitory Action (울금(Curcuma longa L.)의 생리활성 및 지질과산화 저해능에 미치는 영향)

  • The aim was to determine the physiological activity and antioxidant activity by lipid peroxidation inhibitory action of turmeric (Curcuma longa L.). Bioactive compound of total carotenoid $1.581{\pm}0.005mg$ ${\beta}$-carotene equivalents (BCE)/g dry weight. Total phenol content was the highest in the ethyl acetate (EA) extract, followed by chloroform:methanol (CM, 2:1, v/v) and 70% methanol extracts. Antioxidant effects (nitrogen oxide radical scavenging activity, nitrite scavenging activity, ${\beta}$-carotene bleaching assay, and lipid peroxidation inhibition action) of 70% methanol, CM, and EA extract of turmeric. Turmeric extracts yield were 70% methanol 16.54%, CM 5.64%, and EA 4.14%, respectively. Antioxidant activity of the samples exhibited a dose-dependent increase. However, in the current study, none of the samples evaluated showed activity as strong as the BHA (butylated hydroxyanisole) and trolox. Further, nitrite scavenging activity was the highest for the EA extract. As a result of this experiment, indicating their commercial value and potential applications in food and nutraceuticals.

Antioxidative and Antimicrobial Activities of Methanol Extract from Rosmarinus officinalis L. and Their Fractions (로즈마리 추출물 및 분획물의 항균·항산화 활성)

  • This study was performed to evaluate the antioxidant and antimicrobial activity of methanol extract from Rosmarinus officinalis L. and its fractions. The ethyl acetate fraction of rosemary had a higher antioxidant activity in both DPPH ($3.22\;{\mu}g/ml$) and ABTS ($5.05\;{\mu}g/ml$) compared to other extracts and fractions. Based on the results of the FRAP assay, the ethyl acetate fraction of rosemary showed a value of $5.9{\pm}0.3\;{\mu}M/{\mu}g$, and buthanol fraction and rosmarinic acid exhibited values of $4.8{\pm}0.2\;{\mu}M/{\mu}g$ and $5.1{\pm}0.1\;{\mu}M/{\mu}M$, respectively. Measurements of the antimicrobial activities of the extracts, fraction against gram positive, negative bacteria revealed that the methanol extract, hexane, ethyl acetate, and chloroform fraction of rosemary caused Staphylococcus aureus and Escherichia coli to form clear zones greater than 12 mm. Furthermore, the methanol extract and chloroform fraction showed high antibacterial activity, with inhibition zone exceeding 13 mm. The methanol extract and chloroform fraction of rosemary had broad antimicrobial spectrums and low MIC values. Therefore, methanol extracts of rosemary could serve as potential antibacterial agents to inhibit pathogen growth in food and hand sanitizers.

Preparation of Fermented Sauces with Increased ACE Inhibitory and Antioxidative Activities (ACE 저해능 및 항산화능이 향상된 발효소스의 제조)

  • For development of premium sauce, fermented sauces containing ferment (with wheat flour and soybean) and extract(s) of Styela clava (Korean name: miduduk) tunic and/or mulberry leaves and/or onion were prepared, and their angiotensin converting enzyme (ACE) inhibitory and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were evaluated. All sauces containing extract(s) showed increased ACE inhibitory and DPPH radical scavenging activities. Especially, sauces containing extracts of mulberry leaves and onion showed superior activities, with 26.92% and 40.66% relatively increased ACE inhibitory and DPPH radical scavenging activities, respectively, compared to control (no extract was added). These results suggest that extracts of mulberry leaves and onion could improve antihypertensive and antioxidant activities of fermented sauce.

Antioxidative Activities of Anthocyanins in Red Mustard Leaf Kimchi (적갓김치 Anthocyanins의 항산화 특성에 관한 연구)

  • Red mustard leaf (RML) kimchi was prepared with native Brassica juncea czerniak et coss as a major raw material and antioxidative characteristics of crude anthocyanin (CA)/refined anthocyanin (RA) isolated from RML kimchi were studied. Among crude pigment fractions, CA fraction showed a higher inhibition rate of peroxide formation in linoleic acid autoxidation system. These effects seemed to be increased with the increase of CA concentration. CA also was considered to have a role of electron donor by the experiments using DPPH ($\alpha$, $\alpha$'-diphenyl-$\beta$-picrylhydrazyl) reaction. Furthermore, various RA fractions had a higher antioxidative activity and especially RA-fraction 3 demonstrated the highest score of the antioxidative characteristics in terms of the prevention of lipid peroxidation and scavenging of free radicals.

Antioxidant Activities of Solvent Extracts from Pomegranate Endocarp (석류 내피 용매별 추출물의 항산화 활성)

  • This study was designed to investigate antioxidant activities of pomegranate endocarp extracts. Pomegranate endocarp extract contains the highest antioxidant function compared to pomegranate extracts from other parts. Pomegranate endocarp extract was fractionated with hexane, dichloromethane, ethyl acetate, butanol extract, and water, followed by evaluation for antioxidant activity. During this experiment, various antioxidant tests such as nitrite scavenging activity, reducing power, superoxide anion scavenging activity, $ABTS^+$ scavenging activity, SOD like-activity, and DPPH radical scavenging activity were conducted on the $CH_2Cl_2$, EtOAc, BuOH, $H_2O$ fractions of pomegranate endocarp extract. Results showed that the ethyl acetate fraction contained the highest DPPH radical scavenging activity among the samples. For SOD like-activity, the dichloromethane fraction had the highest antioxidant activity. For superoxide anion scavenging activity, the ethyl acetate fraction had the highest antioxidant activity. In the $ABTS^+$ scavenging activity test, the ethyl acetate fraction $IC_{50}$ was 39.26 ${\mu}g$/mL, whereas that of the butanol fraction was 40.95 ${\mu}g$/mL. In testing reduction power at 0.1 mg/mL, the O.D. of the ethyl acetate fraction was highest at 1.404 and showed higher activity than ascorbic acid at 1.332. The results of the nitrite scavenging activity test were very similar to those of the SOD like-activity test. These results suggest that pomegranate endocarp extract may have value as a natural antioxidant.

Changes of Contents of ${\beta}-Carotene$ and Vitamin C and Antioxidative Activities of Juice of Angelica keiskei stored at different conditions (저장조건에 따른 신선초 생즙의 베타카로틴과 비타민 C의 함량 및 항산화능의 변화)

  • Contents of ${\beta}-carotene$ and vitamin C in the vegetable(Angelica keiskei) juice were measured as a function of storing temperature$(-18^{\circ}C{\sim}35^{\circ}C)$ and period$(6{\sim}72\;hours)$. Content of ${\beta}-carotene$ was highest in the fresh juice and the degree of destruction of the ${\beta}-carotene$ was significantly increased as the storage temperature increased. Vitamin C in the juice stored at $4^{\circ}C$ decreased less than that of ${\beta}-carotene$. There was no significant difference in fatty acid compositions among in the fresh, freeze-dried juice and juice stored for 72 hours. Antioxidative activities of components extracted from the juice(fresh and stored for 24 hours) followed by incubating for 1 day were higher than that of ${\alpha}-tocopherol$ and then significantly decreased as incubation prolonged.

Major Compound Analysis and Assessment of Natural Essential Oil on Anti-Oxidative and Anti-Microbial Effects (천연 에센셜 오일의 주요 구성물질 분석과 항산화 및 항균 효과에 관한 연구)

  • We studied the physical, chemical, biological, and antimicrobial effects of eight types of essential oils used in the cosmetics industry: lavender, tea tree, rosemary, juniper berry, Chamaecyparis obtusa, cypress, cedar wood, and pine. Lavender oil had a linalyl acetate (an ester chemical compound) content of 48% and radical scavenging activity of 22.36% at 5,000 ppm. Tea tree oil had radical scavenging activity of 43.94% at 5,000 ppm and antimicrobial activity against S. aureus, S. epidermidis, S. mutans, and C. albicans in each 6, 3.5, 6.5, and 5 mm, respectively. Chamaecyparis obtusa oil had the highest acidity (pH 2.64) compared with the other oils, and sesquiterpene compounds were found to have 19.20%. Cedar wood oil had the highest specific gravity and refractive index compared to the other oils and had a sesquiterpene content of 99.73%. The radical scavenging activity of cedar wood essential oil exceeded 39.68% at 5,000 ppm. The clear zone, indicating antimicrobial activity against P. acnes, P. ovale, and C. albicans, was 3.5, 6, and 6 mm, respectively, at a concentration of 1% cedar wood oil. Results showed that with a high sesquiterpene content, the antioxidant effect was generally, but not always, high, suggesting that this is determined according to composition of the compound rather than presence of each antioxidant. The results indicate that antimicrobial activity is determined by the existence of each antimicrobial ingredient rather than terpene composition.
