Effect of Fermented Natural Materials Mixtures with Lactobacilluse acidophilus on Anti-obesity Activity (유산균 발효 천연소재 혼합물의 항비만 효과)

  • 최근 식생활의 변화로 인한 동물성 포화지방, 고칼로리 식품의 섭취 증가와 운동 부족, 스트레스 등의 여러 요인으로 비만인구 비율이 증가 추세를 보이고 있다. 유자, 석류등 천연소재 추출액에는 플라보노이드 성분이 풍부하여 항암, 항산화, 항염증, 항미생물 활성 등 생리활성이 있는 것으로 보고된 바 있다. 발효유로 대표되는 유산균 제품은 장 건강, 항산화 등 다양한 장점을 지니고 있으나, 짧은 유통기간 및 냉장유통 등 보존성에 있어 취약점을 가지고 있다. 따라서 천연 추출물을 주원료로 Lactobacillus acidophilus로 발효한 천연소재발효물을 활용하여 항비만 효과에 관한 연구를 수행하였다. 유산균 발효유의 풍미 유지하면서 유통기간을 늘릴 수 있는 가공법을 개발하고 건강식품 소재 개발 및 산업화를 통하여 국민의 건강 증진을 도모하고자 본 연구를 수행하였다. 천연소재 프로바이오틱스 발효물을 제조하여 세포독성, NO생성 억제능, 항비만 활성, 관능검사를 수행한 결과는 다음과 같다. 천연소재 추출물 및 천연소재추출발효혼합물을 각각 10, 50, 100, $500{\mu}g/mL$의 농도로 처리한 시험구들의 세포생존율을 측정한 결과 가장 고농도인 $500{\mu}g/mL$의 농도에서 각각 96.2, 95.1, 97.3, 96.2, 98.1, 97.0 %로 확인되어, 높은 수준의 안전성이 확인되었다. 천연소재 추출물 및 천연소재추출발효혼합물의 항염증효과를 분석하고자 수행한 Nitrite 생성저해 효과를 분석한 결과, NO 생성율은 $500{\mu}g/mL$의 농도에서 각각 98.1, 98.2, 96.2, 97.3, 91.2, 95.3 %로 확인되어 시험구 D의 항염증 활성이 가장 높은 것으로 나타났다. 동일한 농도로 처리하였을 때 lipid accumulation은 각각 100.0, 99.1, 98.2, 98.5, 87.6, 91.8 %로 나타나 D시험구와 E시험구에서 지방세포 분화억제효과가 확인되었다. 관능검사 시행 결과, 농축비율에 따른 관능적 기호도는 5 brix > 3 brix > 1 brix > 10 brix 순으로 나타났다.

Evaluation of Lipid Accumulation's Inhibitory Activity on 3T3-L1 Cells with Red Yeast Barley Extracts (홍맥 추출물의 3T3-L1세포에 대한 지방 축적 저해 활성평가)

  • Red yeast rice has been extensively used as food and traditional medicine for thousands of years in East Asian countries. It is produced by the fermentation of a particular yeast (in general, Monascus purpureus) as rice and various cereals (barley, soybean, etc.). Monascus sp. produces many secondary metabolites during its growth, including pigments, monacolins, and γ-aminobutyric acid. Some metabolites―specifically, monacolin K, γ-aminobutyric acid, dimerumic acid, and monascus pigments―have been reported to lower cholesterol and blood pressure while showing anti-obesity effects. In this study, we investigated the anti-obesity effect of ethanol extract from red yeast barley (RYB) fermented with Monascus sp. BHN-MK 2 on 3T3-L1 cells. The anti-obesity effects of RYB extract were examined: its lipid accumulation inhibitory effect was tested by Oil Red O staining, and obesity-related mRNA expression levels were tested by real-time RT-PCR in MDI stimulated 3T3-L1 cells. The intracellular lipid content of MDI-stimulated 3T3-L1 cells decreased significantly to 5.04%, 12.24%, and 23.52% in response to 200, 400, and 800 ㎍/ml RYB, respectively. Moreovers, we evaluated that RYB extract significantly downregulated the expression of C/EBPα, SREBP-1, and PPAR-γ gene in a dose-dependent manner. As a result, red yeast barley ethanol extracts exerted the strongest anti-obesity effects. Also, the results indicate that red yeast barley could be used as a functional anti-obesity food material.

  • Objectives: This study was conducted to analyze anti-obesity effects of the main single medicinal herbs which have effects on obesity or metabolic syndrome. Methods: We selected the 8 main single medicinal herbs, and then 3 databases were searched using search words "obesity", and "each scientific name of selected 8 main herbs". Results and Conclusions: Only Ephedra sinica and Panax ginseng were analyzed through 3 methods, in vitro, animal studies and randomized controlled trial (RCT)s, Ephedra sinica was shown weight loss effects in RCTs, but Panax ginseng couldn't. Other 6 selected medicinal herbs were not verified in RCTs, all had anti-obesity effects in high fat fed mice (or rats) and the others except for Radix notoginseng and Radix puerariae had inhibitory effects on lipid accumulation in 3T3-L1 cells.

Anti-obesity Effect of Sargassum confusum Ethanol Extract in Obese Rats (알쏭이모자반 주정추출물의 항비만 효과)

  • Effect of Sargassum confusum extract on the reduction of body weight gain and lipid contents in obese rats were evaluated to find natural materials with anti-obesity benefits. After inducing obesity by feeding 42.5% high-fat diet for 5 weeks, each 10 Sprague-Dawley rats were randomly assigned to high-fat diet control (HFD) group and high-fat diet group containing 3% Sargassum confusum extract (HFDSC). Weight gain of HFD group ($2.96{\pm}0.31g/day$) was significantly (p<0.05) higher as compared to that of normal diet (ND) group ($2.19{\pm}0.17g/day$). Weights of adipose tissues of HFD group were higher than those of ND group. Body weight gain of HFDSC group, however, was $2.36{\pm}0.24g/day$, which was significantly (p<0.05) lower by 21% than that of HFD group. In addition, weights of epididymal and perirenal adipose tissues were lower by 15% and 16%, respectively, as compared to those of HFD group. Biochemical analyses showed that concentration of triglyceride, total cholesterol, and fatty acids were significantly (p<0.05) lower in HFDSC group. These results suggest that Sargassum confusum extract has a high potential as an anti-obesity material by reducing weight gain and obesity-related factors in serum.

Effects of fermented blueberry liquid in high-fat diet-induced obese C57BL/6J mice (블루베리발효액이 식이유도 비만 Mice의 비만에 미치는 효과)

  • Purpose: The objective of the present study was to determine whether fermentation can increase the protective effects of blueberry liquid in a high-fat diet-induced obese mice model. Methods: Male C57BL/6J mice were fed a high-fat diet (HD, 60% fat, w/w,), HD supplemented with 10 ml/kg BW/day of blueberry liquid (BHD, blueberry high-fat diet), or HD supplemented with 10 ml/kg BW/day of fermented blueberry liquid (FBHD, fermented blueberry high-fat diet) for 10 weeks. Results: There were significant decreases in the body, epididymal adipose tissue, and liver weights of blueberry-fed groups compared to HD, whereas there were no significant differences in food intake among the groups. Furthermore, blueberry liquid groups, especially fermented blueberry liquid, significantly attenuated the contents of hepatic triglycerides and total cholesterol induced by HD. Serum LDL-cholesterol was significantly lower in the BHD and FBHD-fed groups, whereas FBHD significantly increased the serum HDL-cholesterol level compared to the control. Concentrations of aspartate transaminase, alanine transaminase, and leptins in serum were also reduced by blueberry liquid supplementation. The mRNA expression of hepatic acetyl CoA carboxylase was significantly reduced in both the BHD and FBHD groups compared to HD. Furthermore, FBHD altered the mRNA expression level of hepatic lipolysis genes. Conclusion: In conclusion, these results suggest that blueberry, especially fermented blueberry liquid, may improve obesity-related abnormalities.

Effects of the Red Garlic Extract for Anti-Obesity and Hypolipidemic in Obese Rats Induced High Fat Diet (홍마늘 추출물이 고지방식이로 유도된 비만 흰쥐의 항비만 및 지질개선에 미치는 영향)

  • This study tested the anti-obesity and hypolipidemic effects of red garlic extract in obese rats induced by a high fat diet over a period of 4 weeks. Red garlic extract of 15 brix was added in 1, 3, 5 and 7% ratios in diets. The obesity index and body fat content significantly decreased in rats fed a diet with over 3% red garlic extract compared to the control group. There was no significant difference in weight of visceral and epididymal fat in rats fed red garlic extract. Total lipid and triglyceride levels in serum were significantly decreased in a dose-dependent manner, and AI and CRF also fell. ALT and AST activities in groups fed red garlic extract were decreased compared to the control group. Total lipid level in liver tissue of the groups fed 5-7% red garlic extract exhibited a significant decrease compared to the control group. Total cholesterol and triglyceride levels in feces were significantly increased in rats fed a diet with over 5% red garlic extract. Lipid peroxide levels were significantly decreased in the groups fed diets with 5-7% red garlic and antioxidant activity in serum was significantly increased in the group fed a diet with 7% red garlic extract compared to the control group. Our results suggest that red garlic extract could have anti-obesity and hypolipidemic effects for suppressing obesity index and decreasing lipid profiles.
