  • In order to develop a sprayable ethylene antagonist, unlike 1-methylcyclopropene (MCP), we synthesized 2-decylcyclopropene-1-carboxylic acid ethyl ester (DCPE) as a derivative of cyclopropene and tested its effect on the flesh softening of 'Fuyu' persimmon fruits. The fruits on trees were sprayed with $4{\cdot}10^{-4}$ M DCPE solution before harvest. After harvest, the persimmon fruits were stored at a low temperature for 1.5 months. The ripening progress of the fruits was then evaluated during storage at ambient temperature ($20^{\circ}C$). Flesh softening, a measure of ethylene response, was considerably delayed up to 7 days after DCPE treatment. However control fruits was rapidly softened after 3-day storage. The treatment effect of DCPE at $4{\cdot}10^{-4}$ M was also compared to that of 1-MCP at 1 ppm. DCPE was storable at refrigerated conditions for at least one month without any loss. The results show that DCPE could be a potential sprayable agent for the prevention of flesh softening of persimmon fruit.

  • T0901317 is a potent synthetic ligand for liver X receptor (LXR, NR1H2/3), a member of the nuclear receptor superfamily that functions as a transcription factor. However, T0901317 has been also reported to modulate the activity at least four other nuclear receptors (NRs), acting as agonists for farnesoid X receptor (FXR, NR1H4) and pregnane X receptor (PXR, NR1I2) and as antagonists for androgen receptor (AR, NR3C4) and retinoid-related orphan receptor-${\alpha}$ (ROR-${\alpha}$, NR1F1). We report here that T0901317 can also function as an inhibitor for constitutive androstane receptor (CAR, NR1I3). Since CAR is a major player of xenobiotic and cholesterol metabolism in the liver, along with PXR, FXR and LXR, which are reported to be regulated by T0901317, this further complicates the interpretation of potential results with T0901317 in liver cells.

  • To detect the effect of ginseng saponin on the immune response, mice were immunized with a protein antigen (gamma-globulin of chick). Blood was then drawn from them twice, after 10 days of the first immunization and after 10 days of the second immunization respectively, and measurements were made by ELISA method of the antibody titer in antiserum. In addition, mice that has been immunized with the same antigen were treated with immunosuppressor to suppress the immune system of the mice. After the immune system was suppressed, the effect of ginseng saponin on the recovery of immune response was measured by the same method. The experimental groups those were given ginseng saponin (10 mg/kg/day) showed a little variance between-individuals, however showed much higher antibody titer than the control groups those were given the saline solution. Moreover, there was a little recovery from the immune suppression. Although the mechanism of the effect of ginseng saponin on immune response was not well loom, it is believed that ginseng saponin has the effect of increasing the synthesis of serum protein together with its action as one of the immunostimulators.

  • The purpose of this study is to evaluate effect of heavy metals (i.e., $Cu^{2+}$, $Zn^{2+}$, $Cr^{6+}$, $Cd^{2+}$, $Hg^{2+}$, and $Pb^{2+}$) to toxicity of Triclosan as binary mixture. The individual toxicity and combined toxic effects of Triclosan with heavy metals were evaluated by Vibrio fischeri assay. In individual toxicity, the $Hg^{2+}$ was found to be most toxic followed by Triclosan, $Pb^{2+}$, $Cr^{6+}$, $Cu^{2+}$, $Zn^{2+}$, and $Cd^{2+}$, respectively. To evaluate combined toxic effect, correlation analysis of 'predicted value' calculated by Concentration addition (CA) model and Independent action (IA) model with 'experimental value' were performed based on the toxicity of individual compound. As a result, all of the combinations showed that IA model were more correlated with experimental value than CA model. On the basis of the median effect concentration of combination ($EC_{50mix}$) predicted by IA model, experimental $EC_{50mix}$ of Triclosan + Cu, Triclosan + Zn, Triclosan + Pb, Triclosan + Hg, Triclosan + Cd, and Triclosan + Cr were 191%, 226%, 138%, 137%, 209%, and 138% of $EC_{50mix}$ predicted by IA model, respectively, indicating that all of the combinations produced antagonistic effect.

  • This study was performed to investigate the effect of octreotide on the contractility of rat vas deferens. The smooth muscle strips isolated from the prostatic portion were myographied in isolated organ bath, Electric field stimulation (monophasic square wave, duration: 1 mSec, voltage : 50 V, frequency : 5 Hz or 30 Hz, train: 10 Sec) produced reproducible contraction. The contraction was composed of two component, first phasic component (FPC) and second tonic component (STC). These contractions were abolished by tetrodotoxin ($1{\mu}M$). Octreotide inhibited the field stimulation induced contractions both FPC and STC concentration-dependently. The FPC was decreased by a desentization of purinergic receptor by pretreatment of mATP, and the STC was decreased by pretreatment of reserpine(3 mg/kg, IP) 24 hours before experiments. Octreotide reduced the field stimulation induced contraction in the presence of mATP and of reserpinized muscle strips. The inhibitory effect of octreotide was more potent at 5 Hz than at 30 Hz. Octreotide did not affect basal ton and exogenous norepinephrine- or ATP-induced contraction. These results suggest that octreotide inhibit the contractility of the isolated rat vas deferens by inhibition of the release of neurotransmitters, both ATP and norepinephrine from adrenergic nerve terminal.

  • Human neutrophil elastase (HNE, EC 3,4,21, 11), a mediator of tissue breakdown, was inhibited by tetracycline, oxytetracycline and demeclocycline. Among them, oxytetracycline showed the most potent inhibitory effect on the activity of HNE. IC50 of this drug at our specific condition was less than 1 mM. Tetracycline inhibited human neutrophil elastase non-competitively, and oxytetracycline inhibited competitively. Ki values of tetracycline and oxytetracycline were 4.9 mM and 0.39 mM, respectively. Structural modified tetracycline, de-dimethylaminotetracycline, which showed no antibiotic activity since the active dimethylamino radical was removed from the position #4 of the tetracycline, showed similar inhibition effect on the activity of human neutrophil elastase to that of tetracycline. Thus, we speculated that inhibition of human neutrophil elastase by tetracyclines was not depended on the dimethylamino radical which is a critical active site for antibiotic effect, rather it was depended on the hydoxyl radical of tetracyclines. Therefore, the property of inhibiting elastase may be an additional molecular biochemical mechanism of action of these drugs at the inflammatory sites.

  • Response surface methodology (RSM) was used to monitor extraction characteristics of electron donating ability and organoleptic properties for ethanol extracts from Chrysanthemum petals, thereby determining optimum extraction conditions. A central composite design was applied to investigate effects of solvent per sample $(X_1)$, ethanol concentration $(X_2)$ and extraction time $(X_3)\;at\;60^{\circ}C$ on dependent variables such as electron donating ability $(Y_1)$, organoleptic color $(Y_2)$ and organoleptic aroma $(Y_3)$ of the extracts. Second-order models were employed to generate 4-dimensional response surfaces for qualitative and quantitative aspects of ethanol extracts. Coefficients of determination $(R_2)$ of the models for dependent variables were ranged from 0.8180 to 0.9696. Optimum extraction conditions for each variable were 50 mL/g, 61% and 16 hrs in electron donating ability, 88 mL/g, 21% and 16 hrs in organoleptic color, 55 mL/g, 73% and 19 hrs in organoleptic aroma, respectively. The optimum condition ranges for maximized characteristics of ethanol extracts were $65{\sim}78\;mL/g,\;90{\sim}100%\;and\;15{\sim}25\;hrs$. Predicted values at the optimum conditions were in good agreement with experimental values.

  • A solution method is developed for computing the bed shear stress driven by combined wave and current flow on a natural mobile coastal beach. An empirical equation is introduced to determine the shape of ripples formed on the natural sandy beach. The model being based on the Prandtl's mixing length theory, the effect of arbitrarily-angled interaction is included in the estimation of current velocity reduction and all numerical integrations are expressed by explicit approximate equations to improve the computation speed. In addition the computed sediment transport rates were compared with the measured values reported in literature. using the refined bottom friction model considering the ripple formation.

  • 현재는 HTML(Hypertext MarkUp Language) 기반의 웹 페이지가 가장 널리 사용되고 있다. 그러나 사용자의 요구는 갈수록 높아져 2차원의 HTML로서는 표현하지 못하는 3차원 세계의 구현을 갈구하고 있다. 그래서 탄생한 것이 VRML(Virtual Reality Modeling Language)이며 현재 ISO 표준안으로 지정되어 있다.
