Effect of physical and chemical Factors on the Formation and Germination of Chlamydospore of Cylindrocarpon destructans Causing Root Rot of Panax ginseng (인삼 뿌리썩음병균(Cylindrocarpon destructans) 후막포자의 형성 및 발아에 영향을 주는 물리화학적 요인)

  • 인삼뿌리썩음병균(Cylindrocarpon destructans)후막 포자의 형성은 CD(Czapek`s dox broth)와 SNY(Spezieller Nahrtoffarmer yeast extract broth)배지에서 진탕배양(2$0^{\circ}C$, 120 rpm)하면 양호하였다. 특히 CD배지에서 2$0^{\circ}C$로 30일간 진탕배양하면 5.99 log 후막포자/ml로 형성되었다. 후막포자의 발아율은 12.9~23.3%로 CD배지와 SNY배지에서 좋았으며 발아적온은 5~1$0^{\circ}C$였고, 최적 pH는 6이었다. 그러나 2$0^{\circ}C$이상에서는 전혀 발아하지 않았으며 pH 7.0이상에서는 발아율이 급속히 저하되었다. 인삼추출물 3%, GA 10 ppm, IAA 10 ppm, NOVOZYM\ulcorner234 20 ppm을 처리 할 경우 대조구에 비해 발아율이 증가하였다. 특히 NOVOZYM\ulcorner234 20 ppm 처리구는 발아율이 무처리에 비해 2배 증가하였다. CD배지(pH5)에 NOVOZYM\ulcorner234 20 ppm과 GA 10 ppm을 조합처리하고 5$^{\circ}C$로 정치배양 할 때 후막포자의 발아율은 49.4%로 가장 높았다.

  • Conidial germination of Septoria glycines Hemmi, brown spot fungus of soybean, was studied by slide germination test. Poor conidial germination of S. glycines was observed on sterile distilled water, but potato dextrose agar(PDA) and distilled water floated with soybean leaf disc furnished a satisfactory medium for conidial germination. Exogenous supply of carbon source was essential for conidial germination, while phosphorous and potassium were not evident as that for carbon. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $5\times10^{-2}mol$. concentration of glucose. Germination was decreased with increment of conidial concentration and was almost completely suppressed in the density of 10,000 conidia per $mm^2$. It suggested existing a self-inhibitor(s). Non-washed conidia germinated more than washed conidia and this was obvious when the conidia density was over $2\times10^3$ conidia per $mm^2$ on the dry agar block.

  • Effects of incubation temperature and pH on chlamydospore germination of Cylindrocarpon destrcutans (isolate CY-9802) causing root rot of Panax ginseng were studied. Germination rate of the chlamydospores on Czapek solution agar(CSA) was higher than on potato dextrose agar(PDA) at the incubation temperatures tested. The chlamydospores were able to be germinated at range of 5$\^{C}$ to 30$\^{C}$ after 48 hours incubation on CSA. Germination rate was 53.2∼6.27% at range of 15$\^{C}$ to 25$\^{C}$, and the optimum temperature was 20$\^{C}$, whereas they were very low at 30$\^{C}$ on PDA. Germination rate was 43.6% to 47.9% at range of 10$\^{C}$ to 20$\^{C}$, and the optimum temperature was 20$\^{C}$ as well. They were able to be germinated at pH of 5.2 to 8.1 on CSA and 5.2 to 7.2 on PDA. Optimum pHs for the germination on CSA and PDA were from 6.4 to 8.2 and from 5.2 to 6.0, respectively. Mycelial color of the fungus on CSA was pale brown at pH from 5.2 to 6.0 and white from pH 6.4 to 8.1, while it was typical dark brown ar range of pH 5.2 to 7.1 and brown at pH 7.2 on PDA after 21 days incubation.

Effects of Dryness, Moisture Interruption, and Temperature on Germination of Diaporthe citri Pycnidiospores on Yuzu (건조, 습윤중단 및 온도가 유자 검은점무늬병원균 Diaporthe citri 포자 발아에 미치는 영향)

  • Melanose caused by Diaporthe citri is a serious disease on yuzu fruits. Moisture is the most important to infect after the pycnidiospores are released. In order to understand how dryness and moisture interruption affect on germination, the conidial suspensions on slide glass were dried for 0-48 h. For the moisture interruption experiment, moisture was supplied for 10 h then interrupted for 0-6 h depending on the treatments. Germinations on the treatments with longer than 15 h of dryness were less than 10%. And those with longer than 3 h of moisture interruption were less than 30%. Compared with other fungal phytopathogens, D. citri was xero-sensitive. Germination on yuzu feel medium was measured under 15, 20, 25, 30, $35^{\circ}C$ with 3, 6, 9, 12, 15, 18, and 24 h of wetness. The minimum wetness periods achieving 10% and 50% germinations were 4.5 and 13 h, and the optimum temperatures were $29.1^{\circ}C$ and $29.2^{\circ}C$, respectively. From the yuzu medium study, the regression equations to be required wetness period achieving 10% and 50% germinations on various temperatures were $Wh_{50%}=0.1093{\times}T^2-6.3762{\times}+106.08$ and $Wh_{10%}=0.082{\times}T^2-4.8025{\times}T+74.861$.

Effect of Ultrasonic treatment on the Isolation of the Chlamydospores of Cylindrocarpon destructans Causing Root rot of Panax ginseng (초음파처리에 의한 인삼 뿌리썩음병균 Cylindrocarpon destructans 후막포자의 분리)

  • Chlamydospores were isolated from hyphae of Cylindrocanon destmctans by homogenization and/or ultrasonic treatment. Rate of the isolated chlamydospores by the homogenization with glass tissue grinder were 9.8% of all total chlamydospores formed in the culture of C. destructans. The length of mycelial fragments after the homogenization was about 400㎛ They were, however, formed in clusters of the chlamydospores and the mycelia The rate of the isolated chlamydospores from additional ultrasonic treatment after the homogenization of the mycelia were 74.3%. The length of mycelial fragments with the ultrasonic treatment was about 20 fm and chlamydospores seemed to be isolated from the mycelial mats and dispersed evenly in the culture. The numbers of chlamydospore in a catena were 1 to 8 cells after the homogenization on potato dextrose agar (PDA). Meanwhile the numbers of them after added ultrasonic treatment were 1 to 4 cells. Germination percentages of the isolated chlamydospores from the ultrasonic treatment were 46.8% after incubation of 2 days on PDA at 20。C and 60.7% after incubation of 13 days at 5。C, respectively. Germination rate of chlamydospores to the total chlamydospores produced by the ultrasonic treatment was 55.8%. However, it was increased to 74% when it was measured in the germinated catenae to the total catenae.

Germinability of Resting Cysts Associated with Occurrence of Toxic Dinoflagellate Alexandrium Species (유독 와편모조류 Alexandrium속의 출현에 미치는 휴면포자의 발아율)

  • To study the causes of occurring toxic dinoflagellate Alexandriwn species, an experiment was undertaken in Jinhae Bay shellfish harvesting areas. The water and sediment samples were collected to record the abundance of Alexandriwn species, and to study the distribution and the germinability of those benthic cysts from September 1993 to July 1994. Alexandrium species were began to appear at all the sample stations after January, and reached maximum concentration (530 cells/l) at Taekok station (Chilcheon-do) in March 1994. Alexandrium cysts were also found at every station surveyed, of which several sites showed the higher concentration of 700-800 $cysts/cm^3$ at the upper sediment profile (0-4cm), but the concentrations were wide range of 100-800 $cysts/cm^3$. The results of each sampling season showed a great difference in the cyst germination experiments, were potentially high in cold season; $72.5\%$ (Jan.), $68\%$ (Apr.),$44\%$(Jul.), and $9\%$ (Oct.). These results suggested that germination of Alexandrium resting cysts in 15 m depth of coastal waters in Jinhae Bay would be controlled by a seasonal endogenous clock instead of the general environmental factors like temperature. Therefore, it is possible that Alexandrium species could be abundant by the germination of resting cysts in cold season, and contribute to the regional paralytic shellfish poisoning (PSP) toxification.

  • The growth of germ tube of Fusarium oxysporum f. sp. cucumerinum was remarkably inhibited on the water agar treated with 100ppm of Fe-EDDHA, a synthetic iron chelating agent, whereas germination rate of microconidia did not show much differences compare with that of non treated water agar. Both of the germination and the germ tube elongation of microconidia were suppressed significantly in King's B agar by the bacterial siderophores produced by Pseudomonas putida. The highest germination of the chlamydospores was obtained in the soil added with $0.25\%$ of glucose plus $0.05\%$ of asparagine. The chlamydospores of cucumber wil fungus germinated about $14\%$ in rhizosphere soil of 2 day-old cucumber seedlings within 48 hours, and the germination was enhanced notably in rhizosphere soil of 10 day-old seedling. But the rates of germination was not increased according to cucumber growth age after 10 day-old seedling. The effect of P. putida and Fe-EDDHA on the germination on chlamydospores in conducive soil was not pronounced in the non-rhizosphere soil added with nutrient. However, the germination was suppressed significantly both in rhizosphere soil and in rhizosphere soil added with nutrient. The suppression of chlamydospore germination was greater in the bacteria inoculated soil than that in Fe-EDDHA treated soil.

  • Exogenous factor and nutrients affecting for conidial germination of Colletetrichum dematium f. sp. capsicum causing red pepper anthracnose were studied by slide germination test. Optimum temperature of conidial germination was at $28^{\circ}C$, ranging 15 to $35^{\circ}C$. Optimum pH was at 5.5, ranging 4.5 to 8.0, and more than 90% of relative humidity (RH) was optimum. Poor conidial germination of the fungus was observed on sterile distilled water, but potato sucrose broth (PSB), red pepper fruit broth (RPFB), green pepper fruit broth (GPFB) and pepper leaf broth (PLB) furnished a satisfactory nutrients for conidial germination. Exogenous supply of carbon and nitrogen sources were essential for conidial germination, while potassium, phosphorous and sulfur were not evident as that for carbon and nitrogen. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $1{\times}10^4$ conidia per ml. Germination was decreased with increment of conidial concentration, and in the density of $5{\times}10^4$ conida per ml, germination was nearly supipressed. It suggested existing a self-inhibitor. Non-washed conidia germinated more than washed conidia, and conidial germination was also gradually decreased by increasing conidial density.

Germination and Growth of Laminaria japonica (Phaeophyta) Microscopic Stages under Different Temperatures and Photon Irradiances (수온과 광량에 따른 다시마 초기 생활사의 발아와 성장)

  • Germination and growth of Laminaria japonica microscopic stages were investigated under crossed gradients of temperatures and irradiances, and the results related to the seasonal temperature regime in the southeastern coast of Korea. Germination rates of $70\~86\%$ were observed in the temperature range of $5\~20^{\circ}C$, however, at $25^{\circ}C$ no germination of meiospores was observed. The primary factor affecting germination rates at the temperature range of $5\~20^{\circ}C$ was irradiance: germination was significantly reduced at $150 {\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$. Vegetative cell production of female gametophytes was highest at $20^{\circ}C$, but plants were not fertile at the temperature. In the temperature range of $5\~15^{\circ}C$, higher irradiance caused females to reduce cell production, but increased fertility. Cell production was also low at lower temperatures with increased fertility rates. Optimal growth temperature for microsporophytes was $10^{\circ}C$ and their growth rates were light-saturated at $70 {\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$. We have concluded that meiospores released before July could develope to the young sporophytic stage in the southeastern coast of Korea which is off the southern limit of its geographical distribution. However, limiting factor in the development of natural sporophytic population in this region would be the upper temperature limit for the survival of young sporophytes, as water temperature at this area frequently exceeds $25^{\circ}C$ during the summer period.

Establishment of rhizosphere microbes for plant protection on soil-borne diseases -Benificial antagonist and its mode of action toward ginseng root rot pathogen- (근권미생물과 토양병방제 -유용길항균이 인삼근부병원에 미치는 영향-)

  • From soil samples, 380 antagonistic microorgnisms were isolated. Among the isolates, 42 strains had mycelia growing inhibition ability against Fusariun solani, ginseng root rot causing pathogen. Isolates CHA 1 and S-PFHR 6 were proposed as antagonists for this study and they were identified as Promicromonospora sp. and Pseudomonas pseudoalcaligenes respectively. As an antagonism against hyphae of F. solani in dual culture test, CHA 1 and S-PFHR 6 inhibited linear growing, caused abnormal branching, and the membrane projection which formed by cell wall destruction. The secondary metabolites contained in the culture filtrates which prepared from PD broth and Nutrient broth inhibited the spore germination to 14.3%. The culture filtrate of S-PFHR 6 which prepared by a little amount of soil extract addition to nutrient rich medium had more strongly. inhibited the spore germination and spore germination decreased to less than 4.0% in it. The soil used in this study had fungistasis and the germination rate of macroconidia and chlamydospore of F.solani was 19.4% and 17.7% respectively. The steam sterilized soil lost fungistasis and germination rate of conidia increased to more than 97.9%. The soils amended with the propagule of CHA 1 and S-PFHR 6 increased fungistasis and the germination rate of macroconidia decreased to 14.7% and 11.7% respectively in each treatments. But the soil ammended with glucose and asparagine annulled fungistatic ability and the germination rate of macroconidia increased to more than 48.0%. As an antagonistic activity of the secondary metabolites of two antagonistic isolates in soil, the germination rate of macroconidia of F. solani was 9.3% in the soil amended with the culture filtrate of CHA 1 but the culture filtrate of S-PFHR 6 had no such activity. In the soil which treated with antagonist propagule or culture filtrate, the chlamydospore germination rate was lower than that in natural soil. The addition of glucose and asparagine to antagonist propagule treated soil did not enhanced the chlamydospore germination.
