In vitro Fruit Assay for the Evaluation of Fungicide Activity Against Pepper Anthracnose (살균제 효과 검정을 위한 고추 탄저병의 실내 열매 검정법)

  • This study was performed to investigate the effect of conidial density, wetness period and temperature on conidial germination, appressoria formation and disease incidence. While there was not significantly correlated between conidial density and temperature, and conidial germination and appressoria formation, there was a significant correlation between those factors and disease incidence. The longer wetness period was, the higher the ratio of conidial germination, appressoria formation and the disease incidence was. The optimum conidial density, temperature and wetness period was $1{\times}10^6$ conidia $mL^{-1}$, $30^{\circ}C$ and 5 days, respectively. In case the wetness period was more than 5 days, the typical symptom was not found on pepper fruits because of the overgrowth of mycelia. Using this fruit assay method, which the pepper anthracnose pathogens were inoculated by spraying spore suspension on non-wounded or wounded pepper fruits, control effect of three fungicides were evaluated against pepper anthracnose by the protective and/or the curative application. Propineb showed high protective control activity, while it showed curative control activity on unwounded fruits, but did not showed curative control activity on wounded fruits. Tebuconazole, one of curative fungicide, showed higher control activity in non-wound inoculation than wound inoculation. Trifloxystrobin, one of strobilurin group, showed high both protective and control activity against anthracnose. In conclusion, we supposed that the newly developed in vitro pepper fruit assay can be used to evaluate antifungal activity of control agents against pepper anthracnose.

Isolation of Siderophore-producing Pseudomonas fluorescens GL7 and Its Biocontrol Activity against Root-rot Disease (Siderophore 생산성 생물방제균 Pseudomonas fluorescens GL7의 선발 및 식물근부병의 방제)

  • For the development of a multifunctional biocontrol agent, the siderophore-producing strain GL7 was isolated from a rhizosphere on chrome azurol S agar. The GL7 was identified as a strain of Pseudomonas fluorescents on the basis of their reactions to standard physicochemcial tests from Bergey's manual, API diagnostic test, and fatty acid analysis. P. fluorescents GL7 considerably inhibited spore germination and hyphal growth of phytopathogenic fungus Funsarium solani in a dual culture. In pot trials of bean with P. fluorescens GL7, the disease incidence was significantly reduced down to 5% from 70% of incidence in the untreated control. P. fluorescens GL7 also enhanced plant growth to nearly 1.5 times than that of the untreated control, promoting elongation and development of the roots. These results suggest that the plant growth-promoting P. fluorescens GL7 can play an important role in the biological control of soil-borne plant disease in a rhizosphere.

Spore Germination of Some Plant Pathogenic Fungi under Different Soil Conditions in Relation to Soil Fungistasis (토양조건에 따른 몇가지 식물병원균의 포자발아와 토양정균 현상)

  • Some interactions in various soil conditions, numbers of microbial populations, root rot disease development and rates of spore germiation in three different location of soils were investigated. The calcium and magnesium contents were higher in replanted fields of ginseng (Panax ginseng) at Goesan. Potassium contents were high in replanted field at Poonggi and textural class of the soils was silt loam except for silt clay loam in first cultured field of ginseng at Goesan. For the germination process of Fusarium solani, F. moniliforme, F. oxysporum, and Alternaria panax, the percentage germination of fungal spores was high in double distilled water and Pfeffer's solution as media, whereas the lower rate of germination of spores was observed in soil extracts. Numbers of bacteria were high in replanted field soil at Gumsan, and propagules of fungi in replanted fields at Gumsan and Poonggi were higher than other soils, but higher numbers of actinomycetes were found in the first cultured field of ginseng at Goesan and Poonggi. Fungistasis was induced by higher microbial populations present in soil that was initiated when amended with garlic stalk, crushed bean and ginseng leaves. On the other hand, there was no fungistasis in soil amended with wheat and barley straw, and this tendency was a little difference on the soil sample.

Effect of Mepanipyrim on the Resistant Isolates of Gray Mold fungus, Botrytis Cinerea to the Fungicides of Benzimidazole and Dicarboximide (Benzimidazole과 dicarboximide계 살균제 저항성 잿빛곰팡이병원균(Botrytis cinerea)에 대한 mepanipyrim의 효과)

  • Effect of the fungicide mepanipyrim on the resistant and sensitive isolates of Botrytis cinerea was studied in vitro and also tested to control Botrytis rot of strawberry, cucumber and grape in the field. These isolates were selected by relative mycelial growth and spore germination on potato dextrose agar(PDA) incorporated with $100{\mu}g\;a.i./ml$ of benomyl and procymidone, respectively, compared to the unamended PDA. Mycelial growth of the selected resistant isolates was significantly inhibited by mepanipyrim but the inhibition rate was similar to other fungicides belong to benzimidazole or dicarboximide, although spore germination was not inhibited even by the higher concentration of mepanipyrim. When the benomyl and procymidone resistant isolates were inoculated to cucumber leaves, lesion development was significantly inhibited with application of $250{\mu}g\;a.i./ml$ of mepanipyrim but not with that of benomyl and procymidone. In addition, when $250{\mu}g\;a.i./ml$ of mepanipyrim was applied to strawberry, cucumber, and grape in the field, the control of Botrytis rot was significantly different from that of the untreated control(Duncan's multiple range test, p<0.05). The results suggested that mepanipyrim might be an alternative fungicide for the control of benomyl- and procymidone-resistant pathogens of Botrytis rot.

Propagation Method for Sporophyte Formation of Woodsia ilvensis (L.) R.Br. (두메우드풀 포자체 형성을 위한 번식방법)

  • 본 연구는 북방계 식물에 속하는 두메우드풀[Woodsia ilvensis (L.) R.Br.]의 전엽체 증식 및 포자체 형성을 위한 적정방법을 구명하고자 수행되었다. 식물재료는 경상북도 의성군 일대에서 채취한 후 청주의 일반하우스에 이식하여 재배하였다. 포자 성숙기인 8월에 채종하여 발아시켰으며, 포자로부터 획득한 전엽체를 8주간 계대하면서 실험재료로 사용하였다. 전엽체 300mg을 메스로 다진 후 배지농도 및 종류(Knop, 1/4, 1/2, 1 및 2MS)를 달리하여 8주간 배양하였다. 배양실의 온도는 $25{\pm}1.0^{\circ}C$, 광도는 $30{\pm}1.0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(16/8h)로 조절되었다. 연구의 결과, 배지구성물질의 농도가 높았던 2MS배지에서 전엽체의 생체중이 12.4g으로 41배 증가하여 가장 높은 증식율을 나타냈다. 또한 배지구성물질의 농도가 적을수록 생체중의 증가폭도 함께 감소하는 경향을 보였다. 포자체 형성에 적합한 토양조건을 확인하고자, 원예상토, 피트모스, 펄라이트 및 마사토의 비율을 달리한 5종류의 혼합토양을 조성하여 사각분($7.5{\times}7.5{\times}7.5cm$)에 충진하였다. 다음으로 전엽체 1g을 핸드블랜더로 10초간 분쇄 후 충진된 사각분 위에 고르게 분주하여 14주간 재배하였다. 재배환경은 온도 $25{\pm}1.0^{\circ}C$, 광도 $43{\pm}2.0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(16/8h) 및 습도 $72{\pm}2.0%$로 조절되었다. 지베렐린 수용액이 포자체 형성에 미치는 영향을 조사하고자, 선행연구에서 선발된 토양조건을 기준으로 침지농도(0, 50, 100, $200mg{\cdot}L^{-1}$)를 조절하여 1시간 침지 후 상기와 동일한 방법으로 재배하였다. 연구의 결과, 원예상토와 펄라이트 또는 마사토를 2:1(v:v)로 혼용한 토양에서 사각분 당 109.5, 128.8개의 포자체가 형성되었다. 한편, 피트모스가 첨가된 처리는 포자체의 형성이 억제되는 경향을 나타냈다. 지베렐린 50과 $100mg{\cdot}L^{-1}$ 수용액에 침지한 처리에서 무처리에 비해 많은 사각분 당 각 177.0, 181.7개의 포자체가 형성되었으며, 생육도 양호하였다.

Effects of soluble silicon on development powdery mildew(Sphaerotheca fuliginea) in cucumber plants (규소 시용에 의한 오이 흰가루병 발병억제)

  • Effects of silicon application on development of colonies of Sphaerotheca fuliginea were examined. Cucumber plants were applied with nutrient solutions amended with different concentrations of soluble silicon and selected leaves were inoculated with known concentrations of conidia of the pathogen. Colony number per leaf, colony area per leaf, and germination rate of conidia of S. fuliginea collected from the inoculated leaves were reduced as silicate concentrations in the nutrient solutions increased from 0.05 to 4.10 mM. The increase in resistance of plants to mildew infection was apparently due to silicate accumulation in leaves, and there was no correlation between cation or ionic strength effects and the silicate treatments. Silicate treatment in growth medium remarkably suppressed powdery mildew development on cucumber. Colonies of mildew fungus were visible with over approx. 38.3% of the mature leaf surface, while that of the leaves in high Si plants was 2.3% observed at 51 days after transplanting. No significant differences were observed between 1.7 mM and 3.4mM silicate treatments. Conidial germination rates were significantly reduced by increasing Si amendments. Conidial germination ranged from 14.7 to 20.3% for plants grown in low Si solution(<1.40 mM), and from 9.0 to 12.4% for plants grown in high Si solution(>1.8 mM). Foliar applications of Si with ${\geq}$ 17.0 mM decreased the number of powdery mildew colonies. Persistence of Si foliar sprays effects on cucumber demonstrated that the 17 mM Si spray applied 4 days before inoculation with S. fuliginea reduced mildew colony formation. The relationship was positive and linear.

Gamma Irradiation Effects on Conchospores of Porphyra Species 2. The Effects of High Gamma Irradiation on Germination and Growth of Conchospores of Two Varieties (김의 각포자에 대한 r-선의 조사효과 2. 두 품종의 각포자의 발아생장에 미치는 고선량 r-선의 조사효과)

  • For the fundamental studies of radiation breeding in edible marine algae, the biological effects on conchospores of Porphyra species by gamma-irradiation were examined. Two varieties, Keun-cham-gim (Porphyra tenera Kjell. form tamatsuensis Miura) and Saga No.5, were chosen for this study, and their conchospores after r-irradiation($5.0{\sim}20.0$ KR) were cultured for 50 days. The results obtained were summarized as follows. 1. Gamma-irradiation in less than the dose of 20KR did not affect germination of conchospores, and almost all spores grew into two cells germ in 24 hours after irradiation, but withering germs were gradually increased in number according to higher exposure within 5 days old culture. 2. High irradiation caused the induction of giant cells, abnormal useless growth of hold-fast, lumpish thalli and callus-like lumpy tissues. 3. The liberation of neutral spores from young germs and carpospores from mature thalli were observed on the frond exposed at $10{\sim}20$ KR irradiation. All spores were normal in division and its size. 4. The best irradiation effect on growth of Keun-cham-gim was observed at 10 KR dose, whose growth-rates were $140\%$ in wet weight and $108\%$ in mean frond area, but only $48\%$ was recorded in wet weight at 20 KR exposure. Saga No.5 were in contrast with Keun-cham-gim, and their most growth-rate was $400\%$ in wet weight ($258\%$ in frond area) at 10 KR irradiation and the worst was $20\%$ at the dose of 20 KR. 5. The withering phenomenon to death by treatment of gamma-ray presented substantial difference between two varieties. Survival rate compared with control in Keun-cham-gim was $70.7\%$ at 20 KR, but that in Saga No.5 recorded $47.0\%$ at same dose. 6. Synthesizing the results of high and low r-irradiation, it was suggested tat high r-irradiation in more than 5.0 KR inhibited conspicuously the growth of germs derived from conchospores, and about half of them withered at 15.0 KR dose or more.

Occurrence of Botrytis cinerea Resistant to Dicarboximide Fungicides on Strawberries in Greenhouses (시설재배(施設栽培) 딸기 포장(圃場)에서 Dicarboximide 계(系) 약제저항성(藥劑抵抗性) 잿빛곰팡이병균(病菌)의 발생(發生))

  • Isolates of Botrytis cinerea resistant to dicarboximide fungicides were collected from strawberry fields in greenhouses in spring and early summer of 1990. Five out of 9 isolates of B. cinera were resistant, which showed mycerial growth on PDA containing dicarboximide fungicides(procymidone and vinclozolin) with concentrations of 100, 400 and $1,600{\mu}g/ml$. The minimal inhibitory concentration(MIC) values of the dicaboximide-resistant isolates was more than $6,400{\mu}g/ml$, while that of the sensitive isolates was less than $6.25{\mu}g/ml$. The germination ratio of conidia of the resistant isolates on PDA containing procymidone and vincolozolin was more than 95%, while that of the sensitive was less than 15%. The procymidone-resistant isolates were also resistant to vinclozolin, showing cross-resistant between the fungicides, but cross-resistant was not observed between the dicarboximides and dichlofluanid. Resistance to benomyl was also found in all the dicarboximide resistant isolates. Occurrence frequency of dicarboximide-resistant isolates out of 223 isolates was about 40%. The resistant isolates were widely distributed throughout Korea.

Effect of Temperature, Relative Humidity on Germination and Development of Powdery Mildew (Leveillula taurica) on Pepper and Its Inoculation Method (고추 흰가루병균(Leveillula taurica)의 발아특성 및 접종법)

  • Pepper powdery mildew, Leveillula taurica is one of the most important pathogens of pepper in the greenhouses and fields in Korea and is becoming a worldwide disease. These experiments were carried out to investigate the optimal germination, disease development conditions, disease incidence and effective inoculation methods of pepper powdery mildew. The incidence of powdery mildew was investigated throughout the country based on the major pepper growing areas in 2009. The average rate of infected leaves ranged from 15.3% to 81.8% in greenhouses and fields. Powdery mildew incidences were more severe in greenhouses than those of fields. The optimal temperature for conidial germination was $25^{\circ}C$ and showed the highest germination at 6 hs after treatment. The range of temperature for germination was $10^{\circ}C$ to $35^{\circ}C$. Temperature of greater than $30^{\circ}C$ and below $20^{\circ}C$ affected the abnormal germination and germ tube elongation. The optimal relative humidity for germination and germ tube elongation was 85% and germination increased as relative humidity increased. Disease development started 8 days after inoculation and showed the highest disease severity at 15 days in greenhouse and field regardless of varieties. Among three different inoculation methods tested the spore dropping method was better than touching with infected leaves and spraying with spore suspension. However, the method has limitation in mass inoculation due to the amount of time consumed.

Histopathology of Red Pepper Plant Infected with Colletotrichum dematium f. sp. capsicum (탄저병균 Colletotrichum dematium f. sp. capsicum에 감염된 고추의 병태조직학적 관찰)

  • Upon germination, a conidium was septated in accordance with number of germ tubes. The percentages of ppressorial formation were not different between the resistant cultivars (Kumchang NO. 2 and Hongilpum) and the susceptible cultivars (Haneulcho and Saegochu). Appressorial form was various such as round, elliptic or star-shapped. The maximum number of appressoria was shown at 12 hours after inoculation. By 24 hours fter inoculation, hypersensitive tiny brown lesions were observed on the leaves and fruits of the resistant cultivars. Epidermal cells under cuticle layer of the resistant Kumchang NO. 2 fruit showed severe plasmolysis, while on the susceptible cultivars, the lession was largely extended to following incubation. Subcuticular infection hyphae were profusely colonized in the disintegrated tissues. Acervuli and setae on the stromatic structure ere formed at 96 hours. The infected seed coat was not only severely collapsed, but also infection hyphae were observed on the disintegrated seed coat, resulting severe plasmolysis of nucellus and embryo.
