Characteristics of Citrus By-Product Ferment Using Bacillus subtilis as Starter Extracts. (Bacillus subtilis를 이용하여 발효시킨 감귤 가공부산물 추출물의 특성)

  • In this study, we investigated the biological activity of antioxidant and antibacterial activity of citrus by-product ferment. Hot water extracts and 80% methanol extracts from citrus byproduct of ferment were screened for antibacterial activity pathogenic bacteria by paper disc method. Among the various hot water extracts and 80% methanol extracts the Bacillus subtilis showed relatively strong antibacterial activities in the order. The reducing activity on the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and ${\cdot}OH$ radical scavenging potential were sequentially screened, in search for antioxidant activities of citrus by-product ferment.

Antimicrobial Effect of Caesalpinia sappan L. Extract on Foodborne Bacteria (식중독 세균에 대한 소목 추출물의 항균 활성)

  • To develop a natural antimicrobial agent, we investigated the antimicrobial activities of 13 species of edible herbal plant extracts against major Gram-positive foodborne bacteria. Among the 13 screened edible herbal plants, Caesalpinia sappan L. showed the highest antimicrobial activity. In the paper disc agar diffusion assay, Caesalpinia sappan L. extracts had strong antibacterial activities against most Gram-positive bacteria but did not have antibacterial activities against most Gram-negative bacteria. Minimum inhibitory concentrations of the ethanol extract were 0.06 mg/mL against Clostridium difficile and Listeria monocytogenes and 0.03 mg/mL against Staphylococcus aureus. Their inhibitory activities were not reduced by heat treatment or pH adjustment against C. difficile, L. monocytogenes, and S. aureus. Antimicrobial activities were higher in ethanol extract than in distilled water extract. These results support the potential use of Caesalpinia sappan L. ethanol extract as an antimicrobial agent or functional food components against Gram-positive bacteria.

대장균을 이용한 구리 함유 ZrN 박막의 항균성 향상에 관한 연구

  • ZrN 코팅은 TiN과 특성이 유사한 옅은 금색의 질화 박막이나 내식성이 우수하여 의료용, 자동차 부품, 항공부품, 장식용으로 적용되고 있다. 스테인레스계 의료용 기구나 생활기구의 항균성 기능이 부여된 표면처리는 아직 널리 보급되고 있지 않아 장식성과 내부식성이 뛰어난ZrN 박막에 구리를 함유시킴으로서 항균성의 변화를 관찰하고자 하였다. 진공 아크 이온플레이팅 방법으로서 ZrN을 성장시키면서 스퍼터링법에 의하여 Cu를 함유시키는 방법으로 실험을 실시하였다. 기재는 SUS 304를 사용하였고, 공정온도 $400^{\circ}C$에서 질소분압 1-5 Pa, 아크전류 90 A의 조건에서 Cu 스퍼터링 타겟의 전류를 1-7A 까지 변화하여 구리함유 ZrN 박막을 합성하였다. 구리 함류량에 따른 XRD를 통한 결정구조분석과 SEM/EDX를 통한 성분변화분석을 실시하였다. 구리함유 박막에 대해서 시간에 따른 대장균을 성장을 분석하여 기재인 SUS304, ZrN, 구리 함유 ZrN에 대한 대장균 항균성을 조사하였다.

C-3 Quarternized Cephalosporin 화합물 합성 및 항균 활성

  • 4세대 cephalosporin계 약물로 개발되고 있는 cefpirome, cefepime, SCE-2787과 같은 화합물은 항균 spectrum 및 항균력에서 3세대 약물인 cefotaxime, ceftazidime보다 우수하며 구조적으로 C-3 quarternized기률 함유한 특징을 가지고 있다. 본 연구소에서는 기존에 보고된 화합물보다 개선된 약물을 찾으려는 시도로서 benzimidazole, imidazolopyridine등 기타 유사구조를 가진 C-3 quarternized 화합물 합성을 하여 왔으며, 본 연구에서는 최근에 합성된 9종의 신규 화합물에 대한 합성 및 항균활성을 보고한다. 이중 9 종의 화합물은 시험한 균주에서 일반적으로 cefpirome 보다 우수한 항균 profile을 나타내었으나 실제 신약으로의 개발 가능성 평가를 위하여 자세한 연구가 필요한 것이었다 이 화합물에 대한 확립된 실험실적 합성 공정과 물성자료를 함께 보고한다.

Product Quality of Low-fat/salt Sausages Containing Lactoferrin and Antimicrobial Activity Against Escherichia coli O157:H7 (락토페린을 첨가한 저지방/저염 소시지의 품질 및 Escherichia coli O157:H7에 대한 항균 활성에 미치는 효과)

  • This study was performed to determine potential antimicrobial activity of lactoferrin, which incorporated into the low-fat/salt sausages during storage either at 30 or 4℃. First, the model study on the antimicrobial activity of lactoferrin was performed. Based on the model study, more than 0.25% of lactoferrin was required to have a distinctive antimicrobial activity for the growth of E. coli O157:H7 in the broth. However, extended shelf-life was not shown in the low-fat/salt sausages manufactured with lactoferrin during refrigerated storage. In addition, no synergistic effects of lactoferrin with sodium lactate were observed in the sausages. These results indicated that the addition of lactoferrin into the low-fat sausages did not have antimicrobial activity of E. coli O157:H7, due to the denaturation of lactoferrin or the complexity of the sausage, even though it had a distinctive antimicrobial effect in the model study.

The Effects on Antimicrobial and Anticarcinogenic Activity of Momordica Charantia L. (메탄올로 추출한 여주 분획성분의 항균 및 항발암 효과)

  • This study was performed to determine the antimicrobial and anticarcinogenic activities of the Momordica charantia L. (MC) on several microorganisms and human cancer cell lines. In the paper disk test, its antimicrobial activity was increased in proportion to its concentration. Among the various solvent fractions of Momordica charantia L., the ethylether partition layer (MCMEE) showed the strongest antimicrobial activity. Also, the ethylacetate partition layer (MCMEA) and the butanol partition layer (MCMB) showed antimicrobial activity. We also determined the cytotoxicity and chemopreventive effect of Momordica charantia L. extract and fractions on human cancer cells. The experiment was conducted to determine the cytotoxicity of Momordica charantia L. partition layers on HepG2, HeLa and MCF-7 cells by MTT assay. Among the various partition layers of Momordica charantia L., MCMEE and MCMEA showed strong cytotoxic effects on all cancer cell lines. The chemopreventive effect of the quinone reductase induced activities of HepG2 cell, the hexane partition layer (MCMH) at a dose of 50 $\mu\textrm{g}$/mL was 3.62 times more effective compared with the control values of 1.0. Therefore, based on these studies, Momordica charantia L. may be developed into a potentially useful cancer chemopreventive agent.

The development of the natural antimicrobial agents for the quality improvement of the packing pepper powder (포장 고춧가루제품의 품질 향상을 위한 천연 항균 물질 개발)

  • 최근 국산 농산물의 수요급증과 더불어 소비자들은 안전한 제품의 요구가 높아지고 있다. 이러한 요구에 맞는 제품 개발의 일환으로 미생물 제어가 된 고춧가루 포장제품의 품질향상을 위한 천연항균 물질 소재를 탐색하여 제품개발에 응용하였다. 천연물과 한방재료 중 고춧가루 제품에 적합한 소재를 스크리닝하여 항균물질을 탐색하였으며 고춧가루 제품 패킹에 응용하였다. 천연물 항균제의 개발 원칙은 산취 등 이취가 없어야 하며, 제품에 응용할 수 있는 용해성이나 믹싱의 효과를 높이도록 공정을 최적화 하였다. 또한 최종 천연물 항균제의 형태는 분말 형태로 개발하였다. 천연물에 대한 항균활성은 paper disk test를 통하여 비교하였으며, 항균력이 가장 우수한 소재는 황금추출물이었다. 또한 이들 추출물과 비타민 $B_1$ 인산염의 영향을 함께 비교하였다. 이 천연물 소재 추출물로 개발한 항균물질에 대한 효과는 일반세균, 대장균 검사를 실시하였다. 제품의 안정성 확보 및 유통기한 연장 가능성을 실험한 결과, 일반세균은 $1.0{\times}10^5$cfu/g, 대장균군은 $1.0{\times}10^2$cfu/g, 대장균은 불검출 되었다. 이들 추출물의 항균제 개발과 더불어 다양한 식품에 천연물 항균제로서의 적용 가능성을 모색하고자 하며, 안전한 천연 항균제 개발에 대한 기초 자료를 제공하는 기초 연구를 하고자 하였다.

Antifungal activities of sulphamide and dicarboximide fungicides against Botrytis cinerea in several in vitro bioassays (여러 종류의 in vitro 생물검정에서 Botrytis cinerea에 대한 sulphamide계와 dicarboximide계 살균제의 활성 특성)

  • Two sulphamide (dichlofluanid and tolylfluanid) and three dicarboximide fungicides (iprodione, vinclozolin, procymidone) were used to investigate the correlation between in vitro antifungal activities and in vivo disease controlling activities against Botrytis cinerea, a causal agent of tomato gray mold and to develop efficient in vitro assays. They controlled effectively the development of tomato gray mold disease in vivo and their controlling activities were similar one another. However, several in vitro assays revealed that their in vitro antifungal activities were quite different between sulphamide and dicarboximide fungicides; the formers showed stronger inhibition activities for spore germination than the latters, whereas the formers inhibited mycelial growth less severely than the latters. The results indicate that the fungicides having different modes of action can show different in vitro antifungal activities according to in vitro assays, even if they have similar in vivo disease controlling activities. On the other hand, two rapid and efficient in vitro assays named Microtiter plate methods I (MPM I) and II (MPM II) were developed for the evaluation of fungicides for inhibitory activities against spore germination and mycelial growth of B. cinerea, respectively. The antifungal activities of five fungicides of two chemical groups in MPM I and II were correlated with the inhibitory activities against spore germination and mycelial growth using solid media, respectively.

Antimicrobial Activities and Stability of Rhus Javanica L., Cinnamomum Verum and Rosmarinus Officinalis Extracts Used in the Manufacture of Cosmetics (화장품에 적용한 오배자·계피·로즈마리 추출물의 항균활성 및 안정성 검증)

  • This study attempted to investigate the usefulness of Rhus javanica L., Cinnamomum verum and Rosmarinus officinalis extracts which have antimicrobial and antioxidant effects without any harm on human health as natural preservatives. In terms of extraction, extraction by 70% ethanol and hot-water extraction were used for Cinnamomum verum and Rhus javanica L. respectively. For Rosmarinus officinalis, a mixed method (70% ethanol and hot-water extraction) was adopted. In terms of experimental methods, antimicrobial effects, antioxidant activity through DPPH and safety and stability of cosmetics were assessed, and a challenge test was performed, and the results found the followings: According to an antimicrobial test, good antimicrobial effects were found in bacteria (Rhus javanica L. extract) and fungi (Cinnamomum verum extract). In contrast, the Rosmarinus officinalis extract was set aside because of poor antimicrobial activity. In the mixed extract (Rhus javanica L. + Cinnamomum verum), antimicrobial effects were observed in 'complex C (mixed in a 1:1 ratio)' while both inhibitory and sterilizing effects were found in 5 different test strains at minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). In addition, antioxidant effects were detected in non-mixed extract and mixed extract ('complex C'). Furthermore, a test on cosmetics which adopted '10% complex C' instead of synthetic preservative revealed safety and stability. Therefore, this study has confirmed the potential of the Rhus javanica L., Cinnamomum verum and Rosmarinus officinalis extracts as materials for natural preservatives.

Antimicrobial Effect of Rubia akane Nakai Extract on Food-Borne Pathogens (식중독유발 세균의 증식에 미치는 천초근 추출물의 영향)

  • This study was performed to investigate the antimicrobial activity of the Rubia akane Nakai extract against food-borne pathogens. First, the Rubia akane Nakai was extracted with methanol at room temperature and the fractionation of the methanol extract was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol. The antimicrobial activity of the Rubia akane Nakai extract was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extract of Rubia akane Nakai showed the highest antimicrobial activity against Bacillus cereus and Pseudomonas aeruginosa. Synergistic antimicrobial effect was observed when Rubia akane Nakai extract was mixed with Viscum album var. coloratum extract as compared to each extract alone. Finally, the growth inhibition curves were determined by using methanol extract of Rubia akane Nakai against Bacillus cereus and Pseudomonas aeruginosa. The methanol extract of Rubia akane Nakai had strong antimicrobial activity against Pseudomonas aeruginosa at the concentration of 4,000 ppm. At this concentration, the growth of Pseudomonas aeruginosa was retarded more than 72 hours and up to 48 hours for Bacillus cereus. From these results, it was concluded that the methanol extract of Rubia akane Nakai inhibited effectively Bacillus cereus and Pseudomonas aeruginosa.
