Design and Development of Integrated Recirculating Aquaculture System (순환 여과식 양식 시스템의 설계 및 개발)

  • An integrated recirculating aquaculture system suitable to our country's situation was designes and developed, The integrated recirculating aquaculture system consisting of a double drain type rearing tank of the $2.5 m^3$, a sedimentation tank for removal of settleable solids, a floating bead filter for the removal of suspended solids, a foam separator for removal of fine particles and a rotating biological contactor for the nitrification was designed and manufactured. The integrated recirculating aquaculture system was stocked with nile tilapia (Oreochromis niloticus) at $2\%$ rearing density for 2 weeks. Feed coefficient was 1.4 and average daily growth rate was $0.64\%$ in the system for 2 weeks.

  • In order to develop facility for complex farming with aquaculture and vegetable nutrient culture and to investigate growth of tilapia and water quality in the channel type aquaculture system, these experiments were carried out. When tilapia(Tilapia nilotica) was reared in the channel type aquaculture system, Quality of culture water and growth of tilapia were normal. And the growth of tilapia in the channel type and round type aquaculture system was much the same. Channel type aquaculture facilities was applicable to the dual culture system for aquaculture and vegetable culture. Channel type integrated system was composed of aquaculture and sandculture bed, and it was thought that suitable size of the system was about 1.4m(width) $\times$ 1m(height) $\times$ 20m(length).

  • Physiological responses (cortisol, glucose, GPT, GOT, total protein, hematocrit, sodium, chloride, potassium, total calcium and osmolality), growth and survival rates of grey mullet (Mugil cephalus) and Nile tilapia (Oreochromis niloticus) by the manipulation of salinity were studied. Salinity was increased directly from freshwater to $33\%_{\circ}$ and remained for 15 days. Then, salinity was decreased directly from seawater to freshwater and remained for other 15 days. All tilapias were dead at the third day, but $96.5\%$ of grey mullet were survived at the 30th day. The grey mullet showed no significant differences for cortisol and glucose concentrations in the seawater and freshwater. However, those of the Nile tilapia were increased in seawater. The grey mullet showed no significant differences for sodium, chloride and potassium concentrations during experimental period. However, those of the Nile tilapia were significantly increased when exposed to seawater. The osmolality of grey mullet ranged between 293.5 mOsm/kg and 335.0 mOsm/kg for the whole experimental period.

The Effects of a Dietary Edwardsiella tarda Specific Bacteriophage and Bacillus subtilis Mixture on Innate Immune Responses and Antibacterial Activity of Nile tilapia Oreochromis niloticus (Edwardsiella tarda의 특이 Bacteriophage와 Bacillus subtilis가 혼합된 사료급이가 나일 틸라피아(Oreochromis niloticus)의 선천적 면역반응과 항균효과에 미치는 영향)

  • The present study investigated the effects of dietary Edwardsiella tarda (E. tarda) specific bacteriophage (phage) and Bacillus subtilis (B. subtilis) mixture on innate immune responses and antibacterial activity of Nile tilapia, Oreochromis niloticus. In a dietary experiment, tilapia were fed the control diet (C), a phage-only supplemented diet (P), a B. subtilis only supplemented diet (B), or a B. subtilis and phage mixed diet (B+P). A respiratory burst and significant increase in lysozyme activity (P<0.05) were noted in the B+P group, as compared to other groups after 4 days of feeding. The B group showed a significant (P<0.05) increase in respiratory burst and lysozyme activity versus the C and P groups, whereas no significant increases (P<0.05) were observed in the P and C groups. $ACH_{50}$ was significantly up-regulated in the B+P group versus other groups after 8 days of feeding (P<0.05). In vivo antibacterial activity was significantly enhanced in the B+P fed group, as compared to other groups (P<0.05) after 7 days of E. tarda challenge. A significant (P<0.05) increase in antibacterial activity was seen in the B group, as compared to C or P groups after 14 days of feeding. These results suggest that a B. subtilis and phage mixture could be utilized as an alternative to antibiotics in the control of fish diseases caused by E. tarda.

The characteristics of adrenergic responses in tilapis dorsal aorta (틸라피아 배대동맥의 아드레날린성 반응의 특성)

  • The present study was undertaken to investigate the physiological characteristics of the adrenergic responses in the tilapia dorsal aorta. Epinephrine, norepinephrine, clonidine and methoxamine in the presence of propranolol($3{\times}10^{-6}$M), induced only endothelium-independent and concentration-dependent vasocontractions in tilapia dorsal aorta. The rank order of potency of adrenergic agonists inducing vasocontraction was epinephrine>norepinephrine>phenylephrine>clonidine>ethoxamine, Yohimbine produced a parallel shift of the concentration-vascontraction curves of epinephrine, norepinephrine, phenylephrine and clonidine to the right, while prazosin depressed the maximum responses of epinephrine and norepinephrine. Calcium-free physiological solution and verapamil markedly reduced epinephrine or norepinephrine-induced vasocontractions. These results suggest that a-adrenergic agonists produce only on endothelium-inedpenent casoconstrictions in tilapia dorsal aorta and these effect of a-adrenergic agonists, which might be associated with both calcium release from intracellular stores and calcium influx through voltage-dependent calcium channel.

  • Experimental infection method for Edwardsiellosis was studied to imitate histopathologically natural infections in the Tilapia Oreochrimis niloticus. Prior to the bacterial challenge, the intestine of tilapia was damaged by 0.2ml of 30% hydrogen peroxide introduced through a silicon tube which was inserted 4 to 5cm into the intestine from anus. E. tarda was mixed a 10% Gum arabia and administered into the stomach by a cannula 20 hours after the hydrogen peroxide treatment. Bacterial doses used were from $4.2{\times}10^6$ to $6.4{\times}10^\;CFU$/fish. Fish 72 hours after the challenged showed swollen and erosious lesion and focal necrosis with bacterial-eaden inflammatory cell in the Liver, Spleen, Kidney. Fish 96 and 120 hours after bacterial challenge were moribund showing swollen Liver, Spleen, Kidney and focal accumulation of macrophages and production of granulomas in the infected lesions.

  • Physiological responses (cortisol, glucose, GPT, GOT, hematocrit, sodium, chloride, potassium, total protein and osmolality), growth and survival rates of grey mullet and Nile tilapia were tested by the gradual salinity changes for 70 days. Three different sizes of grey mullet, small (MS, $13.3\pm1.8cm$), middle(MM, $28.9\pm3.6cm$) and large (ML, $36.0\pm2.0cm$), and three different sizes of tilapia, small (TS,$16.5\pm1.7cm$), middle (TM, $20.6\pm1.8cm$) and large (TL, $27.2\pm2.7cm$)were used. Salinity of reareing water was increased $\5textperthousand$ in every 5 days until it reached at $0\textperthousand$. The cortisol concentrations in all size groups of grey mullet were increased at both full strength seawater (SW) and fresh water (FW) at the end. The cortisol concentrations of TM and TL groups were higher in SW than FW (0 and 70 days). The GOT values of grey mullet were lower than those of Nile tilapia. The sodium concentrations of grey mullet in SW were ranged 160~184 mEq/$\ell$, while those of Nile tilapia were the highest in the SW. No mortality was observed in MM and ML groups but survival rate of MS group was 76$\textperthousand$. Overall survival rate of Nile tilapia was lower than grey mullet. Survival rate of TS, TM and TL groups were $79\textperthousand$, $29\textperthousand$ and $55\textperthousand$, respectively.

Influences of Parental Pairs on Progeny Sex Ratios of Nile Tilapia Oreochromis niloticus (틸라피아 Oreochromis niloticus의 성비 결정에 미치는 암수어미의 영향)

  • Sex of the Nile tilapia Oreochromis niloticus is mainly determined by an XX/XY system. However, accumulating evidences suggest the existence of additional sex modifying factors including environmental, autosomal and parental influences. In order to investigate the possibility of parental effects on sex ratios of tilapia progenies, in this study, a series of crosses was carried out using gynogenetic clonal fish, neomales, normal males and females, and YY fish. Crosses between clonal XX male and clonal female have yielded only female progenies and no parental influences were observed. However, in the crosses between clonal males and normal females, female parents were significantly associated with the progeny sex ratios ($X^2$=20.046, 7 d.f., p<0.01). Progeny sex ratios from the crosses between neomales and normal females ($X^2$=60.491, 5 d.f and $X^2$=28.072, 2 d.f.) also showed significant association with female parents (P<0.001). The stability of progeny sex ratios from repeated spawns were confirmed by using 6 different parental pairs. In 16 crosses between normal males and normal females, sex ratios of progenies showed clear maternal influences, and further analysis of the results revealed a negative correlation ($r^2$=0.7718, p<0.05) between the sex ratios of progenies from two different males, indicating a strong paternal influence. No statistically significant relationship between survival rates and sex ratios of progenies was observed in any genotypic groups. Taken together, the influence of parental pairs on progeny sex ratios in this species is evident although the cause of this influence is not clear.

Effects of Dietary Supplementation with Nucleotide on Growth Performance, Feed Utilization, and Non-Specific Immune Responses of Nile Tilapia Oreochromis niloticus (사료 내 nucleotide 첨가가 틸라피아(Oreochromis niloticus)의 성장, 사료효율 및 비특이적 면역력에 미치는 영향)

  • The present study examined the effects of dietary supplementation with nucleotide (inosine monophosphate product, IMP) on the growth performance, feed utilization, and non-specific immune responses of juvenile tilapia Oreochromis niloticus. Triplicate groups of tilapia (initial body weight, $7.4{\pm}0.04$ g) were fed experimental diets containing 0%, 0.05%, 0.1%, and 0.2% IMP. Fish were fed six times a day until apparent satiation for 13 weeks. At the end of the feeding trial, final body weight and food utilization of fish fed 0.1% IMP were significantly higher than those of fish fed the control diet. Results of hematological parameters were not affected by dietary IMP. However, blood protein level was significantly higher in the 0.05% treatment, as compared to that of the control and 0.2% IMP diets. Myeloperoxidase activity was higher in fish fed 0.1% IMP than in fish fed the control and 0.2% IMP diets. These results suggest that dietary supplementation with IMP can enhance the growth performance, feed utilization, and innate immune response of juvenile tilapia. The optimal IMP supplementation level appears to be 0.1% in practical feed formulations for tilapia.

The Rapid Diagnosis of $\beta$-Haemolitic Streptococcus sp. by Immunoperoxidase Method (면역효소항체법에 의한 $\beta$-용혈성 연쇄구균 Streptococcus sp.의 신속진단에 대하여)

  • For the rapid diagnosis of bacterial diseases of cultured fishes, the immunoperoxidase method was applied to the detection of $\beta$-haemolitic Streptococcus sp. strain KST-2 isolated from tilapia(Oreochromis niloticus). The suitability of field analysis and the sensitivity of the immunoperoxidase method was compared with those of the counterimmunoelectrophoresis(CIE) and the immunodiffusion(ID). Results of testing cross-reactivity which did not indicate any cross reactivity with other fish pathogens, this method was specific to Streptococcus sp. The sensitivity of this method was $1{\times}10^3CFU/ml$ which was at least $10^2$times greater than the CIE and $10^4$times greater than the ID. The immunoperoxidase method was more suitable for field application and more sensitive than other diagnostic techniques tested on this study.
