Viability test and bulk harvest of natural zooplankton communities to verify the efficacy of a ship's ballast water treatment system based on USCG phase-II (USCG phase-II 선박평형수 처리장치 성능 평가를 위한 자연 해수의 동물플랑크톤 대량 확보 및 생사판별)

  • We investigated >$50-{\mu}m$ marine planktonic organisms (mainly zooplankton) using a bongo net in Masan Bay and Jangmok Bay in order to harvest 75% of natural communities based on Phase-II approval regulations by the United States Coast Guard (USCG). The concentrated volume (in 1 ton) and abundance of zooplankton were $1.8{\times}10^7ind.ton^{-1}$ and $2.3{\times}10^7ind.ton^{-1}$, and their survival rates were 82.6% and 80.1%, respectively. The community structure in Jangmok Bay was similar to that in Masan Bay, and dominant species were adult and immature groups (stage IV) of genus Acartia. Harvested populations were inoculated in a 500-ton test tank. Although the population abundances were $6.0{\times}10^4ind.ton^{-1}$ for both bay samples, the mortality rates were higher in the Masan Bay population (32%) than the Jangmok Bay population (20%). We considered the reason to be that there were 30% more immature individuals of Acartia from Masan Bay than from Jangmok Bay. The younger population may have been greatly stressed by the moving process and netting gear. After applying a Ballast Water Treatment System (BWTS) using a sample form Jangmok Bay, the mortality rates in the treatment groups were found to be 100% after 0 days and 5 days, implying that the BWTS worked well. During the winter season, the zooplankton concentration method alone did not easily satisfy the approval standards of USCG Phase II (> $10{\times}10^4ind.ton^{-1}$ in the 500 ton tank). Increasing the netting frequency and additional fishing boats may be helpful in meeting the USCG Phase II biological criteria.

Cryopreservation of Bovine IVM/IVF/IVC Blastocysts by Vitrification (체외성숙, 체외수정 및 체외배양에서 생산된 소 배반포기배의 초자화 동결)

  • The objective of this study was to examine the effect of developmental stage and embryo age of in vitro produced bovine blastocysts after vitrification and thawing. In vitro cultured day 8 blastocysts after IVF were equilibrated 20% ethylene glycol (EG) for 3 min. and were vitrified using EFS40, which is consisted of 40% EG, 18% ficoll, 0.3M sucrose and 10% FBS added in mDPBS for 30 sec. before being plunged into $LN_2$. Also, survival in vitro was assessed by re-expansion and hatching or hatched at 24 hand 48 h postwarming, respectively. The results obtained in these experiments were summarized as follows; 1) When the embryos were cultured for 8 day after IVF, 41.0% of the cleaved embryos developed to the blastocysts (early; 7.6%, expanded; 22.9%, hatching; 4.6% and hatched; 5.9%). 2) When the embryos were exposed or vitrified to the freezing solution, the re-expansion of vitrified embryos (73.3%) was significantly lower than that of control and exposed embryos (100, 97.0%) (p<0.05). But the formation rate of hatching or hatched blastocysts of vitrified embryos (66.7, 46.7%) at 48h after thawing was similar to that of exposed embryos (66.7, 39.4%) but not control (100, 100%) (p<0.01). However, in the total cell numbers of those developed hatched blastocysts, there were not significantly different among the treatment groups. 3) When the embryo survival rates by different developmental stage were examined, the re-expansion was not different among the groups $(64.5{\sim}75.6%)$. After warming 48 h, the hatching and hatched formation of early blastocysts (25.8, 9.7%) was significantly lower than those of expanded (69.7, 39.4%) and hatching blastocysts (53.3, 43.3%) (p<0.05). 4) In addition, when the expanded blastocysts at day 7, 8 and 9 were vitrified, the re-expansion of day 8 and 9 embryos was significantly lower than that of day 7 (day 7; 93.9%, day 8; 75.8% and day 9; 87.5%) (p<0.05). However, the rates of development to hatched blastocysts were no difference among the groups (day 7; 36.4%, day 8; 36.4% and day 9; 31.3%). These results suggested that in vitro produced expanded or hatching blastocysts can be efficiently cryopreserved by the two-step vitrification method using EFS40.

  • This study was conducted to examine the influence of heat treatment on the $NH_4{^+}$ adsorption by zeolite and to determine the $NH_4{^+}$ loss from $NH_4-$ zeolite during the incubation. The dominant clay minerals of zeolite were clinoptiloiite and mordenite. $NH_4{^+}$ adsorption by zeolite was equilibrated at a 4hrs-shaking. The content of exchangeable $K^+$ of zeolite was increased with increase in the temperature of heat treatment but the amount of $NH_4{^+}$ adsorbed by zeolite was decreased. The amount of $NH_4{^+}$ in the soil treated with ammonium sulfate was decreased during incubation under the submerged condition but the amount of $NH_4{^+}$ adsorbed by zeolite was not decreased in the same course of incubation. It was estimated that $NH_4{^+}$ adsorbed by zeolite was resistant to $NH_4{^+}$-N loss by various reactions.

  • Fixation rate of reactive dyes on silk fibroin showed the same tendency to absorption rate. Fixation rate of Yellow 179 and Blue 71 was quite slow and Black 5, Blue 49, Blue 4, Blue 19 and Blue 21 was relatively fast. Weight increase by MAA graft polymerization onto filk fibroin was decreased, on the whole, according to the the increase of dye fixation, but weight increase of silk fibroin dyed with Blue 71 and Blue 21 was much higher than that of silk fibroin. And weight increase of silk fibroin dyed with Yellow 179 was similar to that of silk fibroin and in case of Blue 19, weight increase was about 12%, constantly. Weight increase of silk fibroin dyed with Blue 71 and Blue 21 was higher and the others were lower than of silk fibroin. And weight increase was diminished according as the dyeing temperature rises. The color was a little changed by MAA graft polymerization.

Study on Adsorption Kinetic Characteristics of Propineb Pesticide on Activated Carbon (활성탄에 대한 프로피네브 농약의 흡착동력학적 특성 연구)

  • The adsorption characteristics of propineb pesticide onto activated carbon has been investigated for the adsorption in aqueous solution with respect to initial concentration, contact time and temperature in batch experiment. The Langmuir and Freundlich adsorption models were applied to described the equilibrium isotherms and isotherm constants were also determined. The Freundlich model agrees with experimental data well. slope of isotherm line indicate that activated carbon could be employed as effective treatment for removal of propineb. The pseudo first order, pseudo second order kinetic models were use to describe the kinetic data and rate constants were evaluated. The adsorption process followed a pseudo second order model, and the adsorption rate constant($k_2$) decreased with increasing initial concentration of propineb. The activation energy, change of free energy, enthalpy, and entropy were also calculated to predict the nature adsorption. The estimated values for change of free energy were -7.28, -8.27 and -11.66 kJ/mol over activated carbon at 298, 308 and 318 K, respectively. The results indicated toward a spontaneous process. The positive value for change of enthalpy, 54.46 kJ/mol, found that the adsorption of propineb on activated carbon is an endothermic process.

Shelf-life Estimation and Sorption Characteristics of Coated Ascorbic Acid by Fluidized Bed Coating (유동층 코팅 처리한 Ascorbic acid의 흡습특성 및 저장기간 예측)

  • This study was performed to investigate the sorption characteristics and shelf-life of coated ascorbic acid Stability of ascorbic acid, which oxidizes easily during storage and processing, was achieved by applying a fluidized bed coating using Zein-DP and HPMC-FCC as covering materials. The monolayer moisture content calculated using the GAB equation showed a higher level of significance than when calculated using the BET equation. The fit to the isotherm curve was in the order of Halsey, Caurie, Oswin and Khun. The equilibrium relative humidity prediction model was established in terms of time and water activity, it had higher significance. The stability of the coated ascoribic acid during storage was investigated in terms of radical-scavenging activity, which decreased with increasing time of storage and was more affected at higher storage temperatures. The quality reduction rate constant (k) was calculated by a first-order reaction rate. The reaction rate constant increased with increasing storage temperature. The shelf-life of Zein-DP-coated ascorbic acid was estimated to be 45.83 days at 20C and 63.19 days at 10C, and the shelf-life for HPMC-FCC-coated ascorbic acid was estimated to be 28.84 days at 20C and 36.14 days at 10, the ascorbic acid was 24.52 days at $20^{\circ}C$ and 27.22 days at $10^{\circ}C$, respectively. Therefore, the fluidized bed coating effectively increased the stability of ascorbic acid.

Effects of Solvent Extraction by Immersion on the Quality and Storage Stability of Rice (용매침지(溶媒浸漬)에 의한 탈지(脫脂)처리가 쌀의 품질(品質) 및 저장성(貯臧性)에 미치는 영향)

  • Effects of solvent extraction by immersion on the quality and storage stability of Korean rice were studied. Proportions of lipid extracted from whole grain of rice by immersing into two volumes(v/wt) of hexane and ethanol for 72 hours at room temperature were 0.41% and 0.38% respectively. Small changes of water content and hardness of rice were observed by solvent treatment. Cooking characteristics; that is, water-uptake ratio. extended volume, total solid, and starch-iodine blue test of rice was markedly changed by ethanol treatment, while little changes were observed by hexane treatment. No considerable differences in moisture sorption isotherm of rice were observed by both solvent treatments. Changes in TBA number and stale flavor appearance of rice treated with or without solvent immersion during storage at $60^{\circ}C$ showed that rice treated with hexane had best storage stability compared to ethanol treatment, while ethanol treatment of rice had better storage stability than no treatment. Similar results were noted in changes of the flavor score of cooked rice samples which were freeze dried.

Removal of Bacteriophage T7 from Artificial groundwater by Layered Double Hydroxide (층상이중수산화물에 의한 인공지하수내의 박테리오파지 T7 제거)

  • The objective of this study was to investigate the virus removal from artificial groundwater using Mg-Fe layered double hydroxide (LDH). Batch experiments were conducted under various experimental conditions to examine bacteriophage T7 removal with Mg-Fe LDH. Results showed that the removal of T7 by Mg-Fe LDH was a fast process, reaching equilibrium within 2~3 hrs. Mg-Fe LDH had the virus removal capacity of $1.57{\times}10^8pfu/g$ with a removal percent of 96%. Results also showed that the effect of solution pH on T7 removal was minimal between pH 6.2 and 9.1. The influence of anions ($SO_4^{2-}$, $CO_3^{2-}$, $HPO_4^{2-}$) on T7 removal was significant due to their competition with bacteriophage at the sorption sites on LDH, while the effect of $NO_3^-$ was negligible. This study demonstrated that Mg-Fe LDH could be applied as adsorbents for virus removal in water treatment.

Characteristics of Isotherm, Kinetic and Thermodynamic Parameters for the Adsorption of Acid Red 66 by Activated Carbon (활성탄에 의한 Acid Red 66의 흡착에 대한 등온선, 동력학 및 열역학적 특성)

  • The kinetic and thermodynamic parameters of Acid Red 66, adsorbed by granular activated carbon, were investigated on areas of initial concentration, contact time, and temperature. The adsorption equilibrium data were applied to Langmuir, Freundlich, Temkin, Redlich-Peterson, and Temkin isotherms. The agreement was found to be the highest in the Freundlich model. From the determined Freundlich separation factor (1/n = 0.125 ~ 0.232), the adsorption of Acid Red 66 by granular activated carbon could be employed as an effective treatment method. Temkin's constant related to adsorption heat (BT = 2.147 ~ 2.562 J mol-1) showed that this process was physical adsorption. From kinetic experiments, the adsorption process followed the pseudo-second order model with good agreement. The results of the intraparticle diffusion equation showed that the inclination of the second straight line representing the intraparticle diffusion was smaller than that of the first straight line representing the boundary layer diffusion. Therefore, it was confirmed that intraparticle diffusion was the rate-controlling step. From thermodynamic experiments, the activation energy was determined as 35.23 kJ mol-1, indicating that the adsorption of Acid Red 66 was physical adsorption. The negative Gibbs free energy change (ΔG = -0.548 ~ -7.802 kJ mol-1) and the positive enthalpy change (ΔH = +109.112 kJ mol-1) indicated the spontaneous and endothermic nature of the adsorption process, respectively. The isosteric heat of adsorption increased with the increase of surface loading, indicating lateral interactions between the adsorbed dye molecules.

Degradation of herbicide paraquat by Fenton reagent and UV light irradiation (Fenton 시약 및 UV 광 조사에 의한 제초제 paraquat의 분해)

  • This study was to investigate the potential degradation of a herbicide paraquat by Fenton reagents(ferric ion and hydrogen peroxide) under UV light irradiation(365 nm) in an aqueous solution. When $10{\sim}500$ mg/L of paraquat was reacted with either ferric ion or hydrogen peroxide in the dark or under UV light, no degradation was occurred. However, the simultaneous application of both ferric ion(0.8 mM) and hydrogen peroxide(0.140 M) in paraquat solution(500 mg/L) caused dramatic degradation of paraquat both in the dark (approximately 78%) and under UV light(approximately 90%). The reaction approached an equilibrium state in 10 hours. In the dark, when $0.2{\sim}0.8$ mM ferric ion was added, $20{\sim}70%$ paraquat of $10{\sim}500$ mg/L was degraded, regardless of hydrogen peroxide concentrations($0.035{\sim}0.140$ M), while under UV light, 95% of 10 and 100 mg/L paraquat was degraded regardless of ferric ion and hydrogen peroxide concentrations. At paraquat concentration of 200 and 500 mg/L, paraquat degradation increased with increasing ferric ion concentrations as in the dark. However the increase in hydrogen peroxide concentration did not affect the extent of paraquat degradation. The initial reaction rate constants(k) for paraquat degradation ranged from 0.0004 to 0.0314, and 0.0023 to 0.0367 in the dark and under UV light, respectively. The initial reaction rate constant increased in proportion to the increase in ferric ion concentration in both conditions. The half-lives of paraquat degradation(t1/2) were 20 - 1,980 and 19 - 303 minutes in the dark and under UV light, respectively. This study indicates that Fenton reagents under UV light irradiation are more potent than in the dark in terms of herbicide paraquat degradation in an aqueous solution.
