Antagonistic Relationships Between Development of Sporangiophores and Phototropism Depending on Monochromatic Light Wavelengths in Phycomyces blakesleeanus (털곰팡이(Phycomyces blakesleeanus)의 생장기간 중 파장별 단색광 조사에 따른 굴광성 반응과 포자낭병 발생간의 길항관계)

  • Six strains of Phycomyces blakesleeanus (DEL, A56, C9, C47, C111, and G5) were cultured in continuous light condition for 6 days. The increase in fresh and dry weight after the culture period was similar in all strains. However, the increase in fresh weight was sustained throughout the culture period in all strains, whereas the increase in dry weight was prominent only during the first 3 days of culture. The development and phototropism of macrophores was observed after 3 days of culture, These results suggest that the development and phototropism is ascribed to the increase in turgor pressure in sporangiophores where water entered. A56 and G5 showed a higher sensitivity to light than C9 and C47, whereas C111 showed no photosensitivity. Antagonistic relations were observed between photosensitivity and development of microphores, suggesting that the development and phototropism of macrophores is regulated by a common recognition factor (photoreceptor).

  • Observations on reproduction of Rhodochorton purpureum (Lightfoot) Rosenvinge (Acrochaetiaceae, Rhodophyta) was carried out with plants from Hokkaido, Japan. This species produces no monosporangia both in nature and culture, even though it does tetrasporangia commonly. The plants from Nemuro produce neither sporangia nor gametangia, and thus vegetative reproduction is the only known way to propagate themselves. It is suggested that the vegetative reproduction occurs in nature by fragmentation of vegetative filaments after development of a rhizoid. Several different modes of rhizoid production are described. The plants from Akkeshi and Oshoro (I) produce tetrasporangia that develop into plants producing only tetrasporangia. The plants from Muroran and Oshoro (II) produce tetrasporangia that develop into gametophytes. Gametophytes of R. purpureum from Muroran produce tetrasporangia as well as spermatangia or carpogonia. Such tetrasporangia on gametophytes are presumed to be mitotic.

Rhizopus Soft Rot on Citrus Fruit Caused by Rhizopus stolonifer in Korea (Rhizopus stolonifer에 의한 감귤 무름병)

  • A fruits soft rot caused by Rhizopus stolonifer found on citrus fruit in Jinjn City Agricultural Products Wholesale Market. The disease infection usually started from wounding. At first, the lesions started water soaked and rapidly softened and diseased lesion gradually expanded to other fruits in the same container. The mycelia grew vigorously on the surface of fruits and formed sporangia, sporangiophores and stolons. Sporangiophores were $900{\sim}4300{\mu}m$ in size. Sporangia were globose or subglobose and $98{\times}346{\mu}m$ in size. The color of sporangia was white at first and then turned black with many spores. Columella were globose or ellipsoid and $60{\sim}216{\times}72{\sim}143{\mu}m$ in size. Sporangiospores were globose or ellipsoid, brownish-black streaked and $8{\sim}18.3{\mu}m$ in diameter. Zygospores were globose, black and $98{\sim}176{\mu}m$ in size. The optimum temperature for mycelial growth of the fungus on PDA was $25^{\circ}C$. The causal fungus was identified as Rhizopus stolonifer. This is the first report of rhizopus soft rot on citrus fruit caused by R. stolonifer in Korea.

  • A Rhizopus soft rot caused by Rhizopus stolonifer occurred on Momordica charantia at Daesan-myon, Chang-won city, Gyeongnam province from 2003 to 2005. The disease usually started from wound on fruit with water-soaking lesions. The lesion rapidly expanded and softened. The fungal mycelia grew vigorously and sporangia, sporangiophores, and stolons were formed on surface of the fruit. Spoyangiophores were $15\~30{\mu}m$ in width. Sporangia were globose or hemispheric and $90\~180{\mu}m$ in size. The color of sporangia was white and cottony at first and turned to brownish black with many spores at maturity. Columella were hemispheric and $80\~150{\mu}m$ in size. Sporangiospores were irregular round or oval, brownish-black streaked and $7\~18{\times}6\~12{\mu}m$ in size. The optimum temperature for mycelial growth of the fungus on PDA was $25^{\circ}C$. On the basis of mycological characteristics and pathogenicity to host plants, the causal fungus was identified as Rhizopus stolonifer (Ehrenberg ex. Fr,) Lind. This is the first report of Rhizopus soft rot on M. charantia caused by R. stolonifer in Korea.

  • 말 원충성척수뇌염(equine protozoal myeloencenphalitis; EPM)은 말에게 심각한 신경성 질병을 일으키는 원충성 질병으로서 Sarcocystis neurona가 원인체이다. 근육포자낭(sarcocyst)을 숨기고 있는 자연중간숙주를 포함한 S. nuerona의 완전한 생활사는 알려져 있지 않으나 주머니쥐의 일종인 opossum(Didelphis virginiana, Didelphis albiventris)이 최종 숙주이며, 말은 정상적 숙주로 알려져 있다. 왜냐하면 말에서는 분열체(schizonts)와 분열소체(merozoites)가 발견되기 때문이다. EPM 유사 질병들이 고양이, 밍크, 너구리, 스컹크, 태평양 점박이 바다표범, 포니, 바다수달 등을 포함한 다양한 포유류에서 발생한다. 고양이는 포자낭(sporocytes) 섭식 후 근육포자낭 단계를 숨기는 실험적인 중간숙주로서 역할을 할 수 있다. 최근 국내에서 신경증상을 나타내는 말에 대한 의심스러운 임상증상이 임상수의사들에 의해 관찰되어졌는, 정확한 진단을 하기위하여 S. nuerona에 의해 발생된 EPM의 역사, 구조, 생활사, 생물학, 병리론, 동물에서 질병 유도, 임상 증상, 진단, 병인론, 생태학, 치료 등에 대하여 알아보고자 한다.

  • Rhizopus soft rot caused by Rhizopus stolonifer on June, 2008 occurred on Rubus crataegifolius Bunge in the box at Jinju City Wholesale Market of Agricultural Products at Gyeongnam province in Korea. The infected fruits were rapidly water-soaked, softened, black and eventually rotted. The symptoms occurred after infecting wounds at harvest time. The colony were white to light brown, formed much sporangiospores. The optimum temperature of the fungus on potato dextrose agar was $30^{\circ}C$. Sporangiophores were $12{\sim}25{\mu}m$ in width. Sporangia were globose or hemispheric, white at first and gradually to black, and $82{\sim}195{\mu}m$ in size. Columella were hemispheric, and $70{\sim}92{\mu}m$ in size. Sporangiospores were irregular round or oval, brownishblack and $9{\sim}21{\times}7{\sim}8{\mu}m$ in size. On the basis of symptom, mycological characteristics and pathogenicity of the fungus, the causal fungus was identified as Rhizopus stolonifer. This is the first report of Rhizopus soft rot by R. stolonifer on R. crataegifolius in Korea.

The colony of the isolate from the infected fruit was white to light brown and formed many sporangiospores. The optimum growth temperature of the fungus on potato dextrose agar was $30^{\circ}C$. Sporangiophores were $12{\sim}26\;{\mu}m$ in width. Sporangia were globose or hemispheric, white at first and changed gradually to black, and $83{\sim}195\;{\mu}m$ in size. Columella were hemispheric, and $72{\sim}125\;{\mu}m$ in size. Sporangiospores were irregular round or oval, brownish-black and $9{\sim}20{\times}7{\sim}8\;{\mu}m$ in size. On the basis of symptom, mycological characteristics and pathogenicity to banana, the causal fungus was identified as Rhizopus stolonifer. This is the first report of Rhizopus soft rot by R. stolonifer on banana in Korea.

Isolation of Mutants in Rhizopus nigricans by Chemical Mutagens (화학적 돌연변이원에 의한 Rhizopus nigricans의 돌연변이주 분리)

  • In order to isolate mutants in Rhizopus nigricans, the optimal treatment conditions for the chemical mutagens, N-Methyl-N'-Nitro-N-Nitrosoguanidine(MNNG) and Ethyl Methane Sulphonate(EMS), were explored. When MNNG was used as the chemical mutagen, the optimum concentration and treatment time for the best mutation frequency were $125{\mu}g/ml$ and 60 minutes, respectively. Under the optimum conditions for MNNG, the survival rate was 0.1-1.0%. The leucine auxotroph could be isolated. The phenotypic characteristics of the three mutants prepared are as follows; shortened sporangiophore, spiral sporangiophore, and reduced size of sporangium and sporangiospore. However, EMS as the chemical mutagen was ineffective for this species.
