Structural Characterization of the Anti-Complementary and Macrophage Activating Polysaccharides Isolated from Agaricus bisporus (양송이에서 분리한 보체계와 대식세포 활성화 다당류의 구조적 특성)

  • Three kinds of anti-complementary system and macrophage activating polysaccharides, AB-20-Ia, AB-20-IIa-2a and AB-20-IVa-2 were isolated from the fruit body of Agaricus bisporus and their structures were characterized. The proteoglycan, AB-20-IVa-2 showing the most potent anti-complementary and macrophage activity was composed of glucose, galactose, mannose, xylose, fucose and arabinose in a molar ratio of 3.48:1.83:1.00:0.79:0.74:0.11 and its main component amino acids were phenylalanine (34.72%) and valine (27.84%). The neutral polysaccharides, AB-20-Ia and AB-20-IIa-2a showing lower activity than AB-20-IVa-2, consisted of xylose, glucose, mannose, fucose and arabinose in molar ratios of <0.05:<0.05:2.07:1.00:2.72 and 2.16:1.58:1.00:0.20:0.14, respectively. The molecular weights of AB-20-Ia, AB-20-IIa-2a and AB-20-IVa-2 were 840,000, 750,000 and 650,000 respectively. In the $^1H-\;and\;^{13}C-NMR$ spectra of AB-20-Ia and AB-20-IIa-2a, AB-20-Ia showed only ${\beta}-configuration\;(^1H:\;4.8\;ppm,\;^{13}C:\;107.0\;ppm)$ in the anomerization of the glycosidic linkages, while AB-20-IIa-2a had both ${\alpha}-anomer\;(^1H:\;5.4\;ppm,\;^{13}C:\;102.0\;ppm)\;and\;{\beta}-anomer$. Especially, AB-20-Ia and AB-20-IIa-2a showed acetyl signals $(^1H:\;2.5\;ppm,\;^{13}C:\;21.0\;ppm)$. In the methylation analysis of the three polysaccharides, high proportion of 1,6-linked glucofuranosyl residues were detected in AB-20-Ia, whereas 1,6-linked glucopyranosyl residues and branches linked at position 4 of those mainly contained in AB-20-IIa-2a. AB-20-IVa-2 consisted mainly of 1,2-linked xylofuranosyl residues and 1,6-linked glucopyranosyl residues and branches linked at position 3 of those.

Pharmacological Effects of Proteoglycans Extracted from Fruiting Bodies of Fomitella fraxinea (장수버섯 자실체의 열탕추출액으로부터 분리한 단백다당체의 약리적 효과)

  • Anti-complementary assay for immuno-stimulating polysaccharide screening, human tumor colony-forming assay for discovering anti-tumor drugs, and toxic assay against mouse were performed to examine pharmacological activities of polysaccharides extracted from fruiting bodies of Jang-soo mushroom (Fomitella fraxinea). Hot water $(100^{\circ}C,\;FF-I)$, 1% ammonium oxalate solution $(80^{\circ}C,\;FF-II)$, and 5% sodium hydroxide solution $(80^{\circ}C,\;FF-III)$ were used for extraction of three polysaccharides from fruiting bodies of it. Anti-complementary activity of FF-I was more effective than the others. FF-I was further fractionated into three groups of polysaccharide by DEAE-Sephadex A25 column chromatography (FF-NP, FF-AP1, and FF-AP2). FF-AP1 showed not only the highest anti-complementary activity but also the growth-inhibitory activity against Snu-I (human stomach cancer cell) among 9 kinds of human tumor cell lines. But FF-AP2 exhibited its activity against Hep-2(larynx cancer) and KB(mouth epidermal cancer) cell lines at $500\;{\mu}g/ml$ although its anti-complementary activity was lower than it of FF-AP1. When FF-I was orally administrated to mice with dosage of 5000 mg/kg, no remarkable changes were observed in viewpoint of tissue-pathology.

Purified Polysaccharide Activating the Complement System from Leaves of Diospyos kaki L. (감잎(Diospyos kaki L.)으로부터 정제한 보체계 활성화 다당류)

  • Cold and hot water fractions of Diospyros kaki were screened to determine its anti-complementary activity. Flour of Diospyros kaki leaf (250 g) was boiled at $100^{\circ}C$ for 3 h and passed through a membrane of 10 kDa molecular weight (DK-0). DK-0 was precipitated with ethanol and refluxed with methanol to obtain the crude polysaccharide (DKC). DKC-1 was isolated by ion exchange chromatography on DEAE-Toyopearl 650C, and DKC-1c was purified from DKC-1 by size exclusion chromatography on Bio gel P-60. The anti-complementary activities of DKC-1c at $1000\;{\mu}g/mL$ were 85.4 and 61.1% via whole and alternative pathways, respectively. DKC-1c was determined as a neutral polysaccharide composed of glucose (29.0 mol.%), arabinose (24.3 mol.%), and galactose (16.2 mol.%) with the molecular weight of 66.6 kDa. Results of agarose gel immunoelectrophoresis revealed DKC-1c, as a complement activator, cleaved C3 into C3a and C3b via both pathways.
