  • 지금까지 많은 항균성 펩타이드가 포유류 (1), 양서류 (2), 곤충 (3) 및 식물 (4)로 부터 단리ㆍ정제되었고, 구조-활성간의 연구가 활발히 진행되고 있지만, 이러한 연구는 육상생물에 제한적으로 수행되어 겼으며 해양생물을 대상으로 한 연구는 극히 제한되어 있는 상태이다. 해양생물로부터 정제된 항균성 펩타이드로서는 Flounder skin 분비물에서 pleurocidin (5), Moses sole fish에서 pardaxins (6), Horseshoe crab에서 Tachyplesin I(7), Tunicates hemocytes에서 Clavanins (8), Mollusc에서 Mytilins (9) 등이 보고되고 있지만 불가사리를 이용한 항균성물질의 연구는 희박한 실정이다.   • The effect of combinations of 8 antimicrobial agents on the growth of Streptococcus mutans and Streptococcus sobrinus was investigated with the minimum inhibitory concentration and fractional inhibitory concentration(FIC) and ${\Sigma}FIC$ index. According to the ${\Sigma}FIC$ values by. The American Society for Microbiology and Berenbaum, Approximately 34% and 82% of the combinations were synergistic respectively. Partial synergy described by Isenberg was also observed in the half of the combinations. There was a tendency for additive antimicrobial effect against cariogenic bacteria though the test results showed difference according to the applied values. It may be beneficial to use combined antimicrobial agents that have various activities against an ecosystem and metabolism of bacteria than using individual agents.

  • - A phthalic acid derivative and basic macrolide antibiotics, with antimicrobial activity against Gram positive bacteria and phytopathogenic fungi, respectively, were found to be produced by a strain 88-GT-161 identified as being a variety of Streptomyces melanosporofaciens. This paper describes an isolation procedure of the active compounds produced by this strain, their in vitro and in vivo (pot test) antimicrobial activites, and structure determination of one of the compounds, bis (2-ethylhexyl) phthalate, a phthalic acid derivative antibiotic. This compounds, upon cornparision with authentic bis (2-ethylhexyl) phthalate, dioctyl phthalate, revealed a difference in antimicrobial activity even though physico-chemical properties of these two compounds seemed indentical. This is the first report that dioctyl phthalate is biosynthetically produced by a Streptomyces sp. and shows antimicrobial activity.

  • Low density polyethylene(LDPE) film was fabricated with the addition of benzoic acid, or/and JP which contained water-soluble silica adsorbed a natural antimicrobial compounds. The used antimicrobial compounds were isolated from culture broths of methylotropic actinomycetes strains MO-16 and MO-17 which was newly isolated from soils. An antimicrobial compound retained antimicrobial activity after heat treatment at 121$^{\circ}C$ for 5 min. JP, which obtained from Japan as a antimicrobial agent, showed antimicrobial activity in the concentration of 100 mg/mL. The silica LDPE film revealed the growth inhibition of total aerobic bacteria in packaged minced pork compared with commercial film and of E. coil on a contained agar plate. In the storage testing of various packaged foods at room temperature for 30 days, the film showed excellent preservation compared with commercial film in case of small tomato and Agaricus bisporus.

  • An oxytetracycline as being a tetracycline-antibiotic substance displayed a general synergism in the antimicrobial activity by the interaction of ginseng saponin and antibiotics, but did not to Sarcina maginata. Penicillin G.Na as being a $\beta$-lactam-antibiotic substance displayed a synergism in the antimicrobial activities by the addition of ginseng saponin to microorganisms used, but changed the effects in the antimicrobial activity of penicillin G.Na against the genus Serratia. An antimicrobial activity by the addition of ginseng saponin to antibiotics showed a non-specificity, and it varied synergism or antagonism to Gram-positive or Gram-negative bacterium. It was assumed that a drug-resistance could be eliminated by the dual administration of ginseng saponin and antibiotics.

  • The possible effects in vivo on the duel usage of sinseng saponin and antibiotics were studied in vitro with microorganisms. Streptomycin.sulfate, kanamycin.sulfate and gentamycin.sulfate as being an aminoglycoside-antibiotic substance showed a general synergism by the interaction of ginseng saponin and these antibiotics. But kanamycin.sulfate and gentamycin.sulfate did not show a synergism in their original antimicrobial activity against Er-winia aroideoe. Chloramphenicol as being a benzene derivative displayed an increased antimicrobial activity by the interactions of ginseng saponin and this antibiotic against Salmonella typhi, Aerobacter aerogenes and the genus Serrotia. This antibiotic also showed the decreased antimicrobial activity against Bacillus subtilis, Bacillus megaterium and Escherichia coli, but did not show an uniform antimicrobial activity against others.

토양균에서 항생물질의 분리와 구조연구

  • 국내의 토양에서 Streptomyces spp.를 분리하였고 이 중 항균활성이 우수한 균주들을 선별하였다. 이들 균주들을 적절한 조건에서 배양하여 얻은 배양액을 이온 교한 수지와 흡착 크로마토그래피를 행하여 항생물질들을 분리하였다. 이들 항생물질들은 Bacillus subtilis, Pyricularia oryzae, Alternaria mail, Candida albicans, Mycobacterium smegmatis, Pseudomonas aeruginosa , 및 Escherichia coli에 강한 항균활성을 나타내었다. 각 항생물질들을 산으로 가수분해하고 흡착관 크로마토그래피로 가수분해 생성물을 분리하였다. 분리한 항생물질과 가수분해 생성물들의 $^1$IH-NMR, $13^{C}$ -NMR, FAB-Mass 스펙트럼을 분석하여 구조를 연구하였다.

Antimicrobial Substances of Distilled Components from Mustard Seed (겨자 증류성분중의 항균성 물질)

  • To investigate the antimicrobial activities of mustard seed(Brassica juncea), a series of solbents were examined for extraction of antimicrobial substances from mustard seed and then antimicrobial substances were identified by simultaneous distillation extraction(SDE) and GC-MS methods. Water and methanol extracts of mustard seed showed antimicrobial activitries against experimental strains while those from hexane extract almost was not observed. The distilled components of mustard seed by SDE method showed strong antimicrobial activities, methanol extract of residues fraction exhibited a little, and water layer with the exception of distilled components showed no antimicrobiol activities. The 30 varieties of distilled components including 3 types of isothiocyanate such as 3-isothiocyanate-1-propene, 4-isothiocyanate-1-butene, and 3-isothiocyanatoethyl benzene were identified from mustard seed.

Antimicrobial Effect of Caesalpinia sappan L. Extract on Foodborne Bacteria (식중독 세균에 대한 소목 추출물의 항균 활성)

  • To develop a natural antimicrobial agent, we investigated the antimicrobial activities of 13 species of edible herbal plant extracts against major Gram-positive foodborne bacteria. Among the 13 screened edible herbal plants, Caesalpinia sappan L. showed the highest antimicrobial activity. In the paper disc agar diffusion assay, Caesalpinia sappan L. extracts had strong antibacterial activities against most Gram-positive bacteria but did not have antibacterial activities against most Gram-negative bacteria. Minimum inhibitory concentrations of the ethanol extract were 0.06 mg/mL against Clostridium difficile and Listeria monocytogenes and 0.03 mg/mL against Staphylococcus aureus. Their inhibitory activities were not reduced by heat treatment or pH adjustment against C. difficile, L. monocytogenes, and S. aureus. Antimicrobial activities were higher in ethanol extract than in distilled water extract. These results support the potential use of Caesalpinia sappan L. ethanol extract as an antimicrobial agent or functional food components against Gram-positive bacteria.
