The Antioxidative and Antimicrobial Effects of Celastrus orbiculatus (노박덩굴 추출물의 항산화 및 항균 효과)

  • This study was carried out to investigate the antioxidative and antimicrobial activities of Celastrus orbiculatus (CO). CO was extracted with methanol and then further fractionated with n-hexane (COMH), butanol (COMB), methanol (COMM) or aqueous (COMA) to get active fractions. The antioxidative activity of fractions from CO was investigated by measuring the scavenging activities of CO against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen speicies (ROS). Among the various solvent fractions, the COMB showed a marked scavenging effect against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species(ROS). The antimicrobial activity was increased in proportion to its concentration by the paper disk method. Among the various solvent fractions, the COMM and COMB fractions of CO showed strong antimicrobial activities. The results suggest that The CO may be suitable for development as a food preservative and alternative antioxidant.

Antimicrobial Activities of Sophorolipids and Its Application for Cosmetics (Sophorolipid의 항균효과와 화장품에의 응용)

  • Some surfactants known as biosurfactants, are produced biologically by yeast and bacteria from various substances. They are more effective and environmentally friendly than many other synthetic surfactants. Antimicrobial activities of sophorolipids produced by Candida bombiocola were investigated against various microorganisms. Minimal Inhibitory concentration (MIC) of sophorolipid against S. mutans, C. xerosis, and P. acnes were 0.005, 0.05, and 0.005%, respectively. The antimicrobiological activities were more effective than those of SLS and APG. Skin exfoliating and moisturizing effects of vehicles with the sophorolipids were tested. The skin turnover time of aqueous solution with 0.25% sophorolipids was similar to that of aqueous solution with 5.0% lactic acid. Higher moisturizing effects on skin showed as the concentration of sophorolipids increased. We suggest that the sophorolipids can be used for cosmetics as an antimicrobiological agent and an active materials of skin moisturizing and exfoliating.

Antibacterial and Antioxidative Activity of Roasted Coffee and Red Ginseng Mixture Extracts (로스팅 커피와 홍삼 혼합추출물의 항균 및 항산화 효과)

  • This study was conducted to investigate the antibacterial and antioxidative activities of water and ethanol extracts from a mixture of roasted coffee and red ginseng. The antibacterial effects of each extract were determined by the classical paper disc method. A water extract of mixture samples inhibited growth of all strains, but antibacterial effects were mostly weakened. Ethanol extracts showed stronger antibacterial effects than water extracts in all strains except Gram negative Escherichia coli and the fungi strain Candida albicans. Also, the antibacterial effect of the Bacillus cereus strain appeared in all samples, and the ES2 sample formed a clear zone of 19 and 20 mm against Pseudomonas aeruginosa and S. Typhimurium respectively (MIC=0.25 and 0.125 mg/mL). Determinations of free radical elimination for the different mixture extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) were compared with ascorbic acid and butylated hyderoxytoluene as positive controls. The water and ethanol extracts of mixture samples (100 ${\mu}g/mL$) showed 55.38~60.01% and 59.37~70.50% DPPH scavenging activities, respectively. DPPH scavenging activities of all mixture samples were slightly higher than roasted coffee and red ginseng samples. However, DPPH scavenging activity decreased when red ginseng extract composed more than 70% of the total extract. The total polyphenol in the mixture samples measured by the Folin-Denis method revealed the highest level of polyphenol content in ethanol extract of sample 3, whereas polyphenol content differed with different mixture ratios, ranging from 105.16~119.79 mg/g in ethanol extract. In the water extract, the polyphenol content was greatest with water extract of sample 1, whereas in other samples the content varied from 93.75~109.18 mg/g.

THE ANTIMICROBIAL EFFECT OF HORSERADISH(ARMORACIA RUSTICANA) ROOT EXTRACTS AGAINST ORAL PATHOGENS (구강내 미생물에 대한 서양산 고추냉이(Armoracia rusticana) 뿌리 추출물의 항균효과)

  • In this study, the antimicrobial effects of Horseradish (Armoracia rusticana) root extracts against oral pathogens were investigated, and also compared with that of chlorhexidine. The following 7 microorganisms were used in this study, Streptococcus mutans ATCC 25175, Streptococcus sobrinus(d) ATCC 27607, Lactobacillus casei ATCC 393, Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 29212, Actinobacillus actinomycetemcomitans ATCC 29522. Candida albicans ATCC 10261. Horseradish root extracts and chlorhexidine were tested to determine their minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC). The results of this study can be summarized as follows : 1. Horseradish root extracts showed antimicrobial effect against the tested oral pathogens. MIC and MBC of this extracts were 30-125, 125-500ppm, respectively. Especially, it was the most effective against C. albicans of other tested microorganisms. 2. Chlorhexidine also showed antimicrobial effect against the tested oral pathogens. MIC of chlorhexidine range between 0.15 and 2.5%, MBC are 0.4-2.5%. In conclusion, it was suggested that AIT had similar antimicrobial effects in the lower concentration, compared with that of chlorhexidine.

Combined Effects of Antibacterial Film and Storage Temperature on Shelf-life and Microbiological Safety of Mackerel (항균 필름과 저장 온도의 고등어 저장성 및 미생물학적 안정성에 대한 병행 효과)

  • The purpose of this study was to investigate the antibacterial effect of films on shelf-life and microbiological safety of mackerel. Effects of antimicrobial films against total aerobic bacteria, Escherichia coli O157:H7, Listeria monocytogenes, Vibrio parahaemolyticus in mackerel were evaluated during storage of 5-14 days at $5^{\circ}C,\;10^{\circ}C\;and\;15^{\circ}C$. Antimicrobial films were developed with addition of a natural substance, wasabi extracts (Wasabia japonica). At $5^{\circ}C$ storage, growth of total aerobic bacteria, Escherichia coli O157:H7, Listeria monocytogenes were inhibited higher than at 10 and $15^{\circ}C$. Especially, the numbers of V. parahaemolyticus were decreased gradually at $5^{\circ}C$ even in the control sample, and about $1\;log_{10}cfu/g\;and\;1.8\;log_{10}cfu/g$ reductions were observed at 1 and 4 days, respectively. After 7 days, V. parahaemolyticus in all samples were not detected. There is a limit of a single treatment of antimicrobial film to prolong shelf-life of mackerel. The synergistic effect of antimicrobial film were shown by addition of $5^{\circ}C$ refrigeration.

  • This study was carried out to measure the antibacterial effect of lactoperoxidase system against L. monocytogenes. When the initial inoculum levels ($10^{2},\;10^{4},\;10^{7}\;CFU/ml$), concentration of LP (10 ppm, 20 ppm, 30 ppm), culture media (TSB-YE, UHT milk) and storage temperatures ($5^{\circ}C,\;10^{\circ}C,\;15^{\circ}C$) were set up differently for the experiment and the antibacterial effect was compared, the highest antibacterial effect of LP system was shown at $10^{2}\;cfu/ml$ of initial inoculum level, 10 ppm of LP concentration and $5^{\circ}C$ of incubaction temperature. The antibacterial effect of LPS in UHT milk was similar to that in Tryptic soy broth.

  • 유산균(Lactic acid bacteria)은 Escherichia coli와 Salmonella typhimurium과 같은 병원균에 항균 활성을 지니며 면역 증강효과를 나타내는 등 건강에 이로운 다양한 역할을 한다. 유산균에 의한 항균 효과는 E. coli와 S. typhiumurium에 대항하는 항균 활성으로 측정되어 졌으며, 면역 증강 효과는 유산균을 처리한 RAW264.7 대식세포의 활성화로 측정하였다. Lactobacillus acidophilus, Streptococcus thermophilus, Bifidobacterium bifidum의 E. coli와 S. typhimurium에 대한 항균활성은 9시간 이상 혼합 배양하였을 때 가장 좋은 항균효과를 나타내었고 E. coli와 S. typhiumurium 두 균주가 9시간 이후에는 모두 콜로니를 형성하지 않았다. RAW264.7 세포는 유산균에 의한 NO와 $TNF-{\alpha}$의 생성과 대식세포의 형태 변화를 알아보기 위한 대식세포로서 이용되었다. NO와 $TNF-{\alpha}$의 생성은 유산균을 처리한 RAW264.7세포의 24,48시간 배양 시 농도 의존적으로 증가하였고 대식세포의 형태 변화 역시 유산균에 의해 영향을 받았음을 확인할 수 있었다. 이를 통하여 (주)쎌바이텍으로부터 분양받은 유산균은 항균활성과 대식세포의 활성을 유도하여 면역을 증강시키는 효과를 지니고 있음을 in vitro 실험을 통해 확인하였다.

Preservative Effect of Natural Antimicrobial Substances Used as Steeping and Packaging Agent on Postharvested Strawberries (천연항균물질을 침지 및 포장소재로 이용한 딸기의 저장효과)

  • Coptis chinensis extract and grapefruit seed extract, natural antimicrobial substances, were applied to the dipping treatment of strawberry and incorporated in the packaging films. Strawberry was steeped in the extract solutions of 50 ppm concentration and packed with the low density polyethylene(LDPE) films incorporated with 1% extracts. During the storage at 5$^{\circ}C$, the qualities of microbial counts, awぶy ratio, texture and chemical attributes were measured for the pretreated strawberry. The LDPE films incorporated with the extracts retarded the growth of aerobic bacteria, lactic acid bacteria and yeast that had been contaminated before the pretreatment, significantly lowed the decay ratio, and gave better retention of textural firmness. The chemical and physical qualities of strawberry were not affected by the packaging films. When strawberry was steeped in the extract solutions, the effects of the packaging film incorporated with the extracts on the qualities of strawberry were accelerated.

Product Quality of Low-fat/salt Sausages Containing Lactoferrin and Antimicrobial Activity Against Escherichia coli O157:H7 (락토페린을 첨가한 저지방/저염 소시지의 품질 및 Escherichia coli O157:H7에 대한 항균 활성에 미치는 효과)

  • This study was performed to determine potential antimicrobial activity of lactoferrin, which incorporated into the low-fat/salt sausages during storage either at 30 or 4℃. First, the model study on the antimicrobial activity of lactoferrin was performed. Based on the model study, more than 0.25% of lactoferrin was required to have a distinctive antimicrobial activity for the growth of E. coli O157:H7 in the broth. However, extended shelf-life was not shown in the low-fat/salt sausages manufactured with lactoferrin during refrigerated storage. In addition, no synergistic effects of lactoferrin with sodium lactate were observed in the sausages. These results indicated that the addition of lactoferrin into the low-fat sausages did not have antimicrobial activity of E. coli O157:H7, due to the denaturation of lactoferrin or the complexity of the sausage, even though it had a distinctive antimicrobial effect in the model study.

Antimicrobial Effect of Lithospermum erythrorhizon Extracts on the Food-borne Pathogens (지치추출물의 식중독성 미생물에 대한 항균효과)

  • Antimicrobial effect of Lithospermum erythrorhizon extracts against food-borne pathogens was investigated. L. erythrorhizon was extracted with methanol at room temperature, and the extraction was sequentially fractionated using petroleum ether, chloroform, ethyl acetate, and methanol. Antimicrobial activity of L. erythrorhizon extracts was determined using paper disc method against food-borne pathogens and food spoilage bacteria. Ethyl acetate extracts of L. erythrorhizon showed the highest activity against Staphylococcus aureus and Shigella dysenteriae. Synergistic effect was found in combined extracts of L. erythrorhizon and Sophora subprostrata as compared with each extract alone. Growth inhibition curve was determined using ethyl acetate extracts of L. erythrorhizon, against S. aureus and S. dysenteriae. Ethyl acetate extract of L. erythrorhizon, showed strong antimicrobial activity against S. aureus at 4,000 ppm, retarding growth of S. aureus more than 48 hr and S. dysenteriae up to 12 hr.
