Discrimination of Korean Agaricus bisporus cultivars using CAPS markers (CAPS 마커를 이용한 국내 개발 양송이 품종 구분)

  • The cleaved amplified polymorphic sequence (CAPS) marker uses a restriction enzyme recognition site resulting from single nucleotide polymorphisms and insertions and deletions on the DNA sequence. This technique does not require expensive equipment, the process is simple, and clear results can be obtained reliably. In this study, Agaricus bisporus cultivars SaeA, SaeDo, SaeHan, SaeYeon, SaeJeong, Dodam, Seolgang, Dahyang, Hogam, and Hadam developed in Korea were discriminated using four CAPS markers. Our results indicated that it is possible to distinguish the ten cultivars and determine the genetic diversity among them.

Development of SCAR markers in Creeping bentgrass(Agrostis palustrics Huds.) cultivars (Creeping bentgrass(Agrostis palustrics Huds.) 품종별 SCAR markers 개발)

  • Creeping bentgrass (Agrostis palustrics Huds.) is cool season turfgrasse that is used for putting green in golf course. Creeping bentgrass cultivars are difficult to distinguish with the same species because of similar morphological characters and low level of genetic diversity. The SCAR markers using the specific DNA can be useful for differentiating between creeping bentgrass cultivars. Five RAPD primers were used for specific band detection among creeping bentgrass cultivars, penncross, penn A-4, crenshaw, L-93, CY-2, T-1. The pairs of SCAR primers for six cultivers were designed by the specific sequences of the bands that amplified by RAPD. Three of the six SCAR primers could not make the use as SCAR primers because the specific false bands were detected in all cultivars. The remaining pairs of SCAR primer, CY850F/R, T700F/R, L2900F/R, amplified the specific band at expected size for three cultivars, CY-2, T-1, L-93, respectively. The CY850F/R primer amplified a band of 850bp in CY-2 cultivar, the T700F/R primer amplified a band of 700bp in T-1 cultivar, and the L2900F/R primer amplified a band of 2.9kb in L-93 cultivar. In this study we developed the SCAR markers to identify and distinguish the inerseeded creeping bentgrass cultivars in a golf course green.

Trends of commercial strain development and spawn industry in mushrooms (버섯의 품종 육성과 종균 산업의 동향)

  • According to UPOV (International Union for Protection of New Varieties of Plants), mushroom spawn market have to be opened by the year 2009. Number of commercial strains distributed by the year 2005 were 179 of 24 species of edible and medicinal mushrooms. Only nine strains of oyster mushrooms were registered as protected variety, which is not compatible with those recorded in other advanced countries. Meaning of spawn in broad sense contains commercial strains. Development of commercial strains faces two main problems in Korea : One is the complicated genetic patterns and sexuality of mushroom species, and the other is expensive experimental equipments and fruiting body growing houses. Resolution of these problems leads to development of mushroom strains. This could be achieved as follows; genetic resources collection and assessment, molecular characterization of useful genetic characters, development of new commercial strains by hybridization using typical genetic resources, strengthening of breeding research using "Mushroom Breeding Group", management of spawn research company by consortium, foundation of mushroom general industry, promotion of consumption, and upgrade of competition ability for other countries. These points are under discussion.

Review on Adaptability of Rice Varieties and Cultivation Technology According to Climate Change in Korea (기후변화에 따른 국내 벼 품종과 재배기술의 적응성에 관한 고찰)

  • In recent years, the temperature of Korea has been rapidly increasing due to global warming. Over the past 40 years, the temperature of Korea has risen by about 1.26℃ compared to that in the early 1980s. By region, the west region of the Gangwon Province was the highest at 1.76℃ and the Jeonnam Province was the lowest at 0.96℃. As the temperature continues to rise, it is expected that the rice yield will decrease in the future using the current standard cultivation method. As a result of global warming, the periods in which rice cultivation could be possible in regions each year has increased compared those to the past, showing a wide variety from 110 days in Taebaek to 180 days in Busan and Gwangyang. In addition, the transplanting time was delayed by 3-5 days in all regions. The average annual yield of rice showed an increasing trend when we analyzed the average productivities of developed varieties for cooked rice since the 1980s, especially in the early 1990s, which showed a rapid increase in productivity. The relationship between the average temperature at the time of development and the rice yield was divided into the periods before and after 1996. The higher the average temperature, the lower the yield of the developed varieties until 1996. However, since 1996, the increase in the average temperature did not show a trend in the productivity of the developed varieties. The climate change adaptability of developed rice varieties was investigated by analyzing the results of growing crops nationwide from 1999 to 2016 and the change in the annual yields of developed varieties and recently developed varieties as basic data to investigate the growth status of the crops in the country. As a result of annual comparisons of the yields of Taebongbyeo (2000) and Ungwangbyeo (2004) developed in the early 2000s for Odaebyeo, which was developed in the 1980s, the annual yields were relatively higher in varieties in the 2000s despite the increase in temperature. The annual yields of Samgwangbyeo (2003) and Saenuribyeo (2007), which were recently developed as mid-late-type varieties, were higher than those of an earlier developed variety called Chucheongbyeo, which was developed in the 1970s. Despite the rapid increase in temperature, rice cultivation technology and variety development are well adapted to climate change. However, since the biological potential of rice could reach its limit, it is necessary to develop continuous response technology.

Development of Suhan Strain-specific SCAR Marker in Pleurotus ostreatus (느타리 버섯에서 수한 품종 특이 SCAR marker 개발)

  • In this study, 81 commercial strains of Pleurotus species cultivated in South Korea were analyzed with randomly amplified polymorphic DNA (RAPD) technique. Sequence characterized amplified region (SCAR) markers were developed by designing from one RAPD polymorhic band specific to Suhan strain. The SCAR primer pair 'S-OPA13-1' amplified a 590-bp fragment in the varieties originated from Suhan strain. The Blast search of S-OPA13-1 showed high homology to the POMFBO1 P. ostreatus cDNA clone MFB02-A05 and Laccaria bicolor S238N-H82. The results showed that this SCAR marker can clearly distinguish Suhan strains from Pleurotus spp.

Plant growth and fruit enlargement among different watermelon (Citrullus lanatus) cultivars in continuous chilling night temperature conditions (지속적인 야간 저온에 의한 수박 품종별 식물체 생장 및 과실 비대 양상)

  • Watermelon (Citrullus lanatus (Thunb.) Matsum. & Nakai) is sensitive to low temperature and shows retarded growth under 10℃. Although early transplanting guarantees higher returns, it requires cost and labor to maintain the appropriate temperature for plant growth. Therefore, cultivars tolerant to chilling stress is necessary to reduce the cost and labor requirements. The purpose of this study is to analyze data on plant growth and fruit enlargement under continuous chilling night temperature to develop new cultivars tolerant to chilling temperature. Two cultivars expected to have chilling tolerance and another cultivar sensitive to chilling temperature were grown in greenhouses with chilling and optimal night temperature conditions. In the early growth stage after transplanting, the cultivars expected to have chilling tolerance showed better vine length, fresh weight and dry weight. However, one of the tolerant cultivars showed significantly lower vine length, leaf length and width, and petiole length than the sensitive cultivar during pollination period and later growth stage, showing genotype specific responses. The fruit length, width, and weight were also significantly lower in the tolerant cultivar. The fruit set ratio was significantly higher in the chilling sensitive cultivar than the two tolerant cultivars. These results suggest that the present chilling tolerant cultivars in watermelon were selected based on their performance in the early growth stage, and further studies on chilling tolerance in different growth and development stages are required to develop cultivars adapted to various forcing cultivation systems.

Analyzing Spillovers of Domestic Varieties Developed by Rural Development Agencies (농촌진흥기관 개발 품종의 경제적 파급효과 분석)

  • This study examined the spillovers (economics) of domestic varieties. The analysis model used the supply-driven of inter-industry analysis (input-output analysis), and the scope of the study was limited to the varieties developed by rural development agencies. The spillovers were analyzed by dividing into the effects on production inducement and employment inducement. First, the effects on production inducement were the largest in Sindongjin, a rice variety, with 49,599.3 billion won. Seolhyang, a strawberry variety, was the second largest with 32,936.9 billion won. On the other hand, Baekma, a flower variety, was small at 87.7 billion won. On the other hand, this is a very large number considering the small area of cultivation of flowers and how most of the flower varieties depend on overseas varieties. Second, the effect on the employment inducement coefficient appeared in a similar order to the effect on production inducement. Sindongjin was the largest with 756,682, and Seolhyang was 701,403. Baekma was analyzed as 1,582 people. The results are of great significance in that it quantitatively analyzed the spillovers of the varieties developed by rural development agencies and ensured a justification for the development of varieties by national institutions through their value.
